1Autoantibody to MOG suggests two distinct clinical subtypes of NMOSD显示文摘We characterized a unique group of patients with neuromyelitis optica spectrum disorder(NMOSD) who carried autoantibodies of aquaporin-4(AQP4) and myelin-oligodendrocyte glycoprotein(MOG). Among the 125 NMOSD patients, 10(8.0%) were AQP4- and MOG-ab double positive, and 14(11.2%) were MOG-ab single positive. The double-positive patients had a multiphase disease course with a high annual relapse rate(P=0.0431), and severe residual disability(P<0.0001). Of the double-positive patients, 70% had MS-like brain lesions, more severe edematous, multifocal regions on spinal magnetic resonance imaging(MRI), pronounced decreases of retinal nerve fiber layer thickness and atrophy of optic nerves. In contrast, patients with only MOG-ab had a higher ratio of monophasic disease course and mild residual disability. Spinal cord MRI illustrated multifocal cord lesions with mild edema, and brain MRIs showed more lesions around lateral ventricles. NMOSD patients carrying both autoantibodies to AQP4 and MOG existed and exhibited combined features of prototypic NMO and relapsing-remitting form of MS, whereas NMOSD with antibodies to MOG only exhibited an 'intermediate' phenotype between NMOSD and MS. 2Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 4Regeneration of commercial selective catalyst reduction catalysts deactivated by Pb and other inorganic elements显示文摘The regeneration of commercial SCR(Selective Catalyst Reduction) catalysts deactivated by Pb and other elements was studied.The deactivated catalyst samples were prepared by chemical impregnation with mixed solution containing K_2SO_4,Na_2SO_4,CaSO_4,Pb(NO_3)_2and NH_4H_2PO_4.A novel method combining Ethylenediaminetetraacetic acid disodium salt(EDTA-2Na) and H_2SO_4solution(viz.catalysts treated by dilute EDTA-2Na and H_2SO_4 solution in sequence) was used to recover the activity of deactivated samples,and the effect was compared with single H_2SO_4,oxalic acid,acetic acid,EDTA or HNO_3 solutions.The surface structure,acidity and reducibility of samples were characterized by N_2adsorption–desorption,inductively coupled plasma optical emission spectrometer(ICP-OES),scanning electron microscopy(SEM),X-ray diffraction(XRD),X-ray fluorescence(XRF),H-2-temperature programmed section(H_2-TPR),NH3-temperature programmed desorption(NH3-TPD) and in situ DRIFTS.Impurities caused a decrease of specific surface area and surface reducibility,as well as Br?nsted acid sites,and therefore led to severe deactivation of the SCR catalyst.The use of an acid solution alone possibly eliminated the impurities on the deactivated catalyst to some extent,and also increased the specific surface area and Br?nsted acid sites and promoted the surface reducibility,thus recovered the activity partially.The combination of EDTA-2Na and H_2SO_4 could remove most of the impurities and improve the activity significantly.The removal of Pb should be an important factor for regeneration.Due to a high removal rate for Pb and other impurities,the combination of EDTA-2Na and H_2SO_4 solutions provided the best efficiency.Yanke Yu Jinxiu Wang Jinsheng Chen Xinjiang He Yujing Wang Kai Song Zongli Xie 2016Journal of Environmental Sciences2016,28,9:11

7Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6
8Ginsenoside Rb1 Reduces D-GalN/LPS-induced Acute Liver Injury by Regulating TLR4/NF-κB Signaling and NLRP3 Inflammasome显示文摘Background and Aims:The effect of ginsenoside Rb1 on D-galactosamine(D-GalN)/lipopolysaccharide(LPS)-induced acute liver injury(ALI)is unknown.The aim of this study was to evaluate the effect of ginsenoside Rb1 on ALI and its underlying mechanisms.Methods:Mice were pretreated with ginsenoside Rb1 by intraperitoneal injection for 3 days before D-GalN/LPS treatment,to induce ALI.The survival rate was monitored every hour for 24 h,and serum biochemical parameters,hepatic index and histopathological analysis were evaluated to measure the degree of liver injury.ELISA was used to detect oxidative stress and inflammatory cytokines in hepatic tissue and serum.Immunohistochemistry staining,RT-PCR and western blotting were performed to evaluate the expression of toll-like receptor 4(TLR4),nuclear factorkappa B(NF-κB),and NLR family,pyrin domain-containing 3 protein(NLRP3)in liver tissue and Kupffer cells(KCs).Results:Ginsenoside Rb1 improved survival with D-GalN/LPS-induced ALI by up to 80%,significantly ameliorated the increased alanine and aspartate transaminase,restored the hepatic pathological changes and reduced the levels of oxidative stress and inflammatory cytokines altered by D-GalN/LPS.Compared to the control group,the KCs were increased in the D-GalN/LPS groups but did not increase significantly with Rb1 pretreatment.D-GalN/LPS could upregulate while Rb1 pretreatment could downregulate the expression of interleukin(IL)-1β,IL-18,NLRP3,apoptosis associated specklike protein containing CARD(ASC)and caspase-1 in isolated KCs.Furthermore,ginsenoside Rb1 inhibited activation of the TLR4/NF-κB signaling pathway and NLRP3 inflammasome induced by D-GalN/LPS administration.Conclusions:Ginsenoside Rb1 protects mice against D-GalN/LPS-induced ALI by attenuating oxidative stress and the inflammatory response through the TLR4/NF-κB signaling pathway and NLRP3 inflammasome activation.Yimei Liu Ninghua Liu Yujing Liu Hongyu He Zhe Luo Wenjun Liu Nan Song Minjie Ju 2022Journal of Clinical and Translational Hepatology2022,10,3:2

11Human cytomegalovirus miR-US5-1 inhibits viral replication by targeting Geminin mRNA显示文摘Viruses commonly create favorable cellular conditions for their survival through multiple mechanisms. Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. 17Cationic telluroviologen derivatives as type-I photosensitizers for tumor photodynamic theranostics显示文摘The hypoxia of the tumor microenvironment(TME)seriously restricts the photodynamic therapy(PDT)effect of conventional type-II photosensitizers,which are highly dependent on O_(2).In this work,a new type-I photosensitizer(TPE-TeVPPh3)consisting of a tetraphenylethylene group(TPE)as a bioimaging moiety,triphenyl-phosphine(PPh3)as a mitochondria-targeting group,and telluroviologen(TeV2+)as a reactive oxygen species(O_(2)•−,•OH)generating moiety is developed.The luminescence intensity of TPE-TeV-PPh3 increased significantly after specific oxidation by excess H2O2 in the TME without responding to normal tissues via the formation of Te═O bond,which can be used for monitoring abnormal H2O2,positioning,and imaging of tumors.TPE-TeV-PPh3 with highly reactive radicals generation and stronger hypoxia tolerance realizes efficient cancer cell killing under hypoxic conditions,achieving 88%tumor growth inhibition.Therefore,TPE-TeV-PPh3 with low phototoxicity in normal tissue achieves tumor imaging and effective PDT toward solid tumors in response to high concentrations of H_(2)O_(2)in the TME,which provides a new strategy for the development of type-I photosensitizers.Qi Sun Qi Su Yujing Gao Kun Zhou Weijie Song Penghe Quan Xiaojian Yang Zhishen Ge Yanmin Zhang Gang He 2023Aggregate2023,4,3:0

19Characteristics and sources of peroxyacetyl nitrate(PAN)in the rural North China Plain:Results from1-year continuous observations显示文摘Peroxyacetyl nitrate(PAN)is an important photochemical pollutant in the troposphere,whereas long-term measurements are scarce in rural areas in North China Plain(NCP),resulting in unclear seasonal variations and sources of PAN in rural NCP.In this study,we conducted a 1-year observation of PAN during 2021-2022 at the rural NCP site.The average concentrations of PAN were 1.10,0.75,0.65,and 0.88 ppbv in spring,summer,autumn,and winter,respectively,with a 1-year average of 0.81±0.60 ppbv.Calculations indicate that the loss of PAN through thermal decomposition in summer accounts for 43.2% of the total formed PAN,which is an important reason for the low concentration of PAN in summer.We speculate that since the correlation between PAN and O_(3) in winter is significantly lower than that in other seasons,the observed regional transport of PAN cannot be ignored in winter.Through budget analysis,regional transport accounted for 12.8% and 55.9% of the observed PAN on the spring and winter pollution days,respectively,which showed that regional transport played key roles during the photochemical pollution of the rural NCP in winter.The potential source contribution function revealed that the transported PAN mainly comes from southern Hebei in spring.In winter,the transported PAN was mainly from Langfang,Hengshui,and southern Beijing.Our findings may aid in understanding PAN variations in different seasons in rural areas and highlight the impact of regional transport on the PAN budget.Xin Liu Xiaowei He Chenglong Zhang Yifei Song Shuyang Xie Chengtang Liu Pengfei Liu Yuanyuan Zhang Yujing Mu Junfeng Liu 2024Journal of Environmental Sciences2024,,4:0
20HAND2-AS1,PRKAA2 and VLDLR predict the risk of peritoneal metastasis in gastric cancer of different Lauren types based on STEPP analysis显示文摘The peritoneum is the most common site of recurrence of gastric cancer(GC).Early occult peritoneal metastasis is difficult to detect by imaging examination.Stratifying the risk of peritoneal metastasis in patients with different Lauren subtypes is of great clinical value.We performed a univariate Cox regression to identify those genes with prognostic value of overall survival(OS)and peritoneal-specified disease-free survival(psDFS)from the Gene Expression Omnibus database.The candidate genes were screened by the Subpopulation Treatment Effect Pattern Plot(STEPP)method.Propensity score matching(PSM)analysis was used to reduce the interference of confounders on the results.Based on the optimal cut-off values determined by the STEPP method,we found overexpression of three genes(HAND2-AS1,PRKAA2,and VLDLR)was correlated with shorter 1-year psDFS among patients with diffuse-type than that of patients with intestinal-type GC,and it is highly significant.Gene Set Enrichment Analysis(GSEA)potentially suggested that the three genes promote the early occurrence of peritoneal metastasis in patients with diffuse-type GC through glucose metabolism-related pathways.These three genes may be potential biomarkers.They can be used to assess the risk of peritoneal metastases to guide treatment decisions and follow-up strategies.HUI QU RUICHUAN SHI BOWEN YANG XI CHEN YUJIA SONG LINGZI HE YITING SUN YUJING YANG ZHI LI XIUJUAN QU 2022BIOCELL2022,46,3:0
