Application of bioactive hydrogels combined with dental pulp stem cells for the repair of large gap peripheral nerve injuries显示文摘Due to the limitations in autogenous nerve grafting or Schwann cell transplantation, large gap peripheral nerve injuries require a bridging strategy supported by nerve conduit.Cell based therapies provide a novel treatment for peripheral nerve injuries. In this study,we first experimented an optimal scaffold material synthesis protocol, from where we selected the 10%GFD formula(10%GelMA hydrogel, recombinant human basic fibroblast growth factor and dental pulp stem cells(DPSCs))to fill a cellulose/soy protein isolate composite membrane(CSM)tube to construct a third generation of nerve regeneration conduit,CSM-GFD.Then this CSM-GFD conduit was applied to repair a 15-mm long defect of sciatic nerve in a rat model. After 12 week post implant surgery,at histologic level,we found CSM-GFD conduit could regenerate nerve tissue like neuron and Schwann like nerve cells and myelinated nerve fibers. At physical level,CSM-GFD achieved functional recovery assessed by a sciatic functional index study. In both levels,CSM-GFD performed like what gold standard, the nerve autograft, could do.Further,we unveiled that almost all newly formed nerve tissue at defect site was originated from the direct differentiation of exogeneous DPSCs in CSM-GFD.In conclusion,we claimed that this third-generation nerve regeneration conduit,CSM-GFD,could be a promising tissue engineering approach to replace the conventional nerve autograft to treat the large gap defect in peripheral nerve injuries. | Lihua Luo Yan He Ling Jin Yanni Zhang Fernando P.Guastaldi Abdullkhaleg A.Albashari Fengting Hu Xiaoyan Wang Lei Wang Jian Xiao Lingli Li Jianming Wang Akon Higuchi Qingsong Ye | 2021 | Bioactive Materials2021,6,3: | 11
A super pan-genomic landscape of rice显示文摘Pan-genomes from large natural populations can capture genetic diversity and reveal genomic complexity.Using de novo longread assembly,we generated a graph-based super pan-genome of rice consisting of a 251-accession panel comprising both cultivated and wild species of Asian and African rice. Our pan-genome reveals extensive structural variations(SVs)and gene presence/absence variations.Additionally, our pan-genome enables the accurate identification of nucleotide-binding leucine-rich repeat genes and characterization of their inter-and intraspecific diversity. Moreover,we uncovered grain weight-associated SVs which specify traits by affecting the expression of their nearby genes. We characterized genetic variants associated with submergence tolerance, seed shattering and plant architecture and found independent selection for a common set of genes that drove adaptation and domestication in Asian and African rice. This super pan-genome facilitates pinpointing of lineage-specific haplotypes for trait-associated genes and provides insights into the evolutionary events that have shaped the genomic architecture of various rice species. | Lianguang Shang Xiaoxia Li Huiying He Qiaoling Yuan Yanni Song Zhaoran Wei Hai Lin Min Hu Fengli Zhao Chao Zhang Yuhua Li Hongsheng Gao Tianyi Wang Xiangpei Liu Hong Zhang Ya Zhang Shuaimin Cao Xiaoman Yu Bintao Zhang Yong Zhang Yiqing Tan Mao Qin Cheng Ai Yingxue Yang Bin Zhang Zhiqiang Hu Hongru Wang Yang Lv Yuexing Wang Jie Ma Quan Wang Hongwei Lu Zhe Wu Shanlin Liu Zongyi Sun Hongliang Zhang Longbiao Guo Zichao Li Yongfeng Zhou Jiayang Li Zuofeng Zhu Guosheng Xiong Jue Ruan Qian Qian | 2022 | Cell Research2022,32,10: | 3
Reliability Analysis for Complex Systems Based on Dynamic Evidential Network Considering Epistemic Uncertainty显示文摘Fault tolerant technology has greatly improved the reliability of modern systems on one hand and makes their failure mechanisms more complex on the other. The characteristics of dynamics of failure, diversity of distribution and epistemic uncertainty always exist in these systems, which increase the challenges in the reliability assessment of these systems significantly. This paper presents a novel reliability analysis framework for complex systems within which the failure rates of components are expressed in interval numbers.Specifically,it uses a dynamic fault tree(DFT)to model the dynamic fault behaviors and copes with the epistemic uncertainty using Dempster-Shafer(D-S)theory and interval numbers.Furthermore,an approach is presented to convert a DFT into a dynamic evidential network(DEN)to calculate the reliability parameters.Additionally,a sorting method based on the possibility degree is proposed to rank the importance of components represented by interval numbers in order to obtain the most critical components, which can be used to provide the guidance for system design, maintenance planning and fault diagnosis. Finally,a numerical example is provided to illustrate the availability and efficiency of the proposed method. | Rongxing Duan Yanni Lin Longfei Hu | 2017 | Computer Modeling in Engineering & Sciences2017,,1: | 1
Estimating the time interval between transmission generations and the presymptomatic period by contact tracing surveillance data from 31 provinces in the mainland of China显示文摘The global pandemic of 2019 coronavirus disease(COVID-19)is a great assault to public health.Presymptomatic transmission cannot be controlled with measures designed for symptomatic persons, such as isolation. This study aimed to estimate the interval of the transmission generation(TG)and the presymptomatic period of COVID-19,and compare the ftting effects of TG and serial interval(S)based on the SEIHR model incorporating the surveillance data of 3453 cases in 31 provinces. These data were allocated into three distributions and the value of AIC presented that the Weibull distribution fitted well. The mean of TG was 5.2 days(95%C:4.6-5.8).The mean of the presymptomatic period was 2.4 days(95%CI:1.5-3.2).The dynamic model using TG as the generation time performed well.Eight provinces exhibited a basic reproduction number from 2.16 to 3.14.Measures should be taken to control presymptomatic transmission in the COVID-19 pandemic. | Zhongxing Ding Kai Wang Mingwang Shen Kai Wang Shi Zhao Wenyu Song Rui Li Zhongjie Li Liping Wang Ganzhu Feng Zhiliang Hu Hongxia Wei Yanni Xiao Changjun Bao Jianli Hu Liguo Zhu Yong Li Xufeng Chen Yi Yin Weiming Wang Yongli Cai Zhihang Peng Hongbing Shen | 2021 | Fundamental Research2021,1,2: | 1
Phase-mediated cobalt phosphide with unique core-shell architecture serving as efficient and bifunctional electrocatalyst for hydrogen evolution and oxygen reduction reaction显示文摘Hydrogen evolution reaction(HER)and oxygen reduction reaction(ORR)have been considered as two critical processes in the field of electrocatalytic water-splitting for hydrogen production and fuel cells. However, the sluggish reaction kinetics of HER and ORR required efficient electrocatalyst such as Pt to promote such process.Transition metal phosphides(TMPs)exhibit great potential to replace noble metal electrocatalysts to accelerate HER and ORR due to their high activity and easy availability.Herein,a highly-efficient bifunctional CoP electrocatalyst for HER and ORR,featuring a unique core-shell structure decorated on nitrogen-doped carbon matrix was designed and constructed via etching a cobalt-based zeolitic imidazolate framework(ZIF-67)with phytic acid(PA)followed by pyrolysis treatment(PA-ZIF-67-900).Experimental results revealed that the pure-phase single-crystalline Co P exhibited outstanding electrocatalytic performance in HER and ORR,superior to Co(PO_(3))_(2) in PA-ZIF-67-700,hybrid phase of Co(PO_(3))_(2) and CoP in PA-ZIF-67-800 and Co_(2)P-doped CoP in PA-ZIF-67-1000.To reach the current density of 10 mA/cm^(2)the as-synthesized Co P required an overpotential of 120 m V for HER in 1 mol/L KOH and half-wave potential of 0.85 V in O_(2)-saturated 0.1 mol/L KOH.This work present new clue for construction of efficient and bifunctional electrocatalyst in the field of energy conversion and storage. | Junsheng Chen Jianfeng Huang Hai Wang Weihang Feng Tianmi Luo Yuzhu Hu Chengke Yuan Liyun Cao Yanni Jie Koji Kajiyoshi Yongqiang Feng | 2022 | Chinese Chemical Letters2022,33,8: | 0
Hybridoma-derived neutralizing monoclonal antibodies against Beta and Delta variants of SARS-CoV-2 in vivo显示文摘Neutralizing monoclonal antibodies(mAb)are a major therapeutic strategy for the treatment of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection.The continuous emergence of new SARS-CoV-2 variants worldwide has increased the urgency for the development of new mAbs.In this study,we immunized mice with the receptor-binding domain(RBD)of the SARS-CoV-2 prototypic strain(WIV04)and screened 35 RBDspecific mAbs using hybridoma technology. Results of the plaque reduction neutralization test showed that 25 of the mAbs neutralized authentic WIV04 strain infection. The 25 mAbs were divided into three categories based on the competitive enzyme-linked immunosorbent assay results.A representative mAb was selected from each category(RD4,RD10,and RD14)to determine the binding kinetics and median inhibitory concentration(IC_(50))of WIV04 and two variants of concern(VOC):B.1.351(Beta)and B.1.617.2(Delta).RD4 neutralized the B.1.617.2 variant with an IC50 of 2.67 ng/mL;however,it completely lost neutralizing activity against the B.1.351 variant.RD10 neutralized both variants with an IC50 exceeding 100 ng/mL;whereas RD14 neutralized two variants with a higher IC50(>1 mg/mL).Animal experiments were performed to evaluate the protective effects of RD4 and RD10 against various VOC infections.RD4 could protect Adv-hACE2 transduced mice from B.1.617.2 infection at an antibody concentration of 25 mg/kg,while RD10 could protect mice from B.1.351 infection at an antibody concentration of 75 mg/kg.These results highlight the potential for future modifications of the mAbs for practical use. | Qianran Wang Lu Peng Yanqiu Nie Yanni Shu Huajun Zhang Zidan Song Yufeng Li Hengrui Hu Liushuai Li Xi Wang Jia Liu Jiang Li Zhengli Shi Fei Deng Yu Guo Yiwu Zhou Bing Yan Zhihong Hu Manli Wang | 2023 | Virologica Sinica2023,38,2: | 0
Exploring the attenuation mechanisms of Dalbergia odorifera leaves extract on cerebral ischemia-reperfusion based on weighted gene co-expression network analysis显示文摘The attenuation function of Dalbergia odorifera leaves on cerebral ischemia-reperfusion(I/R)is little known. The candidate targets for the Chinese herb were extracted from brain tissues through the high-affinity chromatography. The molecular mechanism of D.odorifera leaves on cerebral I/R was investigated. Methods:Serial affinity chromatography based on D.odorifera leaves extract(DLE)affinity matrices were applied to find specific binding proteins in the brain tissues implemented on C57BL/6 mice by intraluminal middle cerebral artery occlusion for 1 h and reperfusion for 24 h.Specific binding proteins were subjected to mass-spectrometry to search for the differentially expressed proteins between control and DLE-affinity matrices. The hub genes were screened based on weighted gene co-expression network analysis(WGCNA).Then, predictive biology and potential experimental verification were performed for the candidate genes. The protective role of DLE in blood-brain barrier damage in cerebral I/R mice was evaluated by the leakage of Evans blue, western blotting, immunohistochemistry, and immunofluorescent staining.Results:952 differentially expressed proteins were classified into seven modules based on WGCNA under soft threshold 6.Based on WGCNA,AKT1,PIK3CA,NOS3,SMAD3,SMAD1,IL6,MAPK1,TGFBR2,TGFBR1,MAPK3,IGF1R,LRG1,mTOR,ROCK1,TGFB1,IL1B,SMAD2,and SMAD518 candidate hub proteins were involved in turquoise module.TGF-β,MAPK,focal adhesion, and adherens junction signaling pathway were associated with candidate hub proteins.Gene ontology analysis demonstrated that candidate hub proteins were related to the TGF-βreceptor signaling pathway,common-partner SMAD protein phosphorylation, etc.DLE could significantly reduce the leakage of Evans blue in mice with cerebral I/R,while attenuating the expression of occludin,claudin-5,and zonula occludens-1.Western blotting demonstrated that regulation of TGF-β/SMAD signaling pathway played an essential role in the protective effect of DLE.Conclusion:Thus,a number of candidate hub proteins were identified based on DLE affinity chromatography through WGCNA.DLE could attenuate the dysfunction of bloodbrain barrier in the TGF-β/SMAD signaling pathway induced by cerebral I/R. | JINFANG HU JIANGEN AO LONGSHENG FU YAOQI WU FENG SHAO TIANTIAN XU MINGJIN JIANG SHAOFENG XIONG YANNI LV | 2023 | BIOCELL2023,47,7: | 0
Characteristics of photosynthesis and vertical canopy architecture of citrus trees under two labor-saving cultivation modes using unmanned aerial vehicle (UAV)-based LiDAR data in citrus orchards显示文摘Analyzing and comparing the effects of labor-saving cultivation modes on photosynthesis,as well as studying their vertical canopy architecture, can improve the tree structure of high-quality and high-yield citrus and selection of labor-saving cultivation modes. The photosynthesis of 1080 leaves of two labor-saving cultivation modes(wide-row and narrow-plant mode and fenced mode)comparing with the traditional mode were measured, and nitrogen content of all leaves and photosynthetic nitrogen use efficiency(PNUE)were determined.Unmanned aerial vehicle(UAV)-based light detection and ranging(LiDAR)data were used to assess the vertical architecture of three citrus cultivation modes. Results showed that for the wide-row and narrow-plant and traditional modes leaf photosynthetic CO_(2) assimilation rate, stomatal conductance, and transpiration rate of the upper layer were significantly higher than those of the middle layer, and values of the middle layer were markedly higher than those of the lower layer. In the fenced mode,a significant difference in photosynthetic factors between the upper and middle layers was not observed.A vertical canopy distribution had a more significant effect on PNUE in the traditional mode.Leaves in the fenced mode had distinct photosynthetic advantages and higher PNUE.UAV-based LiDAR data effectively revealed the differences in the vertical canopy architecture of citrus trees by enabling calculating the density and height percentile of the LiDAR point cloud. The point cloud densities of three cultivation modes were significantly different for all LiDAR density slices, especially at higher canopy heights. The labor-saving modes, particularly the fenced mode, had significantly higher height percentile data. | Yuanyong Dian Xiaoyang Liu Lei Hu Jinzhi Zhang Chungen Hu Yongzhong Liu Jinxin Zhang Wenbo Zhang Qingqing Hu Yahao Zhang Yanni Fang Jingjing Zhou | 2023 | Horticulture Research2023,10,3: | 0

Biochanin A,as the Lrg1/TGF-β/Smad2 pathway blockade, attenuates blood-brain barrier damage after cerebral ischemiareperfusion by modulating leukocyte migration patterns显示文摘Background:Biochanin A is an excellent dietary isoflavone that has the concomitant function of both medicine and foodstuff. The attenuation function of biochanin A on blood-brain barrier(BBB)damage induced by cerebral ischemia-reperfusion remains unclear.Methods:C57BL/6 mice were subjected to 1 h middle cerebral artery occlusion(MCAO)followed by 24 h reperfusion. The infarct volume of the brain was stained by TTC,while leakage of the brain was quantitatively stained by Evans blue, and the neurologic deficit score was measured.Microglial-induced morphologic changes were observed via immunofluorescence staining, and rolling and adhering leukocytes in venules were observed via two-photon imaging, while the inner fluorescein isothiocyanate-albumin of venules were compared with those of surrounding interstitial area through venular albumin leakage. Results:The attenuation effect of biochanin A on tight junction injury was compared in ischemia-reperfusion mice or conventional knockdown of leucine-richα2-glycoprotein 1(Lrg1)mice.Biochanin A could ameliorate BBB injury in mice with cerebral ischemiareperfusion in a dose-dependent manner by strengthening the immunostaining volume of occludin,claudin-5,and zonula occludens-1.The amoeba morphologic changes of microglial combined with the elevated expression of Lrg1 could be relieved under the treatment of biochanin A.Biochanin A played a countervailing role on the rolling leukocytes in the vessel, while the leakage of blood vessels was reduced.Biochanin A diminished its functions to further improved attenuation for tight junction injury on conventional Lrg1-knockout mice,as well as the inhibition effects on TGF-β1,and the phosphorylation of suppressor of mothers against decapentaplegic 2(Smad2)/Smad2 via western blot assay. Conclusion:Biochanin A could alleviate tight junction injury induced by cerebral ischemiareperfusion and blocked the Lrg1/TGF-β/Smad2 pathway to modulate leukocyte migration patterns. | LONGSHENG FU JINFANG HU FENG SHAO YAOQI WU WEI BAI MINGJIN JIANG HAO CHEN LIHUA CHEN YANNI LV | 2023 | BIOCELL2023,47,8: | 0
Ordering Results on Largest Order Statistics from Multiple-Outlier Gamma Variables显示文摘In this article,we carry out stochastic comparisons on the maximum order statistics arising from two batches of multiple-outlier gamma random variables with different shape and scale parameters. It is proved that, under certain conditions, the majorization order between the vectors of shape parameters together with the weak majorization order[p-larger order]between the vectors of scale parameters implies the likelihood ratio order[hazard rate order]between the largest order statistics. The results established here strengthen and generalize some known ones in the literature. | Yiying Zhang Yanni Hu Peng Zhao | 2023 | Communications in Mathematics and Statistics2023,11,2: | 0
Gadolinium-containing semiconducting polymer nanoparticles for magnetic resonance/fluorescence dual-modal imaging and photothermal therapy of oral squamous cell carcinoma显示文摘Oral squamous cell carcinoma(OSCC)is the most common malignant tumor of the oral and maxillofacial region.Due to its unique location, earlier and more accurate diagnosis and more minimally invasive treatment of OSCC is of major importance.Herein,gadolinium-containing semiconductor polymer nanoparticles(SPN-Gd)were designed and prepared. The nanoparticles consist of a near-infrared(NIR)absorption semiconductor polymer(PCPDTBT)served as fluorescence signal source and a photothermal conversion agent(PTA)and a gadolinium-grafted triblock amphiphilic copolymer(F127-DTPA-Gd)served as a magnetic resonance imaging(MRI)contrast agent and nanocarrier. The experiments in vivo showed that SPN-Gd could act as an MRI contrast agent and optical image agent with a long retention time, and it had a significant inhibiting effect on tumors of OSCC mice model through photothermal therapy(PTT).Thus our study provides a simple nanotheranostic platform composed of two components for efficient MR/fluorescence dual-modal imaging-guided PTT. | Xiao Pan Antian Gao Yanni Hu Ziyang Hu Chen Xie Zitong Lin | 2023 | Nano Research2023,16,2: | 0
Regeneration and reuse of anode graphite from spent lithium-ion batteries with low greenhouse gas (GHG) emissions显示文摘Regenerating spent graphite(SG)from retired lithium-ion batteries(LIBs)can effectively avoid resource waste. However, the technology is challenged by the impurity content and energy consumption. In this study,micro-expanded graphite(MEG)was synthesized by one-step oxidation method using waste LIBs anode graphite as material and perchloric acid as intercalation and oxidant agent. Then, its performance as a LIBs anode material were investigated as well as the greenhouse gas(GHG)emissions of the whole process were calculated.Perchloric acid was successfully embedded in the SG during the reaction, which effectively removed the impurities in the graphite.Defects introduced during intercalation and delamination, such as nanopores and intercrystalline cracks. Both provide additional space for Li ions during charging and discharging, thereby promoting capacity enhancement. The prepared MEG expresses a rate capability as high as 340.32 m Ah/g at a current density of 0.1 C and still retains 81.73%of the capacity after 100 cycles at a current density of 1 C.Additionally, the GHG emissions of the synthesis process of this article and other literatures are compared. The results demonstrated that perchloric acid treatment process provides a low-carbon,time-and energy-saving approach for regenerated SG as battery grade material. | Junjie Liu Hui Shi Ke Yu Yanni Geng Xingyu Hu Genping Yi Jianzhi Zhang Xubiao Luo | 2023 | Chinese Chemical Letters2023,34,10: | 0
Characterization and functional analysis of pollen-specific PwSWEET1 in Picea wilsonii显示文摘SWEET transporters play a pivotal role in sugar transport in plants. However, their functions in pollen tube growth, especially in coniferous species remain unknown. Here,we used RT-qPCR to reveal that a SWEET1 gene was specifically expressed in pollen and pollen tubes of Picea wilsonii.A pollen germination assay showed that PwSWEET1 was induced by H3BO3 but not by Ca^2+.In a sugar specificity experiment, sucrose(Suc)and glucose(Glc)were effective sugars for pollen germination and pollen tube growth.PwSWEET1 expression was induced most by Suc and Glc.Heterologous expression of PwSWEET1 in yeast showed that PwSWEET1 can restore the glucose absorption in yeast strain EBY.VW4000,which has a hexose absorption defect, and the absorption of glucose is pH-independent.This evidence supports the involvment of PwSWEET1 in boron-dependent glucose transport in pollen germination and pollen tube growth of Picea wilsonii. | Yanni Zhou Xiaoyue Cui Anni Hu Yahui Miao Lingyun Zhang | 2020 | Journal of Forestry Research2020,31,5: | 0
