| 1 | GRAIN INCOMPLETE FILLING 2 regulates grain filling and starch synthesis during rice caryopsis development显示文摘Rice grain filling determines grain weight,final yield and grain quality.Here,a rice defective grain filling mutant,gif2,was identified.Grains of gif2 showed a slower filling rate and a significant lower final grain weight and yield compared to wild-type.The starch content in gif2 was noticeably decreased and its physicochemical properties were also altered.Moreover,gif2 endosperm cells showed obvious defects in compound granule formation.Positional cloning identified GIF2 to encode an ADP-glucose pyrophosphorylase(AGP) large subunit,AGPL2;consequently,AGP enzyme activity in gif2 endosperms was remarkably decreased.GIF2 is mainly expressed in developing grains and the coded protein localizes in the cytosol.Yeast two hybrid assay showed that GIF2 interacted with AGP small subunits Os AGPS1,Os AGPS2a and Os AGPS2 b.Transcript levels for granule-bound starch synthase,starch synthase,starch branching enzyme and starch debranching enzyme were distinctly elevated in gif2 grains.In addition,the level of nucleotide diversity of the GIF2 locus was extremely low in both cultivated and wild rice.All of these results suggest that GIF2 plays important roles in the regulation of grain filling and starch biosynthesis during caryopsis development,and that it has been preserved during selection throughout domestication of modern rice. | Xiangjin Wei Guiai Jiao Haiyan Lin Zhonghua Sheng Gaoneng Shao Lihong Xie Shaoqing Tang Qingguo Xu Peisong Hu | 2017 | Journal of Integrative Plant Biology2017,59,2: | 20 |
| 2 | Application of autocorrelation method on ionospheric short--term forecasting in China显示文摘Adopting the autocorrelation method in the ionospheric short-term forecasting, we put for- ward a simple and practical forecasting method― the sectional autocorrelation method, that is, for pre- dictions of one hour to four hours ahead the auto- correlation coefficient of RDF with the “iteration” method is selected, for prediction of more than four hours ahead, the autocorrelation coefficient of f0F2 with the “at once” method is used. The prediction precisions have been quantitatively estimated based on the data from Chongqing and Guangzhou Ionos- onde Stations. It is shown that the method is much improved for the predictions of one hour to four hours ahead. For the predictions of more than four hours ahead the prediction error reaches a saturation value, which is still lower than that of the “median” method. | 4 | Aspartic acid in the hippocampus:a biomarker for postoperative cognitive dysfunction显示文摘This study established an aged rat model of cognitive dysfunction using anesthesia with 2% isoflurane and 80% oxygen for 2 hours. Twenty-four hours later, Y-maze test results showed that isoflurane significantly impaired cognitive function in aged rats. Gas chromatography-mass spectrometry results showed that isoflurane also significantly increased the levels of N,N-diethylacetamide, n-ethylacetamide, aspartic acid, malic acid and arabinonic acid in the hippocampus of isoflurane-treated rats. Moreover, aspartic acid, N,N-diethylacetamide, n-ethylacetamide and malic acid concentration was positively correlated with the degree of cognitive dysfunction in the isoflurane-treated rats. It is evident that hippocampal metabolite changes are involved in the formation of cognitive dysfunction after isoflurane anesthesia. To further verify these results, this study cultured hippocampal neurons in vitro, which were then treated with aspartic acid(100 μmol/L). | 7 | Editing of Rice Isoamylase Gene ISA1 Provides Insights into Its Function in Starch Formation显示文摘Isoamylase 1(ISA1) is an isoamylase-type debranching enzyme which plays a predominant role in amylopectin synthesis. In this study, the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated endonuclease 9(CRISPR/Cas9) system was used to edit ISA1 gene in rice via Agrobacterium-mediated transformation. We identified 36 genetic edited lines from 55 T_0 transgenic events, and classified the mutation forms into 7 types. Of those, two homozygous mutants, cr-isa1-1(type 1, with an adenine insertion) and cr-isa1-2(type 3, with a cytosine deletion) were selected for further analysis. Seed sizes of both cr-isa1-1 and cr-isa1-2 were affected, and the two mutants also displayed a shrunken endosperm with significantly lower grain weight. Electron microscopy analysis showed that abnormal starch granules and amyloplasts were found in cr-isa1-1 and cr-isa1-2 endosperm cells. The contents of total starch, amylose and amylopectin in the endosperm of the cr-isa1 mutants were significantly reduced, whereas sugar content and starch gel consistency were observably increased compared to the wild-type. The gelatinization temperature and starch chain length distributions of the cr-isa1 mutants were also altered. Moreover, transcript levels of most starch synthesis-related genes were significantly lower in cr-isa1 mutants. | 10 | Area-oriented coordinated trajectory planning of dual-arm space robot for capturing a tumbling target显示文摘The growing amount of space debris poses a threat to operational spacecraft and the long-term sustainability of activities in outer space. According to the orbital mechanics, an uncontrolled space object will be tumbling, bringing great challenge to capture and remove it. In this paper, a dual-arm coordinated ‘‘Area-Oriented Capture'(AOC) method is proposed to capture a non-cooperative tumbling target. Firstly, the motion equation of the tumbling target is established, based on which, the dynamic properties are analyzed. Then, the ‘‘Area-Oriented Capture'concept is presented to deal with the problem of large pose(position and attitude) deviation and tumbling motion. An area rather than fixed points/devices is taken as the object to be tracked and captured. As long as the manipulators’ end-effectors move to a specified range of the objective areas(not fixed points on the target, but areas), the target satellite will be hugged by the two arms.At last, the proposed method and the traditional method(i.e. fixed-point oriented capture method)are compared and analyzed through simulation. | 12 | Designing near-natural planting patterns for plantation forests in China显示文摘Background: China has a long tradition of managing planted forests. Different species of Populus, Eucalyptus, Larix, Cunninghamia and Pinus are planted to satisfy the local demand for wood products and provide ecological services at the same time. Evidence of the greater resilience of natural forests provides the motivation to develop asymmetric planting patterns, which is the focus of this study. We present a new method for designing plantation patterns that follow those observed in natural ecosystems and to maintain some regularity for operational convenience. Methods: Based on the uniform angle index, we analyzed the spatial structure of six natural forests in different regions of China. The uniform angle index describes the degree of spatial uniformity of the n nearest neighbors of a given reference tree. Accordingly, we identified all possible patterns of a neighborhood group within a regular planting pattern and developed a method to optimize planting point arrangements that contain some randomness as well as a minimum degree of regularity. Results:(1) There are 13 types of structural units in a regular planting, including seven random units, five even units and one cluster unit;(2) Five near-natural arrangements are presented with a minimum proportion of 50% of random units. These five arrangements represent a combination of regularity for operational convenience and asymmetry. Conclusions: The new planting patterns developed in this study are expected to increase the asymmetric competition and resilience of these important ecosystems. | 13 | Synthesis of LiFePO_4 /C as cathode material by a novel optimized hydrothermal method显示文摘Olivine LiFePO 4 , as a cathode material for lithium ion batteries, was prepared by a novel optimized hydrothermal method; afterwards, the product mixed with glucose was two-step (350℃ and 700℃) calcinated under high-purity N 2 atmosphere to obtain the LiFePO 4 /C composite. The study on the hydrothermal preparation method, which focused on the influences of molar ratios, initial pH value, reaction temperature, and duration, was made to promote the resultant performances and to investigate the relations between the performances and the reaction conditions. The resultant samples were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), and electrochemical tests, which include charge-discharge, electrochemical impedance spectroscopy, and cyclic voltammetry. The result shows that the optimal hydrothermal condition is to set the Li:Fe:P molar ratio at 3:1:1 and the reaction temperature at 180℃ for 5 h duration with an initial pH value of 7. | 17 | Derivation of Putative Porcine Embryonic Germ Cells and Analysis of Their Multi-Lineage Differentiation Potential显示文摘Embryonic germ(EG)cells are cultured pluripotent stem cells derived from the primordial germ cells(PGCs)that migrate from the dorsal mesentery of the hindgut to the developing genital ridge.In this study,the morphology of the porcine genital ridge was assessed in embryos harvested on days 22-30 of pregnancy.PGCs from embryos at these stages were cultured to obtain porcine EG cell lines,and EG-like cells were derived from PGCs from embryos harvested on days 24-28 of pregnancy.The EG-like cells expressed Oct4,Sox2,Nanog,SSEA-3,SSEA-4 and alkaline phosphatase(AP).These cells were able to form embryoid bodies(EBs)in suspension culture and differentiate into cells representative of the three germ layers as verified by a-fetoprotein(AFP),a-smooth muscle actin(a-SMA),and Nestin expression.Spontaneous differentiation from the porcine EG-like cells of delayed passage in vitro showed that they could differentiate into epithelial-like cells,mesenchymal-like cells and neuron-like cells.In vitro directed differentiation generated osteocytes,adipocytes and a variety of neural lineage cells,as demonstrated by alizarin red staining,oil red O staining,and immunofluorescence for neuronal classⅢb-tubulin(Tuj1),glial fibrillary protein(GFAP)and galactosylceramidase(GALC),respectively.These results indicate that porcine EG-like cells have the potential for multi-lineage differentiation and are useful for basic porcine stem cell research. | Yimei Cong Jing Ma Ruizhen Sun Jianyu Wang Binghua Xue Jiaqiang Wang Bingteng Xie Juan Wang Kui Hu Zhonghua Liu | 2013 | Journal of Genetics and Genomics2013,40,9: | 4 |
| 18 | Clonogenic colony-forming ability of hepatic stem cells in the spleens of mice显示文摘To confirm the existence of hepatic stem cells (HSCs), fetal liver cells isolated from mice on embryonic day 13 (ED13) were long-term cultured in vitro. Growth of the cells was observed intensively and characteristics were iden-tified by immunocytochemistry. The results showed that some of the cells grew as colonies, in which some cells ex-pressed AFP, CD34 and Albumin. Then the cells were trans-planted intravenously into irradiated syngeneic mice. At day 12 a number of small hyperplasia nodules were seen in the apparently enlarged spleens of recipient mice. | 20 | Low carbon storage of woody debris in a karst forest in southwestern China显示文摘The properties of woody debris(WD) vary across different forests under various soil conditions.Owing to the relatively shallow and low amounts of soils on karst terrains, it is necessary to determine the WD carbon inventory of karst forests. In this study, we recorded WD with a basal diameter for standing snags and the largeend diameter for fallen logs of ≥ 1 cm. The carbon density of WD in a secondary karst mixed evergreen and deciduous broad-leaved forest that had been clear-cut 55 years ago in southwestern China were inventoried in a 2 ha plot. Woody debris carbon density calculated using specific gravity and carbon concentration was 4.07 Mg C ha^-1. Woody debris with diameters ≥ 10 cm(coarse WD) constituted 53.8% of total carbon storage whereas WD < 10 cm in diameters(fine WD) accounted for more pieces of WD(89.9%).Lithocarpus confinis contributed the most WD carbon(26.5%). Intermediate decayed WD was relatively more abundant, but WD with final decay contributed the least to the total pieces of WD(6.7%). The contribution of WD to carbon storage of karst forest was low compared to other forests worldwide. Significant positive correlations were found between WD carbon and biodiversity(R^2= 0.035,p < 0.01) and elevation(R^2= 0.047, p < 0.01) and negative correlations was found in outcrop coverage(R^2= 0.034, p <0.01).
