| 4 | Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. | 5 | Genome-wide Investigation of micro RNAs and Their Targets in Brassica rapa ssp. pekinensis Root with Plasmodiophora brassicae Infection显示文摘Increasing evidence has revealed that micro RNAs play a pivotal role in the post transcriptional regulation of gene expression in response to pathogens in plants. However, there is little information available about the expression patterns of mi RNAs and their targets in Chinese cabbage(Brassica rapa ssp. pekinensis) under Plasmodiophora brassicae stress. In the present study, using deep sequencing and degradome analysis, a genome-wide identification of mi RNAs and their targets during P. brassicae stress was performed. A total of 221 known and 93 potentially novel mi RNAs were successfully identified from two root libraries of one control(635-10CK) and P. brassicae-treated Chinese cabbage samples(635-10T). Of these, 14 known and 10 potentially novel mi RNAs were found to be differentially expressed after P. brassicae treatment. Degradome analysis revealed that the 223 target genes of the 75 mi RNAs could be potentially cleaved. KEGG(Kyoto Encyclopedia of Genes and Genomes)pathway analysis suggested that the putative target genes of the mi RNAs were predominately involved in selenocompound metabolism and plant hormone signal transduction. Then the expression of 12 mi RNAs was validated by quantitative real-time PCR(q RT-PCR). | 10 | 3D microgroove electrical impedance sensing to examine 3D cell cultures for antineoplastic drug assessment显示文摘In recent decades,three-dimensional(3D)cancer cell models have attracted increasing interest in the field of drug screening due to their significant advantages in more accurate simulations of heterogeneous tumor behavior in vivo compared to two-dimensional models.Furthermore,drug sensitivity testing based on 3D cancer cell models can provide more reliable in vivo efficacy prediction.The gold standard fluorescence staining is hard to achieve real-time and label-free viability monitoring in 3D cancer cell models.In this study,a microgroove impedance sensor(MGIS)was specially developed for the dynamic and noninvasive monitoring of 3D cell viability.3D cancer cells were trapped in microgrooves with gold electrodes on opposite walls for in situ impedance measurement.The change in the number of live cells caused inversely proportional changes to the impedance magnitude of the entire cell/Matrigel construct and reflected the proliferation and apoptosis of the 3D cells.It was confirmed that the 3D cell viability detected by the MGIS was highly consistent with the standard live/dead staining by confocal microscope characterization.Furthermore,the accuracy of the MGIS was validated quantitatively using a 3D lung cancer model and sophisticated drug sensitivity testing.In addition,the parameters of the MGIS in the measurement experiments were optimized in detail using simulations and experimental validation.The results demonstrated that the MGIS coupled with 3D cell culture would be a promising platform to improve the efficiency and accuracy of cell-based anticancer drug screening in vitro. | Yuxiang Pan Deming Jiang Chenlei Gu Yong Qiu Hao Wan Ping Wang | 2020 | Microsystems & Nanoengineering2020,6,1: | 2 |
| 11 | Dissimilatory iron reduction contributes to anaerobic mineralization of sediment in a shallow transboundary lake显示文摘Dissimilatory iron reduction(DIR)coupled with carbon cycling is increasingly being recognized as an influential process in freshwater wetland soils and sediments.The role of DIR in organic matter(OM)mineralization,however,is still largely unknown in lake sediment environments.In this study,we clarified rates and pathways of OM mineralization in two shallow lakes with seasonal hydrological connectivity and different eutrophic situations.We found that in comparison with the domination of DIR(55%)for OM mineralization in Lake Xiaoxingkai,the contribution of methanogenesis was much higher(68%)in its connected lake(Lake Xingkai).The differences in rates and pathways of sediment OM mineralization between the two lakes were attributed to higher concentrations of carbonate associated iron oxides(Fecarb)in Lake Xiaoxingkai compared to Lake Xingkai(P=0.002),due to better deposition mixing,more contributions of terrigenous detrital materials,and higher OM content in Lake Xiaoxingkai.Results of structural equation modeling showed that Fecarb and total iron content(TFe)regulated 25%of DIR in Lake Xiaoxingkai and 76%in Lake Xingkai,accompanied by a negative effect of TFe on methanogenesis in Lake Xingkai.The relative abundance and diversity of Fe-reducing bacteria were significantly different between the two lakes,and showed a weak effect on sediment OM mineralization.Our findings emphasize the role of iron minerals and geochemical characterizations in regulating rates and pathways of OM mineralization,and deepen the understanding of carbon cycling in lake sediments. | Yuxiang Yuan Cong Ding Haitao Wu Xue Tian Min Luo Weiyi Chang Lei Qin Liang Yang Yuanchun Zou Kaikai Dong Xiaoyan Zhu Ming Jiang Marinus L.Otte | 2023 | Fundamental Research2023,3,6: | 2 |
