| 2 | A novel mutation in exon 11 of COMP gene in a Chinese family with pseudoachondroplasia显示文摘Pseudoachondroplasia(PSACH)is a relatively common skeletal dysplasia characterized by disproportionate short stature,joint laxity,early-onset osteoarthrosis,and dysplasia of the spine,epiphysis,and metaphysis.It is known as an autosomal dominant disease which results exclusively from mutations in the gene for Cartilage Oligomeric Matrix Protein(COMP).We have identified a five year old Chinese boy who was diagnosed as pseudoachondroplasia according to clinical manifestations and X-ray symptoms.His mother seems like another effected individual because of the apparent short stature.Genomic DNA was extracted from peripheral blood lymphocytes.DNA sequencing analysis of the COMP gene revealed a heterozygous mutation(c.1219 T>C,p.Cys407Arg)in the patient.His mother was also affected with the same genetic change.Mutations in COMP gene is proved to change the Cartilage Oligomeric Matrix Protein.This missense mutation(c.1219 T>C)has not been reported before and it is not belongs to polymorphism sites.Our results extend the spectrum of mutations in COMP gene leading to pseudoachondroplasia. | Jun Chen Wenbing Zhang Jinzhou He Run Zhang Yinqiang Cao Xing Liu | 2019 | Genes & Diseases2019,6,1: | 2 |
| 3 | Experimental and numerical study on the dynamic shear banding mechanism of HfNbZrTi high entropy alloy显示文摘High-entropy alloys(HEAs) have attracted considerable attention in recent years because of their unique mechanical properties.In this work, the mechanism of dynamic shear banding(also called adiabatic shear bands, ASBs) in a BCC HEA HfNbZrTi was investigated combining dynamic experiments and numerical simulations. The temperature evolution during dynamic shear banding, which has been believed to play a dominant role during ASB formation in the literature, was measured using high-speed infrared thermal detectors synchronized with a split Hopkinson pressure bar system. The dynamic mechanical behavior of the BCC HEA was described using the Johnson-Cook model accompanied by damage accumulation. The process of ASB formation,considering potential contributions from thermal softening and damage softening, was numerically investigated by controlling the activation of each softening mechanism separately. | 9 | Effect of operational and geometric parameters on the hydrodynamics of a Wurster coater:A CFD-DEM study显示文摘A coupled CFD-DEM method was used to study the hydrodynamics of a Wurster coater.Firstly,the CFD part of the model was validated by the accurate prediction of the pressure drops over a pseudo-2D fluidized bed under various gas velocities.The effect of gas velocity,gap height,tube length and batch volume of the particles on the cycle time and the residence time of the particles was thoroughly investigated.The central jet gas velocity ui was found to speed up the particle cycle but undermine the coating efficiency.The gas velocity at the horizontal transport zone u2 was able to promote the horizontal transport of the particles but should not be too high,otherwise,it would obstruct the normal falling back of the particles in the downward zone.Big gap heights would decrease the coating efficiency but tube length had little impact on that.The increment of batch volume would commonly abase the cycle time and the working efficiency under a given u1.The de-fluidization problem arose when the batch volume increased to 550 mL.However,this problem could be swept out by the optimization of u1 and u2.In a mixture of different sizes,the coarse particles enjoyed higher coating efficiency and could travel closer to the nozzles.This may shield the fine particles from getting enough coating liquids,and thus coarse particles and fine particles were not recommended to get coated in the same batch. | Yinqiang Song Tuo Zhou Ruiqi Bai Man Zhang Hairui Yang | 2024 | Particuology2024,85,2: | 0 |

| 11 | Targeting autophagy overcomes cancer-intrinsic resistance to CAR-T immunotherapy in B-cell malignancies显示文摘Background:Chimeric antigen receptor T(CAR-T)therapy has substantially revolutionized the clinical outcomes of patients with hematologic malignan-cies,but the cancer-intrinsic mechanisms underlying resistance to CAR-T cells remain yet to be fully understood.This study aims to explore the molecular deter-minants of cancer cell sensitivity to CAR-T cell-mediated killing and to provide a better understanding of the underlying mechanisms and potential modulation to improve clinical efficacy.Methods:The human whole-genome CRISPR/Cas9-based knockout screening was conducted to identify key genes that enable cancer cells to evade CD19 CAR-T-cell-mediated killing.The in vitro cytotoxicity assays and evaluation of tumor tissue and bone marrow specimens were further conducted to confirm the role of the key genes in cancer cell susceptibility to CAR-T cells.In addition,the specific mechanisms influencing CAR-T cell-mediated cancer clearance were elucidated in mouse and cellular models.Results:The CRISPR/Cas9-based knockout screening showed that the enrich-ment of autophagy-related genes(ATG3,BECN1,and RB1CC1)provided protec-tion of cancer cells from CD19 CAR-T cell-mediated cytotoxicity.These findings were further validated by in vitro cytotoxicity assays in cells with genetic and pharmacological inhibition of autophagy.Notably,higher expression of the three autophagy-related proteins in tumor samples was correlated with poorer respon-siveness and worse survival in patients with relapsed/refractory B-cell lymphoma after CD19 CAR-T therapy.Bulk RNA sequencing analysis of bone marrow samples from B-cell leukemia patients also suggested the clinical relevance of autophagy to the therapeutic response and relapse after CD19 CAR-T cell ther-apy.Pharmacological inhibition of autophagy and knockout of RB1CC1 could dramatically sensitize tumor cells to CD19 CAR-T cell-mediated killing in mouse models of both B-cell leukemia and lymphoma.Moreover,our study revealed that cancer-intrinsic autophagy mediates evasion of CAR-T cells via the TNF-α-TNFR1 axis-mediated apoptosis and STAT1/IRF1-induced chemokine signaling activation.Conclusions:These findings confirm that autophagy signaling in B-cell malig-nancies is essential for the effective cytotoxic function of CAR-T cells and thereby pave the way for the development of autophagy-targeting strategies to improve the clinical efficacy of CAR-T cell immunotherapy. | Lu Tang Huan Zhang Fen Zhou Qiuzhe Wei Mengyi Du Jianghua Wu Chenggong Li Wenjing Luo Jie Zhou Xindi Wang Zhaozhao Chen Yinqiang Zhang Zhongpei Huang Zhuolin Wu Yuxi Wen Huiwen Jiang Danying Liao Haiming Kou Wei Xiong Heng Mei Yu Hu | 2024 | Cancer Communications2024,44,3: | 0 |
