| 1 | Panax notoginseng saponins ameliorates experimental hepatic fibrosis and hepatic stellate cell proliferation by inhibiting the Jak2/Stat3 pathways显示文摘OBJECTIVE: To investigate the inhibitory effect of Panax notoginseng saponins(PNS) on liver fibrosis and explore the underlying mechanisms.METHODS: Carbon tetrachloride(CCl4)-treated rats and hepatic stellate cells(HSCs) were used. The effect of PNS on CCl4-induced liver fibrosis was studied with histochemical and biochemical analysis.Transforming growth factor(TGF)-β1, α-smooth muscle actin(α-SMA), and collagen Ι m RNA expression were determined by reverse transcriptase-polymerase chain reaction(RT-PCR). Meanwhile, the protein expression levels of α-SMA, collagen Ι, phosphorylation-Janus activated kinase signal transducer(p-Jak2) / Jak2, and phosphorylation-activator of transcription(p-Stat)3 / Stat3 were determined by immunohistochemistry and / or immunoblotting.RESULTS: PNS treatment significantly improved the liver function of rats as indicated by decreased serum enzymatic activities of alanine aminotransferase and aspartate aminotransferase. Histopathological results indicated that PNS alleviated liver damage and reduced the formation of fibrous septa. | 2 | Fabrication of Periodic Nanostructures Using AFM Tip-Based Nanomachining:Combining Groove and Material Pile-Up Topographies显示文摘This paper presents an atomic force microscopy(AFM)tip-based nanomachining method to fabricate periodic nanostructures.This method relies on combining the topography generated by machined grooves with the topography resulting from accumulated pile-up material on the side of these grooves.It is shown that controlling the distance between adjacent and parallel grooves is the key factor in ensuring the quality of the resulting nanostructures.The presented experimental data show that periodic patterns with good quality can be achieved when the feed value between adjacent scratching paths is equal to the width between the two peaks of material pile-up on the sides of a single groove.The quality of the periodicity of the obtained nanostructures is evaluated by applying one-and two-dimensional fast Fourier transform(FFT)algorithms.The ratio of the area of the peak part to the total area in the normalized amplitude–frequency characteristic diagram of the cross-section of the measured AFM image is employed to quantitatively analyze the periodic nanostructures.Finally,the optical effect induced by the use of machined periodic nanostructures for surface colorization is investigated for potential applications in the fields of anti-counterfeiting and metal sensing. | Yanquan Geng Yongda Yan Jiqiang Wang Emmanuel Brousseau Yanwen Sun Yazhou Sun | 2018 | Engineering2018,4,6: | 3 |

| 4 | Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy. | Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2 |

| 9 | Insight into Atomic-Scale Adhesion at the C-Cu Interface During theInitial Stage of Nanoindentation显示文摘Adhesion is a common phenomenon in nanomachining which affects processing accuracy and repeatability.As material removal approaches the atomic or close-to-atomic scale,quantum mechanics becomes the dominant principle behind the atomic-level interaction.However,atomic-scale effects cannot be properly described by empirical potential function-based molecular dynamics simulations.This study uses a first-principles method to reveal the atomic-scale adhesion between a diamond tip and a copper slab during initial-stage nanoindentation.Using a simplified tip and slab model,adhesion energy,electronic distribution,and density of states are analyzed based on quantum chemistry calculation.Results show that atomic adhesion is primarily due to the covalent bonding interaction between C and Cu atoms,which can induce structural changes to the diamond tip and copper slab.The effects of tip position and angles on adhesion are further studied through a series of simulations.The results show that adhesion between the tip and slab is sensitive to the lattice structure and a variant in angstroms is enough to cause different adhesion and structural changes.The actual determinants of adhesion can only be the atomic and electronic structures at the tip-slab interface.Bond rotation and breakage are observed during simulation and their effects on adhesion are further discussed.To conclude,the first-principles method is important for the analysis of an atomic-scale interaction system,even if only as an aid to describing adhesion at atomic and electronic scales. | Jian Gao Xichun Luo Wenlong Chang Zhengjian Wang Yongda Yan Yanquan Geng | 2022 | Nanomanufacturing and Metrology2022,5,3: | 0 |
