| 3 | Sedimentary and geochemical evidence of Eocene climate change in the Xining Basin, northeastern Tibetan Plateau显示文摘The northeastern Tibetan Plateau began to grow during the Eocene and it is important to understand the climatic history of Asia during this period of so-called ‘doubthouse' conditions. However, despite major advances in the last few decades,the evolutionary history and possible mechanisms of Eocene climate change in the northeastern Tibetan Plateau remain unclear.The Xining Basin in the northeastern Tibetan Plateau contains a continuous sequence of Early to Late Eocene non-marine sediments which provides the opportunity to resolve long-term climate changes during this period. In this study, we report the results of analyses of lithofacies, sediment color and geochemistry of bulk samples collected from the Xijigou section of the Xining Basin. An abrupt lithofacies change between the Early(~52–40 Ma) and Late Eocene(~40–34 Ma) indicates a change in the depositional environment from a shallow lake to a playa lake in response to a significant climatic shift. During ~52–40 Ma,higher values of sediment redness(a*), redness/lightness(a*/L*) and higher modified Chemical Index of Weathering(CIW′)indicate a relatively warm and humid climate, while from ~40–34 Ma the lower values of a*, a*/L*and lower CIW′ imply subhumid to semi-arid climatic conditions. The paleoclimatic records indicate a long-term(~52–34 Ma) trend of decreasing chemical weathering, consistent with global climate change. An abrupt sharp excursion of the proxy records during ~42–40 Ma suggests a relatively brief warm interval, corresponding to the Middle Eocene Climatic Optimum(MECO). | 5 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 6 | Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19. | Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 4 |
| 7 | MKP-1 reduces Aβ generation and alleviates cognitive impairments in Alzheimer’s disease models显示文摘Mitogen-activated protein kinase(MAPK)phosphatase 1(MKP-1)is an essential negative regulator of MAPKs by dephosphorylating MAPKs at both tyrosine and threonine residues.Dysregulation of the MAPK signaling pathway has been associated with Alzheimer’s disease(AD).However,the role of MKP-1 in AD pathogenesis remains elusive.Here,we report that MKP-1 levels were decreased in the brain tissues of patients with AD and an AD mouse model.The reduction in MKP-1 gene expression appeared to be a result of transcriptional inhibition via transcription factor specificity protein 1(Sp1)cis-acting binding elements in the MKP-1 gene promoter.Amyloid-β(Aβ)-induced Sp1 activation decreased MKP-1 expression.However,upregulation of MKP-1 inhibited the expression of both Aβprecursor protein(APP)andβ-site APP-cleaving enzyme 1 by inactivating the extracellular signal-regulated kinase 1/2(ERK)/MAPK signaling pathway.Furthermore,upregulation of MKP-1 reduced Aβproduction and plaque formation and improved hippocampal long-term potentiation(LTP)and cognitive deficits in APP/PS1 transgenic mice.Our results demonstrate that MKP-1 impairment facilitates the pathogenesis of AD,whereas upregulation of MKP-1 plays a neuroprotective role to reduce Alzheimer-related phenotypes.Thus,this study suggests that MKP-1 is a novel molecule for AD treatment. | Yehong Du Yexiang Du Yun Zhang Zhilin Huang Min Fu Junjie Li Yayan Pang Peng Lei Yu Tian Wang Weihong Song Guiqiong He Zhifang Dong | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 2 |
| 8 | Epoxy microcapsules for high-performance self-healing materials using a novel method via integrating electrospraying and interfacial polymerization显示文摘The high-efficiency fabrication of high-quality microcapsules containing epoxy is crucial to the further development of the potential practical self-healing epoxy systems based on microencapsulated two-part epoxy-amine chemistry.Herein,a novel microencapsulation technique based on non-equilibrium droplets via integrating electrospraying and interfacial polymerization(ES-IP)was established to efficiently microencapsulate epoxy monomers.The ES-IP technique,consisting of three continuous steps,i.e.electrospraying to massively generate droplets,enwrapping every single droplet through instant interfacial polymerization,and thickening shell at an elevated temperature,has great flexibility to regulate the microencapsulation process and the microcapsule quality.The fabricated core-shell structured epoxy microcapsules(Ep-MCs)were comprehensively characterized for their properties,showing that they have high cleanness with rare impurities,controllable and tunable size,good thermal stability and tightness,and high effective core fraction.The high-quality Ep-MCs were adopted to formulate a self-healing epoxy based on the microencapsulated epoxy-amine chemistry.The highest healing efficiency,in terms of the recovered mode I fracture toughness,of 110±17%was achieved after being healed at room temperature(~25℃)for 48 h.While the developed ES-IP technique facilitates the microencapsulation technique based on non-equilibrium droplets,the fabricated high-quality Ep-MCs greatly promote the further developments of the practical self-healing materials. | He Zhang Kaibin Xiao Zhilin Lin Shengyu Shi | 2022 | Journal of Materials Science & Technology2022,,17: | 1 |

| 14 | Reconstructing early Eocene(~55 Ma) paleogeographic boundary conditions for use in paleoclimate modelling显示文摘The early Eocene is a period with very high atmospheric CO2 levels, which receives many interests from climate modelling aspects. To simulate the early Eocene paleoclimate, a realistic reconstruction for land-sea distribution, paleotopography and paleobathymetry is the fundamental step. Here, we present global paleogeographic reconstructions for the early Eocene(~55 Ma), based on integrated paleogeographic data set, the plate-tectonic reconstruction software(GPlates) and geographic information system software(ArcGIS). | 16 | Multilayer dielectric grating pillar-removal damage induced by a picosecond laser显示文摘Multilayer dielectric gratings typically remove multiple-grating pillars after picosecond laser irradiation;however,the dynamic formation process of the removal is still unclear.In this study,the damage morphologies of multilayer dielectric gratings induced by an 8.6-ps laser pulse were closely examined.The damage included the removal of a single grating pillar and consecutive adjacent grating pillars and did not involve the destruction of the internal high-reflection mirror structure.Comparative analysis of the two damage morphological characteristics indicated the removal of adjacent pillars was related to an impact process caused by the eruption of localized materials from the left-hand pillar,exerting impact pressure on its adjacent pillars and eventually resulting in multiple pillar removal.A finite-element strain model was used to calculate the stress distribution of the grating after impact.According to the electric field distribution,the eruptive pressure of the dielectric materials after ionization was also simulated.The results suggest that the eruptive pressure resulted in a stress concentration at the root of the adjacent pillar that was sufficient to cause damage,corresponding to the experimental removal of the adjacent pillar from the root.This study provides further understanding of the laser-induced damage behavior of grating pillars and some insights into reducing the undesirable damage process for practical applications. | Kun Shuai Xiaofeng Liu Yuanan Zhao Keqiang Qiu Dawei Li He Gong Jian Sun Li Zhou Youen Jiang Yaping Dai Jianda Shao Zhilin Xia | 2022 | High Power Laser Science and Engineering2022,10,6: | 0 |
