| 3 | Identification of an AFLP marker linked to the stripe rust resistance gene Yr10 in wheat显示文摘AFLP analysis of near-isogenic lines of the stripe rust resistance gene Yr10 was carried out with 6 Pst I -primers and 10 Taq I -primers with the donor parent of Yr1O gene as the check. A total of about 4200 distinguishable bands were amplified, of which 5 were stable. The genetic linkage of the 5 polymorphic DNA fragments with the target gene were tested preliminarily on a segregating F2 population derived from a cross between the gene donor parent 'Moro' and susceptible cultivar 'Mingxian 169'. The DNA fragment PT0502 was found closely linked to the Yr10 gene and cloned and sequenced. Based on the sequence specific primers for PCR were designed and synthesized. Genetic linkage analysis with 195 segregating F2 plants indicated that the genetic distance was 0.5 cM between the main product SC200 fragment produced by PCR with the primers and the Yr10 gene. | 7 | Specific photoacoustic cavitation through nucleus targeted nanoparticles for high-efficiency tumor therapy显示文摘As a new type of cancer treatment,photoacoustic(PA)therapy is based on PA shockwave for rapid,selective and effective killing of cancer cells.The nucleus has been widely used as a target for tumor therapy,which has obtained a very considerable therapeutic effect.In situ destruction of tumor cell nucleus by photoacoustic therapy has not been studied.In this paper,a highly efficient nucleus-targeted photoacoustic theranostic polymer was developed for fluorescence and photoacoustic dual-mode imaging-guided PA therapy.The prepared polymer consists of nucleus targeting TAT peptide(TAT:YGRKKRRQRRR),hydrophilic chain poly(N,N-dimethylacrylamide)(PDMA),and near-infrared(NIR)light absorbing agent(hCyR),which can self-assemble to form nanoparticles of approximately 28 nm(denoted as TAT-PDMA-hCyR NPs).The designed nanoparticles show excellent nucleus targeting and tumor cell death(up to 80%)caused by DNA damage under pulsed laser irradiation compared to non-nucleus target counterpart PDMA-hCyR NPs without TAT peptide in vitro.As expected,the fluorescence and PA dual-mode imaging observed that TAT-PDMA-hCyR NPs were able to passively target and enrich in tumors,providing an experimental basis for in vivo treatment and thus ensuring a significant tumor inhibition rate(about 92%).In conclusion,this study provides a new and practicable method for the development of nucleus-targeting nanoparticles as potential theranostic agent for in vivo cancer imaging and therapy. | Yuan Wang Guangle Niu Shaodong Zhai Wenjia Zhang Da Xing | 2020 | Nano Research2020,13,3: | 3 |
| 8 | RGB WGM lasing woven in fiber braiding cavity显示文摘It is critical to realize woven and wearable red-green-blue(RGB) whispering gallery mode(WGM)lasing that is tunable and flexible in the fiber braiding cavity. However, it is still a challenge to achieve tunable full-color lasing with the large color gamut by a facile approach. Here, we design the braiding cavity to realize RGB WGM lasing with high performance. The braiding cavity consists of three optical fibers coated with dye-doped polymer. Each fiber therein serves as a resonant cavity to support multiple lasing modes. There is no interference owing to the air gap between them;therefore, we can achieve full-color WGM lasing. | 10 | Clinical application of free/total PSA ratio in the diagnosis of prostate cancer in men over 50 years of age with total PSA levels of 2.0-25.0 ng ml^(−1) in Western China显示文摘The goal of this study was to investigate the clinical application of free/total prostate-specific antigen(F/T PSA)ratio,considering the new broad serum total PSA(T-PSA)“gray zone”of 2.0–25.0 ng ml^(−1)in differential diagnosis of prostate cancer(PCa)and benign prostate diseases(BPD)in men over 50 years in Western China.A total of 1655 patients were included,528 with PCa and 1127 with BPD.Serum T-PSA,free PSA(F-PSA),and F/T PSA ratio were analyzed.Receiver operating characteristic curves were used to assess the efficiency of PSA and F/T PSA ratio.There were 47.4%of cancer patients with T-PSA of 2.0–25.0 ng ml^(−1).When T-PSA was 2.0–4.0 ng ml^(−1),4.0–10.0 ng ml^(−1),and 10.0–25.0 ng ml^(−1),the area under the curve(AUC)of F/T PSA ratio was 0.749,0.769,and 0.761,respectively.The best AUC of F/T PSA ratio was 0.811 when T-PSA was 2.0–25.0 ng ml^(−1),with a specificity of 0.732,a sensitivity of 0.788,and an optimal cutoff value of 15.5%.The AUC of F/T PSA ratio in different age groups(50–59 years,60–69 years,70–79 years,and≥80 years)was 0.767,0.806,0.815,and 0.833,respectively,and the best sensitivity(0.857)and specificity(0.802)were observed in patients over 80 years.The T-PSA trend was in accordance with the Gleason score,tumor node metastasis(TNM)stage,and American Joint Committee on Cancer prognosis group.Therefore,the F/T PSA ratio can facilitate the differential diagnosis of PCa and BPD in the broad T-PSA“gray zone”.Serum T-PSA can be a Gleason score and prognostic indicator. | Xue-Dan Gao Qiang Miao Jun-Long Zhang Jian-Zhao Zhai Xue-Mei Gui Yi-Han Cai Qian Niu Bei Cai | 2022 | Asian Journal of Andrology2022,24,2: | 2 |
| 11 | Isolation and characterization of 49 polymorphic microsatellite loci for Decapterus maruadsi using SLAF-seq, and cross-amplifi cation to related species显示文摘Decapterus maruadsi is a commercially important species in China, but has been heavily exploited in some areas. There is a growing need to develop microsatellites promoting its genetic research for the adequate management of this ?shery resources. The recently developed speci?c-locus ampli?ed fragment sequencing(SLAF-seq) is an effcient and high-resolution method for genome-wide microsatellite markers discovery. In this study, 28 905 microsatellites(mono-to hexa-nucleotide repeats) were identi?ed using SLAF-seq technology, of which di-nucleotide was the most frequent(13 590, 47.02%), followed by mono-nucleotide(8 138, 28.15%), tri-nucleotide(5 727, 19.81%), tetra-nucleotide(1 104, 3.82%), pentanucleotide(234, 0.81%), and hexa-nucleotide(112, 0.39%). One hundred and thirty-two microsatellite loci(di-and tri-nucleotide) were randomly selected for ampli?cation and polymorphism, of which 49 were highly polymorphic and well-resolved. The average number of alleles per locus was 13.63, ranging from 4 to 25, and allele sizes varied between 110 bp and 309 bp. The observed heterozygosity( Ho) and expected heterozygosity( He) ranged from 0.233 to 1.000 and from 0.374 to 0.959, with mean values of 0.738 and0.836, respectively. The polymorphism information content(PIC) ranged from 0.341 to 0.941(mean=0.806).However, 12 loci deviated from Hardy-Weinberg equilibrium. Furthermore, transferability tests were also successful in validating the utility of the developed markers in ?ve phylogenetically related species of family Carangidae. | 13 | An Assessment of the Predictability of the East Asian Subtropical Westerly Jet Based on TIGGE Data显示文摘The predictability of the position, spatial coverage and intensity of the East Asian subtropical westerly jet(EASWJ) in the summers of 2010 to 2012 was examined for ensemble prediction systems(EPSs) from four representative TIGGE centers,including the ECMWF, the NCEP, the CMA, and the JMA. Results showed that each EPS predicted all EASWJ properties well, while the levels of skill of all EPSs declined as the lead time extended. Overall, improvements from the control to the ensemble mean forecasts for predicting the EASWJ were apparent. For the deterministic forecasts of all EPSs, the prediction of the average axis was better than the prediction of the spatial coverage and intensity of the EASWJ. ECMWF performed best, with a lead of approximately 0.5–1 day in predictability over the second-best EPS for all EASWJ properties throughout the forecast range. For probabilistic forecasts, differences in skills among the different EPSs were more evident in the earlier part of the forecast for the EASWJ axis and spatial coverage, while they departed obviously throughout the forecast range for the intensity. ECMWF led JMA by about 0.5–1 day for the EASWJ axis, and by about 1–2 days for the spatial coverage and intensity at almost all lead times. The largest lead of ECMWF over the relatively worse EPSs, such as NCEP and CMA, was approximately 3–4 days for all EASWJ properties. | 19 | 14 μm quantum cascade lasers based on diagonal transition and nonresonant extraction显示文摘We report In P-based room-temperature high-average-power quantum cascade lasers emitting at 14 μm. Using a novel active region design, a diagonal bound-to-bound lasing transition is guaranteed by efficient electron injection into the upper laser level and fast nonresonant electron extraction through a miniband from the lower laser level. For a 4 mm long and 40 μm wide double channel ridge waveguide laser with 55 stages of the active region, the threshold current density is only 3.13 k A∕cm^2 at room temperature. At 293 K, the maximum singlefacet peak power and average power are up to 830 m W and 75 m W, respectively. | 20 | Application of an Improved Analog-Based Heavy Precipitation Forecast Model to the Yangtze–Huai River Valley and Its Performance in June–July 2020显示文摘Precipitation extremes,such as the record-breaking Meiyu characterized by frequent occurrences of rainstorms that resulted in severe flooding over the Yangtze-Huai River valley(YHRV)in June-July 2020,are always attracting considerable interest,highlighting the importance of improving the forecast accuracy at the medium-to-long range.To elevate the skill in forecasting heavy precipitation events(HPEs)with both long and short durations,the Key Influential Systems Based Analog Model(KISAM)was further improved and brought into operational application in 2020.Verification and comparison of this newly adapted analog model and ensemble mean forecasts from the ECMWF at lead times of up to 15 days were carried out for the identified 16 HPEs over the YHRV in June-July 2020.The results demonstrate that KISAM is advantageous over ECMWF ensemble mean for forecasts of heavy precipitation≥25 mm day-1 at the medium-to-long(6-15-day)lead times,based on the traditional dichotomous metrics.At short lead times,ECMWF ensemble mean outperforms KISAM due largely to the low false alarm rates(FARs)benefited from an underestimation of the frequency of heavy precipitation.However,at the medium-to-long forecast range,the large fraction of misses induced by the high degree of underforecasting overwhelms the fairly good FARs in the ECMWF ensemble mean,which partly explains its inferiority to KISAM in terms of the threat score.Further assessment on forecasts of the latitudinal location of accumulated heavy precipitation indicates that smaller displacement errors also account for a part of the better performance of KISAM at lead times of 8-12 days. | Baiquan ZHOU Panmao ZHAI Ruoyun NIU | 2021 | Journal of Meteorological Research2021,35,6: | 1 |
