| 1 | Knockdown of HMGB1 improves apoptosis and suppresses proliferation and invasion of glioma cells显示文摘Background: Effective methods for managing patients with solitary pulmonary nodules(SPNs) depend critically on the predictive probability of malignancy.Methods: Between July 2009 and June 2011, data on gender, age, cancer history, tumor familial history, smoking status, tumor location, nodule size, spiculation, calcification, the tumor border, and the final pathological diagnosis were collected retrospectively from 154 surgical patients with an SPN measuring 3-30 mm. Each final diagnosis was compared with the probability calculated by three predicted models—the Mayo, VA, and Peking University(PU) models. The accuracy of each model was assessed using area under the receiver operating characteristics(ROC) and calibration curves.Results: The area under the ROC curve of the PU model [0.800; 95% confidence interval(CI): 0.708-0.891] was higher than that of the Mayo model(0.753; 95% CI: 0.650-0.857) or VA model(0.728; 95% CI: 0.623-0.833); however, this finding was not statistically significant. To varying degrees, calibration curves showed that all three models overestimated malignancy.Conclusions: The three predicted models have similar accuracy for prediction of SPN malignancy, although the accuracy is not sufficient. For Chinese patients, the PU model may has greater predictive power.Background: Here, we introduced our short experience on the application of a new CUSA Excel ultrasonic aspiration system, which was provided by Integra Lifesciences corporation, in skull base meningiomas resection.Methods: Ten patients with anterior, middle skull base and sphenoid ridge meningioma were operated using the CUSA Excel ultrasonic aspiration system at the Neurosurgery Department of Shanghai Huashan Hospital from August 2014 to October 2014. There were six male and four female patients, aged from 38 to 61 years old(the mean age was 48.5 years old). Five cases with tumor located at anterior skull base, three cases with tumor on middle skull base, and two cases with tumor on sphenoid ridge.Results: All the patents received total resection of meningiomas with the help of this new tool, and the critical brain vessels and nerves were preserved during operations. All the patients recovered well after operation.Conclusions: This new CUSA Excel ultrasonic aspiration system has the advantage of preserving vital brain arteries and cranial nerves during skull base meningioma resection, which is very important for skull base tumor operations. This key step would ensure a well prognosis for patients. We hope the neurosurgeons would benefit from this kind of technique.Background: The purposes of this study were to explore the effects of high mobility group protein box 1(HMGB1) gene on the growth, proliferation, apoptosis, invasion, and metastasis of glioma cells, with an attempt to provide potential therapeutic targets for the treatment of glioma. Methods: The expressions of HMGB1 in glioma cells(U251, U-87 MG and LN-18) and one control cell line(SVG p12) were detected by real time PCR and Western blotting, respectively. Then, the effects of HMGB1 on the biological behaviors of glioma cells were detected: the expression of HMGB1 in human glioma cell lines U251 and U-87 MG were suppressed using RNAi technique, then the influences of HMGB1 on the viability, cycle, apoptosis, and invasion abilities of U251 and U-87 MG cells were analyzed using in a Transwell invasion chamber. Also, the effects of HMGB1 on the expressions of cyclin D1, Bax, Bcl-2, and MMP 9 were detected. Results: As shown by real-time PCR and Western blotting, the expression of HMGB1 significantly increased in glioma cells(U251, U-87 MG, and LN-18) in comparison with the control cell line(SVG p12); the vitality, proliferation and invasive capabilities of U251 and U-87 MG cells in the HMGB1 siR NA-transfected group were significantly lower than those in the blank control group and negative control(NC) siR NA group(P<0.05) but showed no significant difference between the blank control group and NC siR NA group. The percentage of apoptotic U251 and U-87 MG cells was significantly higher in the HMGB1 siR NA-transfected group than in the blank control group and NC siR NA group(P<0.05) but was similar between the latter two groups. The HMGB1 siR NA-transfected group had significantly lower expression levels of Cyclin D1, Bcl-2, and MMP-9 protein in U251 and U-87 MG cells and significantly higher expression of Bax protein than in the blank control group and NC siR NA group(P<0.05); the expression profiles of cyclin D1, Bax, Bcl-2, and MMP 9 showed no significant change in both blank control group and NC siR NA group. Conclusions: HMGB1 gene may promote the proliferation and migration of glioma cells and suppress its effects of apoptosis. Inhibition of the expression of HMGB1 gene can suppress the proliferation and migration of glioma cells and promote their apoptosis. | 2 | WAF-Based Chinese Character Recognition for Spam Image Filtering显示文摘We address the problem of filtering image spam, a kind of rapidly spread spam in which the text is embedded into images to defeat text-based spam filter.Particularly, we focus on image spam with Chinese text as 'spam' which is a more challenging task. A popular way to detect image spam is by Optical character recognition(OCR) system, which detects and recognizes the embedded text, then followed by a text classifier that discriminate spam from ham. However, spammers start to obscure image text to prevent OCR system discovering the spam text. To compensate for the shortcomings of OCR system, a novel method which essentially is a keyword reconstruction algorithm based on Word activation force(WAF) model is proposed. It is effective on discovering keywords, hence is benefit for the later classification stage and notably improve the performance of image spam filtering. | 3 | Filtering Chinese Image Spam Using Pseudo-OCR显示文摘For image spam filtering, the Optical character recognition(OCR) based methods often achieve a better performance due to the more complex structure of recognizing corresponding text. However, applying traditional OCR techniques usually introduced shortcomings like the expensive computational cost, vulnerability to image noises and artificial interferences, especially for Chinese image spam filtering. So, by optimizing recognition procedure of traditional OCR, we propose the idea of pseudo-OCR more suitable for Chinese image spam filtering. During which discriminating the potential image spam character features from ham ones is sufficient, instead of recognizing them. What's more, a novel Chinese key-point based character feature specific for pseudo-OCR is also devised and extracted using a carefully designed algorithm, which outperforms classic corner detection methods in finding such key-points. | 9 | Establishment of Indirect ELISA Diagnosis Technique based on the VP1 Protein of Foot and Mouth Disease Virus Serotype A显示文摘The VP1 protein of foot-and-mouth disease virus serotype A was prokaryotically expressed and purified to replace the traditional virus antigen for establishing a fast,safe,effective indirect ELISA method,so as to detecting antibody of foot-and-mouth disease virus serotype A. Western-Blot test showed that the VP1 recombinant protein could be used as detective antigen as it can be specifically recognized by bovine positive serum of FMDV serotype A. By employing matrix titration method,the optimal parameters were obtained as follows: 1 mg / L VP1 protein as coating antigen,Vserum∶ Vblocking solution = 1∶50 dilution for serum and Vsecondary enzyme-linked antibodies∶Vblocking solution = 1∶2 000 for enzyme combined antibodies. The results showed that the sensitivity and specificity of this method were 94. 32% and 99. 09% respectively,the coefficients of variations in intra-assay and inter-assay reproducibility tests was lower than 8%. Compared with liquid phase blocking ELISA kits,the agreement of 201 serum samples reached 92. 54%. | 12 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of | 13 | Mechanism underlying the immune checkpoint inhibitor-induced hyper-progressive state of cancer显示文摘Immune checkpoint inhibitors(ICIs)are gradually replacing chemotherapy as the cornerstone of the treatment of advanced malignant tumors because of their long-lasting and significant effect in different tumor types and greatly prolonging the survival time of patients.However,not all patients can respond to ICIs,and even rapid tumor growth after treatment with ICI has been observed in a number of clinical studies.This rapid progression phenomenon is called hyper-progressive disease(HPD).The occurrence of HPD is not uncommon.Past statistics show that the incidence of HPD is 4%-29% in different tumor types,and the progression-free survival and overall survival of patients with HPD are significantly shorter than those of the non-HPD progressor group.With the deepening of the study of HPD,we have established a preliminary understanding of HPD,but the diagnostic criteria of HPD are still not unified,and the addition of biomarkers may break this dilemma.In addition,quite a few immune cells have been found to be involved in the occurrence and development of HPD in the tumor microenvironment,indicating that the molecular mechanism of HPD may be triggered by a variety of ongoing events at the same time.In this review,we summarize past findings,including case reports,clinical trials,and fundamental research;compare the diagnostic criteria,incidence,and clinical prognostic indicators of HPD in different studies;and explore the molecular mechanism and future research direction of HPD. | Peng Ding Lu Wen Fan Tong Ruiguang Zhang Yu Huang Xiaorong Dong | 2022 | Cancer Drug Resistance2022,5,1: | 0 |
| 14 | Molecular phylogeny of European and African Barbus and their West Asian relatives in the Cyprininae(Teleostei:Cypriniformes)and orogenesis of the Qinghai-Tibetan Plateau显示文摘The phylogenetic relationships of European and African Barbus and their West Asian relatives in Cyprininae remain largely unresolved.Consequently,little is known about the drivers of their evolution,including the possible association of uplifting of the Qinghai-Tibetan Plateau(QTP)with the early divergence of the subfamily.We use complete sequence data of the mitochondrial DNA gene encoding the protein cytochrome b(Cytb)to hypothesize the phylogeny of 85 species belonging to 47 genera in the Cyprininae plus 6 species from the Leuciscinae.We employ 6 other species from Cypriniformes as outgroup taxa and estimate divergence times.Our results indicate that European Barbus sensu stricto lineage including Aulopyge shares a common ancestor with specialized and highly specialized schizothoracins and the genera Cyprinion and Scaphiodonichtys.The common ancestor appears to have originated in the Qinghai-Tibetan Plateau(QTP)region about 19.4–17.8 Ma.Barbus sensu stricto lineage appears to have originated about 16.6–15.5 Ma.Small to medium sized African Barbus sensu lato appear to have had an Oriental origin about 19.1–15.3 Ma and are closely related to Asian Puntius.West Asian Carasobarbus lineage including large African Barbus sensu lato might have originated about 9.94 Ma,also in Oriental Realm and has a close relationship to Asian Neolissochilus and Tor.The large-sized Barbus sensu lato appear to have diverged from Carasobarbus about 7.7 Ma.Finally,the Cyprininae appear to have radiated rapidly into nine lineages and many sublineages from about 27.8 to 17.8 Ma,close to the time of the second-stage tectonic movements of the QTP.Our analyses provide evidence that the uplifting of the QTP drove early diversification of the Cyprininae.Our extensive sampling of species involving all of the important areas results in clear evolutionary scenario for the Cyprininae. | WANG Jing WU XiaoYun CHEN ZiMing YUE ZhaoPing MA Wei CHEN ShanYuan XIAO Heng MURPHY Robert W ZHANG YaPing ZAN RuiGuang LUO Jing | 2013 | Chinese Science Bulletin2013,58,31: | 0 |
