1Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
2Magnetization of microorganism cells by sol-gel method显示文摘Microorganism cells could be used as templates during fabrication of magnetic or conductive microstructures in different standard shapes. In this paper, feasibility of magnetizing microorganism cells by sol-gel method, which is to coat cells of Spirulina (a type of natural micro-helical microorganism) with the ferrite (a kind of magnetic material), was discussed and investigated. Then the cell form, components and the phase structure were observed and analyzed using various tools including optical microscopy, scanning electron microscopy (SEM), energy dispersive X-ray detector (EDX), transmission electron microscopy (TEM), and X-ray diffraction analysis (XRD). Results showed that spirulina cells could be coated with ferrite after the sol-gel process, with the shape of natural helixes well kept, that the components of different sampling points on the surface layer were consistent and the thickness of layer was uniform, and that the type of the surface ferrite layer formed was cubic Fe3O4. It was also observed that there were nano-particles yielded in the cells and certain deposit on the walls between cells. 3Radial propagation of magnetospheric substorm-injected energetic electrons observed using a BD-IES instrument and Van Allen Probes显示文摘In cases where substorm injections can be observed simultaneously by multiple spacecraft,they can help elucidate the potential mechanisms of particle transport and energization,of great importance to understanding and modeling the magnetosphere.In this paper,using data returned from the BeiDa-IES(BD-IES) instrument onboard a satellite in an inclined(55°) geosynchronous orbit(IGSO),in combination with two geo-transfer orbiting(GTO) satellite Van Allen Probes(A and B),we analyze a substorm injection event that occurred on the 16 th of October 2015.During this substorm injection,the IGSO onboard BD-IES was outbound,while both Van Allen Probe satellites(A and B) were inbound,a configuration of multiple trajectories that provides a unique opportunity to simultaneously investigate both the inward and outward radial propagation of substorm injection.Indicated by AE/AL indices,this substorm was closely related to an IMF/solar wind discontinuity that showed a sharp change in IMF Bz direction to the north.The innermost signature of this substorm injection was detected by Van Allen Probes A and B at L-3.7,while the outermost signature was observed by the onboard BD-IES instrument at L~\0.These data indicate that the substorm had a global,rather than just local,effect.Finally,we suggest that electric fields carried by fast-mode compressional waves around the substorm injection are the most likely candidate mechanism for the electron injection signatures observed in the inner- and outermost inner magnetosphere.ZONG QiuGang HAO YiXin ZOU Hong FU SuiYan ZHOU XuZhi REN Jie WANG LingHua YUAN ChongJing LIU ZhiYang JIA XiangHong QUAN Lin LIU Ying WANG YongFu 2016Science China Earth Sciences2016,59,7:8
4Simultaneous extraction of DNA and RNA from Escherichia coli BL 21 based on silica-coated magnetic nanoparticles显示文摘The extraction of nucleic acid is recognized as one of the most essential steps in molecular biology for initiating other downstream applications such as sequencing, amplification, hybridization, and cloning. Many commercial kits and methods are currently available that allow the extraction of only one type of nucleic acids-DNA or RNA. However, in parallel clinical detection of several diseases, a method for simultaneous extraction of both DNA and RNA from the same source is needed in such cases. In this study, a method for simultaneous extraction of DNA and RNA from bacteria based on magnetic nanoparticles(MNPs) was described. Lysis buffers were prepared to help the nucleic acid released and adsorbed to MNPs. Then, two washing buffers were used to remove the contamination of proteins and carbohydrates. The nucleic acids were finally eluted by Deoxyribonuclease(DNase) and Ribonucleases(RNase) free water. Different factors which might affect the purification of the nucleic acid were investigated, and the quantity and quality parameters of the nucleic acid were also recorded. The DNA and RNA extracted from bacteria were then respectively subjected to polymerase chain reaction(PCR) and reverse transcription PCR(RT-PCR) to further confirm its quality. 5Cosmogenic ^(10)Be and ^(26)Al Chronology of the Last Glaciation of the Palaeo-Daocheng Ice Cap,Southeastern Qinghai-Tibetan Plateau显示文摘The glacial landforms of the Qinghai-Tibetan Plateau(QTP) provide a unique opportunity to research hemispheric and global environmental changes. In this study,we focus on the glacial history of the palaeo-Daocheng Ice Cap(p-DIC) in the southeastern QTP during the last glacial cycle. Based on field investigations,morphostratigraphy,and surface exposure dating of roche moutonnée,polished surface and moraine debris through the terrestrial cosmogenic nuclides(TCN) 10 Be and 26 Al. We identify glacial deposits of the last deglaciation,with minimum ages of 14.9±1.3–18.7±1.7 ka,the Last Glacial Maximum(LGM) of 24.7±2.2 ka,and the early part of the last glacial period(marine oxygen isotope stage(MIS) 3) of 37.1±3.4–45.2±3.9 ka. Our results show that in this region,the extent of the glacial advance during MIS 3 was larger than that during the traditional LGM(MIS 2). These ages are consistent with prior chronologies,and the 10 Be age is consistent with the 26 Al age for the same sample. 8Testing and evaluation for fatigue crack propagation of Ti-6Al-4V/ELI and 7050-T7452 alloys at high temperatures显示文摘This paper seeks to evaluate crack propagation properties and residual lives of metallic alloys subjected to fatigue loading at room and high temperatures. Fatigue crack growth tests were performed on Ti-6Al-4V/ELI and 7050-T7452 subjected to constant-amplitude and actual randomspectra loading at room temperature of about 25 ℃ and at high temperatures of 250 ℃ and 150 ℃ to determine their crack growth properties and residual lives. The damage mode and mechanisms at high temperature were compared with those at room temperature on the basis of the results of fractographic analysis. 9High-cycle fatigue behavior of Co-based superalloy 9CrCo at elevated temperatures显示文摘A modified model is developed to characterize and evaluate high-cycle fatigue behavior of Co-based superalloy 9CrCo at elevated temperatures by considering the stress ratio effect. The model is informed by the relationship surface between maximum nominal stress, stress ratio and fatigue life. New formulae are derived to deal with the test data for estimating the parameters of the proposed model. Fatigue tests are performed on Co-based superalloy 9Cr Co subjected to constant amplitude loading at four stress ratios of 1, 0.3, 0.5 and 0.9 in three environments of room temperature(i.e., about 25 °C) and elevated temperatures of 530 °C and 620 °C, and the interaction mechanisms between the elevated temperature and stress ratio are deduced and compared with each other from fractographic studies. Finally, the model is applied to experimental data, demonstrating the practical and effective use of the proposed model. 11Reduced graphene oxide@carbon sphere based metacomposites for temperature-insensitive and efficient microwave absorption显示文摘With the increasing advance of fifth generation(5G)network and the gradual expansion of digital devices,harsh working environment for electronic devices has spawned higher requirements for microwave absorbing materials(MAMs).Since both the electromagnetic response and energy conversion character vary with temperature,to achieve temperature insensitive microwave absorption behaviour in wide temperature range has become extremely challenging.In this work,structured metacomposites containing sub-wavelength reduced graphene oxide(RGO)@carbon spheres were fabricated,and the microwave absorption was further improved through structural and composition design of the RGO@carbon units.Due to the unique anti-reflection effect on microwave of the metacomposites,the temperature-insensitive electromagnetic performance at elevated temperature could be exhibited.Moreover,both the dielectric relaxation behaviour and microwave absorption proformance of the system could be further increased.As a result,the effective absorption bandwidth(reflection loss(RL)<−10 dB)of the metacomposites with only 3.0 wt.%filler content could cover the entire X-band(8.2–12.4 GHz)frequency ranging from 298 to 473K.The metacomposite proposed in this work provides a“de-correlating”strategy to break the linkage between microwave absorption behaviour and temperature,which offers an interesting plateau for fabricating efficient high-temperature microwave absorption structures with tunable and designable advantages.Zhiyang Jiang Haoxu Si Yi Li Dan Li Huihui Chen Chunhong Gong Jingwei Zhang 2022Nano Research2022,15,9:4
12Knockout of butyrophilin subfamily 1 member A1(BTN1A1) alters lipid droplet formation and phospholipid composition in bovine mammary epithelial cells显示文摘Background: Milk lipids originate from cytoplasmic lipid droplets(LD) that are synthesized and secreted from mammary epithelial cells by a unique membrane-envelopment process. Butyrophilin 1 A1(BTN1 A1) is one of the membrane proteins that surrounds LD, but its role in bovine mammary lipid droplet synthesis and secretion is not well known.Methods: The objective was to knockout BTN1 A1 in bovine mammary epithelial cells(BMEC) via the CRISPR/Cas9 system and evaluate LD formation, abundance of lipogenic enzymes, and content of cell membrane phospholipid(PL) species. Average LD diameter was determined via Oil Red O staining, and profiling of cell membrane phospholipid species via liquid chromatography-tandem mass spectrometry(LC-MS/MS).Results: Lentivirus-mediated infection of the Cas9/sg RNA expression vector into BMEC resulted in production of a homozygous clone BTN1 A1^((-/-)). The LD size and content decreased following BTN1 A1 gene knockout. The m RNA abundance of fatty acid synthase(FASN) and peroxisome proliferator-activated receptor-gamma(PPARG) was downregulated in the BTN1 A1^((-/-))clone. Subcellular analyses indicated that BTN1 A1 and LD were co-localized in the cytoplasm. 13In vivo volumetric monitoring of revascularization of traumatized skin using extended depth-of-field photoacoustic microscopy显示文摘Faster and better wound healing is a critical medical issue.Because the repair process of wounds is closely related to revascularization,accurate early assessment and postoperative monitoring are very important for establishing an optimal treatment plan.Herein,we present an extended depth-of-field photoacoustic microscopy system(E-DOF-PAM)that can achieve a constant spatial resolution and relatively uniform excitation efficiency over a long axial range.The superior performance of the system was verified by phantom and in vivo experiments.Furthermore,the system was applied to the imaging of normal and trauma sites of volunteers,and the experimental results accurately revealed the morphological differences between the normal and traumatized skin of the epidermis and dermis.These results demonstrated that the E-DOF-PAM is a powerful tool for observing and understanding the pathophysiology of cutaneous wound healing.Zhongwen CHENG Haigang MA Zhiyang WANG Sihua YANG 2020Frontiers of Optoelectronics2020,13,4:3
14Nanoscale zero-valent iron supported on biochar for the highly efficient removal of nitrobenzene显示文摘The application of nanoscale zero-valent iron (nZVI) in the remediation of contaminated groundwater or wastewater is limited due to its lack of stability, easy aggregation and iron leaching. To address this issue, nZVI was distributed on oak sawdust-derived biochar (BC) to obtain the nZVI/BC composite for the highly efficient reduction of nitrobenzene (NB). nZVI, BC and nZVI/BC were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). For nZVI/BC, nZVI particles were uniformly dispersed on BC. nZVI/BC exhibited higher removal efficiency for NB than the simple summation of bare nZVI and BC. The removal mechanism was investigated through the analyses of UV-Visible spectra, mass balance and XPS. NB was quickly adsorbed on the surface of nZVI/BC, and then gradually reduced to aniline (AN), accompanied by the oxidation of nZVI to magnetite. The effects of several reaction parameters, e.g., NB concentration, reaction pH and nZVI/BC aging time, on the removal of NB were also studied. 17Research on magnetic metallization of microorganism cells using electroplating technique显示文摘Spirulina platensis were chosen as templates to produce microscopic helical soft-core magnetic particles by way of depositing ferromagnetic alloy onto their surface using electroplating technique,and the process of electroplating ferromagnetic alloy onto microorganism cells was studied.The morphology and appearance of the coated Spirulina platensis were analyzed with optical microscopy and scanning electron microscopy,respectively,and the ingredients and phase structure of the alloy coating were analyzed with energy dispersive X-ray detector(EDX) and X-ray diffractive analysis(XRD),respectively.The result showed that the particles were successfully coated with uniform metal coating and their initial helical shape was perfectly replicated.The coating was NiFe alloy,and its phase structure was face-centered cubic structure.The magnetic properties of the coated particles were tested with vibrating sample magnetometer(VSM),and the result showed that the particles were ferro-magnetic,which means the magnetic electroplating of the microorganism cells was successfully achieved.The electrochemical reaction mechanism of the magnetic plating process was also analyzed;the result showed that the deposition of NiFe on the microorganism cells was anomalous codeposition,and that Fe2+ ion was preferential deposited when magnetic stirring was applied.CAI Jun ZHANG DeYuan LAN MingMing LIAN ZhiYang 2011Science China(Technological Sciences)2011,54,6:3
18Quantification of the stiffness and strength of cadherin ectodomain binding with different ions显示文摘The stiffness and strength of extracellular(EC) region of cadherin are proposed to be two important mechanical properties both for cadherin as a mechanotransductor and for the formation of cell-cell adhesion. In this study,we quantitatively characterized the stiffness and strength of EC structure when it binds with different types of ions by molecular dynamics simulations. Results show that EC structure exhibits a rod-like shape with high stiffness and strength when it binds with the bivalent ions of calcium or magnesium. However,it switches to a soft and collapsed conformation when it binds with the monovalent ions of sodium or potassium. 19TiN/BN composite with excellent thermal stability for efciency microwave absorption in wide temperature spectrum显示文摘High-efficiency and temperature-insensitive microwave absorbing materials(MAMs) are ideal for hightemperature electromagnetic attenuation.Herein,the titanium nitride/boron nitride(TiN/BN) composite with TiN as the loss unit and BN as the impedance matching unit has been constructed by the in-situ synthesis method.The insulating BN not only effectively regulates and optimizes the conductivity and impedance matching of the material but also imparts steady cyclic microwave absorption properties.The steady dielectric loss ensures that TiN/BN composite has robust high-temperature absorption properties at X-band,with a minimum reflection loss(RL_(min)) of-16.74 dB at 873 K and an effective absorption bandwidth(EAB) of 3.26 GHz in the temperature range of 293-873 K.Compared with the TiN/SiO2absorbers with a single system,the TiN/BN/SiO2composite keeps reliable high temperature absorbing properties at873 K.This work confirms that wave-transparent materials for dielectric property modulation can resolve poor temperature stability of effective absorption and non-reusability for high-temperature absorbing materials,providing inspirations for designing efficiency high-temperature microwave absorbers.Yupeng Shi Dan Li Haoxu Si Zhiyang Jiang Mengyuan Li Chunhong Gong 2022Journal of Materials Science & Technology2022,,35:3
20Hypoxia response element-directed expression of bFGF in dental pulp stem cells improve the hypoxic environment by targeting pericytes in SCI rats显示文摘Cell-based transplantation strategies possess great potential for spinal cord injury(SCI)repair.Basic fibroblast growth factor(bFGF)has been reported to have multiple neuro-promoting effects on developing and adult nervous system of mammals and considered a promising therapy for nerve injury following SCI.Human dental pulp stem cells(DPSCs)are abundant stem cells with low immune rejection,which can be considered for cell replacement therapy.The purpose of this study was to investigate the roles of DPSCs which express bFGF under the regulation of five hypoxia-responsive elements(5HRE)using an adeno-associated virus(AAV-5HRE-bFGF-DPSCs)in SCI repairing model.In this study,DPSCs were revealed to differentiate into CD13^(+)pericytes and up-regulate N-cadherin expression to promote the re-attachment of CD13^(+)pericytes to vascular endothelial cells.The re-attachment of CD13^(+)pericytes to vascular endothelial cells subsequently increased the flow rate of blood in microvessels via the contraction of protuberance.As a result,increased numbers of red blood cells carried more oxygen to the damaged area and the local hypoxia microenvironment in SCI was improved.Thus,this study represents a step forward towards the potential use of AAV-5HRE-bFGF-DPSCs in SCI treatment in clinic.Sipin Zhu Yibo Ying Yan He Xingxing Zhong Jiahui Ye Zhiyang Huang Min Chen Qiuji Wu Yifan Zhang Ziyue Xiang Yurong Tu Weiyang Ying Jian Xiao Xiaokun Li Qingsong Ye Zhouguang Wang 2021Bioactive Materials2021,6,8:2
