| 1 | The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy. | Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu | 2021 | Cancer Communications2021,41,1: | 5 |
| 2 | Sedimentary Conditions of Evaporites in the Late Jurassic Xiali Formation,Qiangtang Basin:Evidence from Geochemistry Records显示文摘The Qiangtang Basin(QB), located in the central Tibetan Plateau, is a Jurassic marine basin and one of the most important prospective salt resource belts in China. In recent decades, many outcrops of gypsiferous bed have been found in the Jurassic marine strata in the basin. Salt springs with abnormally high sodium(Na^+) contents had been identified in the Late Jurassic Xiali Formation(Fm.) in the basin in the last years. However, to date, no potash or halite deposits have been identified in the QB. Gypsum outcrops and salt springs are very important signs in the investigation of halite and potash deposits. Therefore, the Xiali Fm. is a potentially valuable layer to evaluate for the possible presence of halite and potash deposits in the basin. However, few studies have explored the formation conditions of evaporites in the unit. Here, we present detailed geochemical records from the Yanshiping section related to the study of the formation conditions of evaporites in the Xiali Fm. of the QB. Climate proxies based on the obviously increased anion concentrations of SO_4(2-) and Cl^- and the significant correlation coefficients of Ca2+-SO42-(R = 0.985) and Na^+-Cl^-(R = 0.8974) reveal that the upper member of the Xiali Fm.(the upper Xiali Fm.) formed under an arid climate and evolved into the sulfate phase or early chloride phase. Provenance proxies based on the obviously increased K^+ and Na^+ ion concentrations and the significant correlation coefficient of Na^+-Cl^-(R = 0.8974) suggest that the upper Xiali Fm. featured optimal provenance conditions for the possible formation of halite deposits. The regression and the semi-closed tidal flat environment in the upper Xiali Fm. were favorable for the formation of potash and halite deposits. The low Mg^(2+) /Ca^(2+) values(mean value = 1.82) and significant Na^+-Cl^- correlation coefficient(R = 0.8974) also suggest that the upper Xiali Fm. is the layer most likely to contain potential halite deposits. In addition, the macroscopic correlations of tectonism, provenance, paleoclimate, saliferous strata and sedimentary environment between the QB and the adjoining Amu Darya Basin in Central Asia reveal that the two basins shared similar geologic settings that were favorable for the formation of evaporites during the Late Jurassic. | 3 | Aurora-A is a novel predictor of poor prognosis in patients with resected lung adenocarcinoma显示文摘Background: The Aurora-A(Aur-A) gene, a key regulator of mitosis, has been proved as an oncogene in a variety of cancers. The Aur-A overexpression has been proved correlated with aggressiveness of cancer cells. However, the frequency of Aur-A protein overexpression, as well as its association with clinicopathologic parameters and prognosis remain unclear in lung adenocarcinoma(ADC). This study tried to clarify the clinical significance of Aur-A in patients with resected lung ADC. Patients and methods: A total of 142 informative patients with surgically resected lung ADC and 20 normal lung tissues were enrolled. Western blot and immunohistochemistry(IHC) were utilized to assess protein expression of Aur-A.Result: The expression of Aur-A was elevated in most of tumor tissues compared with the adjacent tissues by western blot. The IHC results showed that Aur-A protein was over-expressed in 98 of 142(69.0%) tumor sections, while Aur-A was low-expressed in all normal lung sections. A positive correlation between Aur-A overexpression rate and ascending pathologic stages was observed(P<0.05). Kaplan-Meier analysis demonstrated that patients with Aur-A high expression had significantly inferior survival compared to those with Aur-A low expression. Both overall survival(OS) and disease-free survival(DFS) of positive overexpression patients were shorter than the negative group(P=0.036, P=0.041, respectively). | 7 | Single molecular insight into steric effect on C-terminal amino acids with various hydrogen bonding sites显示文摘Amino acids are basic units to construct a protein with the assistance of various interactions.During this building process,steric hindrance derived from amino acid side groups or side chains is a factor that could not be ignored.In this contribution,adsorption behaviors of C-terminal amino acid derivatives with amino acid residues fused in 3,4,9,10-perylenetetracarboxylic dianhydride were investigated by scanning tunneling microscopy(STM)and density functional theory(DFT)calculations at various liquid/solid interfaces.STM results at 1-phenyloctane/HOPG interface show that N,N'-3,4,9,10-perylenedicarboximide(GP)and N,N'-methyl-3,4,9,10-perylenedicarboximide(AP)formed linear and herringbone structures,respectively.The driving force could be attributed to different H-bonding sites induced by steric hindrance at side groups.N,N'-Benzyl-3,4,9,10-perylenedicarboximide(PP)generates both linear and herringbone structures because steric hindrance changes the H-bonding sites between PP molecules,whereas N,N'-isopropyl-3,4,9,10-perylenedicarboximide(LP)failed to be imaged because of strong steric hindrance coming from larger side group.To further investigate the impact of steric hindrance,we utilized octanoic acid(OA)as solvent to capture the adsorption details of LP and PP.We found that OA molecules drag PP and LP molecules in a different direction to generate linear structure,impeding the molecular rotation.The structure–solvent relationship shows that the steric hindrance is brought by the large side group,which makes it easier to recognize OA molecules at the interface.These results demonstrate that steric effect plays a significant role in altering interaction sites of the compounds during the adsorption process at the liquid/solid interface. | Yunzhi Xie Chunhua Liu Linxiu Cheng Yulan Fan Huifang Li Wei Liu Lei Zhu Xun Li Ke Deng Qingdao Zeng Shoufa Han | 2022 | Chinese Chemical Letters2022,33,10: | 1 |

| 14 | Applications of EMD to analyses of high-frequency beachface responses to Storm Bebinca in the Qing’an Bay,Guangdong Province,China显示文摘On average,five to six storms occur in the Qiongzhou Strait every year,causing significant damage to coastal geomorphology and several property losses.Tropical Storm Bebinca is the most unusual and complex storm event that has occurred in this region over the last 10 years.To detect the high-frequency beachface responses to the storm,a pressure sensor was deployed in the surf zone to record the free sea surface height,and the heights of grid pile points on the beachface were measured manually to determine beach elevation changes during this storm.Empirical Mode Decomposition and related analysis techniques were used to analyze the high-frequency topography and wave data.The results showed that:(1)the beachface response process occurred in three stages.The first stage was the rapid response stage,wherein the spring tide berm began to erode significantly,and the front edge of the beach berm reacted closely.The two beach sections resisted the harmful energy of the main storm.In the second stage,the beach slope increased after a large sediment loss on the beach berm and its front edge.To adapt to the storm energy,the beach at the low tide line began to erode,and the beach slope decreased.In the third stage,after the storm turned,the wave energy was significantly attenuated,and the beach berm eroded to resist the residual wave energy.The beachface began to oscillate and recover.(2)The main wave surface was the superimposed product of a few internal mode functions.Similar results were observed in beachface changes.High-frequency driving factors determine the local characteristics of beach evolution,and low-frequency driving factors determine the beach evolution trend.(3)The response of sediment to the storm was not a single sea-transportation,but a single-or two-way conversion driven by factors such as wave energy,swash flow,and secondary wave breaking.(4)TheΩ-RTR model is not completely applicable to beach states that undergo rapid changes during storms.Therefore,it is necessary to carry out further research on beach state identification during storms. | Taihuan Hu Zhiqiang Li Chunhua Zeng Gaocong Li Huiling Zhang | 2022 | Acta Oceanologica Sinica2022,41,5: | 0 |

| 16 | Escape of Brownian particles and stochastic resonance with low-temperature quantum fluctuations显示文摘In this paper, we investigate the escape of Brownian particles and stochastic resonance (SR) with low-temperatures quantum fluctuations by using the quantum Smoluchowski equations at low-temperature. Two specific examples have been considered: one is the example of bistable system, and the other is the example of metastable system. The explicit expressions of the mean-first passage time (MFPT) and signal-to-noise ratio (SNR) for both specific examples are obtained, respectively. Based on the numerical computations, we compare the quantum case with its classical counterpart. | 17 | Electrocoagulation coupled with electrooxidation for the simultaneous treatment of multiple pollutants in contaminated sediments显示文摘In situ and simultaneous remediation of a variety of pollutants in sediments remains a challenge.In this study,we report that the combination of electrocoagulation(EC)and electrooxidation(EO)is efficient in the immobilization of phosphorus and heavymetals and in the oxidation of ammonium and toxic organicmatter.The integratedmixed metal oxide(MMO)/Fe anode system allowed the facile removal of ammonium and phosphorus in the overlying water(99% of 10 mg/L NH_(4)^(+)-N and 95% of 10 mg/L P disappeared in 15 and 30 min,respectively).Compared with the controls of the single Fe anode and single MMO anode systems,the dual MMO/Fe anode system significantly improved the removal of phenanthrene and promoted the transition of Pb and Cu from the mobile species to the immobile species.The concentrations of Pb and Cu in the toxicity characteristic leaching procedure extracts were reduced by 99%and 97% after an 8 hr operation.Further tests with four real polluted samples indicated that substantial proportions of acid-soluble fraction Pb and Cu were reduced(30%-31% for Pb and 16%–23% for Cu),and the amounts of total organic carbon and NH_(4)^(+)-N decreased by 56%–71% and 32%–63%,respectively.It was proposed that the in situ electrogenerated Fe(II)at the Fe anode and the active oxygen/chlorine species at the MMO anode are conducive to outstanding performance in the co-treatment of multiple pollutants.The results suggest that the EC/EO method is a powerful technology for the in situ remediation of sediments contaminated with different pollutants. | Qingjun Zeng Yifan Zhang Pingshan Chen Yuting He Congli Yi Chunhua Feng | 2023 | Journal of Environmental Sciences2023,,2: | 0 |
