6Efficient and stable genera-tion of transgenic silkworm, Bombyx mori with the lepi-dopteran derived transposon piggyBac显示文摘The piggyBac transposable element was suc-cessfully used for generation of the transgenic silkworm, Bombyx mori. The EGFP was adopted in this study as a screening marker in transgenic vector under the control of an artificial promoter containing Pax-6 binding sites that can drive eye-specific genes expression in various insect species including B. mori and Drosophila. 111 independent trans-genic lines were obtained among 700 fertile G0 moths. Most of the transgenic lines contained two or more chromosomal insertions of the EGFP marker, which were stably inherited over more than six generations during the time of this pro-ject. PiggyBac-mediated transposition was confirmed by identifying the characteristic TTAA duplication sequence at the insertion sites. 7PCR-based approaches for identification of multi-copy transgene integration sites in mouse genome显示文摘Generation of transgenic mice by DNA microinjection has become one of the most important technologies in studying gene function in vivo. Ran- dom integration of transgene often results in inser- tional mutation which may complicate phenotype analysis of transgenic mice and/or create a good opportunity to study the function of endogenous gene. However,the utilization of these potentially valuable resources is hampered due to lack of efficient ap- proaches for rapid identification of multi-copy trans- gene integration sites. Here we propose two PCR-based approaches to identifying trans- gene/genome junction sequences. The efficiency of these two strategies was tested in 9 independent transgenic mouse lines. Among them,the transgene in 8 mouse lines was clearly localized to certain chromosome regions. 11STAT3 and AKT signaling pathways mediate oncogenic role of NRSF in hepatocellular carcinoma显示文摘Neuron-restrictive silencer factor(NRSF)is a zinc finger protein that acts as a negative transcriptional regulator by recruiting histone deacetylases and other co-factors.It plays a crucial role in nervous system development and is recently reported to be involved in tumorigenesis in a tumor type-dependent manner;however,the role of NRSF in hepatocellular carcinoma(HCC)tumorigenesis remains unclear.Here,we found that NRSF expression was up-regulated in 27 of 49 human HCC tissue samples examined.Additionally,mice with conditional NRSF-knockout in the liver exhibited a higher tolerance against diethylnitrosamine(DEN)-induced acute liver injury and were less sensitive to DEN-induced HCC initiation.Our results showed that silencing NRSF in HepG2 cells using RNAi technology significantly inhibited HepG2 cell proliferation and severely hindered their migration and invasion potentials.Our results demonstrated that NRSF plays a pivotal role in promoting DEN-induced HCC initiation via a mechanism related to the STAT3 and AKT signaling pathways.Thus,NRSF could be a potential therapeutic target for treating human HCC.Ming Ma Yunhe Zhou Ruilin Sun Jiahao Shi Yutong Tan Hua Yang Mengjie Zhang Ruling Shen Leon Xu Zhugang Wang Jian Fei 2020Acta Biochimica et Biophysica Sinica2020,52,10:1
12Rdh13 deficiency weakens carbon tetrachloride-induced liver injury by regulating Spotl4 and Cyp2e1 expression levels显示文摘Mitochondrion-localized retinol dehydrogenase 13 (Rdh13) is a short-chain dehydrogenase/reductase involved in vitamin A metabolism in both humans and mice. We previously generated Rdh13 knockout mice and showed that Rdh13 deficiency causes severe acute retinal light damage. In this study, considering that Rdh13 is highly expressed in mouse liver, we further evaluated the potential effect of Rdh13 on liver injury induced by carbon tetrachloride (CC14). Although Rdh13 deficiency showed no significant effect on liver histology and physiological functions under regular culture, the Rdh13^-/- mice displayed an attenuated response to CCl4-induced liver injury. Their livers also exhibited less histological changes and contained lower levels of liver-related metabolism enzymes compared with the livers of wild-type (WT) mice. Furthermore, the Rdhl3 1 mice had Rdh13 deficiency and thus their liver cells were protected from apoptosis, and the quantity of their proliferative cells became lower than that in WT after CC14 exposure. The ablation of Rdhl3 gene decreased the expression levels of thyroid hormone-inducible nuclear protein 14 (Spot14) and cytochrome P450 (Cyp2el) in the liver, especially after CC14 treatment for 48 h. These data suggested that the alleviated liver damage induced by CC14 in Rdh13^-/- mice was caused by Cyp2el enzymes, which promoted reductive CC14 metabolism by altering the status of thyroxine metabolism. 15Bioluminescence imaging of mouse monocyte chemoattractant protein-1 expression in inflammatory processes显示文摘Monocyte chemoattractant protein-1(MCP-1)plays a crucial role in various inflammatory diseases.To reveal the impact of MCP-1 during diseases and to develop anti-inflammatory agents,we establish a transgenic mouse line.The firefly luciferase gene is incorporated into the mouse genome and driven by the endogenous MCP-1 promoter.A bioluminescence photographing system is applied to monitor luciferase levels in live mice during inflammation,including lipopolysaccharide-induced sepsis,concanavalin A-induced T cell-dependent liver injury,CCl_(4)-induced acute hepatitis,and liver fibrosis.The results demonstrate that the luciferase signal induced in inflammatory processes is correlated with endogenous MCP-1 expression in mice.Furthermore,the expressions of MCP-1 and the luciferase gene are dramatically inhibited by administration of the anti-inflammatory drug dexamethasone in a septicemia model.Our results suggest that the transgenic MCP-1-Luc mouse is a useful model to study MCP-1 expression in inflammation and disease and to evaluate the efficiency of anti-inflammatory drugs in vivo.Fangyang Shao Lei Ci Jiahao Shi Fei Fang Bowen Yan Xijun Liu Xiangyu Yao Mengjie Zhang Hua Yang Zhugang Wang Jian Fei 2022Acta Biochimica et Biophysica Sinica2022,54,10:0
