1Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
2Radial propagation of magnetospheric substorm-injected energetic electrons observed using a BD-IES instrument and Van Allen Probes显示文摘In cases where substorm injections can be observed simultaneously by multiple spacecraft,they can help elucidate the potential mechanisms of particle transport and energization,of great importance to understanding and modeling the magnetosphere.In this paper,using data returned from the BeiDa-IES(BD-IES) instrument onboard a satellite in an inclined(55°) geosynchronous orbit(IGSO),in combination with two geo-transfer orbiting(GTO) satellite Van Allen Probes(A and B),we analyze a substorm injection event that occurred on the 16 th of October 2015.During this substorm injection,the IGSO onboard BD-IES was outbound,while both Van Allen Probe satellites(A and B) were inbound,a configuration of multiple trajectories that provides a unique opportunity to simultaneously investigate both the inward and outward radial propagation of substorm injection.Indicated by AE/AL indices,this substorm was closely related to an IMF/solar wind discontinuity that showed a sharp change in IMF Bz direction to the north.The innermost signature of this substorm injection was detected by Van Allen Probes A and B at L-3.7,while the outermost signature was observed by the onboard BD-IES instrument at L~\0.These data indicate that the substorm had a global,rather than just local,effect.Finally,we suggest that electric fields carried by fast-mode compressional waves around the substorm injection are the most likely candidate mechanism for the electron injection signatures observed in the inner- and outermost inner magnetosphere.ZONG QiuGang HAO YiXin ZOU Hong FU SuiYan ZHOU XuZhi REN Jie WANG LingHua YUAN ChongJing LIU ZhiYang JIA XiangHong QUAN Lin LIU Ying WANG YongFu 2016Science China Earth Sciences2016,59,7:8
3Simultaneous extraction of DNA and RNA from Escherichia coli BL 21 based on silica-coated magnetic nanoparticles显示文摘The extraction of nucleic acid is recognized as one of the most essential steps in molecular biology for initiating other downstream applications such as sequencing, amplification, hybridization, and cloning. Many commercial kits and methods are currently available that allow the extraction of only one type of nucleic acids-DNA or RNA. However, in parallel clinical detection of several diseases, a method for simultaneous extraction of both DNA and RNA from the same source is needed in such cases. In this study, a method for simultaneous extraction of DNA and RNA from bacteria based on magnetic nanoparticles(MNPs) was described. Lysis buffers were prepared to help the nucleic acid released and adsorbed to MNPs. Then, two washing buffers were used to remove the contamination of proteins and carbohydrates. The nucleic acids were finally eluted by Deoxyribonuclease(DNase) and Ribonucleases(RNase) free water. Different factors which might affect the purification of the nucleic acid were investigated, and the quantity and quality parameters of the nucleic acid were also recorded. The DNA and RNA extracted from bacteria were then respectively subjected to polymerase chain reaction(PCR) and reverse transcription PCR(RT-PCR) to further confirm its quality. 7C.elegans-based screen identifies lysosome-damaging alkaloids that induce STAT3-dependent lysosomal cell death显示文摘Lysosomes are degradation and signaling centers within the cell,and their dysfunction impairs a wide variety of cellular processes.To understand the cellular effect of lysosome damage,we screened natural smallmolecule compounds that induce iysosomal abnormality using Caenorhabditis elegans (C.elegans)as a model system.A group of vobasinyl-ibogan type bisindole alkaloids (ervachinines A-D)were identified that caused lysosome enlargement in C.elegans macrophage-like cells.Intriguingly,these compounds triggered cell death in the germ line independently of the canonical apoptosis pathway.In mammalian cells, ervachinines A-D induced lysosomal enlargement and damage,leading to leakage of cathepsin proteases, inhibition of autophagosome degradation and necrotic cell death.Further analysis revealed that this ervachinine-induced lysosome damage and lysosomal cell death depended on STAT3 signaling,but not RIP1 or RIP3 signaling.These findings suggest that lysosomedamaging compounds are promising reagents for dissecting signaling mechanisms underlying lysosome homeostasis and lysosome-related human disorders.Yang Li Yu Zhang Qiwen Gan Meng Xu Xiao Ding Guihua Tang Jingjing Liang Kai Liu Xuezhao Liu Xin Wang Lingli Guo Zhiyang Gao Xiaojiang Hao Chonglin Yang 2018Protein & Cell2018,9,12:2
8Achieving high conversion of syngas to aromatics显示文摘Realizing high CO conversion and high aromatics selectivity simultaneously in syngas-to-aromatics(STA)reaction is still challenging.Herein,we report a 57.5%CO conversion along with 74%aromatics selectivity over a composite catalyst consisting of Fe/ZnCr_(2)O_(4)(Fe modified ZnCr_(2)O_(4)spinel)oxide and H-ZSM-5 zeolite.Impregnation of only 3 wt%of Fe onto ZnCr_(2)O_(4)can remarkably increase CO conversion without sacrificing the aromatics selectivity.Oxygen vacancy concentration is improved after impregnating Fe.The highly dispersed iron carbide species is formed during the reaction over Fe/ZnCr_(2)O_(4)spinel oxide.The synergistic effect of oxygen vacancy and iron carbide results in a rapid formation of abundant oxygenated intermediate species,which can be continuously transformed to aromatics in H-ZSM-5.This study provides a new insight into the design of highly efficient catalyst for syngas conversion.Yi Fu Youming Ni Zhiyang Chen Wenliang Zhu Zhongmin Liu 2022Journal of Energy Chemistry2022,31,3:2

10Catalytic activity of Cu/ZnO catalysts mediated by MgO promoter in hydrogenation of methyl acetate to ethanol显示文摘Hydrogenation of methyl acetate is a key step in ethanol synthesis from dimethyl ether carbonylation and Cu-based catalysts are widely studied.We report here that the hydrogenation activity of Cu/ZnO catalysts can be enhanced by the addition of MgO promoter.The evolution of crystal phases during coprecipitation and the physicochemical properties of calcined and reduced catalysts by X-ray diffraction(XRD),thermogravimetric(TG)-mass spectrometry(MS),Brunauer-Emmett-Teller(BET),transmission electron microscopy(TEM),N_(2)O titration,in situ CO-Fourier transform infrared spectroscopy(FTIR)and H_(2)-temperature programmed reduction(H_(2)-TPR)reveal that the promoter effect likely lies in the presence of Mg^(2+).A proper amount of Mg^(2+)mediates the precipitation process of Cu and Zn,leading to preferable formation of aurichalcite(Cu_(x)Zn_(1-x))5(CO_(3))_(2)(OH)_(6) crystal phase and a small amount of basic carbonates such as hydrozincite Zn_(5)(CO_(3))_(2)(OH)_(6) and malachite Cu_(2) CO_(3)(OH)_(2).The presence of aurichalcite strengthens the interaction between Cu and Zn species,and thus enhances the dispersity of CuO species and helps generation of Cu^(+)species on reduced catalysts.Furthermore,the performance of Cu/ZnO catalysts exhibits an optimal dependence on the Mg loading,i.e.,17.5%.However,too much Mg^(2+)in the precipitation liquid prohibits formation of aurichalcite but enhances formation of basic nitrates,leading to a dramatically reduced hydrogenation activity.These findings may find applications for optimization of other Cu-based catalysts in a wider range of hydrogenation reactions.Fang Zhang Zhiyang Chen Xudong Fang Hongchao Liu Yong Liu Wenliang Zhu 2021Journal of Energy Chemistry2021,30,10:2
11Bioinformatic analysis of chromatin organization and biased expression of duplicated genes between two poplars with a common whole-genome duplication显示文摘The nonrandom three-dimensional organization of chromatin plays an important role in the regulation of gene expression.However,it remains unclear whether this organization is conserved and whether it is involved in regulating gene expression during speciation after whole-genome duplication(WGD)in plants.In this study,high-resolution interaction maps were generated using high-throughput chromatin conformation capture(Hi-C)techniques for two poplar species,Populus euphratica and Populus alba var.pyramidalis,which diverged~14 Mya after a common WGD.We examined the similarities and differences in the hierarchical chromatin organization between the two species,including A/B compartment regions and topologically associating domains(TADs),as well as in their DNA methylation and gene expression patterns.We found that chromatin status was strongly associated with epigenetic modifications and gene transcriptional activity,yet the conservation of hierarchical chromatin organization across the two species was low.The divergence of gene expression between WGD-derived paralogs was associated with the strength of chromatin interactions,and colocalized paralogs exhibited strong similarities in epigenetic modifications and expression levels.Thus,the spatial localization of duplicated genes is highly correlated with biased expression during the diploidization process.This study provides new insights into the evolution of chromatin organization and transcriptional regulation during the speciation process of poplars after WGD.Le Zhang Jingtian Zhao Hao Bi Xiangyu Yang Zhiyang Zhang Yutao Su Zhenghao Li Lei Zhang Brian J.Sanderson Jianquan Liu Tao Ma 2021Horticulture Research2021,8,1:2

15HTR2A agonists play a therapeutic role by restricting ILC2 activation in papain-induced lung inflammation显示文摘Group 2 innate lymphoid cells(ILC2s)are a category of heterogeneous cells that produce the cytokines IL-5 and IL-13,which mediate the type 2 immune response.However,specific drug targets on lung ILC2s have rarely been reported.Previous studies have shown that type 2 cytokines,such as IL-5 and IL-13,are related to depression.Here,we demonstrated the negative correlation between the depression-associated monoamine neurotransmitter serotonin and secretion of the cytokines IL-5 and IL-13 by ILC2s in individuals with depression.Interestingly,serotonin ameliorates papain-induced lung inflammation by suppressing ILC2 activation.Our data showed that the serotonin receptor HTR2A was highly expressed on ILC2s from mouse lungs and human PBMCs.Furthermore,an HTR2A selective agonist(DOI)impaired ILC2 activation and alleviated the type 2 immune response in vivo and in vitro.Mice with ILC2-specific depletion of HTR2A(Il5^(cre/+)·Htr2a^(flox/flox)mice)abolished the DOI-mediated inhibition of ILC2s in a papain-induced mouse model of inflammation.In conclusion,serotonin and DOI could restrict the type 2 lung immune response,indicating a potential treatment strategy for type 2 lung inflammation by targeting HTR2A on ST2+ILC2s.Zhishuo Wang Chenghua Yan Qizhen Du Yuying Huang Xuezhen Li Dan Zeng Ruizhi Mao Rama Krishna Gurram Shipeng Cheng Wangpeng Gu Lin Zhu Weiguo Fan Liyan Ma Zhiyang Ling Ju Qiu Dangsheng Li Enmei Liu Yaguang Zhang Yiru Fang Jinfang Zhu Bing Sun 2023Cellular & Molecular Immunology2023,20,4:1

18Energetic electron detection packages on board Chinese navigation satellites in MEO显示文摘Energetic electron measurements and spacecraft charging are of great significance for theoretical research in space physics and space weather applications.In this paper,the energetic electron detection package(EEDP)deployed on three Chinese navigation satellites in medium Earth orbit(MEO)is reviewed.The instrument was developed by the space science payload team led by Peking University.The EEDP includes a pinhole medium-energy electron spectrometer(MES),a high-energy electron detector(HED)based onΔE-E telescope technology,and a deep dielectric charging monitor(DDCM).The MES measures the energy spectra of 50−600 keV electrons from nine directions with a 180°×30°field of view(FOV).The HED measures the energy spectrum of 0.5−3.0 MeV electrons from one direction with a 30°cone-angle FOV.The ground test and calibration results indicate that these three sensors exhibit excellent performance.Preliminary observations show that the electron spectra measured by the MES and HED are in good agreement with the results from the magnetic electron-ion spectrometer(MagEIS)of the Van Allen Probes spacecraft,with an average relative deviation of 27.3%for the energy spectra.The charging currents and voltages measured by the DDCM during storms are consistent with the highenergy electron observations of the HED,demonstrating the effectiveness of the DDCM.The observations of the EEDP on board the three MEO satellites can provide important support for theoretical research on the radiation belts and the applications related to space weather.YuGuang Ye Hong Zou Qiu-Gang Zong HongFei Chen JiQing Zou WeiHong Shi XiangQian Yu WeiYing Zhong YongFu Wang YiXin Hao ZhiYang Liu XiangHong Jia Bo Wang XiaoPing Yang XiaoYun Hao 2021Earth and Planetary Physics2021,5,2:1
19Printable microlaser arrays with programmable modes for information encryption显示文摘Lasing emissions with multiple and tunable modes are promising in coding field as a novel cryptographic primitive.With the advantages of simple fabrication,full-color and high-quality-factor whispering gallery mode lasing inside a circular cross-section,polymer microfibers are attractive for photonic devices.However,polymer lasing microfibers for information encryption have never been reported.Herein,we propose a design of printable lasing microfiber encryption chip by in-situ tuning the effective refractive index of the microresonator arrays via a facile approach.Through inkjet printing high-refractive-index nanoparticles on the designated position of lasing microfiber arrays,the effective refractive index of the microcavities is regulated,and the ratio of wavenumber spacing between transverse electric and transverse magnetic mode to the free spectral range can be modulated,particularly with neglectable influence by the size factor.Thus,the programmable region selective encoding process can be conducted simply by a printing program within several minutes.Besides,the encoded microfiber arrays are encapsulated into polydimethylsiloxane to reduce the scattering loss and environmental interference,and a printable encryption chip is realized.This work is expected to provide a platform for the printable encrypted devices.Jun Ruan Dan Guo Kun Ge Zhiyang Xu Fangyuan Liu Tianrui Zhai 2023Nano Research2023,16,7:1
