| 2 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. | 3 | Quantifying public health benefits of environmental strategy of PM2.5 air quality management in Beijing–Tianjin–Hebei region,China显示文摘In 2013,China issued 'Air Pollution Prevention and Control Action Plan(Action Plan)' to improve air quality.To assess the benefits of this program in Beijing-Tianjin-Hebei(BTH)region,where the density of population and emissions vary greatly,we simulated the air quality benefit based on Ben MAP to satisfy the Action Plan.In this study,we estimate PM_(2.5) concentration using Voronoi spatial interpolation method on a grid with a spatial resolution of 1 × 1 km^2.Combined with the exposure-response function between PM_(2.5) concentration and health endpoints,health effects of PM_(2.5) exposure are analyzed.The economic loss is assessed by using the willingness to pay(WTP) method and human capital(HC) method.When the PM_(2.5) concentration falls by 25% in BTH and reached 60 μg/m^3 in Beijing,the avoiding deaths will be in the range of 3175 to 14051 based on different functions each year.Of the estimated mortality attributable to all causes,3117 annual deaths were due to lung cancer,1924 – 6318 annual deaths were due to cardiovascular,and343 – 1697 annual deaths were due to respiratory.Based on WTP,the estimated monetary values for the avoided cases of all cause mortality,cardiovascular mortality,respiratory mortality and lung cancer ranged from 1110 to 29632,673 to 13325,120 to 3579,1091 to 6574 million yuan,respectively.Based on HC,the corresponding values for the avoided cases of these four mortalities were 267 to 1178,161 to 529,29 to 143 and 261 million yuan,respectively. | Li Chen Mengshuang Shi Suhuan Li Shuang Gao Hui Zhang Yanling Sun Jian Mao Zhipeng Bai Zhongliang Wang Jiang Zhou | 2017 | Journal of Environmental Sciences2017,29,7: | 6 |
| 4 | Thrombolysis with alteplase 3-4.5 hours after acute ischaemic stroke:the first multicentre,phase III trial in China显示文摘background and purpose Data on the efficacy and safety of alteplase for acute ischaemic stroke(AIS)administered 3-4.5 hours after the onset of stroke symptoms in Chinese patients is limited.We sought to determine whether AIS patients would benefit from thrombolysis with alteplase between 3 and 4.5 hours after the onset of stroke symptoms in a prospective,multicentre,single-arm trial in China.Materials and methods Eligible AIS patients were given 0.9 mg/kg alteplase intravenously.The primary efficacy endpoint was a favourable outcome at 3 months,defined as a score of 0 or 1 on the modified Rankin Scale.Thresholds for the primary efficacy endpoint were determined to be 40%based on the literature review.The primary safety endpoint was symptomatic intracranial haemorrhage(sICH)according to the European Cooperative Acute Stroke Study III(ECASS III)trial definition.Post hoc analysis between this study and the ECASS III trial were compared using the propensity score matching(PSM)method.results A total of 120 eligible AIS patients from 11 sites in China received thrombolysis therapy in this study.The median time from onset of symptoms to needle was 3 hours 54 min.The percentage of patients with a favourable outcome was 63.3%(95%CI 54.4 to 71.4),significantly higher than the predefined threshold(p<0.0001).Three patients(2.5%,95%CI 0.5 to 7.1)had sICH,including two fatal sICH.Six patients died within 3 months after treatment.The post hoc PSM analysis showed a numerically higher rate of the primary efficacy endpoint in this study(63.3%)than the matched placebo arm(56.7%)in the ECASS III trial.Conclusions Intravenous alteplase with a standard dose administered between 3 and 4.5 hours after onset of symptoms is effective and safe for Chinese AIS patients. | Huaguang Zheng Yi Yang Huisheng Chen Chuanling Li Yangkun Chen Fu-Dong Shi Li Yang Xiaoping Cui Zuneng Lu Yanling Liang Songbiao Cui Anding Xu Yiqing Wu Yaqing Sun Yongjun Wang | 2020 | Stroke & Vascular Neurology2020,5,3: | 4 |
| 5 | Effects of shading after pollination on kernel filling and physicochemical quality traits of waxy maize显示文摘Understanding the effects of shading after pollination on kernel filling and physicochemical properties of waxy maize could improve kernel quality.Experiments were conducted to investigate the effects of shading(30% and 50% light deprivation,taken plants without shading as control) on kernel weight,size,and physicochemical properties during kernel development in 2013 and 2014 using two waxy maize varieties(Suyunuo 5 and Yunuo 7).Results indicated that shading reduced kernel filling rate and decreased kernel size and weight,and the influence was large under severe light deprivation conditions.The large reduction in kernel weight and volume of Suyunuo 5 in response to shading indicated that it was more sensitive to shading than Yunuo 7.Starch content was reduced and protein content was increased by shading,especially under severe shading after 22 days after pollination(DAP).The iodine binding capacity of Yunuo 7 was not affected by shading at fresh and maturity stages but was gradually decreased by shading at the newly formed stage,while the values for Suyunuo 5were decreased at 7 and 40 DAP only by moderate shading and were similar among three treatments at 22 DAP.Severe shading decreased crystallinity at all kernel development stages,whereas moderate shading decreased crystallinity at fresh stage and increased it at mature stage for Suyunuo 5.Crystallinity in Yunuo 7 was increased by shading at 7 DAP and decreased by shading at 40 DAP,whereas the value at 22 DAP was increased by moderate shading and reduced by severe shading,respectively.The average gelatinization temperatures at different stages were decreased by shading and showed no difference between two shading levels.The retrogradation percentage at 7 DAP for both varieties was increased by shading.The value at 22 DAP was increased by moderate shading for Suyunuo 5 and decreased by severe shading for Yunuo 7,respectively.The retrogradation percentage at 40 DAP was decreased by shading treatments for Suyunuo 5 and reduced only by moderate shading for Yunuo 7.Peak viscosity was decreased by shading at fresh stage for Yunuo 7 and at maturity for Suyunuo 5.In conclusion,shading after pollination inhibited kernel filling of waxy maize and reduced paste viscosity quality,but kernel retrograde quality,crystallinity and starch iodine binding capacity in response to shading were dependent on stage and variety. | Huan Yang Yanling Shi Renchao Xu Dalei Lu Weiping Lu | 2016 | The Crop Journal2016,4,3: | 4 |

| 7 | Isolation and Characterization of Acid-soluble Collagen and Pepsin-soluble Collagen from the Skin of Hybrid Sturgeon显示文摘By using the wastes fish skin of sturgeon processed as a raw material, a macromolecule biomaterial of collagen was extracted. Acid-soluble collagen(ASC) and pepsin-soluble collagen(PSC) were successfully isolated from the skin of hybrid sturgeon with two extraction methods. The yields of ASC and PSC based on the wet weight of skin were 5.73 ± 0.11% and 10.26 ± 0.39%, respectively. The denaturation and melting points of ASC(26.83 ℃ and 110.49 ℃) and PSC(26.54 ℃ and 102.99 ℃) were assessed by Circular dichroism(CD) and Differential scanning calorimetry(DSC). ASC and PSC appeared to be dense sheet-like film linked by random-coiled filaments under scanning electron microscopy(SEM). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) and Fourier transform infrared spectroscopy(FTIR) confirmed that both the ASC and PSC were Type I collagen and maintained a complete triple helix structure. | 10 | Changes in compressed neurons from dogs with acute and severe cauda equina constrictions following intrathecal injection of brain-derived neurotrophic factor-conjugated polymer nanoparticles显示文摘This study established a dog model of acute multiple cauda equina constriction by experimental constriction injury (48 hours) of the lumbosacral central processes in dorsal root ganglia neurons. The repair effect of intrathecal injection of brain-derived neurotrophic factor with 15 mg encapsulated biodegradable poly(lactide-co-glycolide) nanoparticles on this injury was then analyzed. Dorsal root ganglion cells (L7) of all experimental dogs were analyzed using hematoxylin-eosin staining and immunohistochemistry at 1, 2 and 4 weeks following model induction. Intrathecal injection of brain-derived neurotrophic factor can relieve degeneration and inflammation, and elevate the expression of brain-derived neurotrophic factor in sensory neurons of compressed dorsal root ganglion. Simultaneously, intrathecal injection of brain-derived neurotrophic factor obviously improved neurological function in the dog model of acute multiple cauda equina constriction. | 13 | One-step post-treatment boosts thermoelectric properties of PEDOT:PSS flexible thin films显示文摘Developing high-performance poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate)(PEDOT:PSS)sig-nificantly widens the practical applications of flexible organic thermoelectric devices,while the water-based co-solvent dopants and/or post-treatments are still rarely studied so far.Here,we develop a one-step post-treatment to improve the power factor of PEDOT:PSS films by using a water-based solution,which is composed of co-solvent(polar solvent dimethylacetamide(DMAC)and deionized water)and organic reducing agent L-ascorbic acid(LAA).The 80 vol.%DMAC solution significantly boosts the room-temperature electrical conductivity of the films from 5 to 964 S cm^(−1),while the Seebeck coefficient can be further enhanced from 18.7 to 25μV K−1 by treating with 0.5 mol L−1 LAA,contributing to a sig-nificantly improved power factor of 55.3μW m^(−1)K^(−2).The boosted electrical conductivity is ascribed to the separated PEDOT and PSS phases triggered by the high dielectric constant and polarity of DMAC;while the improved Seebeck coefficient is attributed to the reduced oxidation degree of PEDOT from the reducing agent LAA,both confirmed by the comprehensive structural and morphological characteri-zations.Furthermore,a maximum power factor of 64.4μW m^(−1)K^(−2)can be achieved at 360 K and the observed temperature-dependent electrical transport behavior can be well explained by the Mott variable range hopping model.Besides,a flexible thermoelectric device,assembled by the as-fabricated PEDOT:PSS films,exhibits a maximum output power of∼23 nW at a temperature difference of 25 K,indicating the potential for applying to low-grade wearable electronics. | Xingyu Liu Xiao-Lei Shi Li Zhang Wei-Di Liu Yanling Yang Zhi-Gang Chen | 2023 | Journal of Materials Science & Technology2023,,1: | 2 |
| 14 | Efficacy and safety of single-bolus tenecteplase compared with front-loaded alteplase in Chinese patients with acute myocardial infarction显示文摘Background and Objective Previous study showed tenecteplase and alteplaxe were equovalent for 30-day mortality in the treatment of acute myocardial infarction. The purpose of this open-label,randomized,multi-center,angiographic trial was to assess the efficacy and safety of tenecteplase compared with alteplase in Chinese patients with acute myocardial infarction. Methods We recruited patients with acute ST-elevation myocardial infarction presenting within 6 hours of symptom onset from October,2002 to March,2004,in 5 hospitals in Beijing. After giving informed consent,patients were randomly assigned a single-bolus injection of tenecteplase(30-50 mg according to body weight) or front loaded alteplase(100 mg) ,and underwent coronary angiography at 90 min after starting the study drug. All patients received aspirin and heparin(target activated partial thromboplastin time 50-70 s) . The primary efficacy end point was the rate of TIMI grade 3 flow at 90 minutes. Other efficacy end points included TIMI grade 2/3 flow at 90 minutes. Safety end points included all stroke,intracranial hemorrhage(ICH) ,moderate/severe hemorrhage(except for ICH) ,all-cause mortality at 30-days,and major non-fatal cardiac events at 30 days. Results Overall 110 patients were eligible for statistical analysis,with 58 patients assigned to receive tenecteplase and 52 patients to alteplase. Tenecteplase produced a rate of TIMI grade 3 flow at 90 minutes after the start of thrombolysis(68.4%) similar to that of alteplase(66.7%,P=1.0) ;the rates of TIMI grade 2 or 3 were similar for patients treated with tenecteplase versus alteplase(89.5% versus 80.4%,respectively,P=0.278) . At 30 days,rates for all strokes were similar for the two groups(5.17% for tenecteplase and 1.92% for alteplase,P=0.62) ;rates of ICH were 3.45% and 1.92%(tenecteplase and rt-PA,P=1.00) respectively. The rate of moderate/severe hemorrhage was 8.62% with tenecteplase and 5.77% with alteplase(P=0.72) ;total mortality was almost identical in the two groups(13.8% versus 9.6%,respectively,P=0.565) while the rates of non-fatal cardiac complications were 10.35% and 11.54%(tenecteplase and alteplase,P=1.0) . Conclusions The efficacy of a single-bolus,weight-adjusted tenecteplase fibrinolytic regimen is equivalent to front-loaded alteplase in terms of the rates of TIMI grade 3 flow,and TIMI 2 or 3 flow,but the 30-day mortality and ICH in both groups was so high that the use of tenecteplase is not permitted in China. This paper concerns about a novel mechatronic inerter, which is consisted of a hydraulic piston inerter and linear motor, called hydraulic electric inerter(HEI). Firstly, the structural components and the working principles of two types HEI device are introduced, and the dynamic model of the HEI is established. Then, three classifications of mechatronic inerter, namely, the single motor type, the linear inerter-motor type and the rotary inerter-motor type are presented,and in the meanwhile, some comparisons among the three types mechatronic inerter are analyzed. Subsequently, a methodology of designing and experimental tests of the HEI device is proposed by considering the rated working conditions of the linear motor and the electric elements. At last, the HEI device is conducted, and the force tests of the non-loaded HEI and loaded HEI are tested in order to validate their properties. | 17 | Effectiveness of herb-partitioned moxibustion combined with electroacupuncture on polycystic ovary syndrome in patients with symptom pattern of kidney deficiency and phlegm-dampness显示文摘OBJECTIVE:To observe the effect of herb-partitioned moxibustion combined with electroacupuncture on polycystic ovary syndrome(PCOS)in patients with symptom pattern of kidney deficiency and phlegm-dampness.METHODS:Totally 62 PCOS patients who met the inclusion criteria were randomly divided into intervention group and control group.In the intervention group,acupoints were subjected to(43±1)℃herb-partitioned moxibustion and electroacupuncture treatment.In the control group,acupoints were subjected to(37±1)℃herb-partitioned moxibustion and sham electroacupuncture treatment.Treatment was provided 3 times per week for 12 weeks,and the treatment was stopped during menstruation.After the treatment,the clinical efficacy,the Traditional Chinese Medicine(TCM)symptom pattern score,basal body temperature(BBT)levels,menstrual cycle,and related sex hormone levels of the two groups were evaluated.RESULTS:Sixty patients were eventually included in the study(30 in the intervention group and 30 in the control group).There were not obvious between-group differences in the baseline efficacy parameters(all P>0.05).(a)After treatment,the two groups have improved in terms of clinical efficacy,TCM symptom pattern score,menstrual cycle and,BBT(P<0.01,<0.05),and the intervention group was better than the control group(P<0.01,<0.05).(b)In the intervention group,the estradiol(E2)and P450 aromatase(P450 arom)levels were increased(all P<0.05),the testosterone(T)level was decreased(P<0.05),and the luteotropic hormone(LH)and anti-Müllerian hormone(AMH)levels were significantly decreased(P<0.01).In the control group,E2 level was increased(P<0.05),and LH level was decreased(P<0.05).After treatment,the T,LH,and AMH levels of the intervention group were decreased compared with those of the control group(all P<0.05),and the P450 arom level was increased(P<0.05).CONCLUSIONS:Herb-partitioned moxibustion combined with electroacupuncture can effectively improve the related clinical symptoms of PCOS patients with kidney deficiency and phlegm-dampness.Herb-partitioned moxibustion combined with electroacupuncture may inhibit the overexpression of AMH to increase the expression level of P450 arom in ovarian granulosa cells,thereby reconstructing the dependence of follicular development on FSH,and finally improving abnormal follicular development and hyperandrogenism in PCOS patients with kidney deficiency and phlegmdampness. | ZHAO Qingyi SUN Yi ZHOU Jing GAO Yanling MA Guizhi HU Zhihai WANG Yi SHI Yin | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 1 |

| 20 | Impacts of meteorology and emission variations on the heavy air pollution episode in North China around the 2020 Spring Festival显示文摘Based on the Weather Research and Forecasting model and the Models-3 community multi-scale air quality model(WRF-CMAQ),this study analyzes the impacts of meteorological conditions and changes in air pollutant emissions on the heavy air pollution episode occurred over North China around the 2020 Spring Festival(January to Februray 2020).Regional reductions in air pollutant emissions required to eliminate the PM2.5 heavy pollution episode are also quantified.Our results found that meteorological conditions for the Beijing-Tianjin-Hebei and surrounding'2+26'cities are the worst during the heavy pollution episode around the 2020 Spring Festival as compared with two other typical heavy pollution episodes that occurred after 2015.However,because of the substantial reductions in air pollutant emissions in the'2+26'cities in recent years,and the32%extra reduction in emissions during January to February 2020 compared with the baseline emission levels of the autumn and winter of 2019 to 2020,the maximum PM2.5 level during this heavy pollution episode around the 2020 Spring Festival was much lower than that in the other two typical episodes.Yet,these emission reductions are still not enough to eliminate regional heavy pollution episodes.Compared with the actual emission levels during January to February 2020,a 20%extra reduction in air pollutant emissions in the'2+26'cities(or a 45%extra reduction compared with baseline emission levels of the autumn and winter of 2019 to 2020)could help to generally eliminate regionwide severe pollution episodes,and avoid heavy pollution episodes that last three or more consecutive days in Beijing;a 40%extra reduction in emissions(or a 60%extra reduction compared with baseline emission levels of the autumn and winter of 2019 to 2020)could help to generally eliminate regionwide and continuous heavy pollution episodes.Our analysis finds that during the clean period after the heavy pollution episode around the 2020 Spring Festival,the regionwide heavy pollution episode would only occur with at least a 10-fold increase in air pollutant emissions. | Wenbo XUE Xurong SHI Gang YAN Jinnan WANG Yanling XU Qian TANG Yanli WANG Yixuan ZHENG Yu LEI | 2021 | Science China Earth Sciences2021,64,2: | 1 |
