1Middle segmental pancreatectomy: A safe and organ-preserving option for benign and low-grade malignant lesions显示文摘AIM: To study the feasibility and safety of middle segmental pancreatectomy (MSP) compared with pancreaticoduodenectomy (PD) and extended distal pancreatectomy (EDP). METHODS: We studied retrospectively 36 cases that underwent MSP, 44 patients who underwent PD, and 26 who underwent EDP with benign or low-grade malignant lesions in the mid-portion of the pancreas, between April 2003 and December 2009 in Ruijin Hospital. The perioperative outcomes and long-term outcomes of MSP were compared with those of EDP and PD. Periop-erative outcomes included operative time, intraoperative hemorrhage, transfusion, pancreatic fistula, intraabdominal abscess/infection, postoperative bleeding, reoperation, mortality, and postoperative hospital time. Long-term outcomes, including tumor recurrence, newonset diabetes mellitus (DM), and pancreatic exocrine insufficiency, were evaluated. RESULTS: Intraoperative hemorrhage was 316.1 ± 309.6, 852.2 ± 877.8 and 526.9 ± 414.5 mL for the MSP, PD and EDP groups, respectively (P < 0.05). The mean postoperative daily fasting blood glucose level was significantly lower in the MSP group than in the EDP group (6.3 ± 1.5 mmol/L vs 7.3 ± 1.5 mmol/L, P < 0.05). The rate of pancreatic fistula was higher in the MSP group than in the PD group (42% vs 20.5%, P = 0.039), all of the fistulas after MSP corresponded to grade A (9/15) or B (6/15) and were sealed following conservative treatment. There was no significant difference in the mean postoperative hospital stay between the MSP group and the other two groups. After a mean follow-up of 44 mo, no tumor recurrences were found, only one patient (2.8%) in the MSP group vs five (21.7%) in the EDP group developed new-onset insulin-dependent DM postoperatively (P = 0.029). Moreover, significantly fewer patients in the MSP group than in the PD (0% vs 33.3%, P < 0.001) and EDP (0% vs 21.7%, P = 0.007) required enzyme substitution. 2A comparative study of the characteristics of two Coxsackie A virus type 16 strains (genotype B)显示文摘Coxsackie A virus is one of the major pathogens associated with hand, foot and mouth disease (HFMD). The etiological characteristics of Coxsackie A virus type 16 (CA16) are thought to correlate with the pathological process of its infection. Two CA16 strains that were isolated from a severe HFMD patient presented with different plaque forms. This observation, along with biological analysis, indicated that the differences in the strains' biological characteristics, such as proliferation kinetics and immunogenicity, correlate with differences in their pathogenicity toward neonatal mice. Furthermore, these differences are thought to be associated with the sequence of the 5′ non-coding region of the viral genome and the VP1 structural region sequence. 3Transcriptional regulation of the Herpes Simplex Virus 1α-gene by the viral immediate-early protein ICP22 in association with VP16显示文摘Herpes Simplex Virus 1 (HSV1) is capable of inducing two forms of infection in individuals, and the establishment of which type of infection occurs is linked to the transcriptional activation of viral α genes. One of the HSV1 α genes, ICP22, is known to have multiple functions during virus replication, but its distinct roles are still unclear. This study showed that ICP22 functions as a general repressor for certain viral and cellular promoters, and this transcriptional repression by ICP22 is independent of the specific upstream promoter element, as shown using the CAT enzyme assay system. Further work also found that VP16 interfered with ICP22 mediated transcriptional repression of the viral α4 gene, through interactions with specific elements upstream of the α4 gene promoter. 4Integrated traffic-power simulation framework for electric vehicle charging stations based on cellular automaton显示文摘This paper presents a novel integrated framework for simulating the charging load of an electric vehicle charging station. The framework is built based on the cellular automaton model, including five modules: vehicle generation, charging station, lane change, speed update and boundary clear. The proposed framework can effectively stimulate the system dynamics of the traffic system and charging power process in a charging station. 5Immediate-early gene product ICP22 inhibits the trans-transcription activating function of P53-mdm-2显示文摘As a product of HSVI immediate-early gene, ICP22 is capable of interacting with various cellular tran-scriptive and regulatory molecules during viral infection so as to impact the normal cellular molecular mechanism. ICP22 expressed in transfected cells can push the cells’ entering into S phase with binding to mdm-1 promoter region and impact its trans-transcription activating effect by P53. 10Transcriptome analysis suggests mechanisms for a novel flowering type: Cleistogamous wheat显示文摘Wheat is one of the most important staple crops worldwide.Fusarium head blight severely reduces wheat yield and quality.Cultivation of a novel type of cleistogamous wheat mutant,ZK001,which was created by static magnetic field treatment,is a new strategy for controlling Fusarium head blight.However,little is known about the mechanism of cleistogamy in wheat.The present study demonstrated that anthers of ZK001 were retained on the glumes at all flowering stages,whereas those of YM18 were extruded from the paleae and lemmae.There was a clear difference in the morphological characteristics of lodicules between YM18 and ZK001.Lodicule calcium and potassium contents were significantly higher in YM18 than in ZK001 from white to yellow anther stages.In Fusarium head blight resistance,the diseased kernel rate and deoxynivalenol content of ZK001 were markedly lower than those of YM18 and QM725.Comparative transcriptome analysis of YM18 and ZK001 was performed to identify regulatory mechanisms of cleistogamy.The main differentially expressed genes identified in the spikelets of YM18 and ZK001 at the green anther stage were associated with cell walls,carbohydrates,phytohormones,water channel,and ion binding,transport,and homeostasis.These differentially expressed genes may play an important role in regulating cellular homeostasis,osmotic pressure,and lodicule development.The results indicate that ZK001 lost the ability to push the lemmae and paleae apart during the flowering stage because of the thin lodicules.ZK001 was speculated to provide structural barriers for Fusarium head blight during the flowering stage.The thin lodicule of ZK001 results from low levels of soluble sugar,calcium ions,and potassium ions in the lodicules.These levels are regulated by differentially expressed genes.Caiguo Tang Minghao Li Minghui Cao Ruiju Lu Huilan Zhang Chenghong Liu Shengwei Huang Pingping Zhang Hao Hu Weiwei Zhao Lifang Wu 2020The Crop Journal2020,8,2:1

15Non-Ischemic, Non-Hypoxic Myocardial Injury, and Long-Term Mortality in Patients with Coronavirus Disease 2019: A Retrospective Cohort Study显示文摘Objective:Cardiac damage is commonly reported in patients with coronavirus disease 2019(COVID-19)but its prevalence and impact on the long-term survival of patients remain uncertain.This study aimed to explore the prevalence of myocardial injury and assess its prognostic value in patients with COVID-19.Methods:A single-center,retrospective cohort study was performed at the Affiliated Hospital of Jianghan University.Data from 766 patients with confirmed COVID-19 who were hospitalized from December 27,2019 to April 25,2020 were collected.Demographic,clinical,laboratory,electrocardiogram,treatment data and all-cause mortality during follow-up were collected and analyzed.Results:Of the 766 patients with moderate to critically ill COVID-19,86(11.2%)died after a mean follow-up of 72.8 days.Myocardial injury occurred in 94(12.3%)patients.The mortality rate was 64.9%(61/94)and 3.7%(25/672)in patients with and without myocardial injury,respectively.Cox regression showed that myocardial injury was an independent risk factor for mortality(hazard ratio:8.76,95%confidence interval:4.76–16.11,P<0.001).Of the 90 patients with myocardial injury with electrocardiogram results,sinus tachycardia was present in 29,bundle branch block in 26,low voltage in 10,and abnormal T-wave in 53.Conclusions:COVID-19 not only involves pneumonia but also cardiac damage.Myocardial injury is a common complication and an independent risk factor for mortality in COVID-19 patients.Fajiu Li Xijie Zhu Ziyang Zhu Yinjian Yang Zhuang Tian Duolao Wang Shi Chen Xiaoyan Gao Yalin Xu Bo Zhang Wei Yu Min Liu Xiqi Xu Chenghong Li Shuyang Zhang 2022Cardiology Discovery2022,2,2:0
16Transcriptome Profiling of Barley Cultivar Hua 30 MDEC in Response to Agrobacterium Infection显示文摘Agrobacterium-mediated transformation has been widely used in plants.However,the mechanism in plant cells’response to Agrobacterium infection was very complex.The mechanism of the determinants in host cell remains obscure,especially in barley,which is recalcitrant for Agrobacterium-mediated transformation.In the present study,microspore-derived embryogenic calli(MDEC)from barley elite cultivar were employed as unique subjects to characterize the mechanisms during the Agrobacterium infection process.Hua 30 MDEC can be successfully infected by Agrobacterium.RNA-sequencing at different infection points(0,2,6,12,24 hpi)was performed.The average expressional intensity of the whole genomics increased from 0 to 2 hpi,and then decreased subsequently.More upregulated than downregulated differentially expressed genes(DEGs)were counted at the same time.GO enrichment analysis showed that protein modification was significantly overrepresented in upregulated DEGs.Chromosome-related biological processes,gene expression and cellular metabolic processes were significantly overrepresented in downregulated DEGs.KEGG analysis showed that plant defense responses,phenylpropanoid biosynthesis and biosynthesis of amino acids were significantly enriched across the infection time course.Nine DEGs related to defense responses were identified.All DEGs were upregulated from 2 to 24 hpi.We speculate that these genes are possibly related to Agrobacterium infection.These findings will provide deep insights into the molecular events occurring during the process of Agrobacterium-mediated transformation.Yingbo Li Ting He Guimei Guo Hongwei Xu Yingjie Zong Shuwei Zhang Ruiju Lu Longhua Zhou Chenghong Liu 2022Phyton-International Journal of Experimental Botany2022,91,6:0
17Generating homozygous mutant populations of barley microspores by ethyl methanesulfonate treatment显示文摘Induced mutations are important for genetic research and breeding.Mutations induced by physical or chemical mutagenesis are usually heterozygous during the early generations.However,mutations must be fixed prior to phenotyping or field trials,which requires additional rounds of self-pollination.Microspore culture is an effective method to produce double-haploid(DH)plants that are fixed homozygotes.In this study,we conducted ethyl methanesulfonate(EMS)-induced mutagenesis of microspore cultures of barley(Hordeum vulgare)cultivar‘Hua30’and landrace‘HTX’.The EMS concentrations were negatively correlated with the efficiency of callus induction and the frequency of mutant plant regeneration.The two genotypes showed different regeneration efficiencies.The phenotypic variation of the regenerated M1 plants and the presence of genome-wide nucleotide mutations,revealed by whole-genome sequencing,highlight the utility of EMS-induced mutagenesis of isolated microspore cultures for developing DH mutants.Genome-wide analysis of the mutation frequency in the regenerated plants revealed that a considerable proportion of mutations resulted from microspore culture(somaclonal variation)rather than EMS-induced mutagenesis.In addition to producing a population of 1972 homozygous mutant lines that are available for future field trials,this study lays the foundation for optimizing the regeneration efficiency of DH plants and the richness of mutations(mainly by fine-tuning the mutagen dosage).Linli Huang Guangqi Gao Congcong Jiang Guimei Guo Qiang He Yingjie Zong Chenghong Liu Ping Yang 2023aBIOTECH2023,4,3:0

19Genome-Wide Identification of Candidate Genes Associated with β-glucan Traits in a Hulled and Hulless Barley (Hordeum vulgare L.) Population显示文摘Barley grain is a valuable source ofβ-glucan,which is an important component of dietary fiber with significant human health benefits.Although the genetic basis ofβ-glucan biosynthesis has been widely studied,a genome-wide association study(GWAS)is still required for a scan of the candidate genes related to the complex quantitative trait based on the high-quality barley reference genome.In this study,a GWAS was conducted using a population composed of 87 barley landraces(39 hulled and 48 hulless,β-glucan from 2.07%to 6.56%)with 191,098 nucleotide polymorphisms(SNPs)markers to cover the chromosomes with the highest density.The population was divided into four sub-populations(POP1~POP4),and theβ-glucan content in POP2 was significantly higher than that in other groups,in which most of the hulless barley landraces are from Qinghai-Tibet Plateau in China.Among seven SNP markers identified by GWAS,two(SNP2 and SNP3)of them showed positive correlation toβ-glucan trait and the remaining five(SNP1,SNP4,SNP5,SNP6 and SNP7)showed the negative relationship.Two candidate genes linked to SNP7,HORVU7Hr1G000320 and HORVU7Hr1G000040,belong to the nucleotide triphosphate hydrolase super-family which is probable to affect the activities ofβ-glucan synthase.Another candidate gene associated with SNP1,HORVU1Hr1G000010,is possibly involved in sugar response.In conclusion,our results provide new insights into the genetic basis ofβ-glucan accumulation in barley grains,and the discovery of new SNP markers distributed in each chromosome and the associated candidate genes will be valuable for the breeding of functional barley varieties with high β-glucan.Longhua Zhou Ting He Guimei Guo Jing Li Ruiju Lu Yingbo Li Chenghong Liu 2020Phyton-International Journal of Experimental Botany2020,89,4:0
