1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 3Estimation of aboveground biomass using in situ hyperspectral measurements in five major grassland ecosystems on the Tibetan Plateau显示文摘Aims There are numerous grassland ecosystem types on the Tibetan Plateau.These include the alpine meadow and steppe and degraded alpine meadow and steppe.This study aimed at developing a method to estimate aboveground biomass(AGB)for these grasslands from hyperspectral data and to explore the feasibility of applying air/satellite-borne remote sensing techniques to AGB estimation at larger scales.Methods We carried out a field survey to collect hyperspectral reflectance and AGB for five major grassland ecosystems on the Tibetan Plateau and calculated seven narrow-band vegetation indices and the vegetation index based on universal pattern decomposition(VIUPD)from the spectra to estimate AGB.First,we investigated correlations between AGB and each of these vegetation indices to identify the best estimator of AGB for each ecosystem type.Next,we estimated AGB for the five pooled ecosystem types by developing models containing dummy variables.At last,we compared the predictions of simple regression models and the models containing dummy variables to seek an ecosystem type-independent model to improve prediction of AGB for these various grassland ecosystems from hyperspectral measurements.Important findings When we considered each ecosystem type separately,all eight vegetation indices provided good estimates of AGB,with the best predictor of AGB varying among different ecosystems.When AGB of all the five ecosystems was estimated together using a simple linear model,VIUPD showed the lowest prediction error among the eight vegetation indices.The regression models containing dummy variables predicted AGB with higher accuracy than the simple models,which could be attributed to the dummy variables accounting for the effects of ecosystem type on the relationship between AGB and vegetation index(VI).These results suggest that VIUPD is the best predictor of AGB among simple regression models.Moreover,both VIUPD and the soil-adjusted VI could provide accurate estimates of AGB with dummy variables integrated in regression models.Therefore,ground-based hyperspectral measurements are useful for estimating AGB,which indicates the potential of applying satellite/airborne remote sensing techniques to AGB estimation of these grasslands on the Tibetan Plateau.Miaogen Shen Yanhong Tang Julia Klein Pengcheng Zhang Song Gu Ayako Shimono Jin Chen 2008Journal of Plant Ecology2008,1,4:10
4Patterns of plant diversity at high altitudes on the Qinghai-Tibetan Plateau显示文摘To describe the biodiversity patterns of plants along an altitudinal gradient on the Qinghai-Tibetan Plateau and to clarify the bias in plant specimen records at high altitude.Methods We conducted a large-scale investigation of vegetation at a wide range of altitudes,focusing on a high-altitudinal range(3200-5200 m)at different locations on the Qinghai-Tibetan Plateau.We then compared the altitudinal distribution of plant species obtained from our field investigation with that in plant specimen records from published sources and an online database.Important Findings Our data provide evidence that altitude plays a large role in regulating species composition on the Qinghai-Tibetan Plateau.We could not,however,detect a clear relationship between altitude and species richness,although a weak monotonically increasing trend of richness was detected with increasing altitude.According to specimen records,most species have been sampled at a wide range of altitudes,and the average range of 145 species is>2000 m.Despite this wide range,more than half of the species we observed were at higher altitudes than the specimen records indicate.High-altitude areas have probably been so poorly sampled that only a small fraction of the resident species has been recorded.This study clearly shows the regional bias of specimen records in the Qinghai-Tibetan Plateau.Ayako Shimono Huakun Zhou Haihua Shen Mitsuru Hirota Toshiyuki Ohtsuka Yanhong Tang 2010Journal of Plant Ecology2010,3,1:6
5Altitudinal variation of ecosystem CO_(2) fluxes in an alpine grassland from 3600 to 4200 m显示文摘Aims Recent studies have recognized the alpine grasslands on the Qinghai-Tibetan plateau as a significant sink for atmospheric CO_(2).The carbon-sink strength may differ among grassland ecosystems at various altitudes because of contrasting biotic and physical environments.This study aims(i)to clarify the altitudinal pattern of ecosystem CO_(2) fluxes,including gross primary production(GPP),daytime ecosystem respiration(Redaytime)and net ecosystem production(NEP),during the period with peak above-ground biomass;and(ii)to elucidate the effects of biotic and abiotic factors on the altitudinal variation of ecosystem CO_(2) fluxes.Methods Ecosystem CO_(2) fluxes and abiotic and biotic environmental factors were measured in an alpine grassland at four altitudes from 3600 to 4200 m along a slope of the Qilian Mountains on the northwestern Qinghai-Tibetan Plateau during the growing season of 2007.We used a closed-chamber method combined with shade screens and an opaque cloth to measure several carbon fluxes,GPP,Redaytime and NEP,and factors,light-response curve for GPP and temperature sensitivity of Redaytime.Above-and below-ground biomasses and soil C and N contents at each measurement point were also measured.Important Findings(i)Altitudinal pattern of ecosystem CO_(2) fluxes:The maximum net ecosystem CO_(2) flux(NEPmax),i.e.the potential ecosystem CO_(2) sink strength,was markedly different among the four altitudes.NEPmax was higher at the highest and lowest sites,ap proximately7.460.9 and6.760.6 lmol CO_(2) m^(-2)s^(-1)(mean 6 standard error),respectively,but smaller at the intermediate altitude sites(3800 and 4000 m).The altitudinal pattern of maximum gross primary production was similar to that of NEPmax.The Redaytime,however,was significantly higher at the lowest altitude(3.460.3 lmol CO_(2) m^(-2)s^(-1))than at the other three altitudes.(ii)Altitudinal variation of vegetation biomass:The aboveground biomass was higher at the highest altitude(154627 g DW m2)than at the other altitudes,which we attribute mainly to the large biomass in cushion plants at the highest altitude.The small above-ground biomass at the lower altitudes was probably due to heavy grazing during the growing season.(iii)Features of ecosystem CO_(2) fluxes:Redaytime and GPP were positively correlated with above-ground biomass.The low ratio of Redaytime to GPP at either the measurement point or the site level suggests that CO_(2) uptake efficiency tends to be higher at higher altitudes,which indicates a high potential sink strength for atmospheric CO_(2) despite the low temperature at high altitudes.The results suggest that the effect of grazing intensity on ecosystem carbon dynamics,partly by decreasing vegetation biomass,should be clarified further.Mitsuru Hirota Pengcheng Zhang Song Gu Mingyuan Du Ayako Shimono Haihua Shen Yingnian Li Yanhong Tang 2009Journal of Plant Ecology2009,2,4:6
6Grazing alters warming effects on leaf photosynthesis and respiration in Gentiana straminea,an alpine forb species显示文摘Aims Vast grasslands on the Tibetan Plateau are almost all under livestock grazing.It is unclear,however,what is the role that the grazing will play in carbon cycle of the grassland under future climate warming.We found in our previous study that experimental warming can shift the optimum temperature of saturated photosynthetic rate into higher temperature in alpine plants.In this study,we proposed and tested the hypothesis that livestock grazing would alter the warming effect on photosynthetic and respiration through changing physical environments of grassland plants.Methods Experimental warming was carried by using an infrared heating system to increase the air temperature by 1.2 and 1.7℃ during the day and night,respectively.The warming and ambient temperature treatments were crossed over to the two grazing treatments,grazing and un-grazed treatments,respectively.To assess the effects of grazing and warming,we examined photosynthesis,dark respiration,maximum rates of the photosynthetic electron transport(J_(max)),RuBP carboxylation(V_(cmax))and temperature sensitivity of respiration Q_(10) in Gentiana straminea,an alpine species widely distributed on the Tibetan grassland.Leaf morphological and chemical properties were also examined to understand the physiological responses.Important findings 1)Light-saturated photosynthetic rate(A_(max))of G.straminea showed similar temperature optimum at around 16℃ in plants from all experimental conditions.Experimental warming increased A_(max) at all measuring temperatures from 10 to 25℃,but the positive effect of the warming occurred only in plants grown under the un-grazed conditions.Under the same measuring temperature,A_(max) was significantly higher in plants from the grazed than the un-grazed condition.2)There was significant crossing effect of warming and grazing on the temperature sensitivity(Q_(10))of leaf dark respiration.Under the un-grazed condition,plants from the warming treatment showed lower respiration rate but similar Q_(10) in comparison with plants from the ambient temperature treatment.However,under the grazed condition Q_(10) was significantly lower in plants from the warming than the ambient treatment.3)The results indicate that livestock grazing can alter the warming effects on leaf photosynthesis and temperature sensitivity of leaf dark respiration through changing physical environment of the grassland plants.The study suggests for the first time that grazing effects should be taken into account in predicting global warming effects on photosynthesis and respiration of plants in those grasslands with livestock grazing.Haihua Shen Shiping Wang Yanhong Tang 2013Journal of Plant Ecology2013,6,5:5
7Leaf photosynthesis and simulated carbon budget of Gentiana straminea from a decade-long warming experiment显示文摘Aims Alpine ecosystems may experience larger temperature increases due to global warming as compared with lowland ecosystems.Information on physiological adjustment of alpine plants to temperature changes can provide insights into our understanding how these plants are responding to current and future warming.We tested the hypothesis that alpine plants would exhibit acclimation in photosynthesis and respiration under long-term elevated temperature,and the acclimation may relatively increase leaf carbon gain under warming conditions.Methods Open-top chambers(OTCs)were set up for a period of 11 years to artificially increase the temperature in an alpine meadow ecosystem.We measured leaf photosynthesis and dark respiration under different light,temperature and ambient CO_(2)concentrations for Gentiana straminea,a species widely distributed on the Tibetan Plateau.Maximum rates of the photosynthetic electron transport(J_(max)),RuBP carboxylation(V_(cmax))and temperature sensitivity of respiration Q10 were obtained from the measurements.We further estimated the leaf carbon budget of G.straminea using the physiological parameters and environmental variables obtained in the study.Important findings 1)The OTCs consistently elevated the daily mean air temperature by;1.6℃and soil temperature by;0.5℃during the growing season.2)Despite the small difference in the temperature environment,there was strong tendency in the temperature acclimation of photosynthesis.The estimated temperature optimum of light-saturated photosynthetic CO_(2)uptake(A_(max))shifted;1℃higher from the plants under the ambient regime to those under the OTCs warming regime,and the Amax was significantly lower in the warming-acclimated leaves than the leaves outside the OTCs.3)Temperature acclimation of respiration was large and significant:the dark respiration rates of leaves developed in the warming regime were significantly lower than leaves from the ambient environments.4)The simulated net leaf carbon gain was significantly lower in the in situ leaves under the OTCs warming regime than under the ambient open regime.However,in comparison with the assumed non-acclimation leaves,the in situ warmingacclimated leaves exhibited significantly higher net leaf carbon gain.5)The results suggest that there was a strong and significant temperature acclimation in physiology of G.straminea in response to long-term warming,and the physiological acclimation can reduce the decrease of leaf carbon gain,i.e.increase relatively leaf carbon gain under the warming condition in the alpine species.Haihua Shen Julia AKlein Xinquan Zhao Yanhong Tang 2009Journal of Plant Ecology2009,2,4:5
8MODIS observations of water color of the largest 10 lakes in China between 2000 and 2012显示文摘Forel-Ule(FU)index of water color is an important parameter in traditional water quality investigations.We retrieved the FU index of the largest 10 lakes in China during 2000-2012 from MODerate-resolution Imaging Spectroradiometer surface reflectance product(MOD09)images.Since FU index is an optical parameter,it can be derived from optical remote sensing data by direct formulas,which is invariant with region and season.Based on validation by in situ measured reflectance data,the FU index products are reliable,with average relative error of 7.7%.FU index can be used to roughly assess water clarity:the clearer a water body is,and the bluer it is in color,the smaller its FU index is.FU index can also be used to roughly classify trophic state into three classes:oligotrophic,mesotrophic,and eutrophic.We analyzed the spatial,interannual,and seasonal variations of the FU index and its implications for water clarity and trophic state,and the findings are mostly consistent with the results from related literature.All in all,it might be a feasible way to roughly assess inland water quality by FU index in large region and over long time period.Junsheng Li Shenglei Wang Yanhong Wu Bing Zhang Xiaoling Chen Fangfang Zhang Qian Shen Dailiang Peng Liqiao Tian 2016International Journal of Digital Earth2016,9,8:5

10Influences of air CO2 on hydrochemistry of drip water and implications for paleoclimate study in a stream-developed cave, SW China显示文摘Cave air CO_2 is a vital part of the cave environment. Most studies about cave air CO_2 variations are performed in caves with no streams; there are few studies to date regarding the relationship of cave air CO_2 variations and drip water hydrochemistry in underground stream–developed caves. To study the relationship of underground stream, drip water, and cave air CO_2, monthly and daily monitoring of air CO_2 and of underground stream and drip water was performed in Xueyu Cave from 2012 to 2013.The results revealed that there was marked seasonal variation of air CO_2 and stream hydrochemistry in the cave. Daily variations of cave air CO_2, and of stream and drip water hydrochemistry, were notable during continuous monitoring.A dilution effect was observed by analyzing hydrochemical variations in underground stream and drip water after rainfall. High cave air CO_2 along with low p H and low δ^(13)C DIC in stream and drip water indicated that air CO_2 was one of the dominant factors controlling stream and drip water hydrochemistry on a daily scale. On a seasonal scale, stream flows may promote increased cave air CO_2 in summer; in turn, the higher cave air CO_2 could inhibit degassing of drip water and make calcite δ^(13)C more negative. Variation of calcite δ^(13)C(precipitated from drip water) was in reverse of monthly temperature, soil CO_2, and cave air CO_2. Therefore,calcite δ^(13)C in Xueyu Cave could be used to determine monthly changes outside the cave. 11Characterization of CO_(2) flux in three Kobresia meadows differing in dominant species显示文摘Aims Kobresia meadows,the dominant species of which differ in different habitats,cover a large area of alpine grassland on the QinghaiTibetan Plateau and act as potential CO_(2) sinks.Kobresia meadows with different dominant species may differ in carbon sink strength.We aimed to test the hypothesis and to clarify the differences in CO_(2) sink strength among three major Kobresia meadows on the plateau and the mechanisms underlying these differences.Methods We measured the net ecosystem exchange flux(NEE),ecosystem respiration flux(ER),aboveground biomass(AGB)and environmental variables in three Kobresia meadows,dominated by K.pygmaea,K.humilis,or K.tibetica,respectively,in Haibei,Qinghai.NEE and ER were measured by a closed-chamber method.Environmental variables,including photosynthetic photon flux density(PPFD),air and soil temperature and air and soil moisture,were monitored during the above flux measurements.Important findings The measured peak AGB increased with soil water content and was 365,402 and 434 g dry weight m2 for K.pygmaea,K.humilis and K.tibetica meadow,respectively.From the maximum ecosystem photosynthetic rate in relation to PPFD measured during the growing season,we estimated gross ecosystem photosynthetic potential(GEPmax)as 22.2,29.9 and 37.8 lmol CO_(2) m2 s1 for K.pygmaea,K.humilis and K.tibetica meadow,respectively.We estimated the respective gross primary production(GPP)values as 799,1063 and 1158 g C m^(-2) year^(-1) and ER as 722,914 and 1011 g C m^(-2) year^(-1).Average net ecosystem production(NEP)was estimated to be 76.9,149.4 and 147.6 g C m^(-2) year^(-1) in K.pygmaea,K.humilis and K.tibetica meadows,respectively.The results indicate that(i)the three meadows were CO_(2) sinks during the study period and(ii)Kobresia meadows dominated by different species can differ considerably in carbon sink strength even under the same climatic conditions,which suggests the importance of characterizing spatial heterogeneity of carbon dynamics in the future.Pengcheng Zhang Mitsuru Hirota Haihua Shen Akinori Yamamoto Shigeru Mariko Yanhong Tang 2009Journal of Plant Ecology2009,2,4:4

13Activation of disease resistance against Botryosphaeria dothidea by downregulating the expression of MdSYP121 in apple显示文摘In plants,the vesicle fusion process plays a vital role in pathogen defence.However,the importance of the vesicle fusion process in apple ring rot has not been studied.Here,we isolated and characterised the apple syntaxin gene MdSYP121.Silencing the MdSYP121 gene in transgenic apple calli increased tolerance to Botryosphaeria dothidea infection;this increased tolerance was correlated with salicylic acid(SA)synthesis-related and signalling-related gene transcription.In contrast,overexpressing MdSYP121 in apple calli resulted in the opposite phenotypes.In addition,the results of RNA sequencing(RNA-Seq)and quantitative real-time PCR(qRT-PCR)assays suggested that MdSYP121 plays an important role in responses to oxidation–reduction reactions.Silencing MdSYP121 in apple calli enhanced the expression levels of reactive oxygen species(ROS)-related genes and the activity of ROS-related enzymes.The enhanced defence response status in MdSYP121-RNAi lines suggests that syntaxins are involved in the defence response to B.dothidea.More importantly,we showed that MdSYP121 forms a soluble N-ethylmaleimide-sensitive factor attachment protein receptor(SNARE)complex with MdSNAP33,and the complex may participate in regulating resistance to B.dothidea.In conclusion,by regulating the interaction of SA pathway and oxidation–reduction process,MdSYP121 can influence the pathogen infection process in apple.Xiaowen He Yanhong Huo Xiuxia Liu Qianqian Zhou Shouqian Feng Xiang Shen Baohua Li Shujing Wu Xuesen Chen 2018Horticulture Research2018,5,1:3
14Diagnosis and clinical manifestations of subacute combined degeneration of the spinal cord: Analysis of 21 cases显示文摘BACKGROUND: Subacute combined degeneration of the spinal cord is caused by vitamin B12 deficiency and is a kind of degenerative disease owing the characteristics of nervous system diseases. In addition, different patients have variously clinical manifestations and various prognoses after vitamin B12 therapy. OBJECTIVE: To investigate and analyze diagnosis, clinical manifestations and prognosis of subacute combined degeneration of the spinal cord. DESIGN: Case analysis. SETTING: Department of Neurology, the Third Hospital of Peking University. PARTICIPANTS: A total of 21 subacute combined degeneration of the spinal cord patients including 14 males and 7 females aged from 33 to 82 years were selected from Department of Neurology, the Third Hospital of Peking University from January 1999 to December 2005. Duration from onset to final diagnosis lasted for 1.5–108 months. All patients had typically clinical manifestations; meanwhile, level of serum vitamin B12 was decreased and/or vitamin B12 therapy was effective. All patients provided the confirmed consent. METHODS: Clinical data of 21 subacute combined degeneration of the spinal cord patients were retrospectively analyzed, while general data and clinical characteristics were recorded at the same time. Levels of blood routine, serum vitamin B12 and homocysteine were measured at the phase of hospitalization. Normal value of serum vitamin B12 was 187–1 059 ng/L and normal value of serum homocysteine was 5– 15 μmol/L. All patients received neuroelectrophysiological examination and 15 patients received MRI examinations of spinal cord. After final diagnosis, patients were given vitamin B12 therapy. And follow-up was performed to investigate the prognosis. MAIN OUTCOME MEASURES: ① Levels of blood routine, serum vitamin B12 and homocysteine; ② results of neuroelectrophysiological examination; ③ results of MRI examination of spinal cord; ④ prognosis. RESULTS: Clinical data of 21 patients and follow-up data of 20 patients were involved in the final analysis and 1 patient was lost because of living in the other province. ① Clinical manifestations: All 21 patients had typically clinical manifestations. The original symptoms included numbness of lower and/or upper limbs (5 cases), unstable gait (3 cases), limb asthenia (4 cases), limb numbness combined with light asthenia (5 cases), limb numbness combined with unskillful activity (3 cases), and limb numbness combined with unstable gait (1 case). ② Experimental results: Eight subacute combined degeneration of the spinal cord patients accompanied with mild-severe anemia and mean corpuscular volume of 13 patients were increased. Among 13 subacute combined degeneration of the spinal cord patients not administrating vitamin B12 before hospitalization, the levels of serum vitamin B12 of 2 patients were not measured but those of other patients were decreased. After vitamin B12 therapy,the levels of serum vitamin B12 of 8 patients were normal or increased. In addition, the levels of serum homocysteine of 6 patients were not measured but those of 7 patients were increased. While, the levels of homocysteine of 5 following-up patients were normal. The levels of serum vitamin B12 of 8 patients who received with vitamin B12 therapy before hospitalization were normal or increased. Among them,the levels of homocysteine were not measured in 4 patients, those of 3 patients were increased, and that of 1 patient was normal. ③ Results of neuroelectrophysiological examination: Among all patients, 95% (20/21) patients had abnormal sensory-evoked potential, 89% (8/9) patients had abnormal motor evoked potential, 67% (10/15) patients had abnormal nerve conduction, 13% (2/15) patients had neurogenic muscle injury showed by electromyography (EMG), 70% (7/10) patients had abnormal brain-stem auditory evoked potential, and 40% (4/10) patients had abnormal visual evokedpotential. ④ Results of MRI examination of spinal cord: MRI examination demonstrated that 40% (6/15) patients had spinal cord lesion, but spinal cord lesion disappeared in 2 patients during follow up. In addition, clinical manifestations of patients were improved after standard vitamin B12 therapy. CONCLUSION: Nervous system lesion caused by vitamin B12 deficiency is not only involved in spinal cord, also in peripheral nerve, optic nerve, auditory pathway, etc. Diagnosis of the lesion depends on clinical characteristics and level of serum vitamin B12. 15Study protocol of the Asian XELIRI ProjecT(AXEPT):a multinational,randomized,non-inferiority,phase Ⅲ trial of second-line chemotherapy for metastatic colorectal cancer, comparing the eicacy and safety of XELIRI with or without bevacizumab versus FOLFIRI w显示文摘Background: Capecitabine and irinotecan combination therapy(XELIRI) has been examined at various dose levels to treat metastatic colorectal cancer(m CRC). Recently, in the Association of Medical Oncology of the German Cancer Society(AIO) 0604 trial, tri?weekly XELIRI plus bevacizumab, with reduced doses of irinotecan(200 mg/m^2 on day 1) and capecitabine(1600 mg/m^2 on days 1–14), repeated every 3 weeks, has shown favorable tolerability and eicacy which were comparable to those of capecitabine and oxaliplatin(XELOX) plus bevacizumab. The doses of capecit?abine and irinotecan in the AIO trial are considered optimal. In a phase I/II study, XELIRI plus bevacizumab(BIX) as second?line chemotherapy was well tolerated and had promising eicacy in Japanese patients.Methods: The Asian XELIRI Projec T(AXEPT) is an East Asian collaborative, open?labelled, randomized, phase Ⅲ clinical trial which was designed to demonstrate the non?inferiority of XELIRI with or without bevacizumab versus standard FOLFIRI(5?fluorouracil, leucovorin, and irinotecan combination) with or without bevacizumab as second?line chemo?therapy for patients with m CRC. 16A regional ionospheric TEC mapping technique over China and adjacent areas: GNSS data processing and DINEOF analysis显示文摘A technique is developed to derive two-dimensional maps of total electron content(TEC) over China and adjacent areas using Global Navigation Satellite System(GNSS) data from the Crustal Movement Observation Network of China(CMONOC) and the International Global Navigation Satellite System Service(IGS). A revised self-calibration of pseudo-range errors(SCORE) algorithm is used to derive the TEC and to determine the Differential Code Biases(DCBs) simultaneously. The accuracy and validity of this technique is verified in two ways. Firstly, the estimated TEC is compared with the results derived using DCBs from Center for Orbit Determination of Europe(CODE) under different solar activity conditions and seasons; secondly, sample TEC along the receiver-to-satellite ray paths are simulated by Ne Quick model and are reprocessed by this TEC derivation technique to make the accuracy test. Two-dimensional maps of vertical TEC of ionospheric pierce points(IPPs) are obtained accordingly with a time resolution of 30 s. The data interpolating empirical orthogonal functions(DINEOF) technique is then used to make the extrapolation for the unknown or missing data points. The optimal EOF modes for data reconstruction are specified via cross-validation method. 17Papaya CpbHLH1/2 regulate carotenoid biosynthesis-related genes during papaya fruit ripening显示文摘The ripening of papaya is a physiological and metabolic process associated with accumulation of carotenoids,alternation of flesh color and flavor,which depending on genotype and external factors such as light and hormone.Transcription factors regulating carotenoid biosynthesis have not been analyzed during papaya fruit ripening.RNA-Seq experiments were implemented using different ripening stages of papaya fruit from two papaya varieties.Cis-elements in lycopeneβ-cyclase genes(CpCYC-B and CpLCY-B)were identified,and followed by genome-wide analysis to identify transcription factors binding to these cis-elements,resulting in the identification of CpbHLH1 and CpbHLH2,two bHLH genes.The expressions of CpbHLH1/2 were changed during fruit development,coupled with transcript increase of carotenoid biosynthesis-related genes including CpCYC-B,CpLCY-B,CpPDS2,CpZDS,CpLCY-E,and CpCHY-B.Yeast onehybrid(Y1H)and transient expression assay revealed that CpbHLH1/2 could bind to the promoters of CpCYC-B and CpLCY-B,and regulate their transcriptions.In response to strong light,the results of elevated expression of carotenoid biosynthesis-related genes and the changed expression of CpbHLH1/2 indicated that CpbHLH1/2 were involved in light-mediated mechanisms of regulating critical genes in the carotenoid biosynthesis pathway.Collectively,our findings demonstrated several TF family members participating in the regulation of carotenoid genes and proved that CpbHLH1 and CpbHLH2 individually regulated the transcription of lycopeneβ-cyclase genes(CpCYC-B and CpLCY-B).This study yielded novel findings on regulatory mechanism of carotenoid biosynthesis during papaya fruit ripening.Dong Zhou Yanhong Shen Ping Zhou Mahpara Fatima Jishan Lin Jingjing Yue Xingtan Zhang Li-Yu Chen Ray Ming 2019Horticulture Research2019,6,1:3

19Phylogeography of Potentilla fruticosa,an alpine shrub on the Qinghai-Tibetan Plateau显示文摘Aims Our objectives were(i)to elucidate the phylogeography of chloroplast DNA(cpDNA)in Potentilla fruticosa in relation to Quaternary climate change and postglacial colonization,(ii)to infer historical population range expansion using mismatch distribution analyses and(iii)to locate the refugia of this alpine species on the Qinghai-Tibetan plateau during glacial-interglacial periods.Methods Potentilla fruticosa is a widespread species distributed on the Qinghai-Tibetan Plateau.We sampled leaves of P.fruticosa from 10 locations along a route of;1300 km from the northeastern plateau(Haibei,Qinghai)to the southern plateau(Dangxiong,Tibet).We examined the cpDNA of 15 haplotypes for 87 individuals from the 10 populations based on the sequence data from;1000 base pairs of the trnS-trnG and rpl20-rps12.Phylogenetic relationship of haplotypes was analyzed using the Phylip software package and the program TCS.The diversity of populations indices was obtained using the program ARLEQUIN.Important Findings With the limited samples,we found that(i)higher nucleotide diversity often occurs in high-altitude populations,(ii)the ancestral haplotypes distribute in the populations with higher nucleotide diversity than recent haplotypes,(iii)the expansion time of population in the high altitudes was estimated to be approximately at 52-25 ka BP(1000 years Before Present,where'Present'is AD 1950)and that in the low altitudes to be;5.1-2.5 ka BPand(iv)the source location of P.fruticosa is at the high altitudes,which might provide refugia for the species during the interglacial warm periods.The species expanded from the high-elevated locations on the Tanggula Mountains during the Holocene.Cui Li Ayako Shimono Haihua Shen Yanhong Tang 2010Journal of Plant Ecology2010,3,1:2
20Internal Loads and Bioavailability of Phosphorus and Nitrogen in Dianchi Lake,China显示文摘Sediments have a significant influence on the cycling of nutrient elements in lake environments. In order to assess the distribution characteristics and estimate the bioavailability of phosphorus and nitrogen in Dianchi Lake, organic and inorganic phosphorus and nitrogen forms were analysed. The 210 Pb radiometric dating method was employed to study temporal changes in the phosphorus and nitrogen pools in Dianchi Lake. The result show that the total phosphorus(TP) and total nitrogen(TN) were both at high concentrations, ranging from 697.5–3210.0 mg/kg and 1263.7–7155.2 mg/kg, respectively. Inorganic phosphorus(IP) and total organic nitrogen(TON) were the main constituents, at percentages of 59%–78% and 74%–95%, respectively, in the sediments. Spatially, there was a decreasing trend in phosphorus and nitrogen contents from the south and north to the lake centre, which is related to the distribution pattern of local economic production. The burial rates of the various phosphorus and nitrogen forms increased in same spatially and over time. Particularly in the past two decades, the burial rates doubled, with that TN reached to 1.287 mg/(cm^2·yr) in 2014. As the most reactive forms, nitrate nitrogen(NO_3-N) and ammonia nitrogen(NH_4-N) were buried more rapidly in the south region, implying that the potential for releasing sedimentary nitrogen increased from north to south. Based on their concentrations and burial rates, the internal loads of phosphorus and nitrogen were analysed for the last century. A TP pool of 71597.6 t and a TN pool of 81191.7 t were estimated for Dianchi Lake.
