| 1 | A Domestication-Associated Gene GmPRR3b Regulates the Circadian Clock and Flowering Time in Soybean显示文摘Improved soybean cultivars have been adapted to grow at a wide range of latitudes,enabling expansion of cultivation worldwide.However,the genetic basis of this broad adaptation is still not clear.Here,we report the identification of GmPRR3b as a major flowering time regulatory gene that has been selected during domestication and genetic improvement for geographic expansion.Through a genome-wide association study of a diverse soybean landrace panel consisting of 279 accessions,we identified 16 candidate quantitative loci associated with flowering time and maturity time.The strongest signal resides in the known flowering gene E2,verifying the effectiveness of our approach.We detected strong signals associated with both flowering and maturity time in a genomic region containing GmPRR3b.Haplotype analysis revealed that GmPRR3bH6 is the major form of GmPRR3b that has been utilized during recent breeding of modern cultivars.mRNA profiling analysis showed that GmPRR3bH6 displays rhythmic and photoperiod-dependent expression and is preferentially induced under long-day conditions.Overexpression of GmPRR3bH6 increased main stem node number and yield,while knockout of GmPRR3bH6 using CRISPR/Cas9 technology delayed growth and the floral transition.GmPRR3bH6 appears to act as a transcriptional repressor of multiple predicted circadian clock genes,including GmCCAIa,which directly upregulates J/GmELF3a to modulate flowering time.The causal SNP(Chr12:5520945)likely endows GmPRR3bH6 a moderate but appropriate level of activity,leading to early flowering and vigorous growth traits preferentially selected during broad adaptation of landraces and improvement of cultivars. | Cong Li Ying-hui Li Yanfei Li Hongfeng Lu Huilong Hong Yu Tian Hongyu Li Tao Zhao Xiaowei Zhou Jun Liu Xinan Zhou Scott A.Jackson Bin Liu Li-juan Qiu | 2020 | Molecular Plant2020,13,5: | 11 |
| 2 | Relationship of microstructure transformation and hardening behavior of type 17-4 PH stainless steel显示文摘The relationship between the microstructure transformation of type 17-4 PH stainless steel and the aging hardening be- havior was investigated. The results showed that, when 17-4 PH stainless steel aging at 595°C, the bulk hardness of samples attains its peak value (42.5 HRC) for about 20 min, and then decreases at all time. TEM revealed the microstructure corresponding with peak hardness is that the fine spheroid-shape copper with the fcc crystal structure and the fiber-shape secondary carbide M23C6 precipitated from the lath martensite matrix. Both precipitations of copper and M23C6 are the reasons for strengthening of the alloy at this tem- perature. | 5 | Increased L-arginine Production by Site-directed Mutagenesis of N-acetyl-L-glutamate Kinase and pro B Gene Deletion in Corynebacterium crenatum显示文摘Objective In Corynebacterium crenatum,the adjacent D311 and D312 of N-acetyl-L-glutamate kinase(NAGK),as a key rate-limiting enzyme of L-arginine biosynthesis under substrate regulatory control by arginine,were initially replaced with two arginine residues to investigate the L-arginine feedback inhibition for NAGK.Methods NAGK enzyme expression was evaluated using a plasmid-based method.Homologous recombination was employed to eliminate the pro B.Results The IC50 and enzyme activity of NAGK M4,in which the D311 R and D312 R amino acid substitutions were combined with the previously reported E19 R and H26 E substitutions,were 3.7-fold and 14.6% higher,respectively,than those of the wild-type NAGK.NAGK M4 was successfully introduced into the C.crenatum MT genome without any genetic markers;the L-arginine yield of C.crenatum MT-M4 was 26.2% higher than that of C.crenatum MT.To further improve upon the L-arginine yield,we constructed the mutant C.crenatum MT-M4 ?pro B.The optimum concentration of L-proline was also investigated in order to determine its contribution to L-arginine yield.After L-proline was added to the medium at 10 mmol/L,the L-arginine yield reached 16.5 g/L after 108 h of shake-flask fermentation,approximately 70.1% higher than the yield attained using C.crenatum MT.Conclusion Feedback inhibition of L-arginine on NAGK in C.crenatum is clearly alleviated by the M4 mutation of NAGK,and deletion of the pro B in C.crenatum from MT to M4 results in a significant increase in arginine production. | ZHANG Bin WAN Fang QIU Yu Lou CHEN Xue Lan TANG Li CHEN Jin Cong XIONG Yong Hua | 2015 | Biomedical and Environmental Sciences2015,28,12: | 5 |

| 8 | Polarization-independent all-silicon dielectric metasurfaces in the terahertz regime显示文摘Dielectric metasurfaces have achieved great success in realizing high-efficiency wavefront control in the optical and infrared ranges. Here, we experimentally demonstrate several efficient, polarization-independent, all-silicon dielectric metasurfaces in the terahertz regime. The metasurfaces are composed of cylindrical silicon pillars on a silicon substrate, which can be easily fabricated using etching technology for semiconductors. By locally tailoring the diameter of the pillars, full control over abrupt phase changes can be achieved. To show the controlling ability of the metasurfaces, an anomalous deflector, three Bessel beam generators, and three vortex beam generators are fabricated and characterized. We also show that the proposed metasurfaces can be easily combined to form composite devices with extended functionalities. | 17 | Platinum and Iridium Coatings Obtained by Double Glow Plasma Technology显示文摘Pt and Ir coatings were produced by double glow plasma technology on the surface of Ti alloy substrates. The chemical compositions of the coatings were determined by X-ray diffraction and X-ray photoelectron spectroscopy. The microstructure and morphology of the coatings were observed by scanning electron microscopy. The hardness and elastic modulus of the coatings were estimated by nanoindentation. The measurements of adhesive forces of the coatings were performed with scratch tester. The results indicated that the Pt and Ir coatings displayed the preferred (220) orientation due to the initial nuclei with preferred growth on the surface of the substrates. The interface between the Pt coating and substrate exhibited no evidence of delamination. The Ir coating was composed of irregular columnar grains with many nanovoids at the interface between the coating and substrate. The mean values of hardness for Pt and Ir coatings were 0.9 GPa and 9 GPa, respectively. The elastic modulus of Pt and Ir coatings were 178 GPa and 339 GPa, respectively. The adhesive forces of the Pt and Ir coatings were about 66.4 N and 55 N, respectively.
