3Toxicity of Graphene Quantum Dots in Zebrafish Embryo显示文摘Objective To evaluate the bio-safety of graphene quantum dots(GQDs), we studied its effects on the embryonic development of zebrafish. Methods In vivo, biodistribution and the developmental toxicity of GQDs were investigated in embryonic zebrafish at exposure concentrations ranging from 12.5-200 μg/m L for 4-96 h post-fertilization(hpf). The mortality, hatch rate, malformation, heart rate, GQDs uptake, spontaneous movement, and larval behavior were examined. Results The fluorescence of GQDs was mainly localized in the intestines and heart. As the exposure concentration increased, the hatch and heart rate decreased, accompanied by an increase in mortality. Exposure to a high level of GQDs(200 μg/mL) resulted in various embryonic malformations including pericardial edema, vitelline cyst, bent spine, and bent tail. The spontaneous movement significantly decreased after exposure to GQDs at concentrations of 50, 100, and 200 μg/mL. The larval behavior testing(visible light test) showed that the total swimming distance and speed decreased dose-dependently. Embryos exposed to 12.5 μg/mL showed hyperactivity while exposure to higher concentrations(25, 50, 100, and 200 μg/mL) caused remarkable hypoactivity in the light-dark test. Conclusion Low concentrations of GQDs were relatively non-toxic. 4Passive sensitization increases histamine-stimulated calcium signaling and NF-κB transcription activity in bronchial epithelial cells显示文摘Aim:To find out if the two aspects of asthma(chronic airway inflammation andbronchial hyperresponsiveness)are related to hypersensitivity of calcium signal-ing in bronchial epithelial cells.Methods:Porcine bronchial epithelial cells(PBEC)were divided into sensitized(S)and nonsesitized(N)groups.In group S,the cellswere preincubated with serum from ovalbumin sensitized guinea pigs.In group N,the cells were preincubated with serum from nonsensitized guinea pigs.Singlecell calcium imaging and ELISA-based NF-κ3 activity were used to evaluate thehistamine-stimulated intracellular free calcium level and NF-κB activity,respectively.Results:First,0.1μmol/L histamine could induce [Ca^(2+)]_i oscillationsin PBEC of group S,but not in group N.Second,1/Jmol/L histamine could induce[Ca^(2+)]_i oscillations of PBEC in both group S and group N.The [Ca^(2+)]_i oscillationfrequency of PBEC was significantly higher in group S than in group N,thoughthe[Ca^(2+)]_i oscillation amplitude showed no difference between the two groups.Finally,when 10 μmol/L histamine was used to stimulate PBEC a transient initialincrease followed by a sustained elevation(FSE)of [Ca^(2+)]_i was observed in PBECin both groups.The amplitude of the FSE of [Ca^(2+)]_i in PBEC was significantlyhigher in group S than in group N.The subsequent NF-κB activity was in accor-dance to the calcium oscillation frequency evoked by histamine,but not to theamplitude.Conclusion:It was suggested that the increased sensitivity of calciumsignaling in bronchial epithelial cells might contribute to the exorbitant inflamma-tion or increased susceptibility in asthmatic airway epithelial cells.Si JIN Dan TIAN Jian-guo CHEN Li-ping ZHU Sheng-yuan LIU Di-xun WANG 2006Acta Pharmacologica Sinica2006,27,6:4
5Reduced graphene oxide@carbon sphere based metacomposites for temperature-insensitive and efficient microwave absorption显示文摘With the increasing advance of fifth generation(5G)network and the gradual expansion of digital devices,harsh working environment for electronic devices has spawned higher requirements for microwave absorbing materials(MAMs).Since both the electromagnetic response and energy conversion character vary with temperature,to achieve temperature insensitive microwave absorption behaviour in wide temperature range has become extremely challenging.In this work,structured metacomposites containing sub-wavelength reduced graphene oxide(RGO)@carbon spheres were fabricated,and the microwave absorption was further improved through structural and composition design of the RGO@carbon units.Due to the unique anti-reflection effect on microwave of the metacomposites,the temperature-insensitive electromagnetic performance at elevated temperature could be exhibited.Moreover,both the dielectric relaxation behaviour and microwave absorption proformance of the system could be further increased.As a result,the effective absorption bandwidth(reflection loss(RL)<−10 dB)of the metacomposites with only 3.0 wt.%filler content could cover the entire X-band(8.2–12.4 GHz)frequency ranging from 298 to 473K.The metacomposite proposed in this work provides a“de-correlating”strategy to break the linkage between microwave absorption behaviour and temperature,which offers an interesting plateau for fabricating efficient high-temperature microwave absorption structures with tunable and designable advantages.Zhiyang Jiang Haoxu Si Yi Li Dan Li Huihui Chen Chunhong Gong Jingwei Zhang 2022Nano Research2022,15,9:4
6TiN/BN composite with excellent thermal stability for efciency microwave absorption in wide temperature spectrum显示文摘High-efficiency and temperature-insensitive microwave absorbing materials(MAMs) are ideal for hightemperature electromagnetic attenuation.Herein,the titanium nitride/boron nitride(TiN/BN) composite with TiN as the loss unit and BN as the impedance matching unit has been constructed by the in-situ synthesis method.The insulating BN not only effectively regulates and optimizes the conductivity and impedance matching of the material but also imparts steady cyclic microwave absorption properties.The steady dielectric loss ensures that TiN/BN composite has robust high-temperature absorption properties at X-band,with a minimum reflection loss(RL_(min)) of-16.74 dB at 873 K and an effective absorption bandwidth(EAB) of 3.26 GHz in the temperature range of 293-873 K.Compared with the TiN/SiO2absorbers with a single system,the TiN/BN/SiO2composite keeps reliable high temperature absorbing properties at873 K.This work confirms that wave-transparent materials for dielectric property modulation can resolve poor temperature stability of effective absorption and non-reusability for high-temperature absorbing materials,providing inspirations for designing efficiency high-temperature microwave absorbers.Yupeng Shi Dan Li Haoxu Si Zhiyang Jiang Mengyuan Li Chunhong Gong 2022Journal of Materials Science & Technology2022,,35:3
7Lupeol protects against cardiac hypertrophy via TLR4-PI3K-Akt-NF-κB pathways显示文摘Inflammation and apoptosis are main pathological processes that lead to the development of cardiac hypertrophy.Lupeol,a natural triterpenoid,has shown anti-inflammatory and anti-apoptotic activities as well as potential protective effects on cardiovascular diseases.In this study we investigated whether lupeol attenuated cardiac hypertrophy and fibrosis induced by pressure overload in vivo and in vitro,and explored the underlying mechanisms.Cardiac hypertrophy was induced in mice by transverse aortic constriction(TAC)surgery,and in neonatal rat cardiomyocytes(NRCMs)by stimulation with phenylephrine(PE)in vitro.We showed that administration of lupeol(50 mg·kg^(-1)·d^(-1),i.g.,for 4 weeks)prevented the morphological changes and cardiac dysfunction and remodeling in TAC mice,and treatment with lupeol(50μg/mL)significantly attenuated the hypertrophy of PE-stimulated NRCMs,and blunted the upregulated hypertrophic markers ANP,BNP,andβ-MHC.Furthermore,lupeol treatment attenuated the apoptotic and inflammatory responses in the heart tissue.We revealed that lupeol attenuated the inflammatory responses including the reduction of inflammatory cytokines and inhibition of NF-κB p65 nuclear translocation,which was mediated by the TLR4-PI3K-Akt signaling.Administration of a PI3K/Akt agonist 740 Y-P reversed the protective effects of lupeol in TAC mice as well as in PE-stimulated NRCMs.Moreover,pre-treatment with a TLR4 agonist RS 09 abolished the protective effects of lupeol and restored the inhibition of PI3K-Akt-NF-κB signaling by lupeol in PE-stimulated NRCMs.Collectively,our results demonstrate that the lupeol protects against cardiac hypertrophy via anti-inflammatory mechanisms,which results from inhibiting the TLR4-PI3K-Akt-NF-κB signaling.Dan Li Ying-ying Guo Xian-feng Cen Hong-liang Qiu Si Chen Xiao-feng Zeng Qian Zeng Man Xu Qi-zhu Tang 2022Acta Pharmacologica Sinica2022,43,8:3
8Effects of RNAi Silencing of SSIII Gene on Phosphorus Content and Characteristics of Starch in Potato Tubers显示文摘The sense and antisense fragments of the soluble starch synthase (SSIII) gene and the intron fragment of somatic embryogenesis receptor-like kinase (SERK1) gene were cloned from potato using PCR techniques. The RNAi plant expression vectors pBI-SSIII-RNAi and pBIC-SSIII-RNAi were constructed which containing fusion fragment of sense fragment-intron-antisense fragment'driven by the constitutive expression promoter CaMV 35S and the tuber-specific expression promoter CIPP, respectively. The putative transgenic plants of potato cultivars Kexin-1 and Kexin-4 were obtained using Agrobacterium-mediated transformation method. PCR assay showed that the interference fragment of SSIII gene was integrated into potato genome. The RT-PCR analysis showed that the expression of SSIII gene was repressed apparently on the transcription level. Starch granules of the transgenic potato plants were different in morphology and became cracked in starch granule centre compared with the non-transgenic control plants. The amylose content of starch was increased by 2.68-29.05%, amylopectin to amylose ratio of starch had declined significantly, and the phosphorus content of the starch of the transgenic plants was reduced 9.94-58.36% compared with control plants. 11Investigation of immune escape-associated mutations of hepatitis B virus in patients harboring hepatitis B virus drug-resistance mutations显示文摘It is unclear whether immune escape-associated mutations in the major hydrophilic region of hepatitis B virus surface antigen(HBsAg)are associated with nucleoside/nucleotide analog resistance.AIM To evaluate the association between immune escape-associated mutations and nucleoside/nucleotide analog resistance mutations.METHODS In total,19440 patients with chronic hepatitis B virus infection,who underwent resistance testing at the Fifth Medical Center of Chinese PLA General Hospital between July 2007 and December 2017,were enrolled.As determined by sequence analysis,6982 patients harbored a virus with resistance mutations and 12458 harbored a virus lacking resistance mutations.Phenotypic analyses were performed to evaluate HBsAg production,replication capacity,and drug-induced viral inhibition of patient-derived drug-resistant mutants with or without the coexistence of sA159V.RESULTS The rate of immune escape-associated mutation was significantly higher in 9 of the 39 analyzed mutation sites in patients with resistance mutations than in patients without resistance mutations.In particular,these mutations were sQ101H/K/R,sS114A/L/T,sT118A/K/M/R/S/V,sP120A/L/Q/S/T,sT/I126A/N/P/S,sM133I/L/T,sC137W/Y,sG145A/R,and sA159G/V.Among these,sA159V was detected in 1.95%(136/6982)of patients with resistance mutations and 1.08%(134/12,458)of patients lacking resistance mutations(P<0.05).The coexistence of sA159V with lamivudine(LAM)and entecavir(ETV)-resistance mutations in the same viral genome was identified during follow-up in some patients with drug resistance.HBsAg production was significantly lower and the replication capacity was significantly higher,without a significant difference in LAM/ETV susceptibility,in sA159V-containing LAM/ETV-resistant mutants than in their sA159V-lacking counterparts.CONCLUSION In summary,we observed a close link between the increase in certain immune escape-associated mutations and the development of resistance mutations.sA159V might increase the fitness of LAM/ETV-resistant mutants under environmental pressure in some cases.Bi-Xia Huang Yan Liu Zhen-Ping Fan Lan-Lan Si Rong-Juan Chen Jun Wang Dan Luo Fu-Sheng Wang Dong-Ping Xu Xin-Guang Liu 2020World Journal of Gastroenterology2020,26,35:2

15Economic evaluations of inactivated COVID-19 vaccines in six Western Pacific and South East Asian countries and regions:A modeling study显示文摘Objective:The present study aimed to document the economic profiles of inactivated COVID-19 vaccines in Hong Kong SAR,Indonesia,China's Mainland,Philippines,Singapore,and Thailand,the evidence on which is currently absent.Methods:Decision tree models were developed to assess the cost-effectiveness of two doses of inactivated COVID-19 vaccines at a population vaccination rate of 50%in the base case,which was an estimate of feasible vaccination coverage according to previous studies.Epidemiological,mortality,cost,and health state utility information were sourced from the literature.Vaccine efficacy against COVID-19 cases by severity were estimated using metaanalyses of publicly accessible phase 3 trial results of inactivated vaccines.The health outcomes were quantified as quality-adjusted life years(QALYs)and compared across the vaccination and no vaccination strategies.In scenario analyses,incidence and vaccination rates were changed semi-continuously over spectrums,the results of which were presented as contour lines informing the efficiency frontiers of vaccination strategies.Oneway and probabilistic sensitivity analyses were also conducted.Results:The vaccination strategy was dominant in all jurisdictions in the base case by producing 105.18,98.15,99.70,60.48,112.00,and 103.47 QALYs while saving US$40.26 million,US$5.26 million,US$7.60 million,US$5.91 million,US$21.33 million,and US$7.18 million in Hong Kong SAR,Indonesia,China's Mainland,Philippines,Singapore,and Thailand per every 100,000 vaccinated individuals,respectively.Results were robust in alternative model specifications.Conclusions:Inactivated COVID-19 vaccines may be cost-saving options in Hong Kong SAR,Indonesia,China's Mainland,Philippines,Singapore,and Thailand.Mass vaccination programs using inactivated COVID-19 vaccines should be considered in these jurisdictions.Yawen Jiang Dan Cai Si Shi 2022Infectious Disease Modelling2022,7,1:1

19Mediating role of resilience in the relationship between COVID-19 related stigma and mental health among COVID-19 survivors:a cross-sectional study显示文摘Background The global coronavirus disease 2019(COVID-19)has caused many negative effects on physical and mental health of patients who have survived COVID-19.Apart from some long-lasting physical sequelae,those COVID-19 survivors are also suffering stigma and discrimination at different levels around the world.The current study aims to assess the role resilience played in stigma and mental disorders among COVID-19 survivors.Methods The cross-sectional study was carried out among former COVID-19 patients in Jianghan District(Wuhan,China)from June 10 to July 25,2021.The demographic questions,the Impact of Events Scale-Revised,the Generalized Anxiety Disorder Questionnaire,the Patient Health Questionnaire,the Resilience Style Questionnaire and the Short Version of COVID-19 Stigma Scale of 12 items were used to collect relevant information of the participants.Descriptive analyses,Pearson correlation analysis and Structural Equation Modeling were used to make data description and analysis.Results A total of 1541 out of 1601 COVID-19 survivors(887 females and 654 males)were included in the analysis.Perceived stigma of those COVID-19 survivors correlates significantly with anxiety(r=0.335,P<0.001),depression(r=0.325,P<0.001)and post-traumatic stress disorder(PTSD)(r=0.384,P<0.001).It has a direct effect on COVID-19 survivors’anxiety(β=0.326,P<0.001),depression(β=0.314,P<0.001),PTSD(β=0.385,P<0.001)and their resilience(β=−0.114,P<0.01).Resilience partially mediated the association between perceived stigma and anxiety(β=0.020,P<0.01),depression(β=0.020,P<0.01),and PTSD(β=0.014,P<0.01)among COVID-19 survivors.Conclusion Stigma has a significant negative impact on mental health,while resilience plays a mediator role in the relationship between stigma and mental health among COVID-19 survivors.Based on our study,we suggested that when designing psychological interventions for COVID-19 survivors,consideration should be taken into account to reduce stigma and improve resilience.Weijun Xiao Xiaoyang Liu Hao Wang Yiman Huang Zhenwei Dai Mingyu Si Jiaqi Fu Xu Chen Mengmeng Jia Zhiwei Leng Dan Cui Winnie W.S.Mak Liming Dong Xiaoyou Su 2023Infectious Diseases of Poverty2023,12,2:0
