1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Atomically dispersed Au_1 catalyst towards efficient electrochemical synthesis of ammonia显示文摘Tremendous efforts have been devoted to explore energy-efficient strategies of ammonia synthesis to replace Haber–Bosch process which accounts for 1.4% of the annual energy consumption. In this study,atomically dispersed Au_1 catalyst is synthesized and applied in electrochemical synthesis of ammonia under ambient conditions. A high NH_4^+ Faradaic efficiency of 11.1% achieved by our Au_1 catalyst surpasses most of reported catalysts under comparable conditions. Benefiting from efficient atom utilization, an NH_4^+ yield rate of 1,305 lg h^(-1) mg_(Au)^(-1) has been reached, which is roughly 22.5 times as high as that by supported Au nanoparticles. We also demonstrate that by employing our Au_1 catalyst, NH4+can be electrochemically produced directly from N_2 and H_2 with an energy utilization rate of 4.02 mmol kJ^(-1). 8Analysis of Allergens in 5473 Patients with Allergic Diseases in Harbin, China显示文摘Objective To analyze the allergic status to common inhalant allergens and food allergens in clinical patients in Harbin in northeastern China and provide evidence to develop the prevention strategy of allergic disease. Methods The data were collected from 5473 patients with clinical suspected allergic diseases seeking medical care in the Second Affiliated Hospital of Harbin Medical University. Among these patients, 2530(46.2%) were males aged 0-86 years, the youngest was only 1 month old and 2579(47.1%) were young children and teenagers. The serum specific Immunoglobulin E(sIgE) to 14 kinds of common allergens and serum total IgE were detected by using AllergyScreen test(Mediwiss Analytic GmbH, Moers, Germany). Results In 5473 subjects the positive rate of sIgE was 33.1%(n=1813). Cow milk(6.9%) and wheat(3.1%) were the most common food allergens, followed by house dust mite mix(12.5%) and mould mix(9.4%) and the age and gender specific differences in the positive rate were significant. For the children aged <7 years the positive rates to cow milk, beef-mutton, and egg white/egg yolk were high, but the positive rates to house dust mite mix, ragweed estragon, and mould mix were low(P<0.05). For the adults the positive rates to aeroallergens were high while the rates to food allergens were low. Conclusion The results from this study showed that the food allergens in Harbin had geographic characteristics, which support the viewpoint that the environment factors play an important role in the incidence of allergic diseases. 9Characterizing the optimal operation of photocatalytic degradation of BDE-209 by nano-sized TiO_2显示文摘Brominated flame retardants have been widely used in industry.There is a rapid growing public concern for their availabilities in the environment.Advanced oxidation process(AOP) is a promising and efficient technology which may be used to remove emerging chemicals such as brominated flame retardants.This study aims at investigating optimal operational conditions for the removal of BDE-209 using nano-scaled titanium(IV) oxide.The residual PBDE congeners after photocatalytical degradation of BDE-209 by TiO 2 were analysed by gas chromatography-mass spectrometry(GC-MS).It was found that the degradability of BDE-209 by TiO 2 was attributed to its photocatalytic activity but not the small size of the particles.The half-life of removing BDE-209 by TiO 2 was 3.05 days under visible light.Tetra-and penta-BDEs were the major degraded products of BDE-209.Optimum conditions for photocatalytical degradation of BDE-209 was found to be at pH 12(93% ± 1%),5,10,20 mg/L(93.0% ± 1.70%,91.6% ± 3.21%,91.9% ± 0.952%,respectively),respectively of humic acid and in the form of anatase/rutile TiO 2(82% ± 3%).Hence,the efficiency of removing BDE-209 can be maximized while being cost effective at the said operating conditions.Ka Lai Chow Yu Bon Man Jin Shu Zheng Yan Liang Nora Fung Yee Tam Ming Hung Wong 2012Journal of Environmental Sciences2012,24,9:8
10A Crucial Role of GA-Regulated Flavonol Biosynthesis in Root Growth of Arabidopsis显示文摘Flavonols have been demonstrated to play many important roles in plant growth, development, and communication with other organisms. Flavonol biosynthesis is spatiotemporally regulated by the subgroup 7 R2R3-MYB (SG7 MYB) transcription factors including MYB11/MYB12/MYB111. However, whether SG7- MYB activity is subject to post-translational regulation remains unclear. Here, we show that gibberellic acid (GA) inhibits flavonol biosynthesis via DELLA proteins in Arabidopsis. Protein-protein interaction analyses revealed that DELLAs (RGA and GAI) interacted with SG7 MYBs (MYB12 and MYB111) both in vitro and in vivo, leading to enhanced affinity of MYB binding to the promoter regions of key genes for flavonol biosynthesis and thus increasing their transcriptional levels. We observed that the level of auxin in the root tip was negatively correlated with root flavonol content. Furthermore, genetic assays showed that loss-of-function mutations in MYB 12, which is predominantly expressed in roots, partially rescued the short-root phenotype of the GA-deficient mutant ga1-3 by increasing root meristem size and mature cell size. Consistent with these observations, exogenous application of the flavonol quercetin restored the root meristem size of myb12 ga1-3 to that of ga1-3. 12Long-term postoperative outcomes of bilateral lateral rectus recession vs unilateral recession-resection for intermittent exotropia显示文摘AIM: To discuss the long-term postoperative results of bilateral lateral rectus recession(BLR) and unilateral lateral rectus recession-medial rectus resection(RR) in therapy of intermittent exotropia.METHODS: We retrospectively analyzed 213 cases of intermittent exotropia who underwent surgery between2008 and 2010. The patients were grouped into BLR group and RR group. Motor outcomes were divided into three groups on the basis of the angle of deviation after surgery: overcorrection(esotropia/phoria >5△), orthophoria(esotropia/phoria ≤5△to exotropia/phoria ≤10△), and undercorrection/recurrence(exotropia/phoria >10△).Titmus test was used to evaluate stereoacuity, the stereoacuity <800s of arc meaned the patients had stereopsis. Surgical outcome including motor criteria and sensory status were compared at postoperative 6, 12,24 mo and at 36 mo examination between groups. RESULTS: At 12, 24 mo after surgery, the motor outcomes had no difference(P >0.05) between groups.However, the motor outcomes at 6, 36 mo were signally different in each group, indicating the success rate in RR group at 6mo was higher than that in BLR group(83.02%vs 82.24%, P <0.05) but the result was contrary at the 3y examination(60.75% vs 43.40%, P <0.05). No statistical significance were found in the sensory outcomes between the groups at mean of 3.7y follow-up.CONCLUSION: The motor outcomes in RR group were better than in BLR group at 6mo after surgery, while the3 y outcomes were better in BLR group. 15Application of robotics in gastrointestinal endoscopy:Areview显示文摘Multiple robotic flexible endoscope platforms have been developed based on cross specialty collaboration between engineers and medical doctors. However, significant number of these platforms have been developed for the natural orifice transluminal endo-scopic surgery paradigm. Increasing amount of evidence suggest the focus of development should be placed on advanced endolumenal procedures such as endoscopic submucosal dissection instead. A thorough literature analysis was performed to assess the current status of robotic flexible endoscopic platforms designed for advanced endolumenal procedures. Current efforts are mainly focused on robotic locomotion and robotic instrument control. In the future, advances in actuation and servoing technology, optical analysis, augmented reality and wireless power transmission technology will no doubt further advance the field of robotic endoscopy. 18Epigenetic modification in histone deacetylase deletion strain of Calcarisporium arbuscula leads to diverse diterpenoids显示文摘Epigenetic modifications have been proved to be a powerful way to activate silent gene clusters and lead to diverse secondary metabolites in fungi. Previously, inactivation of a histone H3 deacetylase in Calcarisporium arbuscula had led to pleiotropic activation and overexpression of more than 75% of the biosynthetic genes and isolation of ten compounds. Further investigation of the crude extract of C. arbuscula Δhda A strain resulted in the isolation of twelve new diterpenoids including three cassanes(1-3), one cleistanthane(4), six pimaranes(5-10), and two isopimaranes(11 and 12) along with two know cleistanthane analogues. Their structures were elucidated by extensive NMR spectroscopic data analysis. 20Inorganic Elemental Determinations of Marine Traditional Chinese Medicine Meretricis concha from Jiaozhou Bay:The construction of Inorganic Elemental Fingerprint Based on Chemometric Analysis显示文摘The goal of this paper is to explore the relationship between the inorganic elemental fingerprint and the geographical origin identification of Meretricis concha,which is a commonly used marine traditional Chinese medicine(TCM) for the treatment of asthma and scald burns.For that,the inorganic elemental contents of Meretricis concha from five sampling points in Jiaozhou Bay have been determined by means of inductively coupled plasma optical emission spectrometry,and the comparative investigations based on the contents of 14 inorganic elements(Al,As,Cd,Co,Cr,Cu,Fe,Hg,Mn,Mo,Ni,Pb,Se and Zn) of the samples from Jiaozhou Bay and the previous reported Rushan Bay were performed.It has been found that the samples from the two bays are approximately classified into two kinds using hierarchical cluster analysis,and a four-factor model based on principle component analysis could explain approximately 75% of the detection data,also linear discriminant analysis can be used to develop a prediction model to distinguish the samples from Jiaozhou Bay and Rushan Bay with accuracy of about 93%.The results of the present investigation suggested that the inorganic elemental fingerprint based on the combination of the measured elemental content and chemometric analysis is a promising approach for verifying the geographical origin of Meretricis concha,and this strategy should be valuable for the authenticity discrimination of some marine TCM.SHAO Mingying LI Xuejie ZHENG Kang JIANG Man YAN Cuiiwe LI Yantuan 2016Journal of Ocean University of China2016,15,2:5
