1Cropland redistribution to marginal lands undermines environmental sustainability显示文摘Cropland redistribution to marginal land has been reported worldwide;however,the resulting impacts on environmental sustainability have not been investigated sufficiently.Here we investigated the environmental impacts of cropland redistribution in China.As a result of urbanization-induced loss of high-quality croplands in south China(~8.5 t ha^(-1)),croplands expanded to marginal lands in northeast(~4.5 t ha^(-1)) and northwest China(~2.9 t ha^(-1)) during 1990-2015 to pursue food security.However,the reclamation in these low-yield and ecologically vulnerable zones considerably undermined local environmental sustainability,for example increasing wind erosion(+3.47%),irrigation water consumption(+34.42%),fertilizer use(+20.02%) and decreasing natural habitats(-3.11%).Forecasts show that further reclamation in marginal lands per current policies would exacerbate environmental costs by 2050.The future cropland security risk will be remarkably intensified because of the conflict between food production and environmental sustainability.Our research suggests that globally emerging reclamation of marginal lands should be restricted and crop yield boost should be encouraged for both food security and environmental benefits.Wenhui Kuang Jiyuan Liu Hanqin Tian Hao Shi Jinwei Dong Changqing Song Xiaoyong Li Guoming Du Yali Hou Dengsheng Lu Wenfeng Chi Tao Pan Shuwen Zhang Rafiq Hamdi Zherui Yin Huimin Yan Changzhen Yan Shixin Wu Rendong Li Jiuchun Yang Yinyin Dou Wenbin Wu Liqiao Liang Bao Xiang Shiqi Yang 2022National Science Review2022,9,1:13
2Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12

4Improving Semantic Part Features for Person Re-identification with Supervised Non-local Similarity显示文摘In person re-IDentification (re-ID) task,the learning of part-level features benefits from fine-grained information.To facilitate part alignment,which is a prerequisite for learning part-level features,a popular approach is to detect semantic parts with the use of human parsing or pose estimation.Such methods of semantic partition do offer cues to good part alignment but are prone to noisy part detection,especially when they are employed in an off-the-shelf manner.In response,this paper proposes a novel part feature learning method for re-ID,that suppresses the impact of noisy semantic part detection through Supervised Non-local Similarity (SNS) learning.Given several detected semantic parts,SNS first locates their center points on the convolutional feature maps for use as a set of anchors and then evaluates the similarity values between these anchors and each pixel on the feature maps.The non-local similarity learning is supervised such that:each anchor should be similar to itself and simultaneously dissimilar to any other anchors,thus yielding the SNS.Finally,each anchor absorbs features from all of the similar pixels on the convolutional feature maps to generate a corresponding part feature (SNS feature).We evaluate our method with extensive experiments conducted under both holistic and partial re-ID scenarios.Experimental results confirm that SNS consistently improves re-ID accuracy using human parsing or pose estimation,and that our results are on par with state-of-the-art methods.Yifan Sun Zhaopeng Dou Yali Li Shengjin Wang 2020Tsinghua Science and Technology2020,25,5:2

7Bilirubin inhibits the anticancer activity of sorafenib by blocking MCL-1 degradation in hepatocellular carcinoma cells显示文摘Objective:Sorafenib is a first-line drug for advanced hepatocellular carcinoma(HCC).Unfortunately,most patients with HCC do not respond to sorafenib,mainly because of the frequent development of drug resistance.Bilirubin is an end metabolite of heme catabolism and an indicator of liver function,but its direct role in regulating the anticancer activity of sorafenib in HCC cells is unclear.In the current study,we aimed to investigate the mechanism of action of bilirubin in sorafenib-mediated tumor suppression in HCC.Methods:A retrospective observational cohort of 100 patients receiving sorafenib was conducted to evaluate the potential role of bilirubin in predicting the prognosis of patients with HCC.Human HCC cell lines were treated with sorafenib in the absence or presence of bilirubin,and cell proliferation,apoptosis,and signaling pathways were assayed.The antagonistic effect of bilirubin toward sorafenib was assessed in nude mice bearing HCC xenografts.Results:Serum levels of bilirubin(including total,direct,and indirect bilirubin)negatively correlated with the overall survival of patients with HCC treated with sorafenib(P<0.05).Both in vitro and in vivo analyses demonstrated that bilirubin significantly abrogated sorafenib-mediated proliferation inhibition and apoptosis induction in HCC cells(P<0.05).Mechanically,bilirubin inhibited sorafenib-induced activation of GSK-3βand subsequent downstream MCL-1 degradation.Conclusions:Our study provides experimental evidence of the antagonistic effect of bilirubin toward sorafenib-mediated anticancer activity in HCC,and it suggests that bilirubin could be used to predict the efficacy of sorafenib treatment.Leyi Yao Qian Zhao Ding Yan Ziying Lei Yali Hao Jinghong Chen Qian Xue Xiaofen Li Qingtian Huang Daolin Tang QPing Dou Xin Chen Jinbao Liu 2022Cancer Biology & Medicine2022,19,7:0
8Single-cell transcriptome atlas reveals spatiotemporal developmental trajectories in the basal roots of moso bamboo (Phyllostachys edulis)显示文摘Roots are essential for plant growth and development.Bamboo is a large Poaceae perennial with 1642 species worldwide.However,little is known about the transcriptional atlas that underpins root cell-type differentiation.Here,we set up a modified protocol for protoplast preparation and report single-cell transcriptomes of 14279 filtered single cells derived from the basal root tips of moso bamboo.We identified four cell types and defined new cell-type-specific marker genes for the basal root.We reconstructed the developmental trajectories of the root cap,epidermis,and ground tissues and elucidated critical factors regulating cell fate determination.According to in situ hybridization and pseudotime trajectory analysis,the root cap and epidermis originated from a common initial cell lineage,revealing the particularity of bamboo basal root development.We further identified key regulatory factors for the differentiation of these cells and indicated divergent root developmental pathways between moso bamboo and rice.Additionally,PheWOX13a and PheWOX13b ectopically expressed in Arabidopsis inhibited primary root and lateral root growth and regulated the growth and development of the root cap,which was different from WOX13 orthologs in Arabidopsis.Taken together,our results offer an important resource for investigating the mechanism of root cell differentiation and root system architecture in perennial woody species of Bambusoideae.Zhanchao Cheng Changhong Mu Xiangyu Li Wenlong Cheng Miaomiao Cai Chongyang Wu Jutang Jiang Hui Fang Yucong Bai Huifang Zheng Ruiman Geng Junlei Xu Yali Xie Yuping Dou Juan Li Shaohua Mu Jian Gao 2023Horticulture Research2023,10,8:0
