1Experimental infection of tree shrews(Tupaia belangeri) with Coxsackie virus A16显示文摘Coxsackie virus A16(CA16) is commonly recognized as one of the main human pathogens of hand-foot-mouth disease(HFMD). The clinical manifestations of HFMD include vesicles of hand, foot and mouth in young children and severe inflammatory CNS lesions. In this study, experimentally CA16 infected tree shrews(Tupaia belangeri) were used to investigate CA16 pathogenesis. The results showed that both the body temperature and the percentages of blood neutrophilic granulocytes / monocytes of CA16 infected tree shrews increased at 4-7 days post infection. Dynamic distributions of CA16 in different tissues and stools were found at different infection stages. Moreover, the pathological changes in CNS and other organs were also observed. 2A Comparison of the Biological Characteristics of EV71 C4 Subtypes from Different Epidemic Strains显示文摘The comparative analysis of the biological characterization and the genetic background study of EV71 circulating strains is commonly recognized as basic work necessary for development of an effective EV71 vaccine. In this study, we sequenced five EV71 circulating strains, isolated from Fuyang, Hefei, Kunming and Shenzhen city of China and named them FY-23, FY-22, H44, K9 and S1 respectively. The sequence alignment demonstrated their genotypes be C4. The genetic distance of the VP1 gene from these isolates suggested that they were highly co-related with genetic identity similar to other previously reported EV71 strains in China. Additionally, these strains were identified to display some obvious proliferation dynamics and plaque morphology when propagated in Vero cells. However, a distinctive difference in pathogenic ability in neonatal mice was found. Some differences in cross neutralization test & immunogenic analysis were also found. 4Novel Ru - K/Carbon Nanotubes Catalyst for Ammonia Synthesis显示文摘A novel ammonia synthesis catalyst, potassium-promoted ruthenium supported on carbon nanotubes, was developed. It was found that the Ru-K/carbon nanotubes catalyst had higher activity for ammonia synthesis (20.85 ml NH3/h/g-cat ) than the Ru-K/fullerenes (13.3 ml NH3/h/g-cat ) at atmospheric pressure and 623 K. The catalyst had activity even at 473 K. and had the highest activity (23.46 ml NH3/h/g-cat ) at 643 K. 6A microRNA encoded by HSV-1 inhibits a cellular transcriptional repressor of viral immediate early and early genes显示文摘Viral microRNAs are one component of the RNA interference phenomenon generated during viral infection. They were first identified in the Herpesviridae family, where they were found to regulate viral mRNA translation. In addition, prior work has suggested that Kaposi's sarcoma-associated herpesvirus (KSHV) is capable of regulating cellular gene transcription by miRNA. We demonstrate that a miRNA, hsv1-mir-H27, encoded within the genome of herpes simplex virus 1 (HSV-1), targets the mRNA of the cellular transcriptional repressor Kelch-like 24 (KLHL24) that inhibits transcriptional efficiency of viral immediate-early and early genes. The viral miRNA is able to block the expression of KLHL24 in cells infected by HSV-1. 7Attenuated phenotypes and analysis of a herpes simplex virus 1strain with partial deletion of the UL7, UL41 and LAT genes显示文摘We previously constructed a herpes simplex virus 1(HSV-1) UL7 mutant virus(M1) and showed that a partial deletion mutation of the UL7 gene led to a lower proliferative rate and an attenuated phenotype. Using the M1 mutant, we further modified the UL41 gene, which encodes another tegument protein, and the latency-associated transcript(LAT) gene. Observations of the resulting mutants with modified UL7 and UL41(M2) or UL7, UL41 and LAT(M3) genes indicated attenuated phenotypes, with lower proliferative ratios in various cells, non-lethal infections in mice and lower viral loads in nervous tissues compared with the wild-type strain. Furthermore, no LAT stable intron could be detected in the trigeminal ganglion of M3-infected animals. The results obtained with the three HSV-1 mutants indicate that the M3 mutant is an attenuated strain with low pathogenicity during both acute and latent infections. 8Validity assessment and determination of the cutoff value for the Index of Complexity, Outcome and Need among 12-13 year-olds in Southern Chinese显示文摘To validate the use of the Index of Complexity,Outcome and Need (ICON) in assessing orthodontic treatment need among 12-13 year-olds in southern China,we determined the threshold value of ICON based on Chinese orthodontists' judgments. The samples consisted of 335 students in grade 7 from 16 randomly selected middle schools in Chengdu,China. Three associate professors provided ICON scores for each participant and the results were compared with the gold standard judgments from 25 experts on treatment needs. Based on the gold standard,195 casts belonged to the treatment category,while the rest 140 belonged to the no-treatment category. With the international cutoff point of 43,the sensitivity and specificity of the ICON score were 0.29 and 0.98. The best compromise between sensitivity and specificity in Chengdu,compared with the gold standard,was found at a cutoff point of 29,and the sensitivity and specificity were 0.88 and 0.83. 9A comparative study of the characteristics of two Coxsackie A virus type 16 strains (genotype B)显示文摘Coxsackie A virus is one of the major pathogens associated with hand, foot and mouth disease (HFMD). The etiological characteristics of Coxsackie A virus type 16 (CA16) are thought to correlate with the pathological process of its infection. Two CA16 strains that were isolated from a severe HFMD patient presented with different plaque forms. This observation, along with biological analysis, indicated that the differences in the strains' biological characteristics, such as proliferation kinetics and immunogenicity, correlate with differences in their pathogenicity toward neonatal mice. Furthermore, these differences are thought to be associated with the sequence of the 5′ non-coding region of the viral genome and the VP1 structural region sequence. 10Wheat breeding history reveals synergistic selection of pleiotropic genomic sites for plant architecture and grain yield显示文摘Diversity surveys of crop germplasm are important for gaining insights into the genomic basis for plant architecture and grain yield improvement,which is still poorly understood in wheat.In this study,we exome sequenced 287 wheat accessions that were collected in the past 100 years.Population genetics analysis identified that 6.7%of the wheat genome falls within the selective sweeps between landraces and cultivars,which harbors the genes known for yield improvement.These regions were asymmetrically distributed on the A and B subgenomes with regulatory genes being favorably selected.Genome-wide association study(GWAS)identified genomic loci associated with traits for yield potential,and two underlying genes,TaARF12 encoding an auxin response factor and TaDEP1 encoding the G-proteinγ-subunit,were located and characterized to pleiotropically regulate both plant height and grain weight.Elite single-nucleotide haplotypes with increased allele frequency in cultivars relative to the landraces were identified and found to have accumulated over the course of breeding.Interestingly,we found that TaARF12 and TaDEP1 function in epistasis with the classical plant height Rht-1 locus,leading to propose a“Green Revolution”-based working model for historical wheat breeding.Collectively,our study identifies selection signatures that fine-tune the gibberellin pathway during modern wheat breeding and provides a wealth of genomic diversity resources for the wheat research community.Aili Li Chenyang Hao Zhenyu Wang Shuaifeng Geng Meiling Jia Fang Wang Xiang Han Xingchen Kong Lingjie Yin Shu Tao Zhongyin Deng Ruyi Liao Guoliang Sun Ke Wang Xingguo Ye Chengzhi Jiao Hongfeng Lu Yun Zhou Dengcai Liu Xiangdong Fu Xueyong Zhang Long Mao 2022Molecular Plant2022,15,3:4

12Differential transcriptome analysis between Populus and its synthesized allotriploids driven by second-division restitution显示文摘In this report, we compared transcriptomic differences between a synthetic Populus section Tacamahaca triploid driven by second-division restitution and its parents using a high-throughput RNA-seq method. A total of 4,080 genes were differentially expressed between the high-growth vigor allotriploids(SDR-H) and their parents, and 719 genes were non-additively expressed in SDR-H. Differences in gene expression between the allotriploid and male parent were more signi ficant than those between the allotriploid and female parent, which may be caused by maternal effects. We observed3,559 differentially expressed genes(DEGs) between the SDR-H and male parent. Notably, the genes were mainly involved in metabolic process, cell proliferation, DNA methylation, cell division, and meristem and developmental growth. Among the1,056 DEGs between SDR-H and female parent, many genes were associated with metabolic process and carbon utilization.In addition, 1,789 DEGs between high- and low-growth vigor allotriploid were mainly associated with metabolic process,auxin poplar transport, and regulation of meristem growth.Our results indicated that the higher poplar ploidy level can generate extensive transcriptomic diversity compared with its parents. 13FeCo alloy@N-doped graphitized carbon as an efficient cocatalyst for enhanced photocatalytic H2 evolution by inducing accelerated charge transfer显示文摘Cocatalysts play important roles in improving the activity and stability of most photocatalysts.It is of great significance to develop economical,efficient and stable cocatalysts.Herein,using Na2CoFe(CN)6 complex as precursor,a novel noble-metal-free FeCo@NGC cocatalyst(nano-FeCo alloy@N-doped graphitized carbon) is fabricated by a simple pyrolysis method.Coupling with g-C3 N4, the optimal FeCo@NGC/g-C3N4 receives a boosted visible light driven photocatalytic H2 evolution rate of 42.2 μmol h-1, which is even higher than that of 1.0 wt% Pt modified g-C3N4 photocatalyst.Based on the results of density functional theory(DFT) calculations and practical experiment measurements,such outstanding photocatalytic performance of FeCo@NGC/g-C3N4 is mainly attributed to two aspects.One is the accelerated charge transfer behavior,induced by a photogene rated electrons secondary transfer performance on the surface of FeCo alloy nanoparticles.The other is related to the adjustment of H adsorption energy(approaching the standard hydrogen electrode potential) by the presence of external NGC thin layer.Both factors play key roles in the H2 evolution reaction.Such outstanding performance highlights an enormous potential of developing noble-metal-free bimetallic nano-alloy as inexpensive and efficient cocatalysts for solar applications.Sibo Chen Yun Hau Ng Jihai Liao Qiongzhi Gao Siyuan Yang Feng Peng Xinhua Zhong Yueping Fang Shengsen Zhang 2021Journal of Energy Chemistry2021,30,1:3
14Interfacial characteristics and mechanical properties of additive manufacturing martensite stainless steel on the Cu-Cr alloy substrate by directed energy deposition显示文摘Copper/steel is a typical bimetal functional material,combining the excellent electrical and thermal conductivity of copper alloy and the high strength and hardness of stainless steel.There has been recent interest in manufacturing copper/steel bimetal by directed energy deposition(DED)due to its layer-bylayer method.However,cracks tend to form on the copper/steel interface because of the great difference in thermal expansion coefficient and crystal structure between copper and steel.In this work,interfacial characteristics and mechanical properties of the copper/steel bimetal were studied from one layer to multilayers.The laser power has a great influence on the Cu element distribution of the molten pool,affecting the crack formation dramatically on the solidification stage.Cracks tend to form along columnar grain boundaries because of the Cu-rich liquid films and spherical particles in the cracks.Crack-free and good metallurgical bonding copper/steel interface is formed at a scanning velocity of 800 mm/min and the laser power of 3000 W.The ultimate tensile strength(UTS)and the break elongation(EL)of the vertically combined crack-free copper/steel bimetal are 238.2±4.4 MPa and 20.6±0.7%,respectively.The fracture occurs on the copper side instead of the copper/steel interface,indicating that the bonding strength is higher than that of the Cu-Cr alloy.The UTS of the horizontally combined crack-free copper/steel bimetal is 746.7±22.6 MPa,which is 200%higher than that of the Cu-Cr alloy substrate.The microhardness is 398.6±5.4 HV at the steel side and is 235.3±64.1 HV at the interface,which is400%higher than that of the Cu-Cr alloy substrate.This paper advances the understanding of the interfacial characteristics of heterogeneous materials and provides guidance and reference for the fabrication of multi-material components by DED.Wenqi Zhang Hailong Liao Zhiheng Hu Shasha Zhang Baijin Chen Huanqing Yang Yun Wang Haihong Zhu 2021Journal of Materials Science & Technology2021,,31:2
15Role of LAP^+CD4^+ T cells in the tumor microenvironment of colorectal cancer显示文摘AIM To investigate the abundance and potential functions of LAP^+CD4^+ T cells in colorectal cancer(CRC). METHODS Proportions of LAP^+CD4^+ T cells were examined in peripheral blood and tumor/paratumor tissues of CRC patients and healthy controls using flow cytometry. Expression of phenotypic markers such as forkhead box(Fox)p3, cytotoxic T-lymphocyte-associated protein(CTLA)-4, chemokine CC receptor (CCR)4 and CCR5 was measured using flow cytometry. LAP^-CD4^+ and LAP^+CD4^+ T cells were isolated using a magnetic cellsorting system and cell purity was analyzed by flow cytometry. Real-time quantitative polymerase chain reaction was used to measure expression of cytokines interleukin (IL)-10 and transforming growth factor(TGF)-β.RESULTS The proportion of LAP^+CD4^+ T cells was significantly higher in peripheral blood from patients (9.44% ± 3.18%) than healthy controls (1.49% ± 1.00%, P < 0.001). Among patients, the proportion of LAP^+CD4^+ T cells was significantly higher in tumor tissues(11.76% ± 3.74%) compared with paratumor tissues (3.87% ± 1.64%, P < 0.001). We also observed positive correlations between the proportion of LAP^+CD4^+ T cells and TNM stage(P < 0.001), distant metastasis(P < 0.001) and serum level of carcinoembryonic antigen(P < 0.05). Magnetic-activated cell sorting gave an overall enrichment of LAP^+CD4^+ T cells (95.02% ± 2.87%), which was similar for LAP^-CD4^+ T cells(94.75% ± 2.76%). In contrast to LAP^-CD4^+ T cells, LAP^+CD4^+ T cells showed lower Foxp3 expression but significantly higher levels of CTLA-4, CCR4 and CCR5(P < 0.01). 19Improvement of microalgae lipid productivity and quality in an ion-exchange-membrane photobioreactor using real municipal wastewater显示文摘To improve the productivity and quality of microalgae-based biodiesel when using municipal wastewater(MW)as nutrients source,an ion-exchange-membrane photobioreactor(IEM-PBR)was used in this study to eliminate the negative effects of pollutants in MW on microalgae Chlorella vulgaris and Scenedesmus obliquus.In the IEM-PBR,the real MW and microalgae cultures were separated in two chambers by the ion-exchange-membranes(IEMs).Nutrients(N,P,etc.)in the MW permeated into microalgae cultures through the IEMs,while pollutants(suspended solids,competitors,etc.)in the MW could hardly permeate into microalgae cultures.As a result,the lipid productivity in the IEM-PBR was improved to 85.7 mg/(L·d)for C.vulgaris and 111.8 mg/(L·d)for S.obliquus,which was slightly higher than that in the traditional photobioreactor(T-PBR)with real MW after centrifugation(82.5 mg/(L·d)for C.vulgaris and 105.8 mg/(L·d)for S.obliquus),but much higher than that in the T-PBR with untreated MW and primary MW(with lipid productivity of 20-30 mg/(L·d)).Besides,the lipid quality obtained in the IEM-PBR had higher proportion of cetane number(ca.60%)and lower linolenic acid content(ca.8%),which showed a superior quality in the IEM-PBR to that in the T-PBR.It demonstrated that the IEM-PBR is an effective approach to improve the productivity and quality of microalgae biodiesel.Chang Haixing Fu Qian Huang Yun Xia Ao Liao Qiang Zhu Xun 2017International Journal of Agricultural and Biological Engineering2017,10,1:2
20A cellular response protein induced during HSV-1 infection inhibits viral replication by interacting with ATF5显示文摘Studies of herpes simplex virus type 1(HSV-1)infection have shown that many known and unknown cellular molecules involved in viral proliferation are up-regulated following HSV-1 infection.In this study,using two-dimensional polyacrylamide gel electrophoresis,we found that the expression of the HSV-1 infection response repressive protein(HIRRP,GI 16552881)was up-regulated in human L02 cells infected with HSV-1.HIRRP,an unknown protein,was initially localized in the cytoplasm and then translocated into the nucleus of HSV-1-infected cells.Further analysis showed that HIRRP represses HSV-1proliferation by inhibiting transcription of the viral genome by interacting with the cellular transcription factor,ATF5,via its N-terminal domain.ATF5 represses the transcription of many host genes but can also act as an activator of genes containing a specific motif.We found that ATF5 promotes the proliferation of HSV-1 via a potential mechanism by which ATF5 enhances the transcription of viral genes during the course of an HSV-1 infection;HIRRP then induces feedback repression of this transcription by interacting with ATF5.WU LianQiu ZHANG XueMei CHE YanChun ZHANG Ying TANG SongQing LIAO Yun NA RuiXiong XIONG XiangLin LIU LongDing LI QiHan 2013Science China(Life Sciences)2013,56,12:2
