| 1 | Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. | 3 | Morphology,composition,and sources of individual aerosol particles at a regional background site of the YRD,China显示文摘Aerosol samples were collected at Lin'an, a background site of Yangtze River Delta(YRD).Morphology, size, composition, and mixing state of individual aerosol particles were characterized by transmission electron microscopy(TEM) coupled with energy dispersive X-ray spectroscopy(EDS), and the soluble ions of PM_(1.0) were studied by aerosol mass spectrometer(AMS). The daily average AMS mass concentrations of sulfate, nitrate, and ammonium were about 5.8, 8.6, and 5.6 μg/m^3, respectively. Individual aerosol particles were classified into seven types: S-rich, K-rich, organic matter(OM), soot, fly ash, metal, and mineral. S-rich particles were dominant in all size bins, and 51%(by number) of S-rich particles were internally mixed with other particles. The fraction of organic coating particles was 13.7% in morning, 25.2% in afternoon, and 11% in evening, suggesting that the strong photochemical process during afternoon produced more secondary organic aerosols(SOA) on the surface of inorganic particles. Fly ash and metal particles were abundant during the day, suggesting the influence of emissions from coal-fired power plants and steel plants. The results indicate that the intense industrial emissions in the YRD significantly transported to the background areas. | 5 | Incorporation of silica nanoparticles to PLGA electrospun fibers for osteogenic differentiation of human osteoblast-like cells显示文摘The development of bone tissue engineering scaffolds still remains a challenging field,although various biomaterials have been developed for this purpose.Electrospinning is a promising approach to fabricate nanofibers with an interconnected porous structure,which can support cell adhesion,guide cell proliferation and regulate cell differentiation.The aim of this study is to fabricate composite fibers composed of poly(lactic-co-glycolic acid)(PLGA)and silica nanoparticles(NPs)via electrospinning and investigate the effect of PLGA/SiO_(2)composite fibers on the cellular response of osteoblast-like cells(SaOS-2 cells).SEM and EDX analysis showed that silica NPs were homogenously dispersed in the composite fibers.The mechanical behavior of the fibers showed that silica NPs acted as reinforcements at concentrations of 2.5 and 5 mg/ml.The incorporation of silica NPs led to enhancement of cell attachment and spreading on PLGA/SiO_(2)composite fibers.SaOS-2 cells cultured on PLGA/SiO_(2)composite fibers exhibited increased alkaline phosphatase activity,collagen secretion and bone nodules formation.The bone nodules formation of SaOS-2 cells increased along with the amount of incorporated silica NPs.The present findings indicate that PLGA/SiO_(2)composite fibers can stimulate osteogenic differentiation of SaOS-2 cells and may be a promising candidate scaffold for bone tissue engineering. | Xing Yang Yuanyuan Li Xujie Liu Qianli Huang Ranran Zhang Qingling Feng | 2018 | Regenerative Biomaterials2018,5,4: | 4 |
| 6 | Photoinduced large polaron transport and dynamics in organic-inorganic hybrid lead halide perovskite with terahertz probes显示文摘Organic-inorganic hybrid metal halide perovskites(MHPs)have attracted tremendous attention for optoelectronic applications.The long photocarrier lifetime and moderate carrier mobility have been proposed as results of the large polaron formation in MHPs.However,it is challenging to measure the effective mass and carrier scattering parameters of the photogenerated large polarons in the ultrafast carrier recombination dynamics.Here,we show,in a one-step spectroscopic method,that the optical-pump and terahertz-electromagnetic probe(OPTP)technique allows us to access the nature of interplay of photoexcited unbound charge carriers and optical phonons in polycrystalline CH_(3)NH_(3)PbI_(3)(MAPbI_(3))of about 10μm grain size.Firstly,we demonstrate a direct spectral evidence of the large polarons in polycrystalline MAPbI_(3).Using the Drude-Smith-Lorentz model along with the Frӧhlich-type electron-phonon(e-ph)coupling,we determine the effective mass and scattering parameters of photogenerated polaronic carriers.We discover that the resulting moderate polaronic carrier mobility is mainly influenced by the enhanced carrier scattering,rather than the polaron mass enhancement.While,the formation of large polarons in MAPbI_(3)polycrystalline grains results in a long charge carrier lifetime at room temperature.Our results provide crucial information about the photo-physics of MAPbI3 and are indispensable for optoelectronic device development with better performance. | Zuanming Jin Yan Peng Yuqing Fang Zhijiang Ye Zhiyuan Fan Zhilin Liu Xichang Bao Heng Gao Wei Ren Jing Wu Guohong Ma Qianli Chen Chao Zhang Alexey VBalakin Alexander PShkurinov Yiming Zhu Songlin Zhuang | 2022 | Light(Science & Applications)2022,11,8: | 4 |

| 9 | Evolution and protein interactions of AP2 proteins in Brassicaceae: Evidence linking development and environmental responses显示文摘Plants have evolved a large number of transcription factors(TF), which are enriched among duplicate genes,highlighting their roles in complex regulatory networks. The APETALA2/EREBP-like genes constitute a large plant TF family and participate in development and stress responses. To probe the conservation and divergence of AP2/EREBP genes,we analyzed the duplication patterns of this family in Brassicaceae and identified interacting proteins of representative Arabidopsis AP2/EREBP proteins. We found that many AP2/EREBP duplicates generated early in Brassicaceae history were quickly lost, but many others were retained in all tested Brassicaceae species, suggesting early functional divergence followed by persistent conservation. In addition,the sequences of the AP2 domain and exon numbers were highly conserved in rosids. Furthermore, we used 16 A.thaliana AP2/EREBP proteins as baits in yeast screens and identified 1,970 potential AP2/EREBP-interacting proteins,with a small subset of interactions verified in planta. Many AP2 genes also exhibit reduced expression in an antherdefective mutant, providing a possible link to developmental regulation. The putative AP2-interacting proteins participate in many functions in development and stress responses,including photomorphogenesis, flower development, pathogenesis, drought and cold responses, abscisic acid and auxin signaling. | 11 | Investigation of frost heave prevention using permeable subgrade structure显示文摘This paper set up a series of comprehensive targets based on the concept of 'anti-freeze filler', which include reasonable water retention rate, frost heave characteristics, and compaction characteristics of filling material. Then, a type of permeable graded gravel is proposed, suitable for high-speed railway subgrade. A series of in-door water retention, permeability, and frost heave tests were performed under different graded conditions. Water retention, permeability, and frost heave characteristic of different graded filling materials can be determined, in order to define the gradation range of permeable graded gravel. Relying on the frost-heave monitoring record of high speed railway in Northeast China, a series of experimental studies were performed, which included on-site filler production, compaction test, and the anti-frost effect test, in order to improve the production and compaction techniques of permeable graded gravel. | 12 | Nuclear factor κB represses the expression of latent membrane protein 1 in Epstein-Barr virus transformed cells显示文摘AIM: To investigate the role of nuclear factor κB(NF-κB) in the regulation of Epstein-Barr virus(EBV) latent membrane protein 1(LMP1) in EBV transformed cells. METHODS: LMP1 expression was examined in EBV transformed human B lymphocytes with modulation of NF-κB activity. RESULTS: EBV infection is associated with several human cancers. EBV LMP1 is required for efficient transformation of adult primary B cells in vitro, and is expressed in several pathogenic stages of EBVassociated cancers. Regulation of EBV LMP1 involves both viral and cellular factors. LMP1 activates NF-κB signaling pathway that is a part of the EBV transformation program. However, the relation between NF-κB and LMP1 expression is not well established yet. In this report, we found that blocking the NF-κB activity by Inhibitor of κB stimulated LMP1 expression, while the overexpression of NF-κB repressed LMP1 expression in EBV-transformed IB4 cells. In addition, LMP1 repressed its own promoter activities in reporter assays, and the repression was associated with the activation of NF-κB. Moreover, NF-κB alone is sufficient to repress LMP1 promoter activities.
