| 3 | Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12 |
| 4 | Development of Cryogenic Disinfectants Using in-18℃and-40℃Environments—Worldwide,2021显示文摘Introduction:Since the outbreak of coronavirus disease 2019(COVID-19)in Beijing Xinfadi Wholesale Market,a series of COVID-19 outbreaks has indicated the threat of transmission due to outer surfaces of objects under low temperatures.Therefore,2 kinds of chlorine-containing disinfectants for−18℃and−40℃were developed in order to solve the problem of low temperature disinfection.Methods:The properties of two cryogenic disinfectants were evaluated by low-temperature test.Staphylococcus aureus and Escherichia coli were used as two indicator microorganisms to evaluate the effects of the two cryogenic disinfectants in the laboratory and field tests according to Technical Specifications for Disinfection(2002 version)and Test method for bactericidal effect of disinfectants in laboratory(GB/T 38502-2020).Results:Two disinfectants could remain in the liquid state at−18℃or−40℃in 72 hours without crystallization and precipitation,and the average log values of microbial reduction before and after disinfection(the average killing log value)both on Staphylococcus aureus and Escherichia coli were>3 in the laboratory and field tests.Discussion:The results showed that these two disinfectants remained functional at low temperature to disinfect the packaging of imported cold chain food and surrounding packaging to prevent transmission of COVID-19 from material to people. | Huihui Sun Hongyang Duan Wei Zhang Chen Liang Luyao Li Yuan Lyu Yanyan Chen Jin Shen Liubo Zhang | 2021 | China CDC weekly2021,3,13: | 4 |

| 6 | Development and validation of a CT-based radiomics nomogram for preoperative prediction of tumor histologic grade in gastric adenocarcinoma显示文摘Objectives:To develop and validate a radiomics nomogram for preoperative prediction of tumor histologic grade in gastric adenocarcinoma(GA).Methods:This retrospective study enrolled 592 patients with clinicopathologically confirmed GA(low-grade:n=154;high-grade:n=438)from January 2008 to March 2018 who were divided into training(n=450)and validation(n=142)sets according to the time of computed tomography(CT)examination.Radiomic features were extracted from the portal venous phase CT images.The Mann-Whitney U test and the least absolute shrinkage and selection operator(LASSO)regression model were used for feature selection,data dimension reduction and radiomics signature construction.Multivariable logistic regression analysis was applied to develop the prediction model.The radiomics signature and independent clinicopathologic risk factors were incorporated and presented as a radiomics nomogram.The performance of the nomogram was assessed with respect to its calibration and discrimination.Results:A radiomics signature containing 12 selected features was significantly associated with the histologic grade of GA(P<0.001 for both training and validation sets).A nomogram including the radiomics signature and tumor location as predictors was developed.The model showed both good calibration and good discrimination,in which C-index in the training set,0.752[95%confidence interval(95%CI):0.701-0.803];C-index in the validation set,0.793(95%CI:0.711-0.874).Conclusions:This study developed a radiomics nomogram that incorporates tumor location and radiomics signatures,which can be useful in facilitating preoperative individualized prediction of histologic grade of GA. | Jia Huang Huasheng Yao Yexing Li Mengyi Dong Chu Han Lan He Xiaomei Huang Ting Xia Zongjian Yi Huihui Wang Yuan Zhang Jian He Changhong Liang Zaiyi Liu | 2021 | Chinese Journal of Cancer Research2021,33,1: | 3 |

| 8 | Genome Size of Alexandrium catenella and Gracilariopsis lemaneiformis Estimated by Flow Cytometry显示文摘Flow cytometry(FCM) technique has been widely applied to estimating the genome size of various higher plants. However, there is few report about its application in algae. In this study, an optimized procedure of FCM was exploited to estimate the genome size of two eukaryotic algae. For analyzing Alexandrium catenella, an important red tide species, the whole cell instead of isolated nucleus was studied, and chicken erythrocytes were used as an internal reference. The genome size of A. catenella was estimated to be 56.48 ± 4.14 Gb(1C), approximately nineteen times larger than that of human genome. For analyzing Gracilariopsis lemaneiformis, an important economical red alga, the purified nucleus was employed, and Arabidopsis thaliana and Chondrus crispus were used as internal references, respectively. The genome size of Gp. lemaneiformis was 97.35 ± 2.58 Mb(1C) and 112.73 ± 14.00 Mb(1C), respectively, depending on the different internal references. | 11 | Photooxidation of Methacrolein in Fe(III)-Oxalate Aqueous System and Its Atmospheric Implication显示文摘Iron and oxalic acids are widely distributed in the atmosphere and easily form ferric oxalate complex(Fe(III)-Ox).The tropospheric aqueous-phase could provide a medium to enable the photo-Fenton reaction with Fe(III)-Ox under solar irradiation.Although the photolysis mechanisms of Fe(III)-Ox have been investigated extensively,information about the oxidation of volatile organic compounds(VOC),specifically the potential for Secondary Organic Aerosol(SOA)formation in the Fe(III)-Ox system,is lacking.In this study,a ubiquitous VOC methacrolein(MACR)is chosen as a model VOC,and the oxidation of MACR with Fe(III)-Ox is investigated under typical atmospheric water conditions.The effects of oxalate concentration,Fe(III)concentration,MACR concentration,and pH on the oxidation of MACR are studied in detail.Results show that the oxidation rate of MACR greatly accelerates in the presence of oxalate when compared with only Fe(III).The oxidation rate of MACR also accelerates with increasing concentration of oxalate.The effect of Fe(III)is found to be more complicated.The oxidation rate of MACR first increases and then decreases with increasing Fe(III)concentration.The oxidation rate of MACR increases monotonically with decreasing pH in the common atmospheric water pH range or with decreasing MACR concentration.The production of ferrous and hydrogen peroxide,pH,and aqueous absorbance are monitored throughout the reaction process.The quenching experiments verify that·OH and O_(2)^(+)are both responsible for the oxidation of MACR.MACR is found to rapidly oxidize into small organic acids with higher boiling points and oligomers with higher molecular weight,which contributes to the yield of SOA.These results suggest that Fe(III)-Ox plays an important role in atmospheric oxidation. | Yu WANG Jie ZHAO Huihui LIU Yuan LI Wenbo DONG Yanlin WU | 2021 | Advances in Atmospheric Sciences2021,38,7: | 1 |

| 18 | Identification of microRNAs regulating grain filling of rice inferior spikelets in response to moderate soil drying post-anthesis显示文摘The grain filling of inferior spikelets is much less complete than that of superior spikelets in rice cultivars with large panicles and numerous spikelets and is promoted by moderate soil drying(MD)post-anthesis.A growing body of evidence has shown that microRNAs function in regulating grain development.However,little is known about the mechanism of microRNA control of grain filling of inferior spikelets in response to MD.In this study,grain filling of inferior spikelets was promoted by MD treatment in Nipponbare.Small-RNA profiling at the most active grain-filling stage was conducted in inferior spikelets under control(CK)and MD treatment.Of 521 known and 128 novel miRNAs,38 known and 9 novel miRNAs were differentially expressed between the CK and MD treatments.Target genes of differentially expressed miRNAs were involved in multiple developmental and signaling pathways associated with catalytic activity,carbohydrate metabolism,and other functions.Both miR1861 and miR397 were upregulated by MD,leading to a decrease in OsSBDCP1 and OsLAC,two negative regulators of SSIIIa activity and BR signaling,respectively.In contrast,miR1432 abundance was reduced by MD,resulting in upregulation of OsACOT and thus an elevated content of both ABA and IAA.These results suggest that both starch synthesis and phytohormone biosynthesis are regulated by differentially expressed miRNAs in inferior spikelets in response to MD treatment.Our results suggest the molecular mechanisms by which miRNAs regulate grain filling in inferior spikelets of rice under moderate soil drying,providing potential application in agriculture to increase rice yields by genetic approaches. | Zhenning Teng Yinke Chen Youqing Yuan Yaqiong Peng Yake Yi Huihui Yu Zhenxie Yi Jianchang Yang Yan Peng Meijuan Duan Jianhua Zhang Nenghui Ye | 2022 | The Crop Journal2022,10,4: | 1 |
