1A comparison of two kinds of eastward-moving mesoscale vortices during the mei-yu period of 2010显示文摘During the mei-yu period,the east edge of the Tibetan Plateau and the Dabie Mountain are two main sources of eastward-moving mesoscale vortices along the mei-yu front(MYF).In this study,an eastward-moving southwest vortex(SWV) and an eastward-moving Dabie vortex(DBV) during the mei-yu period of 2010 have been investigated to clarify the main similarities and differences between them.The synoptic analyses reveal that the SWV and DBV were both located at the lower troposphere;however,the SWV developed in a 'from top down' trend,whereas the DBV developed in an opposite way.There were obvious surface closed low centers corresponding to the DBV during its life span,whereas for the SWV,the closed low center only appeared at the mature stage.Cold and warm air intersected intensely after the formation of both the vortices,and the cold advection in the SWV case was stronger than that in the DBV case,whereas the warm advection in the DBV case was more intense than that in the SWV case.The Bay of Bengal and the South China Sea were main moisture sources for the SWV,whereas for the DBV,in addition to the above two moisture sources,the East China Sea was also an important moisture source.The vorticity budget indicates that the convergence was the most important common factor conducive to the formation,development,and maintenance of the SWV and DBV,whereas the conversion from the vertical vorticity to the horizontal one(tilting) was the most important common factor caused the dissipation of both of the vortices.The kinetic energy(KE) budget reveals that the KE generation by the rotational wind was the dominant factor for the enhancement of KE associated with the SWV,whereas for the DBV,the KE transport by the rotational wind was more important than the KE generation.The KE associated with the SWV and the DBV weakened with different mechanisms during the decaying stage.Furthermore,the characteristics of baroclinic and barotropic energy conversions during the life spans of both vortices indicate that the SWV and DBV both belong to the kind of subtropical mesoscale vortices.FU ShenMing YU Fei WANG DongHai XIA RuDi 2013Science China Earth Sciences2013,56,2:18
2Flavonoid content and radical scavenging activity in fruits of Chinese dwarf cherry(Cerasus humilis) genotypes显示文摘The Chinese dwarf cherry(Cerasus humilis(Bge.) Sok.) is a small shrub with edible fruits. It is native to northern and western China. This species was included as a medicinal plant in the ‘‘Chinese Pharmacopeia'' and has emerged as an economically important crop for fresh fruit consumption, processing into juice and wine and nutraceutical products as well. To gain a better understanding of flavonoid biosynthesis and help develop value added products and better cultivars with greater health benefits, we analyzed total flavonoid content(TFC), composition, and radical scavenging activities in fruit extracts of 16 Chinese dwarf cherry genotypes. Fruit peel TFC ranged from 33.5 to72.8 mg/g REáFW(RE: rutin equivalent, FW: fresh weight)while fruit flesh TFC ranged from 4.3 to 16.9 mg/g REáFW.An HPLC analysis revealed that fruit extracts contained 14 flavonoids with considerable variation in their profiles across genotypes. The most abundant flavonoids in most genotypes were proanthocyanidin B1(PA-B1), proanthocyanidin B2(PA-B2), phloretin 20-O-glucoside(PG), and phloretin 20,40-O-diglucoside(PDG). Principal component analysis showed that PG, PA-B1, and PA-B2 had large,positive factor loading values in the first principal component for each genotype. Increased scavenging activity of2,2-diphenyl-1-picrylhydrazyl(DPPH) radicals was apparent in genotypes ‘Nongda 4', ‘Nongda 3', ‘Nongda 6',‘Wenfenli', and '10-32', suggesting promising applications in the production of nutraceutical products. 4Hemodynamic analysis of intracranial aneurysms using phase-contrast magnetic resonance imaging and computational fluid dynamics显示文摘Additional hemodynamic parameters are highly desirable in the clinical management of intracranial aneurysm rupture as static medical images cannot demonstrate the blood flow within aneurysms. There are two ways of obtaining the hemodynamic information—by phase-contrast magnetic resonance imaging(PCMRI) and computational fluid dynamics(CFD). In this paper, we compared PCMRI and CFD in the analysis of a stable patient's specific aneurysm.The results showed that PCMRI and CFD are in good agreement with each other. 6Gastrin attenuates sepsis-induced myocardial dysfunction by down-regulation of TLR4 expression in macrophages显示文摘Myocardial dysfunction is the most serious complication of sepsis.Sepsis-induced myocardial dysfunction(SMD)is often associated with gastrointestinal dysfunction,but its pathophysiological significance remains unclear.The present study found that patients with SMD had higher plasma gastrin concentrations than those without SMD.In mice,knockdown of the gastrin receptor,cholecystokinin B receptor(Cckbr),aggravated lipopolysaccharide(LPS)-induced cardiac dysfunction and increased inflammation in the heart,whereas the intravenous administration of gastrin ameliorated SMD and cardiac injury.Macrophage infiltration plays a significant role in SMD because depletion of macrophages by the intravenous injection of clodronate liposomes,48 h prior to LPS administration,alleviated LPSinduced cardiac injury in Cckbr-deficient mice.The intravenous injection of bone marrow macrophages(BMMs)overexpressing Cckbr reduced LPS-induced myocardial dysfunction.Furthermore,gastrin treatment inhibited toll-like receptor 4(TLR4)expression through the peroxisome proliferator-activated receptor a(PPAR-a)signaling pathway in BMMs.Thus,our findings provide insights into the mechanism of the protective role of gastrin/CCKBR in SMD,which could be used to develop new treatment modalities for SMD.Dandong Fang Yu Li Bo He Daqian Gu Mingming Zhang Jingwen Guo Hongmei Ren Xinyue Li Ziyue Zhang Ming Tang Xingbing Li Donghai Yang Chunmei Xu Yijie Hu Hongyong Wang Pedro AJose Yu Han Chunyu Zeng 2023Acta Pharmaceutica Sinica B2023,13,9:1
7Effect of low dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via VEGF and mTOR显示文摘Objective To investigate the mechanism of low-dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via vascular endothelial growth factor(VEGF) and mammalian target of rapamycin(mTOR) in vivo.Methods Human cisplatin-resistant ovarian carcinoma cells(SKOV3/DDP) were injected into nude mice to establish ovarian cancer xenografts. The mice were randomly divided into three groups: a control group, a low-dose fractionated radiation(LDRFT) group, and a conventional-dose radiation group. Each group was exposed to 0 cGy, 50 cGy, and 200 cGy radiation, respectively, for 4 weeks, up to a total of 8.0 Gy. Mice in the LDFRT group were irradiated twice daily with 6 hour intermissions on day 1 and 2 of every week for a total of 4 weeks. Conventional-dose group mice were given a single 200 cGy radiation dose on the first day each week for a total of 4 weeks. Maximum horizontal and vertical diameters of the tumors were measured every other day and used to create a tumor growth curve. After 4 weeks of irradiation, we dissected the tumor tissue and calculated the tumor inhibition rate. RT-PCR detected the expression of VEGF and m TOR, and Western blots detected the expression of corresponding proteins.Results Both LDRFT and conventional-dose radiation inhibited the growth of tumor cells, and growth of tumors in the two radiation groups compared with growth in the control group were significantly different(P < 0.05). The rate of tumor inhibition in the LDFRT group(37.5603%) was lower than in the conventionaldose group(47.4446%), but there was no significant difference(P 0.05). Compared with the other two groups, the m RNA expression of VEGF was significantly lower in the LDFRT group(P < 0.05), but there was no obvious difference between the conventional-dose and control groups. There was no obvious difference in the m RNA expression of m TOR among the three groups, but the expression of the protein p-m TOR was lower in the LDFRT group(P < 0.05), as confirmed by Western blotting.Conclusion LDFRT is as effective at inhibiting the growth of tumor cells as conventional-dose radiation. 10Identification of potential immune-related prognostic biomarkers of lung cancer using gene co-expression network analysis显示文摘Objective The objective of this study was to identify new carcinogenetic hub genes and develop the integration of differentially expressed genes to predict the prognosis of lung cancer.Methods GSE139032 microarray data packages were downloaded from the Gene Expression Omnibus for planning,testing,and review of data.We identified KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV from a key module for validation.Results We found that the five genes were related to a poor prognosis,and the expression levels of these genes were associated with tumor stage.Furthermore,Kaplan-Meier plotter showed that the five hub genes had better prognostic values.The mean levels of methylation in lung adenocarcinoma(LUAD)were significantly lower than those in healthy lung tissues for the hub genes.However,gene set enrichment analysis(GSEA)for single hub genes showed that all of them were immune-related.Conclusion Our findings demonstrated that KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV are all candidate diagnostic and prognostic biomarkers for LUAD.They may have clinical implications in LUAD patients not only for the improvement of risk stratification but also for therapeutic decisions and prognosis prediction.Aixia Chen Shengnan Zhao Fei Zhou Hongying Lv Donghai Liang Tao Jiang Rui Liu Lijin Zhu Jingyu Cao Shihai Liu Hongsheng Yu 2020Oncology and Translational Medicine2020,6,6:1

19Effects of Soil Hydraulic Properties on Soil Moisture Estimation显示文摘Accurate quantification of soil moisture is essential to understand the land surface processes.Soil hydraulic properties influence water transport in soil and thus affect the estimation of soil moisture.However,some soil hydraulic properties are only observable at a few field sites.In this study,the effects of soil hydraulic properties on soil moisture estimation are investigated by using the one-dimensional(1-D)Richards equation at ELBARA,which is part of the Maqu monitoring network over the Tibetan Plateau(TP),China.Soil moisture assimilation experiments are then conducted with the unscented weighted ensemble Kalman filter(UWEnKF).The results show that the soil hydraulic properties significantly affect soil moisture simulation.Saturated soil hydraulic conductivity(Ksat)is optimized based on its observations in each soil layer with a genetic algorithm(GA,a widely used optimization method in hydrology),and the 1-D Richards equation performs well using the optimized values.If the range of Ksat for a complete soil profile is known for a particular soil texture(rather than for arbitrary layers within the horizon),optimized Ksat for each soil layer can be obtained by increasing the number of generations in GA,although this increases the computational cost of optimization.UWEnKF performs well with optimized Ksat,and improves the accuracy of soil moisture simulation more than that with calculated Ksat.Sometimes,better soil moisture estimation can be obtained by using optimized saturated volumetric soil moisture content Ksat.In summary,an accurate soil profile can be obtained by using soil moisture assimilation with optimized soil hydraulic properties.Xiaolei FU Haishen LYU Zhongbo YU Xiaolei JIANG Yongjian DING Donghai ZHENG Jinbai HUANG Hongyuan FANG 2023Journal of Meteorological Research2023,37,1:1
20Cefotetan-bound human RKIP involves in Ras/Raf1/MEK/ERK signaling pathway显示文摘Cefotetan is widely used to treat bacterial infections in the clinic owing to its broad spectrum of antibacterial activity.In the present study,we demonstrate that cefotetan can bind to the conserved ligand-binding pocket of human Raf1 kinase inhibitory protein(hRKIP),which acts as a negative regulator of the Ras/Raf1/MEK/ERK signaling pathway.The cefotetan-bound hRKIP adopts a rigid structure with insufficient space for binding Raf1 kinase,thereby reliving the inhibitory activity of hRKIP in the Ras/Raf1/MEK/ERK signaling pathway and enhancing the phosphorylation level of ERK.Both NMR titration and molecular docking approaches show that several residues(P74,Y81,W84,P111,P112,K113,S142,G143,D144,W173,P178,Y181 and L184)play crucial roles in hRKIP binding cefotetan.NMR dynamics analysis reveals that the binding of cefotetan with hRKIP promotes ps-ns internal motion but reducesμs-ms conformational exchange for residues in the cefotetan-binding pocket of hRKIP.Our results not only disclose the structural basis of cefotetan upregulating the Ras/Raf1/MEK/ERK signaling pathway but also benefit developing novel drugs against diseases caused by the impaired Ras/Raf1/MEK/ERK pathway.Chenyun Guo Hao Xu Yu Zhou Zhihua Wu Bin Jiang Hanyu Chen Donghai Lin 2022Acta Biochimica et Biophysica Sinica2022,54,12:0
