| 1 | Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12 |
| 2 | Shuganjieyu capsule increases neurotrophic factor expression in a rat model of depression显示文摘Shuganjieyu capsule has been approved for clinical treatment by the State Food and Drug Administration of China since 2008. In the clinic, Shuganjieyu capsule is often used to treat mild to moderate depression. In the rat model of depression established in this study, Shuganjieyu capsule was administered intragastrically daily before stress. Behavioral results confirmed that depressive symptoms lessened after treatment with high-dose(150 mg/kg) Shuganjieyu capsule. Immunohistochemistry results showed that high-dose Shuganjieyu capsule significantly increased phosphorylation levels of phosphorylation cyclic adenosine monophosphate response element binding protein and brain-derived neurotrophic factor expression in the medial prefrontal cortex and hippocampal CA3 area. | 3 | Protist Interactions and Seasonal Dynamics in the Coast of Yantai, Northern Yellow Sea of China as Revealed by Metabarcoding显示文摘Facilitated by the high-throughput sequencing(HTS)technique,the importance of protists to aquatic systems has been widely acknowledged in the last decade.However,information of protistan biotic interactions and seasonal dynamics is much less known in the coast ecosystem with intensive anthropic disturbance.In this study,year-round changes of protist community composition and diversity in the coastal water of Yantai,a city along the northern Yellow Sea in China,were investigated using HTS for the V4 region of 18S rDNA.The interactions among protist groups were also analyzed using the co-occurrence network.Data analyses showed that Alveolata,Chlorophyta,and Stramenopiles are the most dominant phytoplanktonic protists in the investigated coastal area.The community composition displayed strong seasonal variation.The abundant families Dino-Group-I-Clade-1 and Ulotrichales_X had higher proportions in spring and summer,while Bathycoccaceae exhibited higher ratios in autumn and winter.Alpha diversities(Shannon and Simpson)were the highest in autumn and the lowest in spring(ANOVA test,P<0.05).Nutrients(SiO42−,PO43−),total organic carbon(TOC),and pH seemed to drive the variation of alpha diversity,while temperature,PO43−and TON were the most significant factors influencing the whole protist community.Co-variance network analyses reveal frequent co-occurrence events among ciliates,chlorophytes and dinoflagellate,suggesting biotic interactions have been induced by predation,parasitism and mixotrophy. | FU Yingjun ZHENG Pengfei ZHANG Xiaoli ZHANG Qianqian JI Daode | 2020 | Journal of Ocean University of China2020,19,4: | 3 |
| 4 | Face and object visual working memory deficits in first-episode schizophrenia correlate with multiple neurocognitive performances显示文摘Background Working memory(WM)deficit is considered a core feature and cognitive biomarker in patients with schizophrenia.Several studies have reported prominent object WM deficits in patients with schizophrenia,suggesting that visual WM in these patients extends to non-spatial domains.However,whether non-spatial WM is similarly affected remains unclear.Aim This study primarily aimed to identify the processing of visual object WM in patients with first-episode schizophrenia.Methods The study included 36 patients with first-episode schizophrenia and 35 healthy controls.Visual object WM capacity,including face and house WM capacity,was assessed by means of delayed matching-to sample visual WM tasks,in which participants must distribute memory so that they can discriminate a target sample.We specifically examined their anhedonia experience by the Temporal Experience of Pleasure Scale and the Snaith-Hamilton Pleasure Scale.Cognitive performance was measured by the Repeatable Battery for the Assessment of Neuropsychological Status(RBANS).Results Both face and house WM capacity was significantly impaired in patients with schizophrenia.For both tasks,the performance of all the subjects was worse under the high-load condition than under the low-load condition.We found that WM capacity was highly positively correlated with the performance on RBANS total scores(r=−0.528,p=0.005),RBANS delayed memory scores(r=−0.470,p=0.013),RBANS attention scores(r=−0.584,p=0.001),RBANS language scores(r=−0.448,p=0.019),Trail-Making Test:Part A raw scores(r=0.465,p=0.015)and simple IQ total scores(r=−0.538,p=0.005),and correlated with scores of the vocabulary test(r=−0.490,p=0.011)and scores of the Block Diagram Test(r=−0.426,p=0.027)in schizophrenia.No significant correlations were observed between WM capacity and Positive and Negative Syndrome Scale symptoms.Conclusions Our research found that visual object WM capacity is dramatically impaired in patients with schizophrenia and is strongly correlated with other measures of cognition,suggesting a mechanism that is critical in explaining a portion of the broad cognitive deficits observed in schizophrenia. | Yi Liu Taiyong Bi Bei Zhang Qijie Kuang Haijing Li Kunlun Zong Jingping Zhao Yuping Ning Shenglin She Yingjun Zheng | 2021 | General Psychiatry2021,34,1: | 2 |
| 5 | Study of hot electrons generated from intense laser-plasma interaction employing Image Plate显示文摘Image Plate (IP) is convenient to be used and very suitable for radiation detection because of its advantages such as wide dynamic range, high detective quantum efficiency, ultrahigh sensitivity and superior linearity. The function mechanism and characteristics of IP are introduced in this paper. IP was employed in the study of hot electrons generated from intense laser-plasma interaction. The angular distri- bution and energy spectrum of hot electrons were measured with IP in the experi- ments.
