12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 2Effect of suspended sediment grain size on channel sedimentation in the lower Yellow River and some implications显示文摘Based on the data of suspended sediment transport and channel sedimentation in various grain size fractions in the period of 1962―1985, the relationship between channel sedimentation in the lower Yellow River and sediment input has been plotted with respect to each grain size fraction. Several fill-scour thresholds in sediment input have been identified from these graphs. It was found that the fill-scour threshold in sediment input decreases with the increase in fraction grain size. The correlation coefficient between channel sedimentation and sediment input becomes larger with the increasing fraction grain size, indicating that channel sedimentation depends more on coarser grain size fractions than on smaller ones. The fraction channel sedimentation induced by unit change of fraction sediment input increases with grain size. Of the input of sediment larger than 0.025 mm, 43.73% was deposited on the channel, and for inputs of sediments larger than 0.05 mm and larger than 0.10 mm, 76.61% and 97.68% were deposited on the channel, respectively. Thus, for reduction of each ton of sediment larger than 0.10 mm from the drainage basin, the resultant reduction in channel sedimentation in the lower Yellow River would be 1.275 times that for the sediment larger than 0.10 mm, and 2.234 times that for the sediment larger than 0.025 mm. 3White Matter Deficits Underlying the Impaired Consciousness Level in Patients with Disorders of Consciousness显示文摘In this study, we aimed to(1) identify white matter(WM) deficits underlying the consciousness level in patients with disorders of consciousness(DOCs) using diffusion tensor imaging(DTI), and(2) evaluate the relationship between DTI metrics and clinical measures of the consciousness level in DOC patients. With a cohort of 8 comatose, 8 unresponsive wakefulness syndrome/vegetative state, and 14 minimally conscious state patients and 25 patient controls, we performed group comparisons of the DTI metrics in 48 core WM regions of interest(ROIs), and examined the clinical relevance using correlation analysis. We identified multiple abnormal WM ROIs in DOC patients compared with normal controls, and the DTI metrics in these ROIs were significantly correlated with clinical measures of the consciousness level. 4Hepatitis B virus X protein modulates the apoptosis of hepatoma cell line induced by TRAIL显示文摘The purpose of this study is to observe the effects of HBx on the apoptosis of hepatoma cells induced by TNF-related apoptosis-inducing ligand (TRAIL) and to study prelimi- nary molecular mechanisms for its effects. In order to set up a model in vitro, BEL7402-HBx cell line, stably expressing HBx mRNA, was established by stable transfection of pcDNA-HBx, which contains HBx gene, into hepatoma cell line BEL7402. Control cell line BEL7402-cDNA3, stably transfected with pcDNA3, was set up simultaneously as a control. Trypan blue exclusion test, caspase 3 activity detection and TUNEL assay were performed to detect the apoptosis of BEL7402, BEL7402-cDNA3, BEL7402-HBx induced by TRAIL. The expression of TRAIL recep- tors in three groups was analyzed by Flow cytometry. In addition, phosphorothioated antisense oligonucleotide against the translation initial region of HBx gene (PS-asODNs/HBx) was used to block the expression of HBx in HepG2.2.15 cells and to further confirm the effects of HBx on TRAIL-induced apoptosis. Trypan blue exclusion test indicated that TRAIL had a dose-dependent cytotoxicity on BEL7402, BEL7402-cDNA3 and BEL7402-HBx cells. Under treatment of the same concentration of TRAIL, BEL7402-HBx had a higher apoptosis rate and a higher level of Caspase 3 activation than BEL7402 and BEL7402-cDNA3. TUENL assay showed that the apoptosis rate of BEL7402-HBx induced by 10 μg/L TRAIL was 41.4%±7.2%, signifi- cantly higher than that of BEL7402 and BEL7402-cDNA3 cells. Blockade of HBx expression in Hep G2.2.15 cells partly inhibited the apoptosis induced by TRAIL. The introduction or blockade of HBx did not change the expression pattern of TRAIL receptors. The present study firstly con- firms the effects of HBx on TRAIL- induced apoptosis from two different points and it is not re- lated with the expression level of TRAIL receptors. 6Cytotoxicity of liver targeted drug-loaded alginate nanoparticles显示文摘In this study, novel liver targeted doxorubicin (DOX) loaded alginate (ALG) nanoparticles were prepared by CaCl2 crosslinking method. Glycyrrhetinic acid (GA, a liver targeted molecule) modified alginate (GA-ALG) was synthesized in a heterogeneous system, and the structure of GA-ALG and the substitu-tion degree of GA were analyzed by 1H NMR, FT-IR and elemental analysis. The drug release profile under the simulated physiological condition and cytotoxicity experiments of drug-loaded GA-ALG nanoparticles were carried out in vitro. Transmission electron micrographs (TEM) and dynamic light scattering (DLS) analysis showed that drug-loaded GA-ALG nanoparticles have spherical shape structure with the mean hydrodynamic diameter around 214 ± 11 nm. 7Anti-PD-L1/TGF-BR fusion protein (SHR-1701) overcomes disrupted lymphocyte recovery-induced resistance to PD-1/PD-L1 inhibitors in lung cancer显示文摘Background:Second-generation programmed cell death-protein 1/pro-grammed death-ligand 1(PD-1/PD-L1)inhibitors,such as bintrafusp alfa(M7824),SHR-1701,and YM101,have been developed to simultaneously block PD-1/PD-L1 and transforming growth factor-beta/transforming growth factor-beta receptor(TGF-P/TGF-βR).Consequently,it is necessary to identify predictive factors of lung cancer patients who are not only resistant to PD-1/PD-LI inhibitors but also sensitive to bifunctional drugs.The purpose of this study was to search for such predictors.Methods:Multivariable Cox regression was used to study the association between the clinical outcome of treatment with PD-1/PD-L1 inhibitors and lym-phocyte recovery after lymphopenia in lung cancer patients.Murine CMT167 lung cancer cells were engineered to express the firefly luciferase gene and implanted orthotopically in the lung of syngeneic mice.Bioluminescence imag-ing,flow cytometry,and immunohistochemistry were employed to detrmine response to immunotherapy and function of tumor-infiltrating immune cells.Results:For lung cancer patients treated with anti-PD-1/PD-LI antibodies,poor lymphocyte recovery was associated with a shorter progression-free survival(PFS;P<0.001),an accumulation of regulatory T cells(Tregs),and an elimi-nation of CD8+T cells in the peripheral blood.Levels of CD8+T cells and Treg cells were also imbalanced in the tumors and peripheral immune organs of mice with poor lymphocyte recovery after chemotherapy.Moreover,these mice failed to respond to anti-PD-1 antibodies but remained sensitive to the anti-PD-LI/TGF-βR fusion protein(SHR-1701).Consistently,SHR-1701 but not anti-PD-1 antibod-ies,markedly enhanced IFN-γproduction and Ki-67 expression in peripheral CD8+T cells from patients with impaired lymphocyte recovery.Conclusions:Lung cancer patients with poor lymphocyte recovery and suffer-ing from persistent lymphopenia after previous chemotherapy are resistant to anti-PD-1/PD-L1 antibodies but might be sensitive to second-generation agents such as SHR-1701.Bo Cheng Kaikai Ding Pengxiang Chen Jianxiong Ji Tao Luo Xiaofan Guo Wei Qiu Chunhong Ma Xue Meng Jian Wang Jinming Yu Yuan Liu 2022Cancer Communications2022,42,1:5
8Reduced graphene oxide@carbon sphere based metacomposites for temperature-insensitive and efficient microwave absorption显示文摘With the increasing advance of fifth generation(5G)network and the gradual expansion of digital devices,harsh working environment for electronic devices has spawned higher requirements for microwave absorbing materials(MAMs).Since both the electromagnetic response and energy conversion character vary with temperature,to achieve temperature insensitive microwave absorption behaviour in wide temperature range has become extremely challenging.In this work,structured metacomposites containing sub-wavelength reduced graphene oxide(RGO)@carbon spheres were fabricated,and the microwave absorption was further improved through structural and composition design of the RGO@carbon units.Due to the unique anti-reflection effect on microwave of the metacomposites,the temperature-insensitive electromagnetic performance at elevated temperature could be exhibited.Moreover,both the dielectric relaxation behaviour and microwave absorption proformance of the system could be further increased.As a result,the effective absorption bandwidth(reflection loss(RL)<−10 dB)of the metacomposites with only 3.0 wt.%filler content could cover the entire X-band(8.2–12.4 GHz)frequency ranging from 298 to 473K.The metacomposite proposed in this work provides a“de-correlating”strategy to break the linkage between microwave absorption behaviour and temperature,which offers an interesting plateau for fabricating efficient high-temperature microwave absorption structures with tunable and designable advantages.Zhiyang Jiang Haoxu Si Yi Li Dan Li Huihui Chen Chunhong Gong Jingwei Zhang 2022Nano Research2022,15,9:4

10Loss of scleraxis leads to distinct reduction of mineralized intermuscular bone in zebrafish显示文摘Intermuscular bones(IBs)are ossified from tendons and only occur in lower teleosts.Positive association between the regulation of scleraxis gene(scx)and tendon development gave us reasons to speculate that the scx gene may play a potential role in regulating the development of IBs.A phylogenetic analysis conducted for this study revealed potential functional differentiation between two scx orthologues,scxa and scxb.The scxa^(-/-) and scxb^(-/-) zebrafish were generated through CRISPR-Cas9 technology to study the role of scx in the IB and rib development.The results showed a significant reduction of the number of IBs in adult scxa-1^(-/-) zebrafish,with almost 70%reduction(15-25 IBs)compared to the wild type scxa-1^(+/+)zebrafish(76-80 IBs).In the scxa-1^(+/+)adults,IBs were observed in both dorsal and tail segments;however,in scxa-1^(-/-)fish IBs were observed only in the tail segment(none in the dorsal segment).Although scxa-1^(-/-) zebrafish had rib defects,the mutants were viable and fertile as adult fish.The scxb^(-/-) zebrafish had the same number of IBs and same skeletal phenotype as the wild-type fish.This suggests that only scxa has a crucial role in the IB development,and confirms functional differentiation of scx orthologues.To further clarify the molecular mechanism by which scxa affects the IB development,we conducted comparative transcriptome analysis of dorsal tissue samples of scxa-1^(+/+)(with IBs)and scxa-1^(-/-) (without IBs),and further verified the expression of key genes via qPCR.This is the first study to identify a gene that controls the amount of IBs in fish,and it provides a new sight into the effects of scxa on the molecular mechanism of IB development in fish.Chunhong Nie Shiming Wan Yulong Chen Dejie Zhu Xudong Wang Xiaoru Dong Ze-Xia Gao 2021Aquaculture and Fisheries2021,6,2:3
11Recent Advances on Electrospun Nanofiber Materials for Post-lithium Ion Batteries显示文摘Lithium ion batteries(LIBs)have dominated the portable electric market over decades;however,the limited and unevenly distributed lithium resources induce concerns on their future large-scale applications.Increasing efforts have been endeavored on exploring post-Li ion batteries,such as Na-ion,K-ion,Al-ion and Mg-ion batteries,due to the high abundance of the corresponding elements in Earth crust.Manufacturing reliable electrode materials is the key to develop these new battery systems.Facile and scalable electrospinning has been widely utilized in preparing mechanically stable,flexible and conductive nanofiber electrodes as successfully proven in LIBs.In recent years,tremendous efforts have been devoted to electrospinning nanofiber electrodes for post-Li ion batteries and discernible progress in the electrochemical performance has been witnessed.Herein,we aim to review the-state-of-the-art advances made in electrospun nanofiber materials in optimizing post-Li ion battery technology by surveying the correlations among the morphology,the surface chemistry,the structure of electrospun nanofibers,and the post-Li ion batteries performance.Based on intensive investigations and insightful understandings,perspectives to the future design of electrospun nanofiber electrodes are also presented.Fangyi Shi Chunhong Chen Zheng-Long Xu 2021Advanced Fiber Materials2021,3,5:3
12Reactive MnO_(2) template-assisted synthesis of double-shelled PPy hollow nanotubes to boost microwave absorption显示文摘Microwave absorbing materials(MAMs)with wide effective absorption bandwidth(EAB)and low filling ratio are highly desirable for practical applications.Rational design in components and structures is one of the effective strategies to achieve MAMs with high performance.Herein,double-shelled hollow(DSH)polypyrrole(PPy)nanotubes were synthesized with hydrochloric acid(HCl)and sodium pstyrene sulfonate(SS)co-doping polymerization process using manganese dioxide(MnO_(2))nanorods as a self-sacrifice template.With the increase of HCl concentration,the 1D MnO_(2) core diminishes gradually to form the MnO_(2)@PPy coaxial nanostructures and finally the DSH PPy nanotube,which tunes the microwave absorption performance.Importantly,the DSH PPy nanotubes exhibit excellent microwave absorption of an optimal reflection loss of–50.4 dB and a wide EAB of 7.7 GHz with a low filling ratio of 5 wt%in a paraffin wax matrix.The excellent microwave absorption is believed to be mainly attributed to the enhanced synergistic effects of interfacial polarization and conduction loss arising from the unique DSH structure and the co-doping polymerization.Huiling Gu Ji Huang Na Li Hua Yang Gang Chen Chengjun Dong Chunhong Gong Hongtao Guan 2023Journal of Materials Science & Technology2023,,15:3

14OTUD5 promotes innate antiviral and antitumor immunity through deubiquitinating and stabilizing STING显示文摘Stimulator of interferon genes(STING)is an adaptor protein that is critical for effective innate antiviral and antitumor immunity.The activity of STING is heavily regulated by protein ubiquitination,which is fine-tuned by both E3 ubiquitin ligases and deubiquitinases.Here,we report that the deubiquitinase OTUD5 interacts with STING,cleaves its K48-linked polyubiquitin chains,and promotes its stability.Consistently,knockout of OTUD5 resulted in faster turnover of STING and subsequently impaired type I IFN signaling following cytosolic DNA stimulation.More importantly,Lyz2-Cre Otud5^(fl/Y) mice and CD11-Cre Otud5^(fl/Y) mice showed more susceptibility to herpes simplex virus type 1(HSV-1)infection and faster development of melanomas than their corresponding control littermates,indicating that OTUD5 is indispensable for STING-mediated antiviral and antitumor immunity.Our data suggest that OTUD5 is a novel checkpoint in the cGAS-STING cytosolic DNA sensing pathway.Yunyun Guo Fei Jiang Lingli Kong Haifeng Wu Honghai Zhang Xiaorong Chen Jian Zhao Baoshan Cai Yanqi Li Chunhong Ma Fan Yi Lei Zhang Bingyu Liu Yi Zheng Lingqiang Zhang Chengjiang Gao 2021Cellular & Molecular Immunology2021,18,8:2
15Pig target tracking algorithm based on multi-channel color feature fusion显示文摘In the process of tracking the target of the pig,with the change of the size of the tracking target in the video image,the estimated tracking target scale cannot be adaptively updated in real-time,resulting in the low accuracy of the tracking target.In this study,a multi-channel color feature adaptive fusion algorithm was proposed,and the target scale of the pig was updated in real-time by utilizing the contour information of the target pig.Experiments show that the proposed algorithm had a distance precision of 89.7%and an overlap precision of 87.5%,and the average running speed of this algorithm was 50.1 fps.The robustness of the proposed algorithm in tracking target deformation and scale variation were significantly improved,which satisfies the accuracy and real-time requirements of pig target tracking.Longqing Sun Shuaihua Chen Ting Liu Chunhong Liu Yan Liu 2020International Journal of Agricultural and Biological Engineering2020,13,3:2
16Clinical analysis of CHD2 gene mutations in pediatric patients with epilepsy显示文摘Importance:CHD2 is a member of the chromodomain helicase DNA-binding(CHD)family of proteins,which have important roles in the regulation of gene expression.Dysregulation of this protein may lead to various disorders.Objective:To delineate the genotypes and phenotypes of CHD2-related epilepsy.Methods:We analyzed the medical history,magnetic resonance imaging findings,and video-electroencephalogram recordings of 17 patients withCHD2 mutations in the Neurology Department of Beijing Children’s Hospital from June 2016 to June 2021.Results:Age at seizure onset ranged from 6 months to 10 years;the median age at onset was 4 years.Generalized tonic-clonic,myoclonic,eyelid myoclonic,atonic,atypical absence,myoclonic-atonic,and spasm seizures were observed.Ten of the 17 patients had multiple types of seizures.One patient exhibited photosensitivity epilepsy and one patient exhibited grid image-induced visual reflex epilepsy.Developmental disability was present in 14 patients,while autism features were present in five patients.Sixteen patients hadde novo mutations ofCHD2;one patient had an inherited variant.Eleven mutations were novel.One patient had two mutations;that patient exhibited development delay and refractory epilepsy.Seizures were controlled in eight patients,improved in seven patients,and resistant to treatment in two patients.Interpretation:Phenotype severity in patients withCHD2 variants ranged from drug-responsive seizures to severe epileptic encephalopathy.Most patients exhibited developmental disorders.Weixing Feng Fang Fang Xiaohui Wang Chunhong Chen Junlan Lu Jie Deng 2022Pediatric Investigation2022,6,2:2
17Ferroptosis:An emerging therapeutic opportunity for cancer显示文摘Ferroptosis,a new form of non-apoptotic,regulated cell death characterized by iron dependency and lipid peroxidation,is involved in many pathological conditions such as neurodegenerative diseases,heart ischemia/reperfusion injury,acute renal failure,and cancer.While metabolic dysfunctions can lead to excessive lipid peroxidation culminating in ferroptotic cell death,glutathione peroxidase 4(GPX4)resides in the center of a network that functions to prevent lipid hydroperoxides from accumulation,thereby suppressing ferroptosis.Indeed,RSL3 and other small-molecule GPX4 inhibitors can induce ferroptosis in not only cultured cancer cells but also tumor xenografts implanted in mice.Similarly,erastin and other system Xc−inhibitors can deplete intracellular glutathione required for GPX4 function,leading to lipid peroxidation and ferroptosis.As therapy-resistant cancer cells are sensitive to GPX4-targeted therapeutic regimens,the agents capable of inducing ferroptosis hold great promises to improve current cancer therapy.This review will outline the molecular basis of ferroptosis,but focus on the strategies and the agents developed in recent years for therapeutic induction of ferroptosis.The potentials of these ferroptosis-inducing agents,which include system Xc−inhibitors,GPX4 inhibitors,and iron-based nanoparticles,in cancer therapy will be subsequently discussed.Liyuan Wang Xiaoguang Chen Chunhong Yan 2022Genes & Diseases2022,9,2:2

20An integrated microfluidic device for long-term culture of isolated single mammalian cells显示文摘We developed an integrated microfluidic chip for long-term culture of isolated single cells. This polydimethylsiloxane (PDMS) based device could accurately seed each single cell into different culture chambers, and isolate one chamber from each other with monolithically integrated pneumatic valves. We optimized the culture conditions, including the frequency of medium replacement and the introduction of conditioned medium, to keep the single cells alive for 4 days. We cultured a few hundred cells in a separated chamber on the same chip to continuously supply the conditioned medium into the culture chambers for single cells. This approach greatly facilitated the growth of single cells, and created a suitable microenvironment for observing cells' autonomous process in situ without the interference of other adjacent cells.
