| 1 | Chromosomal level assembly and population sequencing of the Chinese tree shrew genome显示文摘Chinese tree shrews (Tupaia belangeri chinensis) have become an increasingly important experimental animal in biomedical research due to their close relationship to primates. An accurately sequenced and assembled genome is essential for understanding the genetic features and biology of this animal. In this study, we used long-read single-molecule sequencing and high-throughput chromosome conformation capture (Hi-C) technology to obtain a high-qualitychromosome-scale scaffolding of the Chinese tree shrew genome. The new reference genome (KIZ version 2: TS_2.0) resolved problems in presently available tree shrew genomes and enabled accurate identification of large and complex repeat regions, gene structures, and species-specific genomic structural variants. In addition, by sequencing the genomes of six Chinese tree shrew individuals, we produced a comprehensive map of 12.8 M single nucleotide polymorphisms and confirmed that the major histocompatibility complex (MHC) loci and immunoglobulin gene family exhibited high nucleotide diversity in the tree shrew genome. We updated the tree shrew genome database (TreeshrewDB v2.0: http://www.treeshrewdb.org) to include the genome annotation information and genetic variations. | 4 | A high-quality chromosome-level genome assembly reveals genetics for important traits in eggplant显示文摘Eggplant(Solanum melongena L.)is an economically important vegetable crop in the Solanaceae family,with extensive diversity among landraces and close relatives.Here,we report a high-quality reference genome for the eggplant inbred line HQ-1315(S.melongena-HQ)using a combination of Illumina,Nanopore and 10X genomics sequencing technologies and Hi-C technology for genome assembly.The assembled genome has a total size of~1.17 Gb and 12 chromosomes,with a contig N50 of 5.26 Mb,consisting of 36,582 protein-coding genes.Repetitive sequences comprise 70.09%(811.14 Mb)of the eggplant genome,most of which are long terminal repeat(LTR)retrotransposons(65.80%),followed by long interspersed nuclear elements(LINEs,1.54%)and DNA transposons(0.85%).The S.melongena-HQ eggplant genome carries a total of 563 accession-specific gene families containing 1009 genes.In total,73 expanded gene families(892 genes)and 34 contraction gene families(114 genes)were functionally annotated.Comparative analysis of different eggplant genomes identified three types of variations,including singlenucleotide polymorphisms(SNPs),insertions/deletions(indels)and structural variants(SVs).Asymmetric SV accumulation was found in potential regulatory regions of protein-coding genes among the different eggplant genomes.Furthermore,we performed QTL-seq for eggplant fruit length using the S.melongena-HQ reference genome and detected a QTL interval of 71.29–78.26 Mb on chromosome E03.The gene Smechr0301963,which belongs to the SUN gene family,is predicted to be a key candidate gene for eggplant fruit length regulation.Moreover,we anchored a total of 210 linkage markers associated with 71 traits to the eggplant chromosomes and finally obtained 26 QTL hotspots.The eggplant HQ-1315 genome assembly can be accessed at http://eggplant-hq.cn.In conclusion,the eggplant genome presented herein provides a global view of genomic divergence at the whole-genome level and powerful tools for the identification of candidate genes for important traits in eggplant. | Qingzhen Wei Jinglei Wang Wuhong Wang Tianhua Hu Haijiao Hu Chonglai Bao | 2020 | Horticulture Research2020,7,1: | 12 |

A number of viral proteins are known to modify and/or be modified by the SUMOylation system to exert their function, to create a cellular environment more favorable for virus survival and propagation, and to prevent host antiviral responses. The inevitable machining and assembly errors, as well as the complicated error propagation mechanism, cause overproof or non-uniform tip clearances. However, it is generally accepted that tip clearances are difficult to predict, even under assembly state. In this paper, a tip clearance prediction model is proposed based on measured error data. Some 3 D error propagation sub-models, regarding rotors, supports and casings, are built and combined. The complex error coupling relationship is uncovered using mathematical methods. Rotor and stator tip clearances are predicted and analyzed in different phase angles. The maximum, minimum and average tip clearances can be calculated. The proposed model is implemented by a computer program,and a case study illustrates its performance and verifies its feasibility. | 16 | Study on the relative radiometric correction of CBERS satellite CCD image显示文摘The main payload on CBERS-01/02 of China-Brazil Earth Resources Satellite (CBERS) is a push-broom CCD camera with moderate spatial and radiant resolution. Because at lab the data for calibration at satellite assembly stage were unable to be collected, and also because the onboard calibrator after launch was in a different state from imaging, the calibration of CCD image got a series of difficulties involved. In practice, two methods are used in the processing on the ground station: One is extracting calibration data by statistics from the image itself, and the other is the method of histogram match. It was proved that the latter can calibrate the image much better, because it can remove the effect of unstable response of the camera largely and also can overcome the nonlinearity of the camera basically by using Look-Up Table (LUT) calculated from histogram statistics of different temporal images. Considering the problems of CBERS-01, a lot of calibration tests were done before the launch of CBERS-02, in which a set of lab coefficients for relative calibration was formulated after the data collection by using integration-hemisphere in the stage of satellite assembly test. During the on-orbit test, it was found that the calibration result from such coefficients was not satisfying, especially there being response difference between 3 detector arrays, which was attributed to the unstable dark currents of the CCD camera. This paper comes up with a statistic method to remove such response difference. | 19 | Preparation and characterization of a novel strong-fluorescent hydrotalcite-like compound (Al-HTLc)显示文摘A novel strong-fluorescent hydrotalcite-like compound (Al-HTLc) was synthesized by coprecipitation. In the sample, the content of aluminum(III) in the layers was decreased to a proper value. The Al3+ ions coordinated with 8-hydroxyquinolines (8-HQ) which were dispersed into the anions in the interlayer region. The sample was characterized by XRD, XPS, FT-IR, TG-DSC, UV-Vis, and fluorescence spec- troscopy; its composition and structure were also determined. The results indicate that the sample can emit fluorescence (487 nm) with a strong fluorescence intensity (4.9×105 (a.u.)). The fluorescent lifetime and fluorescence quantum yield of Al-HTLc were measured to be 21.24 ns and 67%, respectively, higher than those of pure 8-hydroxyquinoline aluminum (Alq3). The result of TG-DSC measurement clearly shows the enhanced thermal stability of Al-HTLc compared with that of MgAl-LDH and pure Alq3. | 20 | Formin-like 3 regulates RhoC/FAK pathway and actin assembly to promote cell invasion in colorectal carcinoma显示文摘AIM To clarify the underlying mechanism of formin-like 3(FMNL3)in the promotion of colorectal carcinoma(CRC)cell invasion.METHODS The in vitro biological function analyses of FMNL3 were performed by gain-and loss-of function approaches.Changes in the F-actin cytoskeleton were detected by the technologies of phalloidin-TRITC labeling and confocal microscopy.The signaling pathway mediated by FMNL3 was explored by western blot,gelatin zymograph assay,co-immunoprecipitation(co-IP),immunofluorescence colocalization,and glutathione S-transferase(GST)pulldown assay.RESULTS The in vitro experimental results showed that FMNL3 significantly promoted the proliferation,invasion,and migration of CRC cells(P<0.05 and P<0.01).Moreover,FMNL3regulated the remodeling of actin-based protrusions such as filopodia and lamellipodia in a RhoC-dependent manner.The western blot and gelatin zymograph assay results indicated that FMNL3 was involved in the RhoC/focal adhesion kinase(FAK)pathway and acted as an effector of RhoC to activate the downstream signaling of p-FAK as well as p-MAPK and p-AKT.This resulted in the increased expression of matrix metalloproteinase 2(MMP2),matrix metalloproteinase 9(MMP9)and vascular endothelial growth factor(VEGF),and the subsequent promotion of CRC cell invasion.The results of TAE226,U0126 or Ly294002 treatment confirmed an essential role of FMNL3 in activation of the RhoC/FAK pathway and the subsequent promotion of CRC invasion.Co-IP,colocalization and GST pull-down assays showed the direct interaction of FMNL3 with RhoC in vivo and in vitro.CONCLUSION FMNL3 regulates the RhoC/FAK signaling pathway and RhoC-dependent remodeling of actin-based protrusions to promote CRC invasion. | Yuan-Feng Zeng Yi-Sheng Xiao Yong Liu Xiao-Jiang Luo Li-Dan Wen Qian Liu Min Chen | 2018 | World Journal of Gastroenterology2018,24,34: | 5 |
