2Isolation and genetic charac-terization of a fragile plant mutant in rice (Oryza sativa L.)显示文摘The fragile rice mutant was isolated from an M2 population of indica variety Shuang Ke Zao (SKZ) treated with γ-rays, and designated as fp1 {fragile plant 1) because of its fragile leaves and culms. To map FP1 locus, an F2 mapping population was derived from a cross between the fp1 and C-bao, a polymorphic japonic variety. The primary mapping result places the FP1 locus in an interval between two molecular markers, microsatellite marker RM16 (3.1 cM proximal to FP1) and STS marker G144a (9.1 cM distal to FP1) in the centromere region of chromosome 3. A CAPS marker C524a was further developed between RM16 and G144a, with 0.4 cM genetic distances to the FP1 locus, providing a practical starting point for constructing a BAC con-tig spanning the FP1 locus and cloning the fp1 gene. 4Gene editing and its applications in biomedicine显示文摘The steady progress in genome editing, especially genome editing based on the use of clustered regularly interspaced short palindromic repeats(CRISPR) and programmable nucleases to make precise modifications to genetic material, has provided enormous opportunities to advance biomedical research and promote human health. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges. 5Methane dehydroaromatization with periodic CH_4-H_2 switch:A promising process for aromatics and hydrogen显示文摘Long-term stability test of Mo/HZSM-5-N catalysts(HZSM-5-N stands for nano-sized HZSM-5) in methane dehydroaromatization(MDA)reaction has been performed with periodic CH4-H2 switch at 1033-1073 K for more than 1000 h.During this test,methane conversion ranges from 13% to 16%,and mean yield to aromatics(i.e.benzene and naphthalene) exceeds 10%.N2-physisorption,XRD,NMR and TPO measurements were performed for the used Mo/HZSM-5 catalysts and coke deposition,and the results revealed that the periodic hydrogenation can effectively suppress coke deposition by removing the inert aromatic-type coke,thus ensuring Mo/HZSM-5 partly maintained its activity even in the presence of large amount of coke deposition.The effect of zeolite particle size on the catalytic activity was also explored,and the results showed that the nano-sized zeolite with low diffusion resistance performed better.It is recognized that the size effect was enhanced by reaction time,and it became more remarkable in a long-term MDA reaction even at a low space velocity.Changyong Sun Guangzong Fang Xiaoguang Guo Yuanli Hu Shuqi Ma Tianhua Yang Jie Han Hao Ma Dali Tan Xinhe Bao 2015Journal of Energy Chemistry2015,24,3:6
6Regulation of phenylpropanoid biosynthesis by MdMYB88 and MdMYB124 contributes to pathogen and drought resistance in apple显示文摘MdMYB88 and MdMYB124 have been demonstrated to be responsible for lignin accumulation in apple under drought stress.In this study,using a metabolomic approach,we identified differentially accumulated phenylpropanoid and flavonoid metabolites in MdMYB88/124 transgenic RNAi plants under control and long-term drought stress conditions in apple roots.We confirmed the regulation of phenylalanine by MdMYB88 and MdMYB124 via UPLC-MS in apple roots under both control and drought conditions.Using Electrophoretic Mobility Shift Assay(EMSA)and ChIPquantitative PCR(qPCR)analyses,we found that MdMYB88 positively regulates the MdCM2 gene,which is responsible for phenylalanine biosynthesis,through binding to its promoter region.Under long-term drought conditions,MdMYB88/124 RNAi plants consistently accumulated increased amounts of H2O2 and MDA,while MdMYB88 and MdMYB124 overexpression plants accumulated decreased amounts of H2O2 and MDA.We also examined the accumulation of metabolites in the phenylpropanoid biosynthesis pathway in the leaves of MdMYB88 and MdMYB124 transgenic apple plants after long-term drought stress.We found that metabolites responsible for plant defense,including phenylpropanoids and flavonoids,accumulated less in the RNAi plants but more in the overexpression plants under both control and drought conditions.We further demonstrated that MdMYB88/124 RNAi plants were more sensitive to Alternaria alternata f.sp.mali and Valsa mali,two pathogens that currently severely threaten apple production.In contrast,MdMYB88 and MdMYB124 overexpression plants were more tolerant to these pathogens.The cumulative results of this study provided evidence for secondary metabolite regulation by MdMYB88 and MdMYB124,further explained the molecular roles of MdMYB88 and MdMYB124 in drought resistance,and provided information concerning molecular aspects of their roles in disease resistance.Dali Geng Xiaoxia Shen Yinpeng Xie Yusen Yang Ruiling Bian Yuqi Gao Pengmin Li Liying Sun Hao Feng Fengwang Ma Qingmei Guan 2020Horticulture Research2020,7,1:5

8Extraction kinetics of neodymium from chloride medium using HEH/EHP saponified with magnesium bicarbonate solution显示文摘Magnesium bicarbonate solution is considered as an environmentally friendly extractant saponification agent for the solvent extraction of rare earth elements due to its advantage of minimum water pollution.In order to reveal the extraction regularity, optimize production-process and guide the use of this new extraction system, the extraction of Nd(Ⅲ) in chloride medium with HEH/EHP saponified by magnesium bicarbonate solution was investigated with the self-designed constant interfacial area cell. Besides, the effects of stirring rate, temperature, specific interfacial area and concentration of Mg-HEH/EHP on the extraction rate of Nd(Ⅲ) were systematically investigated. Results show that, the rate of extraction is governed by both diffusion and chemical reaction, and the extraction reaction takes place at the interface. The apparent activation energy of the extraction reaction is 16.88 kJ/mol. The corresponding rate equation is deduced. 9miR-155 aggravates impaired autophagy of pancreatic acinar cells through targeting Rictor显示文摘The aim of this study was to investigate the role and mechanism of miR-155 in regulating autophagy in a caerulein-induced acute pancreatitis(AP)cellular model.GFP-LC3 immunofluo-rescence assay was performed to detect autophagy vesicle formation in pancreatic acinar cell line AR42J.AR42J cells were transfected with miR-155 mimic,inhibitor,and corresponding controls to explore the effect of miR-155 on autophagy.The protein levels of LC3-I,LC3-11,Beclin-1,and p62 were analyzed by western blot analysis.Dual-luciferase reporter assay was performed to verify the interaction between miR-155 and Rictor(RPTOR independent companion of MTOR complex 2).The results showed that caerulein treatment induced impaired autophagy as evi-denced by an increase in the accumulation of p62 together with LC3-11 in AR42J cells,accompanied by miR-155 upregulation.Furthermore,miR-155 overexpression aggravated,whereas miR-155 silencing reduced the caerulein-induced impairment of autophagy.Mechanistically,Rictor was confirmed to be a direct target of miR-155,which could rescue the miR-155 overexpression-mediated aggravation of im paired autophagy.Collectively,these findings indicate that miR-155 aggravates impaired autophagy in caerulein-treated pancreatic acinar cells by targeting Rictor.Xueming Zhang Jiangtao Chu Haijun Sun Dali Zhao Biao Ma Dongbo Xue Weihui Zhang Zhituo Li 2020Acta Biochimica et Biophysica Sinica2020,52,2:4

11Tonoplast-associated calcium signaling regulates manganese homeostasis in Arabidopsis显示文摘Manganese(Mn)is an essential micronutrient in plants.However,excessive Mn absorption in acidic and waterlogged soils can lead to Mn toxicity.Despite their essential roles in Mn homeostasis,transcriptional and post-transcriptional modifications of Mn transporters remain poorly understood.Here,we demonstrated that high-Mn stress induces an obvious Ca^(2+) signature in Arabidopsis.We identified four calcium-dependent protein kinases,CPK4/5/6/11,that interact with the tonoplast-localized Mn and iron(Fe)transporter MTP8 in vitro and in vivo.The cpk4/5/6/11 quadruple mutant displayed a dramatic high-Mn-sensitive phenotype similar to that of the mtp8 mutant.CPKs phosphorylated the N-terminal domain of MTP8 primarily at the Ser31 and Ser32 residues.Transport assays combined with multiple physiological experiments on phospho-dead variant MTP8^(S31/32A)and phospho-mimetic variant MTP8^(S31/32D)plants under different Mn and Fe conditions suggested that Ser31 and Ser32 are crucial for MTP8 function.In addition,genetic analysis showed that CPKs functioned upstream of MTP8.In summary,we identified a tonoplast-associated calcium signaling cascade that orchestrates Mn homeostasis and links Mn toxicity,Ca^(2+) signaling,and Mn transporters.These findings provide new insight into Mn homeostasis mechanisms and Ca^(2+) signaling pathways in plants,providing potential targets for engineering heavy metal toxicity-tolerant plants.Zhenqian Zhang Dali Fu Zhihui Sun Chuanfeng Ju Cuicui Miao Zhangqing Wang Dixiang Xie Liang Ma Zhizhong Gong Cun Wang 2021Molecular Plant2021,14,5:2

13Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. 15Geochronology,geochemistry and Hf isotope of monzogranite in Niubiziliang of Qinghai显示文摘Zircon U-Pb dating, whole-rock geochemical analyses and Hf isotope are undertaken for the monzogranite in Niubiziliang area with the aim of constraining its formation time,petrogenesis and the regional tectonic setting. The zircons from monzogranite are euhedral-subhedral in shape,and display rhythm growth zoning,indicating a magmatic origin. LA-ICP-MS zircon U-Pb dating indicates the monzogranite formed most probably in the Late Devonian( 359. 0 ± 2. 6 Ma). The monzogranite has Si O2= 74. 69%--76. 11%,Al2O3=12. 07%--12. 81%,Na2 O + K2 O = 8. 24%--8. 70%,Na2 O / K2 O = 0. 60--0. 68,A / CNK > 1,which shows that it belongs to high-Si and high-K weakly peraluminous calc-alkaline series. The monzogranite is enriched in K,Rb,Th( LILEs) and La,Ce,Sm,Nd( LREEs); and depleted in Ba,U( HREEs) and Ta,Nb( HFSEs).Their zircon εHf( t) values range from 1. 21 to 3. 46,in response to their two-stage Hf model ages( TDM2) ranging from 1 034 Ma to 1 159 Ma,respectively,indicating that the primary magma was derived from the young crust in Meso-Neoproterozoic. 18Zircon U-Pb geochronology and geochemistry of syenogranitesin western Niubiziliang,northwestern Qaidam,Qinghai显示文摘The study presents the results of U-Pb dating of zircons and whole-rock geochemical analyses of a syenogranite located in the western Niubiziliang area,China with the aim of determining its formation time,petrogenesis and the regional tectonic setting. Zircons within the syenogranite are euhedral-subhedral and display rhythmic growth zoning,indicating a magmatic origin. Zircon U-Pb data obtained by LA-ICP-MS indicate the syenogranite formed in the Late Permian (260.7±1.5 Ma). The w(SiO_2) of syenogranites is 70.82%--73.59%,w(Al_2O_3) is 13.49%--14.82%,and w(Na_2O + K_2O) is 7.85%--8.52%,and yield K_2O/Na_2O ratios of 1.06--1.26. Therefore,the syenogranites belong to the high-K calc-alkaline and metaluminous (A/CNK< 1,A/NK > 1) series which display I-type granites similarly. The syenogranites also show the geochemical characteristics of volcanic arc rocks,being enriched in large-ion lithophile elements (LILEs; K,Rb) and light rare-earth elements (LREEs; La,Ce,Sm,Nd),but depleted in high field strength elements( HFSEs; Ta,Nb,P,Ti). On the whole,trace element ratios are close to the mean of the Earth's crust,indicating a lowercrust magma source. The low Sr (161--214) ×10^(-6) and Yb (1.08--1.80)×10^(-6) concentrations indicate that plagioclase and hornblende are residual mineral phases in the source.
