2Combination of chemotherapy and immunotherapy for colon cancer in China: A meta-analysis显示文摘AIM:To investigate whether autologous dendritic cell(DC)-cytokine-induced killer(CIK)cell therapy is able to improve the therapeutic efficacy of chemotherapy in colon cancer.METHODS:We conducted a systematic review of published papers from the sources of MEDLINE,the Cochrane Central Register of Controlled Trials,EMBASE,the Wanfang Database,the China Science and Technology Periodical Database and China Journal Net.Published data were extracted independently by two authors using predefined database templates.The quality of the data from individual papers was also assessed.The effects of chemotherapy were compared with those of chemotherapy in combination with DC-CIK immunotherapy.The pooled analysis was performed using the data from random or fixed-effect models.RESULTS:Seven trials matched our inclusion criteria(n=533).The overall analysis showed significant survival benefit[one-year overall survival(OS),P<0.0001;twoyear OS,P=0.009;three-year OS,P=0.002]in favor of DC-CIK immunotherapy combined with chemotherapy.Disease-free survival(DFS)rate was improved after the combination of DC-CIK immunotherapy and chemotherapy(one-year DFS,P<0.0001;two-year DFS,P=0.002;three-year DFS,P=0.02).An improved overall response rate(P=0.009)was also observed in patients who received DC-CIK therapy.Furthermore,the analysis of T-lymphocyte subsets in peripheral blood indicated that the number of CD4+T cells significantly increased in the DC-CIK plus chemotherapy group(P<0.05).CONCLUSION:The combination of DC-CIK immunotherapy and chemotherapy was superior in prolonging the survival time and enhancing immunological responses.Zheng-Xu Wang Jun-Xia Cao Zhi-Ping Liu Yu-Xin Cui Chun-Yun Li Duo Li Xiao-Yan Zhang Jin-Long Liu Jun-Li Li 2014World Journal of Gastroenterology2014,20,4:30

5Effect of minor Sc addition on microstructure and stress corrosion cracking behavior of medium strength Al–Zn–Mg alloy显示文摘Influence of Sc content on microstructure and stress corrosion cracking behavior of medium strength Al–Zn–Mg alloy have been investigated by optical microscopy, scanning electron microscopy, electron backscatter diffraction, transmission electron microscopy and slow strain rate test. The results indicate that the addition of Sc results in the formation of the quaternary coherent Al_3(Sc, Zr, Ti) dispersoids during homogenization treatment, which will inhibit the dynamic recrystallization behavior. The number density of Al_3(Sc, Zr, Ti) particles increases with the increase of Sc content, and thus the recrystallization fraction of hot-extruded alloy is reduced and the peak strength in two-stage artificial aging sample is enhanced. At the same time, the wide of precipitation free zone is reduced, and the content of Zn and Mg in grain boundary particles and precipitation free zone is increased with the increase of Sc content. In peak-aged state, the 0.06 wt% Sc added alloy shows the better stress corrosion cracking resistance than the Sc-free alloy because of the reduction of recrystallization fraction and the interrupted distribution of grain boundary precipitates along grain boundary. 6Climate change in Mt. Qomolangma region since 1971显示文摘Using monthly average, maximum, minimum air temperature and monthly precipitation data from 5 weather stations in Mt. Qomolangma region in China from 1971 to 2004, climatic linear trend, moving average, low-pass filter and accumulated variance analysis methods, the spatial and temporal patterns of the climatic change in this region were analyzed. The main findings can be summarized as follows: (1) There is obvious ascending tendency for the interannual change of air temperature in Mt. Qomolangma region and the ascending tendency of Tingri, the highest station, is the most significant. The rate of increasing air temperature is 0.234 oC/decade in Mt. Qomolangma region, 0.302 oC/decade in Tingri. The air temperature increases more strongly in non-growing season. (2) Compared with China and the global average, the warming of Mt. Qomolangma region occurred early. The linear rates of temperature increase in Mt. Qomolangma region exceed those for China and the global average in the same period. This is attributed to the sensitivity of mountainous regions to climate change. (3) The southern and northern parts of Mt. Qomolangma region are quite different in precipitation changes. Stations in the northern part show increasing trends but are not statistically significant. Nyalam in the southern part shows a decreasing trend and the sudden decreasing of precipitation occurred in the early 1990s. (4) Compared with the previous studies, we find that the warming of Mt. Qomolangma high-elevation region is most significant in China in the same period. The highest automatic meteorological comprehensive observation station in the world set up at the base camp of Mt. 7Biomechanical comparison of locking plate and crossing metallic and absorbable screws fixations for intra-articular calcaneal fractures显示文摘The locking plate and percutaneous crossing metallic screws and crossing absorbable screws have been used clinically to treat intra-articular calcaneal fractures, but little is known about the biomechanical differences between them. This study compared the biomechanical stability of calcaneal fractures fixed using a locking plate and crossing screws. Three-dimensional finite-element models of intact and fractured calcanei were developed based on the CT images of a cadaveric sample. Surgeries were simulated on models of Sanders type III calcaneal fractures to produce accurate postoperative models fixed by the three implants. A vertical force was applied to the superior surface of the subtalar joint to simulate the stance phase of a walking gait. This model was validated by an in vitro experiment using the same calcaneal sample. The intact calcaneus showed greater stiffness than the fixation models. Of the three fixations, the locking plate produced the greatest stiffness and the highest von Mises stress peak. The micromotion of the fracture fixated with the locking plate was similar to that of the fracture fixated with the metallic screws but smaller than that fixated with the absorbable screws. Fixation with both plate and crossing screws can be used to treat intra-articular calcaneal fractures. 8Preoperative diagnosis and operative effect of intracranial aneurysm with three-dimensional computed tomography angiography显示文摘BACKGROUND: Digital subtraction angiography (DSA) is always regarded as the golden standard for diagnosis of intracranial aneurysm; however, the procedure is complex, traumatic, expensive and easy to induce vascular complication. Three-dimensional computed tomography angiography (3D-CTA) can make up deficiencies of DSA; therefore, it is used in clinical therapy wider and wider. OBJECTIVE: To evaluate the clinical effect of 3D-CTA on disruption and hemorrhage of intracranial aneurysm pre- and post-operation and compare with the effect of DSA.DESIGN: Auto-control contrast observation.SETTING: Department of Neurosurgery, Shengjing Hospital of China Medical University.PARTICIPANTS: A number of 106 patients with disruption and hemorrhage of intracranial aneurysm were selected from the Department of Neurosurgery, Shengjing Hospital of China Medical University from January 2003 to April 2006. All patients were diagnosed with cranial operation and consent. There were 47 males and 59 females aged from 3-76 years with the mean age of (47±13) years. Among them, 82 patients had extensive subarachnoid hemorrhage (SAH), 7 had hemorrhage at longitudinal fissure, and 17 had hemorrhage at ambiens cisterna and lateral fissure. Moreover, intraventricular hematocele was accompanied on 13 patients and hematom on 9 patients. METHODS: ① 3D-CTA examination: Siemens SOMATOM Sensation 64 CT was used in this study. The thickness was 1 mm and interval of reconstruction was 0.8 mm. Localizing section was plainly scanned as the standard of canthus line. Scan ranged from 30 mm below sella to 50 mm above sella. Non-ion contrast medium of Omnipaque 350 (concentration of iodine was 350 g/L) was inserted into anterior vein of elbow with 18G trochar retained with high-pressured injectoc pum. The speed was 4.5 mL/s and the total volume was 80-100 mL with the means of 90 mL. Scan started at 10-20 s after injection of contrast medium. Original image was dealt with Leonardo workstation and retreated with Syngo software. Volume rendering and maximum intensity projection were used to reconstructed images. ② All 106 patients suffered from occlusion of aneurysm clamp. Before operation, 3D-CTA was undertaken and DSA was followed. After operation, patients were rechecked with 3D-CTA. MAIN OUTCOME MEASURES: Comparisons between 3D-CTA and DSA.RESULTS: All 106 patients were involved in the final analysis. ① Examination of 3D-CTA and DSA: Among 118 patients with aneurysm, 110 were checked with 3D-CTA and the detected rate was 93.2% (110/118). Among other 8 cases, 3 were negative and checked again with DSA; 1 had pericallosal aneurysm, 1 ophthalmic aneurysm, and 1 anterior choroidal artery of aneurysm. 3D-CTA results of other 5 cases were suspicious, and then, they were regarded as having aneurysm with DSA. Before operation, correlation among site, body, neck of aneurysm and peripheral anatomic structure were shown sufficiently. After operation, 82 patients were rechecked with 3D-CTA, which was complete occlusion, precise, unobvious constriction, emphraxis or remains as compared with 3D-CTA those pre-operation. ② Characteristics of 3D-CTA: With multiple vessels and angles, 3D-CTA observed the relationship between aneurysm neck and carried artery and showed thrombosis in cavity of aneurysm, calcification of aneurysm wall and peripheral structure of vessel at the same time. However, DSA could not detect the reactions mentioned above. It could delete image of cranium, simulate image of operative route, eliminate artifact induced by metal, but not distinguish blood stream direction. Meanwhile, posterior communicating artery was always poor during circle of Willis artery showing. CONCLUSION: ① 3D-CTA is characterized by simple operation and non-invasive showing vascular stereo structure and correlation. 9Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 10Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
11Vacuum brazing of GH99 superalloy using graphene reinforced BNi-2 composite filler显示文摘A novel graphene reinforced BNi-2 composite filler was developed for brazing GH99 superalloy. The interfacial microstructure of brazed joints was analyzed by field emission scanning electron microscope and a transmission electron microscope. The effects of graphene addition on the microstructure evolution and mechanical properties of brazed joints were investigated, and the strengthening mechanism of graphene was analyzed. The results revealed that due to the addition of graphene, M23(C,B)6 compounds were synthesized in the solid solution and brittle boride precipitates near the brazing seam decreased.Graphene was effective in retarding solute atoms diffusion thus impeding the precipitation of borides.Furthermore, the low coefficient of thermal expansion(CTE) of graphene was conducive to relieve stress concentration of the brazed joints during the cooling process. The shear strengths of brazed joints were significantly improved by exerting the strengthening effect of graphene. 12Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients.Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo 2021Cancer Biology & Medicine2021,18,1:10

14Observations of atmospheric pollutants at Lhasa during 2014-2015:Pollution status and the influence of meteorological factors显示文摘Atmospheric pollutants including SO_2, NO_2, CO, O_3 and inhalable particulate matter(PM2.5 and PM10) were monitored continuously from March 2014 to February 2015 to investigate characteristics of air pollution at Lhasa, Tibetan Plateau. Species exhibited similar seasonal variations except O_3, with the peaks in winter but low valleys in summer. The maximum O_3 concentration was observed in spring, followed by summer, autumn, and winter. The positive correlation between O_3 and PM10 in spring indicated similar sources of them, and was assumed to be turbulent transport. Temperature was the dominant meteorological factor for most species in spring. High temperature accelerates O_3 photochemistry, and favors air disturbance which is conductive to dust resuspension in spring. Relative humidity(RH) and atmospheric pressure were the main meteorological factors in summer. RH showed negative correlations with species, while atmospheric pressure posed opposite situation. Wind speed(WS) was the dominant meteorological factor in autumn, the negative correlations between WS and species indicated diffusion by wind. Most species showed non-significant correlations with meteorological factors in winter, indicating the dependence of pollution on source emission rather than restriction by meteorology. Pollution weather character indicated that emissions were from biomass burning and dust suspension, and meteorological factors also played an important role. Air stream injection from the stratosphere was observed during O_3 pollution period. Air parcels from Southwest Asia were observed during air pollution period in winter. 15Engineering the Biosynthesis of Caffeic Acid in Saccharomyces cerevisiae with Heterologous Enzyme Combinations显示文摘Engineering the biosynthesis of plant-derived natural products in microbes presents several challenges, especially when the expression and activation of the plant cytochrome P450 enzyme is required. By recruiting two enzymes—HpaB and HpaC—from several bacteria, we constructed functional 4- hydroxyphenylacetate 3-hydroxylase (4HPA3H) in Saccharomyces cerevisiae to take on a role similar to that of the plant-derived cytochrome P450 enzyme and produce caffeic acid. Along with a common tyrosine ammonia lyase (TAL), the different combinations of HpaB and HpaC presented varied capabilities in producing the target product, caffeic acid, from the substrate, L-tyrosine. The highest production of caffeic acid was obtained with the enzyme combination of HpaB from Pseudomonas aeruginosa and HpaC from Salmonella enterica, which yielded up to (289.4 ± 4.6) mg-L1 in shake-flask cultivation. The compatibility of heterologous enzymes within a yeast chassis was effectively improved, as the caffeic acid production was increased by 40 times from the initial yield. 16The origin of Wx^(la) provides new insights into the improvement of grain quality in rice显示文摘Appearance and taste are important factors in rice(Oryza sativa) grain quality. Here, we investigated the taste scores and related eating-quality traits of533 diverse cultivars to assess the relationships between—and genetic basis of—rice taste and eating-quality. A genome-wide association study highlighted the Wx gene as the major factor underlying variation in taste and eating quality. Notably, a novel waxy(Wx) allele, Wx^(la), which combined two mutations from Wx^(b) and Wx^(in), exhibited a unique phenotype. Reduced GBSSI activity conferred Wx^(la) rice with both a transparent appearance and good eating quality. Haplotype analysis revealed that Wx^(la) was derived from intragenic recombination. In fact,the recombination rate at the Wx locus was estimated to be 3.34 kb/c M, which was about 75-fold higher than the genome-wide mean, indicating that intragenic recombination is a major force driving diversity at the Wx locus. Based on our results, we propose a new network for Wx evolution, noting that new Wx alleles could easily be generated by crossing genotypes with different Wx alleles. 17Gangba sheep in the Tibetan plateau:Validating their unique meat quality and grazing factor analysis显示文摘Gangba sheep are known for having typical sensory characteristics attributed to free range conditions and grazing on wild plants.The genuine Gangba mutton was selected as the experimental group,and the commercial Tibetan mutton was selected as the control group,the nutritive composition of basic chemical components,amino acids and fatty acids in mutton were investigated to correlate its unique meat quality and eating satisfaction.The results showed that fatty acids were significantly higher(P<0.05)in Gangba mutton than in commercial mutton,and the higher content of flavoring amino acids(glutamic acid and aspartame)were primarily responsible for the taste attributes umami of meat juices.Moreover,the trace elements analysis in mutton and grazing factors(forage,water source and soil)were conducted,to explain the source of essential trace elements in mutton.The concentrations of essential trace elements show that the Gangba mutton was a valuable source for highly available Cu and Zn in human nutrition,and well managed with few detected of toxicity metal.The concentrations of essential trace elements in mutton are closely related to the trace elements in environmental grazing factors.In conclusion,the congenital grazing conditions(a highly mineralized water resource,natural forages and clean soils)were shown to contribute to the unique meat characteristics of Gangba sheep.Qiangying Zhang Ming Que Wei Li Shuang Gao Xin Tan Duo Bu 2021Journal of Environmental Sciences2021,33,3:8

19Lithium promotes recovery of neurological function after spinal cord injury by inducing autophagy显示文摘Lithium promotes autophagy and has a neuroprotective effect on spinal cord injury(SCI); however, the underlying mechanisms remain unclear. Therefore, in this study, we investigated the effects of lithium and the autophagy inhibitor 3-methyladenine(3-MA) in a rat model of SCI. The rats were randomly assigned to the SCI, lithium, 3-MA and sham groups. In the 3-MA group, rats were intraperitoneally injected with 3-MA(3 mg/kg) 2 hours before SCI. In the lithium and 3-MA groups, rats were intraperitoneally injected with lithium(LiCl; 30 mg/kg) 6 hours after SCI and thereafter once daily until sacrifice. At 2, 3 and 4 weeks after SCI, neurological function and diffusion tensor imaging indicators were remarkably improved in the lithium group compared with the SCI and 3-MA groups. The Basso, Beattie and Bresnahan locomotor rating scale score and fractional anisotropy values were increased, and the apparent diffusion coefficient value was decreased. Immunohistochemical staining showed that immunoreactivities for Beclin-1 and light-chain 3 B peaked 1 day after SCI in the lithium and SCI groups. Immunoreactivities for Beclin-1 and light-chain 3 B were weaker in the 3-MA group than in the SCI group, indicating that 3-MA inhibits lithium-induced autophagy. Furthermore, NeuN+ neurons were more numerous in the lithium group than in the SCI and 3-MA groups, with the fewest in the latter. Our findings show that lithium reduces neuronal damage after acute SCI and promotes neurological recovery by inducing autophagy. 20Feasibility of 3.0 T diffusion-weighted nuclear magnetic resonance imaging in the evaluation of functional recovery of rats with complete spinal cord injury显示文摘Diffusion tensor imaging is a sensitive way to reflect axonal necrosis and degeneration, glial cell regeneration and demyelination following spinal cord injury, and to display microstructure changes in the spinal cord in vivo. Diffusion tensor imaging technology is a sensitive method to diagnose spinal cord injury; fiber tractography visualizes the white matter fibers, and directly displays the structural integrity and resultant damage of the fiber bundle. At present, diffusion tensor imaging is restricted to brain examinations, and is rarely applied in the evaluation of spinal cord injury. This study aimed to explore the fractional anisotropy and apparent diffusion coefficient of diffusion tensor magnetic resonance imaging and the feasibility of diffusion tensor tractography in the evaluation of complete spinal cord injury in rats. The results showed that the average combined scores were obviously decreased after spinal cord transection in rats, and then began to increase over time. The fractional anisotropy scores after spinal cord transection in rats were significantly lower than those in normal rats(P < 0.05); the apparent diffusion coefficient was significantly increased compared with the normal group(P < 0.05). Following spinal cord transection, fractional anisotropy scores were negatively correlated with apparent diffusion coefficient values(r = –0.856, P < 0.01), and positively correlated with the average combined scores(r = 0.943, P < 0.01), while apparent diffusion coefficient values had a negative correlation with the average combined scores(r = –0.949, P < 0.01). Experimental findings suggest that, as a non-invasive examination, diffusion tensor magnetic resonance imaging can provide qualitative and quantitative information about spinal cord injury.
