1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Pore throat characteristics of tight sandstone of Yanchang Formation in eastern Gansu,Ordos Basin显示文摘An important factor to evaluate reservoir quality is the pore-throat size.However,the strong heterogeneity makes it difficult to characterize the pore-throat distribution in tight reservoirs.The field emission scanning electron microscope(FESEM),high pressure mercury injection and rate-controlled mercury injection are used to investigate the pore-throat size distribution in tight sandstone reservoirs of Member 7 of the Yanchang Formation in eastern Gansu,Ordos Basin,and studies of the pore throat size controlling on physical property of the tight sandstone reservoirs are also carried out.The result shows that the pore type is mainly dominated by the residual intergranular pore,dissolution pore,micropore and a few micro-fractures;the high-pressure mercury injection experiment indicates that the pore-throat size ranges from 0.0148 μm to 40mm,the pore throat more than 1 mm is less;the ratecontrolled mercury injection experiment reveals that for samples with different physical properties,the pore radius mainly varies from 80 μm to 350 μm;the throat radius exhibits the strong heterogeneity,and is from 0.12 μm to 30μm;the pore-throat size can be effectively characterized by combination of high-pressure and rate-controlled mercury injections,and it varies from 0.0148 μm to 350 μm.The permeability is mainly controlled by the large pore throat(>R_(50))which accounts for a small proportion;in the tight sandstone with the permeability greater than 0.1 mD,the permeability is mainly controlled by the micropore and mesopore;in the tight sandstone with the permeability smaller than 0.1 mD,the permeability is mainly controlled by the nanopore and micropore;the proportion of small pore throat increases with reduction of permeability,it is important that the small pore throat influences the reservoir storage property though its effect on permeability are small.Hao Wu Chunlin Zhang Youliang Ji Rui'e Liu Shang Cao Sheng Chen Yunzhao Zhang Ye Wang Wei Du Gang Liu 2018Petroleum Research2018,3,1:7
4Combined remediation of DDT congeners and cadmium in soil by Sphingobacterium sp.D-6 and Sedum alfredii Hance显示文摘Combined pollution of 1,1,1-trichloro-2,2-bis (4-chlorophenyl) ethane (DDT) and cadmium (Cd) in agricultural soils is of great concern because they present serious risk to food security and human health.In order to develop a cost-effective and safe method for the removal of DDTs and Cd in soil,combined remediation of DDTs and Cd in soil by Sphingobacterium sp.D-6 and the hyperaccumulator,Sedum alfredii Hance was investigated.After treatment for 210 days,the degradation half-lives of DDTs in soils treated by strain D-6 decreased by 8.1% to 68.0% compared with those in the controls.The inoculation of strain D-6 into soil decreased the uptake of DDTs by pak choi and S.alfredii.The shoots/roots ratios of S.alfredii for the Cd accumulation ranged from 12.32 to 21.75.The Cd concentration in soil decreased to 65.8%-71.8% for S.alfredii treatment and 14.1%-58.2% for S.alfredii and strain D-6 combined treatment,respectively,compared with that in the control.The population size of the DDTs-degrading strain,Simpson index (1/D) and soil respiratory rate decreased in the early stage of treatment and then gradually increased,ultimately recovering to or exceeding the initial level.The results indicated that synchronous incorporation of strain D-6 and S.alfredii into soil was found to significantly (p 0.05) enhance the degradation of DDTs in soil and the hyperaccumulation of Cd in S.alfredii.It was concluded that strain D-6 and S.alfredii could be used successfully to control DDTs and Cd in contaminated soil.Hua Fang Wei Zhou Zhengya Cao Feifan Tang Dandan Wang Kailin Liu Xiangwei Wu Xiao'e Yang Yongge Sun Yunlong Yu 2012Journal of Environmental Sciences2012,24,6:5

6Influence of particle composition on thorium scavenging in the marginal China seas显示文摘Thorium-234 and particle composition (organic matter, biogenic silica, carbonate and lithogenic component) were examined in the East China Sea (ECS) and the northern South China Sea (NSCS) in order to constrain the particle types scavenging thorium isotopes. Good positive correlations be-tween particulate organic matter (POM) or carbonate and thorium-234 in suspended particulate matter (SPM) indicates that POM and carbonate are efficient to scavenge thorium-234. No relation-ship between biogenic silica and thorium-234 suggests that geochemical behavior of thorium-234 may be not influenced by biogenic silica. A simple model was used to evaluate the affinity of thorium-234 to different particle components. The results show that POM is the most efficient scavenger for thorium-234 in the ECS and the NSCS, followed by carbonate. The authors’ re-sults lend support to the utility of thorium-234 as a proxy of POC and carbonate in the upper layer. 7Trace detection of Ce^(3+) by adsorption strip voltammetry at a carbon paste electrode modified with ion imprinted polymers显示文摘To develop a convenient method for sensitive and selective determination of Ce3+ in aqueous phase with complicated matrices, a carbon paste electrode(CPE) modified with ion imprinted polymers(IIPs) were fabricated. The polymers were prepared by precipitation polymerization using Ce3+ as template, allyl phenoxyacetate(APA) as monomer, ethylene glycol dimethacrylate(EGDMA) as crosslinker and azobisisobutyronitrile(AIBN) as initiator under the molar ratio of Ce3+, APA and EGDMA as 1:4:40, respectively. Ce^(3+) was detected directly by differential pulse adsorptive stripping voltammetry(DPASV) and its oxidation peak appears at about 0.93 V. All parameters affecting the sensor's response are optimized and a calibration curve is plotted at a linear range of 1.0 × 10^(-6)-1.0 x 10^(-5) mol/L and 1.0×10^(-5)-2.0 × 10^(-4)mol/L with the detection limit of 1.5 × 10^(-7) mol/L. 8全颗粒饲料对奶牛生产性能的影响显示文摘饲料中的中性洗涤纤维(NDF)含量和饲料颗粒直径是影响瘤胃功能的重要因素。试验评估了以颗粒饲料为主的奶牛瘤胃健康情况、NDF消化率和生产性能。选取8头荷斯坦母牛(336±30 d,体重346±35 kg),随机分配到交叉重复试验组中,奶牛安置于牛栏,自由采食。试验期为3周,前两周是适应期,最后一周为数据收集期。各组饲粮原料相同,但物理形式不同,分为全混合日粮(TMR)和颗粒料(直径=8 mm)。两组间物理有效NDF(pe NDF)不同,TMR组为39.8%,颗粒料组为11.8%。在试验过程中,干物质采食量(DMI)、饮水量、瘤胃反刍时间、瘤胃温度和pH情况均记录。3周后收集粪样,确定潜在的可消化NDF总消化率(pdNDF)。每阶段结束后计算平均日增重和饲料转化率。颗粒料组的DMI、DMI/体重和饮水量更高。而两组间平均日增重和饲料转化率无显著差异。颗粒料组的瘤胃反刍时间为241 min·d-1低于TMR组的507 min·d-1。饲料的不同物理外形对瘤胃温度、瘤胃pH无显著影响。TMR组的pdNDF总消化率为90.25%高于颗粒料组的86.82%。试验结果表明,动物易接受完整的颗粒饲料,DMI较高,且反刍时间减少,但瘤胃pH无显著变化。TMR组的pdNDF消化率显著增加。尽管两组奶牛在DMI和纤维消化方面表现不同,但总体表现相似,两组饲料的保存时间与其物理形式有关。在短期内,提供能满足NDF需求的完全颗粒饲料可改善瘤胃健康状况和动物生产性能,而较长时间的数据则需要进一步试验。Wang J Yang M Cao M Lin Y Che L Q Duraipandiyan V Al-Dhabi N A Fang Z F Xu S Y Feng B Liu G Wu D Bonfante E Palmonari A Mammi L Canestrari G Fustini M Formigoni A 2016饲料博览2016,0,12:2
9Experimental Study on Heterograft of Glomus Cells of Carotid Body for Hemiparkinsonian Rats显示文摘Summary: To observe the effects of heterograft of glomus cells of carotid body on hemiparkinsonian rat models, rats with unilateral 6-hydroxydopamine (6-OHDA)-induced lesions of the right dopaminergic neurons of substantia nigra received intrastriatal glomus cells heterograft. Apomorphine-induced rotation was monitored for 30 min at various time points after grafting. The striata were cut and examined for dopamine content by HPLC and for immunohistochemical staining of tyrosine hydroxylase positive neurons (TH +) at the end of the experiments. The results showed that apomorphine-induced rotational behavior was significantly reduced for 12 weeks and the dopamine contents were significantly elevated after grafting (P<0.01), and TH + cells survived better. 10The Alzheimer's disease-associated gene TREML2 modulates inflammation by regulating microglia polarization and NLRP3 inflammasome activation显示文摘Triggering receptor expressed on myeloid cells-like 2(TREML2)is a newly identified susceptibility gene for Alzheimer's disease(AD).It encodes a microglial inflammation-associated receptor.To date,the potential role of mic roglial TREML2 in neuroinflammation in the context of AD remains unclear.In this study,APP/PS1 mice were used to investigate the dynamic changes of TREML2 levels in brain during AD progression.In addition,lipopolysaccharide(LPS)stimulation of primary microglia as well as a lentivirus-mediated TREML2 overexpression and knockdown were employed to explore the role of TREML2 in neuroinflammation in the context of AD.Our res ults show that TREML2 levels gradually increased in the brains of AP P/PS1 mice during disease progression.LPS stimulation of primary microglia led to the release of inflammato ry cytokines including interleukin-1β,inte rleukin-6,and tumor necrosis factor-a in the culture medium.The LPS-induced mic roglial release of inflammatory cytokines was enhanced by TREML2 overexpression and was attenuated by TREML2 knoc kdown.LPS increased the levels of mic roglial M1-type polarization marker inducible nitric oxide synthase.This effect was enhanced by TREML2 overexpression and ameliorated by TREML2 knockdown.Furthermore,the levels of microglial M2-type polarization markers CD206 and ARG1 in the primary microglia were reduced by TREML2 overexpression and elevated by TREML2 knockdown.LPS stimulation increased the levels of NLRP3 in primary microglia.The LPS-induced increase in NLRP3 was further elevated by TREML2 overexpression and alleviated by TREML2 knockdown.In summary,this study provides the first evidence that TREML2 modulates inflammation by regulating microglial polarization and NLRP3 inflammasome activation.These findings reveal the mechanisms by which TREML2 regulates microglial inflammation and suggest that TREML2 inhibition may represent a novel therapeutic strategy for AD.Si-Yu Wang Xin-Xin Fu Rui Duan Bin Wei Hai-Ming Cao Yan E Shuai-Yu Chen Ying-Dong Zhang Teng Jiang 2023Neural Regeneration Research2023,18,2:2
