Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. Taken together, our findings reveal the direct protein targets and molecular mechanisms via which celastrol ameliorates hepatic fibrosis, thus supporting the further development of celastrol as a promising therapeutic agent for liver fibrosis. | Piao Luo Dandan Liu Qian Zhang Fan Yang Yin-Kwan Wong Fei Xia Junzhe Zhang Jiayun Chen Ya Tian Chuanbin Yang Lingyun Dai Han-Ming Shen Jigang Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,5: | 14
Multiaxial electrospun generation of hollow graphene aerogel spheres for broadb and high-performance microwave absorption显示文摘Although graphene aerogels(GA)have been attracted great attention, the easy-operation and large-scale production of GA are still challenges.Further, most GA have a monolith-like appearance, limiting their application-specific needs.Herein,we highlight graphene aerogel spheres with controllable hollow structures(HGAS)that are delicately designed and manufactured via coaxial electrospinning coupled with freeze-drying and calcination. The HGAS exhibit a spherical configuration at the macroscale, while the construction elements of graphene on the microscale showing an interconnected radial microchannel structure.Further ball-in-ball graphene aerogel spheres(BGAS)are obtained by reference to the triaxial electrospinning technology. The as-prepared spheres possess the controllable integrated conductive networks, leading to the effective dielectric loss and impedance matching thus bringing on high-performance microwave absorption. The as-obtained HGAS shows a minimum reflection loss of-52.7 dB and a broad effective absorption bandwidth(f)of 7.0 GHz with thickness of 2.3 mm.Further, the fe reaches 9.3 GHz for BGAS with thickness of 3.4 mm.Aforementioned superior microwave absorption of HGAS and BGAS confirms combination of multiaxial electrospinning and freeze-drying on the multiscale is an effective strategy for scalable fabrication of advanced microwave.absorbing functional graphene aerogel spheres. | Tian Li Dandan Zhi Yao Chen Bing Li Zuowan Zhou Fanbin Meng | 2020 | Nano Research2020,13,2: | 13
Preparation of modified Ce-SAPO-34 catalysts and their catalytic performances of methanol to olefins显示文摘The modified Ce-SAPO-34 catalysts were prepared with three methods, i.e., the liquid ion exchange with air calcination, impregnation with air calcination and impregnation with steam calcination methods. The catalytic performances of the catalysts for methanol to olefins were investigated. The properties of the catalysts were characterized using XRD, BET, XRF, FT-IR and NH3-TPD. The results indicated that compared to the SAPO-34 catalyst the catalyst prepared with the impregnation and air calcination prolonged the lifetime by 40 min and improved the selectivity to ethylene by 5% (mol) and the catalyst prepared with the impregnation and steam calcination showed the best modification effect, prolonging the lifetime by 70 min and improving the ethylene selectivity by 10% (mol). The catalyst prepared with the liquid ion exchange showed similar behaviour as the SAPO-34 catalyst. It was verified that the porous structure and surface acidity of these catalysts determined their catalytic behaviors. | Shuxun Tian Shengfu Ji Dandan Lü Bingyang Bai Qi Sun | 2013 | Journal of Energy Chemistry2013,22,4: | 10

The shallow Longmenshan crustal S-velocity structure of the fault zone using ambient noise tomography of a seismic dense array显示文摘The Longmenshan fault zone(LMSF),characterized by complex structures and strong seismicity,is located at the junction between the eastern margin of the Tibetan Plateau and the north-western Sichuan basin.Since the Wenchuan earthquake on May 12,2008,abundant studies of the formation mechanism of earthquakes along the LMSF were performed. In this study,a short-period dense seismic array deployed across the LMSF was applied by ambient noise tomography.Fifty-two 3-D seismic instruments were used for data acquisition for 26 days.We calculated the empirical Green's functions(EGFs)between different station-pairs and extracted 776 Rayleigh-wave dispersion curves between 2 and 7 s.And then,we used the direct-inversion method to obtain the fine shallow crustal S-wave velocity structure within 6 km depth in the middle section of the Longmenshan fault zone and nearby areas. Our results show that the sedimentary layer(>5 km)exists in the northwest margin of Sichuan Basin with a low S-wave velocity(~1.5-2.5 km/s)which is much thicker than that beneath the Longmenshan fault zone and the Songpan-Garze block. The high-velocity structures with clear boundaries below the middle of Longmenshan fault zone(~2-4 km)and the Songpan-Garze block(~4.5-6 km)probably reveal the NW-SE distribution patterns of both the Pengguan complex and the high-density belt hidden in the northwest of the Pengguan complex. And the obviously high-velocity anomalies observed at the depth of^1-2 km in the southeastern margin of the Songpan-Garze block can be considered as the Laojungou granites. Our results provide a high-resolution shallow velocity structure for detailed studies of the Longmenshan fault zone. | Dandan Li Gaochun Wang Ruihua Lin Kai Deng Xiaobo Tian | 2019 | Earthquake Science2019,32,5: | 3
Impacts of exercise intervention on various diseases in rats显示文摘Background:Exercise is considered as an important intervention for treatment and prevention of several diseases, such as osteoarthritis, obesity, hypertension, and Alzheimer's disease. This review summarizes decadal exercise intervention studies with various rat models across 6 major systems to provide a better understanding of the mechanisms behind the effects that exercise brought. Methods:PubMed was utilized as the data source. To collect research articles,we used the following terms to create the search:(exercise[Title]OR physical activity[Title]OR training[Title])AND(rats[Title/Abstract]OR rat[Title/Abstract]OR rattus[Title/Abstract]).To best cover targeted studies, publication dates were limited to'within 11 years.'The exercise intervention methods used for different diseases were sorted according to the mode, frequency, and intensity of exercise. Results:The collected articles were categorized into studies related to 6 systems or disease types:motor system(17 articles),metabolic system(110 articles),cardiocerebral vascular system(171 articles),nervous system(71 articles),urinary system(2 articles),and cancer(21 articles). Our review found that, for different diseases, exercise intervention mostly had a positive effect. However, the most powerful effect was achieved by using a specific mode of exercise that addressed the characteristics of the disease. Conclusion:As a model animal, rats not only provide a convenient resource for studying human diseases but also provide the possibility for exploring the molecular mechanisms of exercise intervention on diseases. This review also aims to provide exercise intervention frameworks and optimal exercise dose recommendations for further human exercise intervention research. | Ruwen Wang Haili Tian Dandan Guo Qianqian Tian Ting Yao Xingxing Kong | 2020 | Journal of Sport and Health Science2020,9,3: | 3
Sensing carboxylesterase 1 in living systems by a practical and isoformspecific fluorescent probe显示文摘Carboxylesterase 1(CES1), one of the most abundant serine hydrolases in mammals, has drawn much attentions in recent years, owing to this enzyme involves in many physiological processes via hydrolysis of both endogenous esters and xenobiotic esters. Herein, to real-time monitor the activities of CES1 in various biological systems, a practical and iso form-specific fluorescent probe was developed on the basis of the substrate preference of CES1, as well as the structural and optical properties of BODIPY dyes. After screening of a panel of BODIPY ester derivatives, probe 1 displayed the best combination of specificity, sensitivity, enzymatic kinetics and applicability for monitoring CES1 activities in real samples. This probe was successfully used to detect CESl activities in several biological systems including tissue preparations, living cells, tissue slices and zebrafish. Furthermore, the biomedical applications of probe 1 for screening of CES1 inhibitors were also demonstrated using tissue preparations or living cells as enzyme sources. In summary, a practical and broadly applicable tool for real-time monitoring CES1 in biological systems was developed and well-characterized, which held great promise for further investigations on CES1-associated drug discovery, clinical practice and fundamental research. | Lele Ding Zhenhao Tian Jie Hou Tongyi Dou Qiang Jin Dandan Wang Liwei Zou Yadi Zhu Yunqing Song Jingnan Cui Guangbo Ge | 2019 | Chinese Chemical Letters2019,30,3: | 2
Inhibitory effect of Sumu(Lignum Sappan) plus Fuzi(Radix Aconiti Lateralis Praeparata) on a lung carcinoma model显示文摘OBJECTIVE: To investigate the inhibitory effect of Sumu(Lignum Sappan) plus Fuzi(Radix Aconiti Lateralis Praeparata)(SF) on the growth and metastasis of Lewis lung carcinoma.METHODS: A lung carcinoma model was established by subcutaneously inoculating Lewis lung carcinoma cells into C57 BL/6 mice. The mice were randomly divided into four groups of 13 mice(control, low-dose, moderate-dose, and high-dose), and gavaged once-daily for 21 consecutive days. The rates of tumor inhibition, metastasis, and metastasis inhibition were observed. The differential expressions of s P-selectin and vascular endothelial growth factor C(VEGFC) were compared between the treatment groups and the control group.RESULTS: The tumor weights differed significantly between the treatment groups and the control group(P < 0.05). Administration of SF in the moderate-dosage and low-dose groups significantly inhibited the expression of s P-selectin and VEGFC(both P < 0.05), suggesting anti-tumor activity.CONCLUSION: SF can inhibit the growth and metastasis of Lewis lung carcinoma. | Zhang Hewei Ma Shuran Tian Tian Xu Ya Zhao Dandan Wang Tong Hou Chunying Zhang Shujing | 2018 | Journal of Traditional Chinese Medicine2018,38,2: | 1
Orally administered BZL-s RNA-20 oligonucleotide targeting TLR4 effectively ameliorates acute lung injury in mice显示文摘The global COVID-19 pandemic emerged at the end of December 2019.Acute respiratory distress syndrome(ARDS)and acute lung injury(ALI)are common lethal outcomes of bacterial lipopolysaccharide(LPS),avian influenza virus, and SARS-Co V-2.Toll-like receptor 4(TLR4)is a key target in the pathological pathway of ARDS and ALI.Previous studies have reported that herbal small RNAs(s RNAs)are a functional medical component.BZL-s RNA-20(Accession number:B59471456;Family ID:F2201.Q001979.B11)is a potent inhibitor of Toll-like receptor 4(TLR4)and pro-inflammatory cytokines.Furthermore,BZLs RNA-20 reduces intracellular levels of cytokines induced by lipoteichoic acid(LTA)and polyinosinic-polycytidylic acid(poly(I:C)).We found that BZL-s RNA-20 rescued the viability of cells infected with avian influenza H5N1,SARS-Co V-2,and several of its variants of concern(VOCs).Acute lung injury induced by LPS and SARS-Co V-2 in mice was significantly ameliorated by the oral medical decoctosome mimic(bencaosome;sphinganine(d22:0)+BZL-s RNA-20).Our findings suggest that BZLs RNA-20 could be a pan-anti-ARDS/ALI drug. | Dandan Zhao Yuhao Qin Jiaqi Liu Kegong Tang Shuaiyao Lu Zirui Liu Yexuan Lin Cong Zhang Fengming Huang Jiahui Chang Chang Li Mingyao Tian Yiming Ma Xiaoyun Li Congzhao Zhou Xiao Li Xiaozhong Peng Ningyi Jin Chengyu Jiang | 2023 | Science China(Life Sciences)2023,66,7: | 1

Two-Dimensional Tunable and Temperature-Insensitive Lyot Filter for FM-to-AM Compensation显示文摘Utilizing polarization maintaining photonic crystal fiber(PM-PCF)with the low temperature coefficient of birefringence,a two-dimensional tunable and temperature-insensitive Lyot filter aiming to compensate the frequency modulation to amplitude modulation(FM-to-AM)conversion in high power laser facility is demonstrated. The Jones matrix is applied to analyze the relationship between optical characteristics of the filter and physical parameters(including amplitude ratio, phase delay, and susceptibility of the birefringence to temperature)of the polarization optical field. Both the transmission peak wavelength and extinction ratio of the spectral transmission are able to be changed simultaneously, hence,it shows more efficient FM-to-AM compensation ability.Besides, the transmission peak shift is about 18pm/°C with the PM-PCF configuration, which is about two orders of magnitude less than the normal polarization maintaining fiber(PMF)configuration. The demonstrated filter presents a practical application potential in large scale laser driven facility. | Mengqiu Fan Xiaocheng Tian Dandan Zhou Jiatian Wei Handing Xia Hongwei Lv Hao Zhao Dangpeng Xu Wanguo Zheng | 2021 | Photonic Sensors2021,11,3: | 1

Biomimetic AgNPs@antimicrobial peptide/silk fibroin coating for infection-trigger antibacterial capability and enhanced osseointegration显示文摘Endowing implant surfaces with combined antibacterial and osteogenic properties by drug-loaded coatings has made great strides, but how to achieve the combined excellence of infection-triggered bactericidal and in vivo-proven osteogenic activities without causing bacterial resistance still remains a formidable challenge.Herein, antimicrobial peptides(AMPs)with osteogenic fragments were designed and complexed on the surface of silver nanoparticle(AgNP)through hydrogen bonding, and the collagen structure-bionic silk fibroin(SF)was applied to carry AgNPs@AMPs to achieve infection-triggered antibacterial and osteointegration. As verified by TEM,AMPs contributed to the dispersion and size-regulation of AgNPs,with a particle size of about 20 nm,and a clear protein corona structure was observed on the particle surface. The release curve of silver ion displayed that the SF-based coating owned sensitive pH-responsive properties. In the antibacterial test against S.aureus for up to 21 days, the antibacterial rate had always remained above 99%.Meanwhile,the underlying mechanism was revealed, originating from the destruction of the bacterial cell membranes and ROS generation. The SF-based coating was conducive to the adhesion, diffusion, and proliferation of bone marrow stem cells(BMSCs)on the surface, and promoted the expression of osteogenic genes and collagen secretion. The in vivo implantation results showed that compared with the untreated Ti implants,SF-based coating enhanced osseointegration at week 4 and 8.Overall,the AgNPs@AMPs-loaded SF-based coating presented the ability to synergistically inhibit bacteria and promote osseointegration, possessing tremendous potential application prospects in bone defects and related-infection treatments. | Wenhao Zhou Tian Bai Lan Wang Yan Cheng Dandan Xia Sen Yu Yufeng Zheng | 2023 | Bioactive Materials2023,,2: | 1
Fusarium-produced vitamin B6 promotes the evasion of soybean resistance by Phytophthora sojae显示文摘Plants can be infected by multiple pathogens concurrently in natural systems. However, pathogen–pathogen interactions have rarely been studied. In addition to the oomycete Phytophthora sojae, fungi such as Fusarium spp. also cause soybean root rot. In a 3-year field investigation, we discovered that P. sojae and Fusarium spp. frequently coexisted in diseased soybean roots. Out of 336 P. sojae–soybean–Fusarium combinations, more than 80% aggravated disease. Different Fusarium species all enhanced P. sojae infection when co-inoculated on soybean. Treatment with Fusarium secreted non-proteinaceous metabolites had an effect equal to the direct pathogen coinoculation. By screening a Fusarium graminearum mutant library, we identified Fusarium promoting factor of Phytophthora sojae infection 1(Fpp1),encoding a zinc alcohol dehydrogenase. Fpp1 is functionally conserved in Fusarium and contributes to metabolite-mediated infection promotion, in which vitamin B6(VB6) produced by Fusarium is key. Transcriptional and functional analyses revealed that Fpp1 regulates two VB6 metabolism genes, and VB6 suppresses expression of soybean disease resistance-related genes. These results reveal that co-infection with Fusarium promotes loss of P. sojae resistance in soybean, information that will inform the sustainable use of diseaseresistant crop varieties and provide new strategies to control soybean root rot. | Shuchen Wang Xiaoyi Zhang Zhichao Zhang Yun Chen Qing Tian Dandan Zeng Miao Xu Yan Wang Suomeng Dong Zhonghua Ma Yuanchao Wang Xiaobo Zheng Wenwu Ye | 2023 | Journal of Integrative Plant Biology2023,65,9: | 1
