1Heterotrimeric G protein α subunit is involved in rice brassinosteroid response显示文摘Heterotrimeric G proteins are known to function as messengers in numerous signal transduction pathways.The nullmutation of RGA(rice heterotrimeric G protein α subunit),which encodes the α subunit of heterotrimeric G proteinin rice,causes severe dwarfism and reduced responsiveness to gibberellic acid in rice.However,less is known aboutheterotrimeric G protein in brassinosteroid(BR)signaling,one of the well-understood phytohormone pathways.In thepresent study,we used root elongation inhibition assay,lamina inclination assay and coleoptile elongation analysis todemonstrated reduced sensitivity of dl mutant plants(caused by the null mutation of RGA)to 24-epibrassinolide(24-epiBL),which belongs to brassinosteroids and plays a wide variety of roles in plant growth and development.Moreover,RGA transcript level was decreased in 24-epiBL-treated seedlings in a dose-dependent manner.Our results show thatRGA is involved in rice brassinosteroid response,which may be beneficial to elucidate the molecular mechanisms of Gprotein signaling and provide a novel perspective to understand BR signaling in higher plants.Lei Wang Yun-Yuan Xu Qi-Bin Ma Dan Li Zhi-Hong Xu Kang Chong 2006Cell Research2006,16,12:29
2Enhanced CT and CT virtual endoscopy in diagnosis of heterotopic pancreas显示文摘AIM:To improve the diagnosis of heterotopic pancreas by the use of contrastenhanced computed tomography(CT)imaging and CT virtual endoscopy(CTVE).METHODS:A total of six patients with heterotopic pancreas,as confirmed by clinical pathology and immunohistochemistry in the Sixth Affiliated People's Hospital of Shanghai Jiao Tong University,Shanghai,China,were included.Nonenhanced CT and enhanced CT scanning were performed,and the resulting images were reviewed and analyzed using threedimensional postprocessing software,including CTVE.RESULTS:Four males and two females were enrolled.Several heterotopic pancreas sites were involved;three occurred in the stomach,including the gastric antrum(n =2)and lesser curvature(n=1),and two were in the duodenal bulb.Only one case of heterotopic pancreas lesion occurred in the mesentery.Four cases had a solid yet soft tissue density that had a homogeneous pattern when viewed by enhanced CT.Additionally,their CT values were similar to that of the pancreas.The ducts of the heterotopic pancreas tissue,one of the characteristic CT features of heterotopic pancreas tissue,were detected in the CT images of two patients.CTVE images showed normal mucosa around the tissue,which is also an important indicator of a heterotopic pancreas.However,none of the CTVE images showed the typical signs of central dimpling or umbilication.CONCLUSION:CT,enhanced CT and CTVE techniques provide useful information about the location,growth pattern,vascularity,and condition of the gastrointestinal wall around heterotopic pancreatic tissue.Dan Wang Xiao-Er Wei Lei Yan Yu-Zhen Zhang Wen-Bin Li 2011World Journal of Gastroenterology2011,17,33:28
3Pathways of China's PM;air quality 2015–2060 in the context of carbon neutrality显示文摘Clean air policies in China have substantially reduced particulate matter(PM;) air pollution in recent years,primarily by curbing end-of-pipe emissions.However,reaching the level of the World Health Organization(WHO) guidelines may instead depend upon the air quality co-benefits of ambitious climate action.Here,we assess pathways of Chinese PM;air quality from 2015 to 2060 under a combination of scenarios that link global and Chinese climate mitigation pathways(i.e.global 2℃-and 1.5℃-pathways,National Determined Contributions(NDC) pledges and carbon neutrality goals) to local clean air policies.We find that China can achieve both its near-term climate goals(peak emissions) and PM;air quality annual standard(35 μg/m;) by 2030 by fulfilling its NDC pledges and continuing air pollution control policies.However,the benefits of end-of-pipe control reductions are mostly exhausted by 2030,and reducing PM;exposure of the majority of the Chinese population to below 10 μg/m;by 2060 will likely require more ambitious climate mitigation efforts such as China’ s carbon neutrality goals and global1.5℃-pathways.Our results thus highlight that China’s carbon neutrality goals will play a critical role in reducing air pollution exposure to the level of the WHO guidelines and protecting public health.Jing Cheng Dan Tong Qiang Zhang Yang Liu Yu Lei Gang Yan Liu Yan Sha Yu Ryna Yiyun Cui Leon Clarke Guannan Geng Bo Zheng Xiaoye Zhang Steven JDavis Kebin He 2021National Science Review2021,8,12:23
4Diel and seasonal variation of methane and carbon dioxide fluxes at Site Guojiaba, the Three Gorges Reservoir显示文摘In order to investigate the CH4and CO2fluxes across the water-gas interface and identify their controlling factors, four diel field campaigns and one monthly sampling campaign during June 2010–May 2011 were carried out at a site near the Three Gorges Dam,China. The averaged CH4and CO2fluxes across the air-water interface from the site were much less than those reported from reservoirs in tropic and temperate regions, and from the natural river channels of the Yangtze River. CH4Fluxes at the site were very low compared to most other reservoirs or natural lakes. One of the most important reasons may be due to the oxidation of CH4in the water column owing to the great water depth and high DO in water in the Three Gorges Reservoir. The averaged monthly CH4and CO2fluxes at the site during the observation year were 0.05 mg/(m2·hr) and 104.43 mg/(m2·hr) respectively with the maximum occurred in July 2010. The monthly CO2fluxes during the observation year were positively correlated to the surface water temperature, and negatively correlated to the air pressure and the surface water pH. The CO2flux showed a positive correlation with DOC to some extent, although not significantly, which indicated that allochthonous organic C was a major source of CO2and biogeochemical processes in this reservoir were C-limited. 6Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 7Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE.Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu 2019Cellular & Molecular Immunology2019,16,3:15
8Structured illumination microscopy for super-resolution and optical sectioning显示文摘Optical microscopy plays an essential role in biological studies due to its capability and compatibility of non-contact,minimally invasive observation and measurement of live specimens.However,the conventional optical microscopy only has a spatial resolution about200 nm due to the Abbe diffraction limit,and also lacks the ability of three-dimensional imaging.Super-resolution farfield optical microscopy based on special illumination schemes has been dramatically developed over the last decade.Among them,only the structured illumination microscopy(SIM)is of wide-field geometry that enables it easily compatible with the conventional optical microscope.In this article,the principle of SIM was introduced in terms of point spread function(PSF)and optical transform function(OTF)of the optical system.The SIM for super-resolution(SIM-SR)proposed by Gustafsson et al.and the SIM for optical sectioning(SIM-OS)proposed by Neil et al.are the most popular ones in the research community of microscopy.They have the same optical configuration,but with different data postprocessing algorithms.We mathematically described the basic theories for both of the SIMs,respectively,and presented some numerical simulations to show the effects of super-resolution and optical sectioning.Various approaches to generation of structured illumination patterns were reviewed.As an example,a SIM system based on DMDmodulation and LED-illumination was demonstrated.A lateral resolution of 90 nm was achieved with gold nanoparticles.The optical sectioning capability of the microscope was demonstrated with Golgi-stained mouse brain neurons,and the sectioning strength of 930 nm was obtained.Dan Dan Baoli Yao Ming Lei 2014Chinese Science Bulletin2014,59,12:14
9CapOX as neoadjuvant chemotherapy for locally advanced operable colon cancer patients: a prospective single-arm phase Ⅱ trial显示文摘Objective: The aim of this prospective, single-arm phase II trial was to confirm the safety and efficacy of neoadjuvant chemotherapy(NAC) using oxaliplatin plus capecitabine(Cap OX) for patients with operable locally advanced colon cancer(CC).Methods: Patients with computed tomography-defined T4 or lymph node-positive CCs were enrolled. After radiological staging, patients were treated with at least 2 cycles of NAC consisting of 130 mg/m2 oxaliplatin on d 1,plus 1,000 mg/m2 capecitabine twice daily for 14 d every 3 weeks, followed by surgery, and then with the rest cycles of adjuvant chemotherapy. Radiological response was evaluated after 2 cycles of NAC. Tumor response, treatment toxicity, and surgical complications were recorded. The pathological response to therapy was evaluated according to the tumor regression grade(TRG) score. The primary endpoint was pathologic tumor response. This trial is registered in Clinical Trials.gov(No: NCT02415829).Results: Forty-seven patients were enrolled in the study. Forty-two patients completed the planned treatments.The total radiological response rate was 68%(32/47), including complete and partial response rates of 2%(1/47)and 66%(31/47), respectively. Stable disease was observed in 32%(15/47) and progressive disease was observed in none. Complete pathologic response, major regression, and at least moderate regression were achieved in 1(2%), 2(4%), and 29(62%) patients, respectively. Four patients developed grade 3 treatment toxicities. One patient with wound infection occurred after operation(1/47, 2%). 13Development of a core set of SNP markers for the identifi cation of upland cotton cultivars in China显示文摘Considering the advantages of single nucleotide polymorphisms(SNP) in genotyping and variety identification, the first set public SNP markers at Cotton Marker Database(http://www.cottonmarker.org/) were validated and screened across standard varieties of cotton distinctness, uniformity and stability(DUS) test, aiming to obtain an appropriate set of core SNP markers suitable for upland cotton cultivars in China. A total of 399 out of 1 005 SNPs from 270 loci including 170 insertions-deletions(In Dels) were evaluated for their polymorphisms among 30 standard varieties using Sanger sequencing. As a result, 147 loci were sequenced successfully, 377 SNPs and 49 In Dels markers were obtained. Among the 377 SNP markers, 333 markers(88.3%) were polymorphic between Gossypium hirsutum and G. barbadense, while 164 markers(43.5%) were polymorphic within upland cotton. As for In Del markers, the polymorphic rate is relatively lower than that of SNP both between species and within species. The homozygous DNA locus ratio of 121 SNPs was higher than 86.2% while that of other 43 SNPs was less than 70%. Only 64 SNPs displayed completely homozygous genotypes among all of the detected upland cotton varieties with 100% homozygous DNA locus ratio. At last, a set of 23 pairs of core SNPs were achieved in view of avoidance of linkage, with polymorphism information content(PIC) values varying from 0.21 to 0.38 with an average of 0.28. Genotype characteristics and genetic diversity were analyzed based on the set of core markers, while 40 pairs of core simple-sequence repeats(SSR) primers comprised of 10 sets of four multiplex PCR combinations were also used for analysis based on fluorescence detection system. Comparison results indicated that the genetic diversity level was almost equal, while various varieties were significantly different from each other. Genetic relationship revealed by SSR markers is related to geographic source to a certain extent. 14Typical atmospheric haze during crop harvest season in northeastern China:A case in the Changchun region显示文摘This study presents the mass concentrations of PM_(2.5),O_3,SO_2 and NOxat one urban,one suburban and two rural locations in the Changchun region from September 25 to October 272013. Major chemical components of PM_(2.5)at the four sites were daily sampled and analyzed. Most of daily concentrations of SO_2(7–82 μg/m^3),O_3(27–171 μg/m^3) and NOx(14–213 μg/m^3) were below the limits of the National Ambient Air Quality Standard(NAAQS)in China. However,PM_(2.5)concentrations(143–168 μg/m^3) were 2-fold higher than NAAQS.Higher PM_(2.5)concentrations(~ 150 μg/m^3) were measured during the pre-harvest and harvest at the urban site,while PM_(2.5)concentrations significantly increased from 250 to400 μg m^(-3) at suburban and rural sites with widespread biomass burning. At all sites,PM_(2.5)components were dominated by organic carbon(OC) and followed by soluble component sulfate(SO_4^(2-)),ammonium(NH_4^+) and nitrate(NO_3^-). Compared with rural sites,urban site had a higher mineral contribution and lower potassium(K^+and K) contribution to PM_(2.5).Severe atmospheric haze events that occurred from October 21 to 23 were attributed to strong source emissions(e.g.,biomass burning) and unfavorable air diffusion conditions.Furthermore,coal burning originating from winter heating supply beginning on October 18 increased the atmospheric pollutant emissions. 16Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin 2014Science China(Life Sciences)2014,57,2:9

19Removal of Pb(Ⅱ) from aqueous solution by hydrous manganese dioxide:Adsorption behavior and mechanism显示文摘Hydrous manganese dioxide (HMO) synthesized by redox of potassium permanganate and hydrogen peroxide was used as an adsorbent for Pb(Ⅱ) removal.The specific surface area,pore volume and BJH pore diameter of the HMO were 79.31m2/g,0.07cm3/g and 3.38 nm,respectively.The adsorption equilibrium at 298K could be well described by the Langmuir isotherm equation with q max value of 352.55mg/g.The negative values of G and the positive values of H and S indicated the adsorption process was spontaneous and endothermic.The pseudo second-order equation could best fit the adsorption data.The value of the calculated activation energy for Pb(Ⅱ) adsorption onto the HMO was 38.23 kJ/mol.The uptake of Pb(Ⅱ) by HMO was correlated with increasing surface hydroxyl group content and the main adsorbed speciation was PbOH+.The final chemical state of Pb(Ⅱ) on the surface of HMO was similar to PbO.HMO was a promising candidate for Pb(Ⅱ) removal from aqueous solution.Meng Xu Hongjie Wang Di Lei Dan Qu Yujia Zhai Yili Wang 2013Journal of Environmental Sciences2013,25,3:9
20Numerical simulation of land subsidence and verification of its character for an iron mine using sublevel caving显示文摘Combined with a digital bored photography system and in-situ statistics concerning the joints and fissures of both ore-body and surrounding rock,a 2D discrete model was constructed using UDEC.The stress field and displacement field changes of different sublevel stoping systems were also studied.Changes in the overlying rock strata settlement pattern has been analyzed and validated by in-situ monitoring data.The results show that:in the caving process,there exists an obvious delay and jump for the overlying rock strata displacement over time,and a stable arch can be formed in the process of caving,which leads to hidden goafs.Disturbed by the mining activity,a stress increase occurred in both the hanging wall and the foot wall,demonstrating a hump-shaped distribution pattern.From the comparison between simulation results and in-situ monitoring results,land subsidence shows a slow-development,suddenfailure,slow-development cycle pattern,which leads eventually to a stable state.This pattern validates the existence of balanced arch and hidden goafs.Cao Shuai Song Weidong Deng Dan Lei Yuankun Lan Jianqiang 2016International Journal of Mining Science and Technology2016,26,2:9
