1Responses of Soil Enzyme Activities and Microbial Community Composition to Moisture Regimes in Paddy Soils Under Long-Term Fertilization Practices显示文摘The effects of fertilization on activity and composition of soil microbial community depend on nutrient and water availability;however,the combination of these factors on the response of microorganisms was seldom studied.This study investigated the responses of soil microbial community and enzyme activities to changes in moisture along a gradient of soil fertility formed within a long-term(24 years)field experiment.Soils(0–20 cm)were sampled from the plots under four fertilizer treatments:i)unfertilized control(CK),ii)organic manure(M),iii)nitrogen,phosphorus,and potassium fertilizers(NPK),and iv)NPK plus M(NPK+M).The soils were incubated at three moisture levels:constant submergence,five submerging-draining cycles(S-D cycles),and constant moisture content at 40%water-holding capacity(low moisture).Compared with CK,fertilization increased soil organic carbon(SOC) by 30.1%–36.3%,total N by 27.3%–38.4%,available N by 35.9%–56.4%,available P by 61.4%–440.9%,and total P by 28.6%–102.9%.Soil fertility buffered the negative effects of moisture on enzyme activities and microbial community composition.Enzyme activities decreased in response to submergence and S-D cycles versus low moisture.Compared with low moisture,S-D cycles increased total phospholipid fatty acids(PLFAs)and actinomycete,fungal,and bacterial PLFAs.The increased level of PLFAs in the unfertilized soil after five S-D cycles was greater than that in the fertilized soil.Variations in soil microbial properties responding to moisture separated CK from the long-term fertilization treatments.The coefficients of variation of microbial properties were negatively correlated with SOC,total P,and available N.Soils with higher fertility maintained the original microbial properties more stable in response to changes in moisture compared to low-fertility soil.LI Weitao WU Meng LIU Ming JIANG Chunyu CHEN Xiaofen Yakov KUZYAKOV Jorg RINKLEBE LI Zhongpei 2018Pedosphere2018,28,2:16
2Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. 3Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells.Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu 2019Cell Research2019,29,11:14

5Effect of N fertilization rate on soil alkalihydrolyzable N, subtending leaf N concentration,fiber yield, and quality of cotton显示文摘Soil alkali-hydrolyzable nitrogen, which is sensitive to N fertilization rate, is one of the indicators of soil nitrogen supplying capacity. Two field experiments were conducted in Dongtai(120°19″ E, 32°52″ N), Jiangsu, China in 2009 and Dafeng(120°28″ E, 33°12″ N), Jiangsu province, China in 2010. Six nitrogen rates(0, 150, 300, 375, 450, and 600 kg ha^(-1)) were used to study the effect of N fertilization rate on soil alkali-hydrolyzable nitrogen content(SAHNC), subtending leaf nitrogen concentration(SLNC), yield, and fiber quality. In both Dongtai and Dafeng experiment station, the highest yield(1709 kg ha^(-1)), best quality(fiber length 30.6 mm, fiber strength 31.6 c N tex^(-1), micronaire 4.82), and highest N agronomic efficiency(2.03 kg kg^(-1)) were achieved at the nitrogen fertilization rate of 375 kg ha^(-1). The dynamics of SAHNC and SLNC could be simulated with a cubic and an exponential function,respectively. The changes in SAHNC were consistent with the changes in SLNC. Optimal average rate(0.276 mg day^(-1)) and duration(51.8 days) of SAHNC rapid decline were similar to the values obtained at the nitrogen rate of 375 kg ha^(-1)at which cotton showed highest fiber yield, quality, and N agronomic efficiency. Thus, the levels and strategies of nitrogen fertilization can affect SAHNC dynamics. 6Vitamin B12 modulates Parkinson’s disease LRRK2 kinase activity through allosteric regulation and confers neuroprotection显示文摘Missense mutations in Leucine-Rich Repeat Kinase 2(LRRK2)cause the majority of familial and some sporadic forms of Parkinson’s disease(PD).The hyperactivity of LRRK2 kinase induced by the pathogenic mutations underlies neurotoxicity,promoting the development of LRRK2 kinase inhibitors as therapeutics.Many potent and specific small-molecule LRRK2 inhibitors have been reported with promise.However,nearly all inhibitors are ATP competitive—some with unwanted side effects and unclear clinical outcome—alternative types of LRRK2 inhibitors are lacking.Herein we identify 5′-deoxyadenosylcobalamin(AdoCbl),a physiological form of the essential micronutrient vitamin B12 as a mixed-type allosteric inhibitor of LRRK2 kinase activity.Multiple assays show that AdoCbl directly binds LRRK2,leading to the alterations of protein conformation and ATP binding in LRRK2.STD-NMR analysis of a LRRK2 homologous kinase reveals the contact sites in AdoCbl that interface with the kinase domain.Furthermore,we provide evidence that AdoCbl modulates LRRK2 activity through disrupting LRRK2 dimerization.Treatment with AdoCbl inhibits LRRK2 kinase activity in cultured cells and brain tissue,and prevents neurotoxicity in cultured primary rodent neurons as well as in transgenic C.elegans and D.melanogaster expressing LRRK2 disease variants.Finally,AdoCbl alleviates deficits in dopamine release sustainability caused by LRRK2 disease variants in mouse models.Our study uncovers vitamin B12 as a novel class of LRRK2 kinase modulator with a distinct mechanism,which can be harnessed to develop new LRRK2-based PD therapeutics in the future.Adam Schaffner Xianting Li Yacob Gomez-Llorente Emmanouela Leandrou Anna Memou Nicolina Clemente Chen Yao Farinaz Afsari Lianteng Zhi Nina Pan Keita Morohashi Xiaoluan Hua Ming-Ming Zhou Chunyu Wang Hui Zhang Shu G.Chen Christopher J.Elliott Hardy Rideout Iban Ubarretxena-Belandia Zhenyu Yue 2019Cell Research2019,29,4:7

8Hyaluronic acid facilitates bone repair effects of calcium phosphate cement by accelerating osteogenic expression显示文摘Calcium phosphate cements(CPC)are widely anticipated to be an optimum bone repair substitute due to its satisfied biocompatibility and degradability,suitable to be used in minimally invasive treatment of bone defects.However the clinical application of CPC is still not satisfied by its poor cohesiveness and mechanical properties,in particular its osteoinductivity.Hyaluronic acid reinforced calcium phosphate cements(HA/CPC)showed extroadinary potential not only enhancing the compressive strength of the cements but also significantly increasing its osteoinductivity.In our study,the compressive strength of HA/CPC increased significantly when the cement was added 1%hyaluronic acid(denoted as 1-HA/CPC).In the meantime,hyaluronic acid obviously promoted ALP activity,osteogenic related protein and mRNA expression of hBMSCs(human bone marrow mesenchymal stem cells)in vitro,cement group of HA/CPC with 4%hyaluronic acid adding(denoted as 4-HA/CPC)showed optimal enhancement in hBMSCs differentiation.After being implanted in rat tibial defects,4-HA/CPC group exhibited better bone repair ability and bone growth promoting factors,comparing to pure CPC and 1-HA/CPC groups.The underlying biological mechanism of this stimulation for HA/CPC may be on account of higher osteogenic promoting factors secretion and osteogenic genes expression with hyaluronic acid incorporation.These results indicate that hyaluronic acid is a highly anticipated additive to improve physicochemical properties and osteoinductivity performance of CPCs for minimally invasive healing of bone defects.Xu Cui Chengcheng Huang Zhizhen Chen Meng Zhang Chunyu Liu Kun Su Jianyun Wang Li Li Renxian Wang Bing Li Dafu Chen Changshun Ruan Deping Wang William W.Lu Haobo Pan 2021Bioactive Materials2021,6,11:6

11Effect of a combination of calorie-restriction therapy and Lingguizhugan decoction on levels of fasting blood lipid and inflammatory cytokines in a high-fat diet induced hyperlipidemia rat model显示文摘OBJECTIVE:To combine calorie-restriction therapy(CRT) with Traditional Chinese Medicine(TCM) using Lingguizhugan Decoction and note the effects on expression of inflammatory cytokines[tumor necrosis factor(TNF)-α,high-sensitivity C-reactive protein(hs-CRP),interleukin(IL)-6]in high-fat-diet induced hyperlipidemia in rats.METHODS:Forty rats were divided randomly and equally into four groups:control group A(regular feeding for 5 weeks);model group B(high-fat diet for 5 weeks),calorie-limited group C(high-fat diet for 5 weeks) and TCM calorie-limit group D(high-fat diet for 5 weeks).After modeling,groups C and D were fasted for 3 days:group C with simple fasting,and group D with TCM fasting.The motion as well as changes in color,body weight,food intake,plasma lipids[low-density lipoprotein-cholesterol(LDL-C),high-density lipoprotein-cholesterol,total cholesterol(TC) and triglyceride(TG) along with TNF-α,hs-CRP and IL-6]were measured before and after intervention.RESULTS:Modeled rats were established after five weeks.After 3 days of fasting,compared with group B,blood lipid levels(TG,TC,LDL-C) of groups C and D decreased dramatically.Those of group D decreased more obviously than those of group C.However,the values of TNF-α,hs-CRP and IL-6 showed no obvious difference between each of the three treatment groups.CONCLUSION:CRT can reduce fasting blood lipid levels in rats,but not by reducing the levels of inflammatory cytokines.Wang Yuanyuan Jin Minghua Zhang Lina Li Suhua Zhai Jiayu Shen Yongzhi Chen Chunyu Qin Jian 2015Journal of Traditional Chinese Medicine2015,35,2:4
12The role of bone marrow-derived cells in the origin of liver cancer revealed by single-cell sequencing显示文摘Objective:Epithelial cancers often originate from progenitor cells,while the origin of hepatocellular carcinoma(HCC)is still controversial.HCC,one of the deadliest cancers,is closely linked with liver injuries and chronic inflammation,which trigger massive infiltration of bone marrow-derived cells(BMDCs)during liver repair.Methods:To address the possible roles of BMDCs in HCC origination,we established a diethylnitrosamine(DEN)-induced HCC model in bone marrow transplanted mice.Immunohistochemistry and frozen tissue immunofluorescence were used to verify DENinduced HCC in the pathology of the disease.The cellular origin of DEN-induced HCC was further studied by single cell sequencing,single-cell nested PCR,and immunofluorescence-fluorescence in situ hybridization.Results:Studies by using single cell sequencing and biochemical analysis revealed that HCC cells in these mice were coming from donor mice BMDCs,and not from recipient mice.Furthermore,the copy numbers of mouse orthologs of several HCC-related genes previously reported in human HCC were also altered in our mouse model.DEN-induced HCCs exhibited a similar histological phenotype and genomic profile as human HCCs.Conclusions:These results suggested that BMDCs are an important origin of HCC,which provide important clues to HCC prevention,detection,and treatments.Lu Chen Xianfu Yi Piao Guo Hua Guo Ziye Chen Chunyu Hou Lisha Qi Yongrong Wang Chengwen Li Peng Liu Yucun Liu Yuanfu Xu Ning Zhang 2020Cancer Biology & Medicine2020,17,1:4
13Transcriptome analysis of three cotton pests reveals features of gene expressions in the mesophyll feeder Apolygus lucorum显示文摘The green mirid bug Apolygus lucorum is an agricultural pest that is known to cause damage to more than 150 plant species.Here,we report the transcriptomes of A.lucorum at three different developmental stages(the second and fifth instar nymphs and adults).A total of 98,236 unigenes with an average length of l,335 nt was obtained,of which 50,640 were annotated,including those encoding digestive enzymes and cytochrome P450 s.Comparisons with cotton bollworm and cotton aphid transcriptomes revealed distinct features of A.lucorum as a mesophyll feeder.The gene expression dynamics varied during development from young nymphs to adults.The high-quality transcriptome data and the gene expression dynamics reported here provide valuable data for a more comprehensive understanding of the physiology and development of mirid bugs,and for mining targets for their control.Dianyang Chen Fangyan Chen Chunyu Chen Xiaoya Chen Yingbo Mao 2017Science China(Life Sciences)2017,60,8:4
14Toward dendrite-free and anti-corrosion Zn anodes by regulating a bismuth-based energizer显示文摘Aqueous rechargeable zinc metal batteries display high theoretical capacity along with economical effectiveness,environmental benignity and high safety.However,dendritic growth and chemical corrosion at the Zn anodes limit their widespread applications.Here,we construct a Zn/Bi electrode via in-situ growth of a Bi-based energizer upon Zn metal surface using a replacement reaction.Experimental and theoretical calculations reveal that the Bi-based energizer composed of metallic Bi and ZnBi alloy contributes to Zn plating/stripping due to strong adsorption energy and fast ion transport rates.The resultant Zn/Bi electrode not only circumvents Zn dendrite growth but also improves Zn anode anti-corrosion performance.Specifically,the corrosion current of the Zn/Bi electrode is reduced by 90%compared to bare Zn.Impressively,an ultra-low overpotential of 12​mV and stable cycling for 4000​h are achieved in a Zn/Bi symmetric cell.A Zn–Cu/Bi asymmetric cell displays a cycle life of 1000 cycles,with an average Coulombic efficiency as high as 99.6%.In addition,an assembled Zn/Bi-activated carbon hybrid capacitor exhibits a stable life of more than 50,000 cycles,an energy density of 64​Wh kg−1,and a power density of 7​kW​kg−1.The capacity retention rate of a Zn/Bi–MnO_(2)full cell is improved by over 150%compared to a Zn–MnO_(2)cell without the Bi-based energizer.Our findings open a new arena for the industrialization of Zn metal batteries for large-scale energy storage applications.Mingming Wang Yahan Meng Ke Li Touqeer Ahmad Na Chen Yan Xu Jifei Sun Mingyan Chuai Xinhua Zheng Yuan Yuan Chunyue Shen Ziqi Zhang Wei Chen 2022eScience2022,2,5:4
15Whole-genome sequencing of monozygotic twins discordant for schizophrenia indicates multiple genetic risk factors for schizophrenia显示文摘Schizophrenia is a common disorder with a high heritability, but its genetic architecture is still elusive.We implemented whole-genome sequencing(WGS) analysis of 8 families with monozygotic(MZ) twin pairs discordant for schizophrenia to assess potential association of de novo mutations(DNMs) or inherited variants with susceptibility to schizophrenia. Eight non-synonymous DNMs(including one splicing site) were identified and shared by twins, which were either located in previously reported schizophrenia risk genes(p.V24689 I mutation in TTN, p.S2506 T mutation in GCN1L1, IVS3+1G > T in DOCK1) or had a benign to damaging effect according to in silico prediction analysis. By searching the inherited rare damaging or loss-of-function(LOF) variants and common susceptible alleles from three classes of schizophrenia candidate genes, we were able to distill genetic alterations in several schizophrenia risk genes, including GAD1, PLXNA2, RELN and FEZ1. Four inherited copy number variations(CNVs; including a large deletion at 16p13.11) implicated for schizophrenia were identified in four families, respectively. Most of families carried both missense DNMs and inherited risk variants, which might suggest that DNMs, inherited rare damaging variants and common risk alleles together conferred to schizophrenia susceptibility. 16Classification of Northeast China Cold Vortex Activity Paths in Early Summer Based on K-means Clustering and Their Climate Impact显示文摘The classification of the Northeast China Cold Vortex(NCCV)activity paths is an important way to analyze its characteristics in detail.Based on the daily precipitation data of the northeastern China(NEC)region,and the atmospheric circulation field and temperature field data of ERA-Interim for every six hours,the NCCV processes during the early summer(June)seasons from 1979 to 2018 were objectively identified.Then,the NCCV processes were classified using a machine learning method(k-means)according to the characteristic parameters of the activity path information.The rationality of the classification results was verified from two aspects,as follows:(1)the atmospheric circulation configuration of the NCCV on various paths;and(2)its influences on the climate conditions in the NEC.The obtained results showed that the activity paths of the NCCV could be divided into four types according to such characteristics as the generation origin,movement direction,and movement velocity of the NCCV.These included the generation-eastward movement type in the east of the Mongolia Plateau(eastward movement type or type A);generation-southeast longdistance movement type in the upstream of the Lena River(southeast long-distance movement type or type B);generationeastward less-movement type near Lake Baikal(eastward less-movement type or type C);and the generation-southward less-movement type in eastern Siberia(southward less-movement type or type D).There were obvious differences observed in the atmospheric circulation configuration and the climate impact of the NCCV on the four above-mentioned types of paths,which indicated that the classification results were reasonable.Yihe FANG Haishan CHEN Yi LIN Chunyu ZHAO Yitong LIN Fang ZHOU 2021Advances in Atmospheric Sciences2021,38,3:4
17Air pollution and inhalation exposure to particulate matter of different sizes in rural households using improved stoves in central China显示文摘Household air pollution is considered to be among the top environmental risks in China.To examine the performance of improved stoves for reduction of indoor particulate matter(PM) emission and exposure in rural households, individual inhalation exposure to size-resolved PM was investigated using personal portable samplers carried by residents using wood gasifier stoves or improved coal stoves in a rural county in Central China.Concentrations of PM with different sizes in stationary indoor and outdoor air were also monitored at paired sites. The stationary concentrations of size-resolved PM in indoor air were greater than those in outdoor air, especially finer particles PM0.25. The daily averaged exposure concentrations of PM0.25, PM1.0, PM2.5 and total suspended particle for all the surveyed residents were 74.4 ± 41.1, 159.3 ± 74.3, 176.7 ± 78.1 and 217.9 ± 78.1 μg/m3,respectively. Even using the improved stoves, the individual exposure to indoor PM far exceeded the air quality guideline by WHO at 25 μg/m3. Submicron particles PM1.0 were the dominant PM fraction for personal exposure and indoor and outdoor air. Personal exposure exhibited a closer correlation with indoor PM concentrations than that for outdoor concentrations. Both inhalation exposure and indoor air PM concentrations in the rural households with gasifier firewood stoves were evidently lower than the reported results using traditional firewood stoves. 18Free-standing ZnO nanorod arrays modified with single-walled carbon nanotubes for betavoltaics and photovoltaics显示文摘A full-duplex radiant energy converter based on both betavoltaic and photovoltaic effects in an easyto-implement way is an attractive alternative for the autonomous wireless sensor microsystem.Here,we report a novel beta/photovoltaic cell based on free-standing Zn O nanorod arrays(ZNRAs)modified with metallic single-walled carbon nanotubes(m-SWCNTs),using radioisotope63 Ni as beta-emitting source.The ZNRAs were grown on Al-doped Zn O(AZO)conductive glass using hydrothermal method.The optimum length and diameter of Zn O nanorods were determined by Monte Carlo simulation for beta energy deposition in ZNRAs.The m-SWCNTs were anchored into the ZNRAs to form a three-dimensional(3-D)Schottky junction structure for effectively separating the beta/photo-excited electron-hole pairs.Experimentally,the betavoltaic and photovoltaic effects were confirmed through the I-V measurements of beta/photovoltaic cells under beta/UV/Vis irradiations,respectively.It is suggested that the m-SWCNTs play key role for the enhancement of beta/photovoltaic performance through the formation of extensive3-D Schottky junction,the conductive network for hole transport,and the surface plasmon resonance exciton absorption for visible light.Changsong Chen Jiang Chen Zhen Wang Jian Zhang Haisheng San Shichao Liu Chunyu Wu Werner Hofmann 2020Journal of Materials Science & Technology2020,54,19:3
19Fractional Modeling and Analysis of Coupled MR Damping System显示文摘The coupled magnetorheological(MR) damping system addressed in this paper contains rubber spring and magnetorheological damper. The device inherits the damping merits of both the rubber spring and the magnetorheological damper.Here a fractional-order constitutive equation is introduced to study the viscoelasticity of the combined damper. An introduction to the definitions of fractional calculus, and the transfer function representation of a fractional-order system are given. The fractional-order system model of a magnetorheological vibration platform is set up using fractional calculus, and the function of displacement is presented. 20Agricultural Risk Factors Influence Microbial Ecology in Honghu Lake显示文摘Agricultural activities, including stock-farming, planting industry, and fish aquaculture,can affect the physicochemical and biological characters of freshwater lakes. However, the effects of pollution producing by agricultural activities on microbial ecosystem of lakes remain unclear.Hence, in this work, we selected Honghu Lake as a typical lake that is influenced by agriculture activities. We collected water and sediment samples from 18 sites, which span a wide range of areas from impacted and less-impacted areas. We performed a geospatial analysis on the composition of microbial communities associated with physicochemical properties and antibiotic pollution of samples. The co-occurrence networks of water and sediment were also built and analyzed. Our results showed that the microbial communities of impacted and less-impacted samples of water were largely driven by the concentrations of TN, TP, NO_3^--N, and NO_2^--N, while those of sediment were affected by the concentrations of Sed-OM and Sed-TN. Antibiotics have also played important roles in shaping these microbial communities: the concentrations of oxytetracycline and tetracycline clearly reflected the variance in taxonomic diversity and predicted functional diversity between impacted and less-impacted sites in water and sediment samples, respectively. Furthermore, for samples from both water and sediment, large differences of network topology structures between impacted and less-impacted were also observed.
