| 2 | Loss of Lkb1 impairs Treg function and stability to aggravate graft-versus-host disease after bone marrow transplantation显示文摘Accumulating evidence suggests that a reduction in the number of Foxp3^(+) regulatory T cells(Tregs)contributes to the pathogenesis of acute graft-versus-host disease(aGVHD),which is a major adverse complication that can occur after allogeneic hematopoietic stem cell transplantation(allo-HSCT).However,the precise features and mechanism underlying the defects in Tregs remain largely unknown.In this study,we demonstrated that Tregs were more dramatically decreased in bone marrow compared with those in peripheral blood from aGVHD patients and that bone marrow Treg defects were negatively associated with hematopoietic reconstitution.Tregs from aGVHD patients exhibited multiple defects,including the instability of Foxp3 expression,especially in response to IL-12,impaired suppressor function,decreased migratory capacity,and increased apoptosis.Transcriptional profiling revealed the downregulation of Lkb1,a previously identified critical regulator of murine Treg identity and metabolism,and murine Lkb1-regulated genes in Tregs from aGVHD patients.Foxp3 expression in human Tregs could be decreased and increased by the knockdown and overexpression of the Lkb1 gene,respectively.Furthermore,a loss-of-function assay in an aGVHD murine model confirmed that Lkb1 deficiency could impair Tregs and aggravate disease severity.These findings reveal that Lkb1 downregulation contributes to multiple defects in Tregs in human aGVHD and highlight the Lkb1-related pathways that could serve as therapeutic targets that may potentially be manipulated to mitigate aGVHD. | Xiuhua Su Qianqian Wang Wei Guo Xiaolei Pei Qing Niu Maolan Liu Yuanyuan Liu Song Chen Sizhou Feng Yi He Donglin Yang Rongli Zhang Qiaoling Ma Weihua Zhai Aiming Pang Jialin Wei Yong Huang Yuechen Luo Mingzhe Han Xiaoming Feng Erlie Jiang | 2020 | Cellular & Molecular Immunology2020,17,5: | 3 |
| 3 | In vitro cytocompatibility evaluation of poly(octamethylene citrate) monomers toward their use in orthopedic regenerative engineering显示文摘Citrate based polymer poly(octamethylene citrate)(POC)has shown promise when formulated into composite material containing up to 65 wt%hydroxylapatite(HA)for orthopedic applications.Despite significant research into POC,insufficient information about the biocompatibility of the monomers 1,8-Octanediol and Citrate used in its synthesis is available.Herein,we investigated the acute cytotoxicity,immune response,and long-term functionality of both monomers.Our results showed a cell-type dependent cytotoxicity of the two monomers:1,8-Octanediol induced less acute toxicity to 3T3 fibroblasts than Citrate while presenting comparable cytotoxicity to MG63 osteoblast-like cells;however,Citrate demonstrated enhanced compatibility with hMSCs compared to 1,8-Octanediol.The critical cytotoxic concentration values EC30 and EC50,standard for comparing cytotoxicity of chemicals,were also provided.Additionally,Citrate showed slower and less inhibitory effects on long-term hMSC cell proliferation compared with 1,8-Octanediol.Furthermore,osteogenic differentiation of hMSCs exposure to Citrate resulted in less inhibitory effect on alkaline phosphatase(ALP)production.Neither monomer triggered undesired pro-inflammatory responses.In combination with diffusion model analysis of monomer release from cylindrical implants,based on which the maximum concentration of monomers in contact with bone tissue was estimated to be 2.2104 mmol/L,far lower than the critical cytotoxic concentrations as well as the 1,8-Octanediol concentration(0.4 mg/mL or 2.7 mmol/L)affecting hMSCs differentiation,we provide strong evidence for the cytocompatibility of the two monomers degraded from citrate-based composites in the orthopedic setting. | Chuying Ma Ethan Gerhard Qiaoling Lin Silun Xia April Dawn Armstrong Jian Yang | 2018 | Bioactive Materials2018,3,1: | 2 |
| 4 | Risk factors associated with hemorrhagic cystitis after allogeneic hematopoietic stem cell transplantation显示文摘Hemorrhagic cystitis(HC)is a common complication of allogeneic hematopoietic stem cell transplantation(HSCT).The incidence is about 7%to 68%,and some patients have to suffer a long period of frequent,urgent,and painful urination,which brings great pain.This study aimed to analyze risk factors of HC and its effect on patient survival.We collected the medical records of 859 patients who underwent HSCT at our hospital between August 2016 and August 2020.Patients with and without HC were matched using propensity score matching at a 1:1 ratio based on sex,age,and diagnosis,and logistic regression analyses were used to identify factors associated with HC.We used Kaplan–Meier curves to analyze the survival rates of patients in the HC and non-HC groups.We also analyzed the relationship between BK viral load and the occurrence of HC using receiver operating characteristic curve(ROC)analysis.After propensity score matching,there were 131 patients each in the HC and non-HC groups.In the HC group,89 patients(67.9%)had mild HC(stage II°)and 43(32.1%)had severe HC(stage III–IV).The median interval between stem cell transplantation and HC development was 31(3–244)days.Univariate analysis indicated that donor age,hematopoietic stem cell source,HLA,acute graft-versus-host disease,busulfan,anti-thymocyte globulin(ATG),total body irradiation,cytomegalovirus(CMV)(urine),and BK polyomavirus(BKV)(urine)were significantly associated with HC.ATG,CMV(urine),and BKV(urine)were independent risk factors for HC based on the multivariate analysis.The Kaplan–Meier survival analysis showed no significant difference between the HC and non-HC groups(P=0.14).The 1-and 2-year survival rates in the HC group were 78.4%and 69.6%,respectively,and the corresponding rates in the non-HC group were 84.4%and 80.7%,respectively.ROC analysis indicated that a urine BKV load of 1×10^(7) copies/mL was able to stratify the risk of HC.In conclusion,when the BKV load is>1×10^(7),we needtobe aware of the potential for the development of HC. | Biao Shen Yueshen Ma Haixiao Zhang Mingyang Wang Jia Liu Jiaxin Cao Wenwen Guo Dan Feng Donglin Yang Rongli Zhang Xin Chen Qiaoling Ma Weihua Zhai Sizhou Feng Mingzhe Han Aiming Pang Erlie Jiang 无 | 2022 | Blood Science2022,4,2: | 2 |

| 10 | Bismuth stabilized by ZIF derivatives for electrochemical ammonia production:Proton donation effect of phosphorus dopants显示文摘N2 electroreduction reaction(NRR)offers a feasible and promising alternative for NH_(3)production by using clean energy sources.However,it is still obstructed by the pretty low NH3 yield rate and Faradaic efficiency(FE)primarily due to the undesired competing hydrogen evolution reaction and the extremely stable N≡N bond.Herein,bismuth nanoparticles were successfully embedded in N and P co-doped carbon nanoflakes(Bi/NPC)by high-temperature pyrolyzation of Bi-zeolitic imidazole frameworks(ZIF)followed by phosphorization,and used as a high-efficiency catalyst toward N2 electroreduction to NH3.In 0.1 M KHCO_(3)electrolyte,Bi/NPC exhibits excellent NRR performances,including a high NH3 yield rate of 3.12μg·h^(−1)·cm^(−2)(−0.6 V vs.reversible hydrogen electrode(RHE)),an outstanding FE of 13.58%(−0.4 V vs.RHE),and a remarkable stability up to 36 h under ambient conditions.This outstanding NRR catalytic activity is mainly attributed to the intrinsic electrocatalytic NRR activity combined with the inert hydrogen evolution reaction(HER)activity of Bi,the adsorption and activation of N2 facilitated by N dopants,as well as the superior conductivity and the large specific surface area of the two-dimensional layered carbon matrix.Notably,the hydrogen source provided by P dopant promotes the hydrogenation of the adsorbed N,which further boosts the NRR performance in alkaline electrolyte.The ultralong durability of Bi/NPC is attributed to the highly dispersed bismuth catalytic active centers confined in the skeleton of N and P co-doped carbon nanoflakes,which inhibits the agglomeration of bismuth centers.This work presents a novel avenue for designation and fabrication of high-performance Bi-based electrocatalysts for NRR. | Qiaoling Wu Ying Sun Qin Zhao Hui Li Zhengnan Ju Yu Wang Xiaodong Sun Baohua Jia Jieshan Qiu Tianyi Ma | 2023 | Nano Research2023,16,4: | 0 |
