1Photocatalytic properties of BiOX (X = Cl, Br, and Ⅰ)显示文摘A novel series of photocatalysts,bismuth oxyhalide(BiOX,X =Cl,Br,and I),were synthesized by a hydrolysis method.The powder sam- pies were characterized by the use of X-ray diffraction(XRD),scanning electron microscope,and UV-Vis spectrophotometer.The XRD pattern showed that all the BiOX were well crystallized in the tetragonal structure.The band gaps of the sheet-shaped compounds BiOX(X= Cl,Br,and I)were 3.44,2.76,and 1.85 eV,respectively.BiOBr showed the highest photocatalytic activity in degrading rhodamine B(RhB) and evolving O_2 for its proper valence band(VB).BiOI has no photocatalytic activity.BiOCl showed the highest activity in decomposing isopropanol because of electron-hole pair separation through trapping electrons by oxygen vacancies.AN Huizhong DU Yi WANG Tianmin WANG Cong HAO Weichang ZHANG Junying 2008Rare Metals2008,27,3:43
2Notch signaling mediated by TGF-β/Smad pathway in concanavalin A-induced liver fibrosis in rats显示文摘AIM To explore the exact interaction between Notch and transforming growth factor(TGF)-β signaling in liver fibrosis. METHODS We established a rat model of liver fibrosis induced by concanavalin A. Peripheral blood mononuclear cells(PBMCs) were isolated from the modeled rats, and cultured with γ-secretase inhibitor DAPT and TGF-β inhibitor for 24 h. The m RNA levels of Notch and TGF-β signaling were detected by quantitative real-time polymerase chain reaction. Expression of Notch and TGF-β proteins was analyzed by western blotting.RESULTS Compared to control rats, Notch and TGF-β signaling was activated in PBMCs of model rats. Administration of DAPT and TGF-β inhibitor suppressed Notch and TGF-β signal transducer in PBMCs of model rats. DAPT reduced m RNA and protein expression of TGF-β signaling, such as TGF-β1 and Smad3. TGF-β inhibitor also downregulated Notch1, Hes1 and Hes5, and m RNA and protein expression of the Notch signaling pathway.CONCLUSION Notch and TGF-β signaling play a role in liver fibrosis. 4Biosynthesis of Triacylglycerol Molecules with a Tailored PUFA Profile in Industrial Microalgae显示文摘The composition of polyun saturated fatty acids (PUFAs) in triacylglycerols (TAGs) is key to health ben efits and for oil applications, yet the underlying genetic mechanism remains poorly understood. In this study, by in si Iico, ex vivo, and in vivo profili ng of type-2 diacylglycerol acyltransferases (DGAT2s) in Nannochloropsis oceanica we revealed two novel PUFA-preferring enzymes that discriminate individual PUFA species in TAG assembly, with NoDGAT2J for linoleic acid (LA) and NoDGAT2K for eicosapentaenoic acid (EPA). The LA and EPA composition of TAG molecules is mediated in vivo via the functional partitioning between NoDGAT2J and 2K, both of which are localized in the chloroplast envelope. By modulating transcript abundance of the DGAT2s, an N. oceanica strain bank was created, where proportions of LA and EPA in TAG vary by 18.7-fold (between 0.21% and 3.92% dry weight) and 34.7-fold (between 0.09% and 3.12% dry weight), respective!y. 5Rosmarinic acid elicits neuroprotection in ischemic stroke via Nrf2 and heme oxygenase 1 signaling显示文摘Rosmarinic acid(RA) can elicit a neuroprotective effect against ischemic stroke, but the precise molecular mechanism remains poorly understood. In this study, an experimental ischemic stroke model was established in CD-1 mice(Beijing Vital River Laboratory Animal Technology, Beijing, China) by occluding the right middle cerebral artery for 1 hour and allowing reperfusion for 24 hours. After intraperitoneally injecting model mice with 10, 20, or 40 mg/kg RA, functional neurological deficits were evaluated using modified Longa scores. Subsequently, cerebral infarct volume was measured using TTC staining and ischemic brain tissue was examined for cell apoptosis with TUNEL staining. Superoxide dismutase activity and malondialdehyde levels were measured by spectrophometry. Expression of heme oxygenase-1(HO-1), nuclear factor erythroid 2-related factor 2(Nrf2), Bcl-2, Bax, Akt, and phospho-Ser473 Akt proteins in ischemic brain tissue was detected by western blot, while mRNA levels of Nrf2, HO-1, Bcl-2, and Bax were analyzed using real time quantitative PCR. In addition, HO-1 enzyme activity was measured spectrophotometrically. RA(20 and 40 mg/kg) greatly improved neurological function, reduced infarct volume, decreased cell apoptosis, upregulated Bcl-2 protein and mRNA expression, downregulated Bax protein and mRNA expression, increased HO-1 and Nrf2 protein and mRNA expression, increased superoxide dismutase activity, and decreased malondialdehyde levels in ischemic brain tissue of model mice. However, intraperitoneal injection of a HO-1 inhibitor(10 mg/kg zinc protoporphyrin IX) reversed the neuroprotective effects of RA on HO-1 enzyme activity and Bcl-2 and Bax protein expression. The PI3 K/Akt signaling pathway inhibitor LY294002(10 mM) inhibited Akt phosphorylation, as well as Nrf2 and HO-1 expression. 6Successive inverse and normal magnetocaloric effects in the Mn-vacancy compound Mn0.95Co0.75Cu0.25Ge显示文摘Ferromagnetic-structural transformation has been studied widely in MnCoGe-based materials. However, the magnetostructural transition(MST) from antiferromagnetic(AFM) orthorhombic phase to ferromagnetic(FM) hexagonal phase, which may lead to a large inverse magnetocaloric effect(MCE), has rarely been reported. Here, the introduction of Mn vacancy lowers the structural transition temperature while retains the AFM state in the orthorhombic phase, thus successfully realizing the AFM-FM MST in Mn0.95Co0.75Cu0.25Ge. Moreover, successive inverse and normal MCEs are observed around the first-order AFM-FM MST and the second-order FM-paramagnetic(PM) transition, respectively. A thermostat is proposed based on this special feature, which could release heat above the critical temperature while absorb heat below the critical temperature by simply applying the same magnetization/demagnetization cycles. 7In silico genome-wide identification,phylogeny and expression analysis of the R2R3-MYB gene family in Medicago truncatula显示文摘The R2R3-MYB genes make up one of the largest transcription factor families in plants, and play regulatory roles in various biological processes such as development, metabolism and defense response. Although genome-wide analyses of this gene family have been conducted in several species, R2R3-MYB genes have not been systematically analyzed in Medicago truncatula, a sequenced model legume plant. Here, we performed a comprehensive, genome-wide computational analysis of the structural characteristics, phylogeny, functions and expression patterns of M. truncatula R2R3-MYB genes. DNA binding domains are highly conserved among the 155 putative Mt R2R3-MYB proteins that we identified. Chromosomal location analysis revealed that these genes were distributed across all eight chromosomes. Results showed that the expansion of the Mt R2R3-MYB family was mainly attributable to segmental duplication and tandem duplication. A comprehensive classification was performed based on phylogenetic analysis of the R2R3-MYB gene families in M. truncatula, Arabidopsis thaliana and other plant species. Evolutionary relationships within clades were supported by clade-specific conserved motifs outside the MYB domain. Species-specific clades have been gained or lost during evolution, resulting in functional divergence. Also, tissue-specific expression patterns were investigated. The functions of stress response-related clades were further verified by the changes in transcript levels of representative R2R3-MYB genes upon treatment with abiotic and biotic stresses. 10Antioxidant Effect of Human Selenium-containing Single-chain Fv in Rat Cardiac Myocytes显示文摘Reactive oxygen species(ROS) plays a key role in human heart diseases. Glutathione peroxidase(GPX) functions as an antioxidant as it catalyzes the reduction of hydroperoxide. In order to investigate the antioxidant effect of human selenium-containing single-chain Fv(Se-scFv-B3), a new mimic of GPX, a model system of hydrogen peroxide(H2O2)-induced rat cardiac myocyte damage was established. The cardiac myocyte damage was characterized in terms of cell viability, lipid peroxidation, cell membrane integrity, and intracellular H2O2 level. The Se-scFv-B3 significantly reduced H2O2-induced cell damage as shown by the increase of cell viability, the decline of malondialdehyde(MDA) production, lactate dehydrogenase(LDH) release, and intracellular H2O2 level. 13IL-33 induces immunosuppressive neutrophils via a type 2 innate lymphoid cell/IL-13/STAT6 axis and protects the liver against injury in LCMV infection-induced viral hepatitis显示文摘Viral hepatitis is still a public health problem affecting several million people around the world.Neutrophils are polymorphonuclear cells that have a critical role in antibacterial infection.However,the role of neutrophils in viral infection is not fully understood.By using a mouse model of lymphocytic choriomeningitis virus infection-induced viral hepatitis,we observed increased neutrophil recruitment in the liver accompanied by enhanced CD8+T-cell responses.Liver neutrophils expressed high levels of immunomodulatory cytokines,such as C-X-C chemokine ligand 2,arginase-1,inducible nitric oxide synthase and interleukin(IL)-10,demonstrating immunosuppressive properties.Depletion of neutrophils in vivo by a neutralizing antibody resulted in the exacerbation of liver injury and the promotion of T-cell responses at the immune contraction stage.IL-33 significantly induced neutrophil recruitment in the liver and attenuated liver injury by limiting effector T-cell accumulation.Mechanistically,we found that IL-33 promoted the expression of arginase-1 in neutrophils through the type 2 innate lymphoid cell(ILC2)-derived IL-13.Additionally,IL-13 increased the inhibitory effect of neutrophils on CD8+T-cell proliferation in vitro,partially through arginase-1.Finally,we found that IL-13 induced arginase-1 expression,depending on signal transducer and activator of transcription factor 6(STAT6)signaling.Therefore,IL-33 induced immunosuppressive neutrophils via an ILC2/IL-13/STAT6 axis.Collectively,our findings shed new light on the mechanisms associated with IL-33-triggered neutrophils in the liver and suggest potential targets for therapeutic investigation in viral hepatitis.Yuejin Liang Panpan Yi Denley Ming Kee Yuan Zuliang Jie Zakari Kwota Lynn Soong Yingzi Cong Jiaren Sun 2019Cellular & Molecular Immunology2019,16,2:7
14Heterosis and combining ability evaluation for growth traits of blunt snout bream(Megalobrama amblycephala)when crossbreeding three strains显示文摘The blunt snout bream(Megalobrama amblycephala)is a major aquaculture species in the Chinese freshwater polyculture system.In this study,complete diallel crossing of three strains(i.e.,Liangzi[LZ],Poyang[PY]and Yuni[YN])was used to evaluate the combining ability and heterosis effect of intraspecific crossbreeding on the growth performance(i.e.,body weight,total length,body length,body height,and Fulton’s coefficient of condition).The offspring produced from the three strains were reared in a communal pond for 20 months,and nine microsatellites were used to assess their pedigree.Of the749 offspring,708(94.53%)could be assigned directly to a single parental pair.Significant differences were observed in body weight(BW)among the different combinations,indicating that the combining ability of BW from each strain was significantly different.The general combining ability(GCA)of BW from the sire was much higher than that from the dam.The GCA for the dam and sire of the LZ and YN were the largest and the smallest,respectively.The special combining ability of YN$9 PY#was the largest,and YN$9 PY#showed significantly positive heterosis effects on all F1growth traits,except for Fulton’s condition coefficient(K)(P\0.05).In addition,significant positive linear correlations were found between the mean BW of corresponding progeny and the H E of the parental generation(y=600.7x-29.472,r=0.8651,P=0.0026),H O(y=1206.9x-518.14,r=0.7436,P=0.0216),and the polymorphism information content(y=1021.3x-331.31,r=0.8245,P=0.0063)as determined by a correlative test.The findings obtained from the study will be important for future considerations of M.amblycephala stock improvement programs.Wei Luo Cong Zeng Shaokui Yi Nicholas Robinson Weimin Wang Zexia Gao 2014Chinese Science Bulletin2014,59,9:7
15Toxoplasma ROP16Ⅰ/Ⅲ ameliorated inflammatory bowel diseases via inducing M2 phenotype of macrophages显示文摘BACKGROUND Inflammatory bowel disease(IBD)is characterized by chronic and non-specific inflammation of the intestinal mucosa and mainly includes ulcerative colitis and Crohn's disease.AIM To explore the beneficial effect of ToxoROP16I/III-induced M2 phynotype macrophages in homeostasis of IBDs through downregulation of M1 inflammatory cells.METHODS RAW264.7 macrophages stimulated by lipopolysaccharide(LPS)(M1 cells)were co-cultured with Caco-2 cells as an inflammatory model of IBD in vitro.The expression of ToxoROP16I/III was observed in RAW264.7 macrophages that were transfected with pEGFP-rop16I/III.The phenotypes of M2 and M1 macrophage cells were assessed by quantitative real-time reverse transcriptase polymerase chain reaction and the expression of tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,transforming growth factor(TGF)-β1,IL-10,inducible nitric oxide synthase(iNOS),and arginase-1(Arg-1)was detected.The expression of iNOS,Arg-1,signal transducer and activator of transcription 3(Stat3),p-Stat3,Stat6,p-Stat6,programmed death ligand-2(PD-L2),caspase-3,-8,and-9 was analyzed by Western blotting,and Griess assays were performed to detect nitric oxide(NO).TNF-α,IL-1β,IL-6,TGF-β1,and IL-10 expression in the supernatants was detected by enzyme-linked immunosorbent assay,and Caco-2 cell apoptosis was co-culture system.RESULTS M1 cells exhibited significantly increased production of iNOS,NO,TNF-α,IL-1β,and IL-6,while ToxoROP16I/III induced macrophage bias to M2 cells in vitro,showing increased expression of Arg-1,IL-10 and TGF-β1 and elevated production of p-Stat3 and p-Stat6.The mixed M1 and M2 cell culture induced by ToxoROP16I/III exhibited decreased production of NO and iNOS and upregulated expression of Arg-1 and PD-L2.Accordingly,Caco-2 cells became apoptotic,and apoptosis-associated proteins such as caspase-3,-8 and-9 were dampened during co-culture of M1 and M2 cells.Flow cytometry analysis showed that co-culture of M1 cells with Caco-2 cells facilitated the apoptosis of Caco-2 cells,but co-culture of M1 and M2 cells alleviated Caco-2 cell apoptosis.CONCLUSION ToxoROP16I/III-induced M2 macrophages inhibited apoptosis of Caco-2 cells caused by M1 macrophages.This finding may help gain a better understanding of the underlying mechanism and represent a promising therapeutic strategy for IBDs.Yong-Wei Xu Rui-Xin Xing Wen-Hui Zhang Lu Li Yi Wu Jing Hu Cong Wang Qing-Li Luo Ji-Long Shen Xi Chen 2019World Journal of Gastroenterology2019,25,45:7
16Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 19The effects of air temperature and precipitation on the net primary productivity in China during the early 21st century显示文摘Research on how terrestrial ecosystems respond to climate change can reveal the complex interactions between vegetation and climate,net primary productivity (NPP),an important vegetation parameter and ecological indicator,fluctuates within any given ecological environment or regional carbon budget.In this study, spatial interpolation was used to generate a spatial dataset, with 1-km spatial resolution,with meteorological data from 736 observation stations across China.An improved CASA model was used to simulate NPP over the period of 2001-2013 by taking into account land-cover change in every year during the same period.We propose the grid- based spatial patterns and dynamics of annual NPP,annual average temperature,and annual total precipitation based on the model.We also used the model to demonstrate the spatial correlation between NPP,temperature,and pre- cipitation in the study area with special focus on the impact of climate change in the early 21st century.Results showed that the spatial pattern of NPP over all of China is characterized by higher values in the southeast and lower values in the northwest.The spatial pattern of temperature indicates substantial latitudinal differences across the country,and the spatial pattem of precipitation shows a ribbon of decline from the southeast coast to the northwest inland.Most areas show an upward trend in NPP. 20Association between Serum Alkaline Phosphatase and Carotid Atherosclerosis in a Chinese Population: A Community-based Cross-sectional Study显示文摘Objective This study aimed to investigate the relationship between alkaline phosphatase(ALP) and common carotid intima media thickness(IMT), carotid plaque, and extracranial carotid artery stenosis(ECAS). Methods A total of 3,237 participants aged ≥ 40 years were recruited from Jidong community in 2013-2014. Participants were divided into five quintile groups based on their serum ALP levels. Carotid atherosclerosis was assessed using ultrasound. Abnormal IMT, carotid plaque, and ECAS were defined as IMT > 0.9 mm, IMT > 1.5 mm, and ≥ 50% stenosis in at least one extracranial carotid artery, respectively. Results Common carotid IMT values and the prevalence of carotid plaque increased across serum ALP quintiles. Higher ALP quintiles were correlated with an increased risk of abnormal IMT [fourth quintile: odds ratio(OR) 1.78, 95% confidence interval(CI) 1.13-2.82, P = 0.0135;fifth quintile: OR = 1.82, 95% CI: 1.15-2.87, P = 0.0110] and ECAS compared to the lowest quintile(fifth quintile: OR = 1.47, 95% CI: 1.09-1.97, P = 0.0106). The association between ALP and prevalence of carotid plaque became insignificant after adjustment for confounders. Conclusion Serum ALP levels were independently associated with abnormal common carotid IMT and ECAS.
