1E17110 promotes reverse cholesterol transport with liver X receptor β agonist activity in vitro显示文摘Liver X receptor(LXR) plays an important role in reverse cholesterol transport(RCT), and activation of LXR could reduce atherosclerosis. In the present study we used a cell-based screening method to identify new potential LXRβ agonists. A novel benzofuran-2-carboxylate derivative was identified with LXRβ agonist activity: E17110 showed a significant activation effect on LXRβ with an EC50 value of 0.72 μmol/L. E17110 also increased the expression of ATP-binding cassette transporter A1(ABCA1) and G1(ABCG1) in RAW264.7 macrophages. Moreover, E17110 significantly reduced cellular lipid accumulation and promoted cholesterol efflux in RAW264.7 macrophages. Interestingly, we found that the key amino acids in the LXRβ ligand-binding domain had distinct interactions with E17110 as compared to TO901317. 8Remote control of the recruitment and capture of endogenous stem cells by ultrasound for in situ repair of bone defects显示文摘Stem cell-based tissue engineering has provided a promising platform for repairing of bone defects.However,the use of exogenous bone marrow mesenchymal stem cells(BMSCs)still faces many challenges such as limited sources and potential risks.It is important to develop new approach to effectively recruit endogenous BMSCs and capture them for in situ bone regeneration.Here,we designed an acoustically responsive scaffold(ARS)and embedded it into SDF-1/BMP-2 loaded hydrogel to obtain biomimetic hydrogel scaffold complexes(BSC).The SDF-1/BMP-2 cytokines can be released on demand from the BSC implanted into the defected bone via pulsed ultrasound(p-US)irradiation at optimized acoustic parameters,recruiting the endogenous BMSCs to the bone defected or BSC site.Accompanied by the daily p-US irradiation for 14 days,the alginate hydrogel was degraded,resulting in the exposure of ARS to these recruited host stem cells.Then another set of sinusoidal continuous wave ultrasound(s-US)irradiation was applied to excite the ARS intrinsic resonance,forming highly localized acoustic field around its surface and generating enhanced acoustic trapping force,by which these recruited endogenous stem cells would be captured on the scaffold,greatly promoting them to adhesively grow for in situ bone tissue regeneration.Our study provides a novel and effective strategy for in situ bone defect repairing through acoustically manipulating endogenous BMSCs.Yanni He Fei Li Peng Jiang Feiyan Cai Qin Lin Meijun Zhou Hongmei Liu Fei Yan 2023Bioactive Materials2023,,3:1
9Estimating the time interval between transmission generations and the presymptomatic period by contact tracing surveillance data from 31 provinces in the mainland of China显示文摘The global pandemic of 2019 coronavirus disease(COVID-19)is a great assault to public health.Presymptomatic transmission cannot be controlled with measures designed for symptomatic persons,such as isolation.This study aimed to estimate the interval of the transmission generation(TG)and the presymptomatic period of COVID-19,and compare the ftting effects of TG and serial interval(S)based on the SEIHR model incorporating the surveillance data of 3453 cases in 31 provinces.These data were allocated into three distributions and the value of AIC presented that the Weibull distribution fitted well.The mean of TG was 5.2 days(95%C:4.6-5.8).The mean of the presymptomatic period was 2.4 days(95%CI:1.5-3.2).The dynamic model using TG as the generation time performed well.Eight provinces exhibited a basic reproduction number from 2.16 to 3.14.Measures should be taken to control presymptomatic transmission in the COVID-19 pandemic.Zhongxing Ding Kai Wang Mingwang Shen Kai Wang Shi Zhao Wenyu Song Rui Li Zhongjie Li Liping Wang Ganzhu Feng Zhiliang Hu Hongxia Wei Yanni Xiao Changjun Bao Jianli Hu Liguo Zhu Yong Li Xufeng Chen Yi Yin Weiming Wang Yongli Cai Zhihang Peng Hongbing Shen 2021Fundamental Research2021,1,2:1
10Hybridoma-derived neutralizing monoclonal antibodies against Beta and Delta variants of SARS-CoV-2 in vivo显示文摘Neutralizing monoclonal antibodies(mAb)are a major therapeutic strategy for the treatment of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection.The continuous emergence of new SARS-CoV-2 variants worldwide has increased the urgency for the development of new mAbs.In this study,we immunized mice with the receptor-binding domain(RBD)of the SARS-CoV-2 prototypic strain(WIV04)and screened 35 RBDspecific mAbs using hybridoma technology.Results of the plaque reduction neutralization test showed that 25 of the mAbs neutralized authentic WIV04 strain infection.The 25 mAbs were divided into three categories based on the competitive enzyme-linked immunosorbent assay results.A representative mAb was selected from each category(RD4,RD10,and RD14)to determine the binding kinetics and median inhibitory concentration(IC_(50))of WIV04 and two variants of concern(VOC):B.1.351(Beta)and B.1.617.2(Delta).RD4 neutralized the B.1.617.2 variant with an IC50 of 2.67 ng/mL;however,it completely lost neutralizing activity against the B.1.351 variant.RD10 neutralized both variants with an IC50 exceeding 100 ng/mL;whereas RD14 neutralized two variants with a higher IC50(>1 mg/mL).Animal experiments were performed to evaluate the protective effects of RD4 and RD10 against various VOC infections.RD4 could protect Adv-hACE2 transduced mice from B.1.617.2 infection at an antibody concentration of 25 mg/kg,while RD10 could protect mice from B.1.351 infection at an antibody concentration of 75 mg/kg.These results highlight the potential for future modifications of the mAbs for practical use.Qianran Wang Lu Peng Yanqiu Nie Yanni Shu Huajun Zhang Zidan Song Yufeng Li Hengrui Hu Liushuai Li Xi Wang Jia Liu Jiang Li Zhengli Shi Fei Deng Yu Guo Yiwu Zhou Bing Yan Zhihong Hu Manli Wang 2023Virologica Sinica2023,38,2:0

12Alterations in DNA damage response and repair genes as potential biomarkers for immune checkpoint blockade in gastrointestinal cancer显示文摘Objective:Immune checkpoint inhibitors(ICIs)have achieved remarkable results in cancer treatments.However,there is no effective predictive biomarker for gastrointestinal(GI)cancer.Methods:We conducted integrative analyses of the genomic and survival data of ICI-treated GI cancer patients from the Memorial Sloan Kettering Cancer Center cohort(MSK-GI,n=227),the Janjigian cohort(n=40),and the Peking University Cancer Hospital&Institute cohort(PUCH,n=80)to determine the possible associations between DNA damage response and repair(DDR)gene mutations and clinical outcomes.Data from The Cancer Genome Atlas database were analyzed to determine the possible correlations between DDR gene mutations and the tumor microenvironment.Results:In the MSK cohort,the presence of≥2 DDR gene mutations was correlated with prolonged overall survival(OS).The Janjigian and PUCH cohorts further confirmed that subgroups with≥2 DDR gene mutations displayed a prolonged OS and a higher durable clinical benefit.Furthermore,the DDR gene mutation load could be considered as an independent prognostic factor,and exhibited a potential predictive value for survival in GI cancer patients treated with ICIs.Mechanistically,we showed that the presence of≥2 DDR gene mutations was correlated with higher levels of tumor mutation burden,neoantigen,and T cell infiltration.Conclusions:The DDR gene mutation status was correlated with favorable clinical outcomes in GI cancer patients receiving ICIs,which could serve as a potential biomarker to guide patient selection for immunotherapy.Yujiao Wang Xi Jiao Shuang Li Huan Chen Xin Wei Chang Liu Jifang Gong Xiaotian Zhang Xicheng Wang Zhi Peng Changsong Qi Zhenghang Wang Yanni Wang Na Zhuo Jianling Zou Henghui Zhang Jian Li Lin Shen Zhihao Lu 2022Cancer Biology & Medicine2022,19,8:0

14Visual abstraction of dynamic network via improved multi-class blue noise sampling显示文摘Massive sequence view (MSV) is a classic timeline-based dynamic network visualization approach. However, it is vulnerable to visual clutter caused by overlapping edges, thereby leading to unexpected misunderstanding of time-varying trends of network communications. This study presents a new edge sampling algorithm called edge-based multi-class blue noise (E-MCBN) to reduce visual clutter in MSV. Our main idea is inspired by the multi-class blue noise (MCBN) sampling algorithm, commonly used in multi-class scatterplot decluttering. First, we take a node pair as an edge class, which can be regarded as an analogy to classes in multi-class scatterplots. Second, we propose two indicators, namely, class overlap and inter-class conflict degrees, to measure the overlapping degree and mutual exclusion, respectively, between edge classes. These indicators help construct the foundation of migrating the MCBN sampling from multi-class scatterplots to dynamic network samplings. Finally, we propose three strategies to accelerate MCBN sampling and a partitioning strategy to preserve local high-density edges in the MSV. The result shows that our approach can effectively reduce visual clutters and improve the readability of MSV. Moreover, our approach can also overcome the disadvantages of the MCBN sampling (i.e., long-running and failure to preserve local high-density communication areas in MSV).
