13Revealing CO_(2)-Fixing SAR11 Bacteria in the Ocean by Raman-Based Single-Cell Metabolic Profiling and Genomics显示文摘The majority of marine microbes remain uncultured,which hinders the identification and mining of CO_(2)-fixing genes,pathways,and chassis from the oceans.Here,we investigated CO_(2)-fixing microbes in seawater from the euphotic zone of the Yellow Sea of China by detecting and tracking their ^(13)C-bicarbonate(^(13)C-HCO_(3)^(-))intake via single-cell Raman spectra(SCRS)analysis.The target cells were then isolated by Raman-activated Gravity-driven Encapsulation(RAGE),and their genomes were amplified and sequenced at one-cell resolution.The single-cell metabolism,phenotype and genome are consistent.We identified a notyet-cultured Pelagibacter spp.,which actively assimilates ^(13)C-HCO_(3)^(-),and also possesses most of the genes encoding enzymes of the Calvin-Benson cycle for CO_(2) fixation,a complete gene set for a rhodopsin-based light-harvesting system,and the full genes necessary for carotenoid synthesis.The four proteorhodopsin(PR)genes identified in the Pelagibacter spp.were confirmed by heterologous expression in E.coli.These results suggest that hitherto uncultured Pelagibacter spp.uses light-powered metabolism to contribute to global carbon cycling.Xiaoyan Jing Yanhai Gong Teng Xu Paul ADavison Craig MacGregor-Chatwin CNeil Hunter La Xu Yu Meng Yuetong Ji Bo Ma Jian Xu Wei E.Huang 2022BioDesign Research2022,,1:1

19Observatory science with eXTP显示文摘In this White Paper we present the potential of the enhanced X-ray Timing and Polarimetry(eXTP) mission for studies related to Observatory Science targets. These include flaring stars, supernova remnants, accreting white dwarfs, low and high mass X-ray binaries, radio quiet and radio loud active galactic nuclei, tidal disruption events, and gamma-ray bursts. eXTP will be excellently suited to study one common aspect of these objects: their often transient nature. 20Trade-off between microbial carbon use efficiency and specific nutrient-acquiring extracellular enzyme activities under reduced oxygen显示文摘Mangroves are one of the most ecologically sensitive ecosystems to global climate change,which have cascading impacts on soil carbon(C),nitrogen(N)and phosphorus(P)cycling.Moreover,mangroves are experiencing increasing N and P loadings and reduced oxygen availability due to intensified climate change and human activities.However,both direct and interactive effects of these perturbations on microbially mediated soil C,N and P cycling are poorly understood.Here,we simultaneously investigated the effects of N and P loadings and reduced oxygen on microbial biomass,microbial respiration,and extracellular enzyme activities(EEAs)in mangrove soils.We calculated the microbial metabolic quotient(qCO_(2)),which is regarded as a useful inverse metric of microbial C use efficiency(CUE).Our results show that reduced oxygen significantly increases both qCO_(2) and microbial specific EEAs(enzyme activity per unit of microbial biomass)for C-,N-and P-acquisition regardless of N or P loadings.Furthermore,we found that qCO_(2) positively correlated with microbial specific EEAs under reduced oxygen,whereas no clear relationship was detected under ambient oxygen.These results suggest that reduced oxygen increases microbial specific EEAs at the expense of increasing microbial respiration per unit biomass,indicating higher energy cost per unit enzyme production.Ji Chen Irene Cordero Daryl L.Moorhead Jennifer K.Rowntree Lorae T.Simpson Richard D.Bardgett Hayley Craig 2023Soil Ecology Letters2023,5,2:0
