2Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. 3Inflammation and tumor progression:signaling pathways and targeted intervention显示文摘Cancer development and its response to therapy are regulated by inflammation,which either promotes or suppresses tumor progression,potentially displaying opposing effects on therapeutic outcomes.Chronic inflammation facilitates tumor progression and treatment resistance,whereas induction of acute inflammatory reactions often stimulates the maturation of dendritic cells(DCs)and antigen presentation,leading to anti-tumor immune responses.In addition,multiple signaling pathways,such as nuclear factor kappa B(NF-kB),Janus kinase/signal transducers and activators of transcription(JAK-STAT),toll-like receptor(TLR)pathways,cGAS/STING,and mitogen-activated protein kinase(MAPK);inflammatory factors,including cytokines(e.g.,interleukin(IL),interferon(IFN),and tumor necrosis factor(TNF)-a),chemokines(e.g.,C-C motif chemokine ligands(CCLs)and C-X-C motif chemokine ligands(CXCLs)),growth factors(e.g.,vascular endothelial growth factor(VEGF),transforming growth factor(TGF)-P),and inflammasome;as well as inflammatory metabolites including prostaglandins,leukotrienes,thromboxane,and specialized proresolving mediators(SPM),have been identified as pivotal regulators of the initiation and resolution of inflammation.Nowadays,local irradiation,recombinant cytokines,neutralizing antibodies,small-molecule inhibitors,DC vaccines,oncolytic viruses,TLR agonists,and SPM have been developed to specifically modulate inflammation in cancer therapy,with some of these factors already undergoing clinical trials.Herein,we discuss the initiation and resolution of inflammation,the crosstalk between tumor development and inflammatory processes.We also highlight potential targets for harnessing inflammation in the treatment of cancer.Huakan Zhao Lei Wu Guifang Yan Yu Chen Mingyue Zhou Yongzhong Wu Yongsheng Li 2021Signal Transduction and Targeted Therapy2021,6,8:11

5Expression of human soluble TRAIL in Chlamydomonas reinhardtii chloroplast显示文摘Tumor necrosis factor-related apop-tosis-inducing ligand (TRAIL) induces selectively apoptosis in various tumor cells and virus-infected cells, but rarely in normal cells. A chloroplast expres-sion vector, p64TRAIL, containing the cDNA coding for the soluble TRAIL (sTRAIL), was constructed with clpP-trnL-petB-chlL-rpl23-rpl2 as Chlamydomonas reinhardtii plastid homologous recombinant frag-ments and spectinomycin-resistant aadA gene as a select marker. The plasmid p64TRAIL was trans-ferred into the chloroplast genome of C. reinhardtii by the biolistic method. Three independently trans-formed lines were obtained by 100 mg/L spectino-mycin selection. PCR amplification, Southern blot analysis of the sTRAIL coding region DNA and culti-vation cells in the dark all showed that the exogenous DNA had been integrated into chloroplast genome of C. reinhardtii. Western blot analysis showed that human soluble TRAIL was expressed in C. reinhardtii chloroplast. The densitometric analysis of Western blot indicated that the expressed human sTRAIL protein in the chloroplasts of C. reinhardtii accounted for about 0.43%―0.67% of the total soluble proteins. 6Underwater Laser Welding/Cladding for High-performance Repair of Marine Metal Materials:A Review显示文摘With the rapid developments of marine resource exploitation,mounts of marine engineering equipment are settled on the ocean.When it is not possible to move the damaged equipment into a dry dock,welding operations must be performed in underwater environments.The underwater laser welding/cladding technique is a promising and advanced technique which could be widely applied to the maintenance of the damaged equipment.The present review paper aims to present a critical analysis and engineering overview of the underwater laser welding/cladding technique.First,we elaborated recent advances and key issues of drainage nozzles all over the world.Next,we presented the underwater laser processing and microstructural-mechanical behavior of repaired marine materials.Then,the newly developed powder-feeding based and wire-feeding based underwater laser direct metal deposition techniques were reviewed.The differences between the convection,conduction,and the metallurgical kinetics in the melt pools during underwater laser direct metal deposition and in-air laser direct metal deposition were illustrated.After that,several challenges that need to be overcame to achieve the full potential of the underwater laser welding/cladding technique are proposed.Finally,suggestions for future directions to aid the development of underwater laser welding/cladding technology and underwater metallurgical theory are provided.The present review will not only enrich the knowledge in the underwater repair technology,but also provide important guidance for the potential applications of the technology on the marine engineering.Guifang Sun Zhandong Wang Yi Lu Mingzhi Chen Kun Yang Zhonghua Ni 2022Chinese Journal of Mechanical Engineering2022,35,1:4
7High flux photocatalytic self-cleaning nanosheet C_(3)N_(4) membrane supported by cellulose nanofibers for dye wastewater purification显示文摘Hazardous dye substances discharged from the textile and dyestuff industries not only threaten local the surrounding ecosystems but are also hard to degraded.We report the preparation of process for a photocatalytic membrane device that can degrade dye pollution under visible light.This filtration membrane,with a well-organized multilayer structure,simultaneously achieves continuous and flow-through separation of degradation products.Cellulose nanofibers(CNFs)were used as a template for nanosheet C_(3)N_(4)(NS C_(3)N_(4))preparation;the performance for the photocatalytic degradation of dyes improved as the morphology changed from bulking to nanosheet.NS C3N4 was then attached to the surface of a prepared CNF membrane via vacuum filtration.This device exhibited high efficiency(the degradation rates of both Rhodamine B and Methylene blue both reached 96%),high flux(above 160 L·h^(-1)·m^(-2)·bar^(-1))and excellent stability(maintaining steady flux and high separation were maintained after 4 h).This easy-preparation,easy-scale-up,and low-cost process provides a new method of fabricating photocatalytic membrane devices for dye wastewater treatment.Lilong Zhang Ge Meng Guifang Fan Keli Chen Yulong Wu Jian Liu 2021Nano Research2021,14,8:3

11Interaction between curcumin and mimetic biomembrane显示文摘Curcumin,a major bioactive compound in turmeric,has a broad spectrum of antioxidant,anticarcinogenic,antimutagenic and anti-inflammatory properties.At the molecular level,curcumin modulates many structurally unrelated membrane proteins through several signaling pathways.Curcumin has been suggested to change the properties of cell membranes and affect the membrane-bound proteins indirectly;however,the detailed mechanism has yet to be investigated.In this paper,self-assembled bilayer lipid membranes are artificially constructed on the surface of a gold electrode to mimic biomembranes,and interaction between the supported membranes and curcumin is studied electrochemically.Results show that curcumin interacts with the membranes strongly,in a concentration-dependent manner.At low concentrations,curcumin tends to insert into the outer monolayer only,while at high concentrations,it may also begin to penetrate the inner monolayer.The results obtained in this work may enhance our understanding of the effect of curcumin,and possibly flavonoids,on cell membranes and membrane proteins.CHEN GuiFang CHEN YangYang YANG NaNa ZHU XueJun SUN LiZhou LI GenXi 2012Science China(Life Sciences)2012,55,6:2
12Efficacy of Yinzhihuang Granules and Entecavir on Hepatitis B-induced Cirrhosis显示文摘[Objectives]This study aimed to observe the efficacy and safety of Yinzhihuang granules and entecavir in the treatment of hepatitis B-induced cirrhosis.[Methods]Total 80 patients with decompensated hepatitis B-induced cirrhosis were evenly and randomly divided into two groups,treatment group and control group.The patients in the treatment group were given orally with 0.25 mg entecavir and 5000 mg Yinzhihuang granules once a day,and those in the control group were given orally with 0.5 mg entecavir once a day.The levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),hyaluronic acid(HA)and HBV DNA,the Child-Pugh score,and the positive situation of HBeAg in different groups were observed before treatment and 12,24,48 and 96 weeks after the treatment,respectively.[Results]The serum ALT,AST,HA and HBV DNA levels,the HBeAg positive rate,and the Child-Pugh score in the treatment and control groups were significantly improved after the treatment(P<0.05).The improvement of the above indicators in the treatment group at week 96 was significantly better than that in the control group(P<0.05).[Conclusions]The combination of Yinzhihuang granules and entecavir is superior to entecavir in the treatment of decompensated hepatitis B-induced cirrhosis.Fen TANG Guifang TANG Ruiling CHEN Zhenyi LIU Riming WEI 2019Medicinal Plant2019,10,5:2
13Control of de novo root regeneration efficiency by developmental status of Arabidopsis leaf explants显示文摘De novo root regeneration(DNRR) has wide applications in agriculture such as those related to cutting technology. Detached Arabidopsis thaliana leaf explants can regenerate adventitious roots without added hormones. The regenerative ability is highly dependent on the developmental status of the leaf. An immature leaf has a higher regenerative ability, while a mature leaf is difficult to regenerate. Using RNASeq analysis, we showed that the expression levels of many genes, including those in the auxin network,changed during leaf maturation. Particularly, the expression levels of many YUCCA(YUC) genes in the auxin biosynthesis pathway are responsive to leaf maturation. Overexpression of YUC1 in the yuc-1 D dominant mutant rescued the rooting defects caused by leaf maturation. In addition, YUC4 expression levels were also affected by circadian rhythms. The regenerative ability was reduced in both immature and mature mutant leaf explants from the new wuschel-related homeobox 11-3(wox11-3) and wox12-3 mutant alleles created by the CRISPR/Cas9 method. Overall, the transcriptome and genetic data, together with the auxin concentration analysis, indicate that the ability to upregulate auxin levels upon detachment may be reduced during leaf maturation. 18The RNA N^(6)-methyladenosine demethylase ALKBH9B modulates ABA responses in Arabidopsis显示文摘The mRNA modification N^(6)-methyladenosine(m^(6)A)plays vital roles in plant development and biotic and abiotic stress responses.The RNA m^(6)A demethylase ALKBH9 B can remove m^(6)A in alfalfa mosaic virus RNA and plays roles in alfalfa mosaic virus infection in Arabidopsis.However,it is unknown whether ALKBH9 B also exhibits demethylation activity and has a biological role in endogenous plant mRNA.We demonstrated here that mRNA m^(6)A modification is induced by the phytohormone abscisic acid(ABA)and that ALKBH9 B has m^(6)A demethylation activity on endogenous mRNA.Knocking out ALKBH9 B led to hypersensitivity to ABA treatment during seed germination and early seedling development.We further showed that ALKBH9 B removes the m^(6)A modification in the ABA INSENSITIVE 1(ABI1)and BRI1-EMS-SUPPRESSOR 1(BES1)transcripts following ABA treatment,affecting the stability of these mRNAs.Furthermore,we determined that ALKBH9 B acts genetically upstream of the transcription factors ABI3 and ABI5,and its regulatory function in ABA responses depended on ABI3 and ABI5.Our findings reveal the important roles of the m^(6)A modification in ABA responses and highlight the role of ALKBH9 Bmediated m^(6)A demethylation in regulating ABA responses post-transcriptionally.Jun Tang Junbo Yang Qiang Lu Qian Tang Shuyan Chen Guifang Jia 2022Journal of Integrative Plant Biology2022,64,12:1
