1Polymorphisms in PPARD,PPARG and APM1 associated with four types of Traditional Chinese Medicine constitutions显示文摘Based on the theory of constitution of Traditional Chinese Medicine (TCM), the human population is divided into nine constitutions including one balanced constitution (Normality) and eight unbalanced constitutions (Yang-deficiency, Yin-deficiency, Phlegm-wetness, Qi-deficiency, Wetness-heat, Blood stasis, Depressed constitution, and Inherited special constitution). Different constitutions have specific metabolic features and different susceptibility to certain diseases. However, whether a genetic basis accounts for such constitution classifica-tion is yet to be determined. Here we performed a genetic study to assess the association between genetic variations of metabolic genes in-cluding PPARD, PPARG and APM1 and the constitutions. A total of 233 individuals of the Han population in China were classified into four groups, Normality, Yang-deficiency, Yin-deficiency and Phlegm-wetness with whom 23 single nucleotide polymorphisms (SNPs) in the three genes were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Biased distri-bution of PPARD rs2267669 and rs2076167, APM1 rs7627128 and rs1063539 in Yang-deficiency, PPARG Pro12Ala in Yin-deficiency and PPARD rs2076167, APM1 rs266729 and rs7627128 in Phlegm-wetness were observed. The frequencies of Haplotype13 (Hap13) of PPARG in Yin-deficiency, Hap25 of APM1 in Yang-deficiency and Hap2 of PPARD and Hap14 of PPARG in Phlegm-wetness, were significantly different from those in Normality, suggesting those might be group-associated haplotypes. 2A possible important CO_2 sink by the global water cycle显示文摘The locations,magnitudes,variations and mechanisms responsible for the global CO2 sink are uncer-tain and under debate.Here,we show,based on theoretical calculations and evidences from field monitoring results,that there is a possible important CO2 sink(as DIC-dissolved inorganic carbon)by the global water cycle.The sink constitutes up to 0.8013 Pg C/a(or 10.1% of the total anthropogenic CO2 emission,or 28.6% of the missing CO2 sink),and is formed by the CO2 absorption of water and subsequent enhanced consumption by carbonate dissolution and aquatic plant photosynthesis.Of the sink,0.5188 Pg C/a goes to sea via precipitation over sea(0.2748 Pg C/a)and continental rivers(0.244 Pg C/a),0.158 Pg C/a is released to the atmosphere again,and 0.1245 Pg C/a is stored in the continental aquatic ecosystem.Therefore,the net sink could be 0.6433 Pg C/a.This sink may increase with the global-warming-intensified global water cycle,the increase in CO2 and carbonate dust in atmosphere,and reforestation/afforestation,the latter increasing soil CO2,and thus the concentration of the DIC in water.LIU ZaiHua Wolfgang DREYBRODT WANG HaiJing 2008Chinese Science Bulletin2008,53,3:22

4Ethylene Emission as a Potential Indicator of Fuji Apple Flavor Quality Evaluation Under Low Temperature显示文摘Fuji apple(Malus×domestica Borkh.)is a popular fruit cultivar and occupies an important position in the fruit market due to its excellent flavor and storage quality.Ethylene emission can induce respiration,which would reduce postharvest quality and increase storage losses.In order to maintain fruit quality and extend shelf life,apples are usually stored in a low-temperature environment after harvest to reduce the formation of ethylene.Volatile aroma components are regarded as one of the most important aspects of flavor quality and a key factor for apple quality grading and customer satisfaction.Ethylene emission and its relationship with volatile aromas during low temperature,however,have not been determined.In this study,the dynamic changes of volatile aroma compounds of Fuji apples stored at 4°C were detected and analyzed for 42 days.The variation of ethylene released from Fuji apple was studied correspondingly.The results showed that ethyl butyrate,ethyl caproate,ethyl 2-methylbutyrate and 2-methylbutyl acetate were the main aroma components of Fuji apple.The change in ethylene emission and the normalized peak area of volatile aroma compounds exhibited a similar tendency that increased significantly from the 3rd day to the peak on the 14th day and then decreased;there was a positive linear correlation between them with a correlation coefficient of 0.79(P<0.05).Therefore,ethylene release can be used as a potential indicator for evaluation of volatile aroma compounds in apples.Using ethylene emission as an indicator could reduce the difficulty and complexity of volatile aroma evaluation,which could be a new non-destructive inspection choice for apple flavor quality assessment.Weiyan Qi Haijing Wang Zhen Zhou Peng Yang Wenbin Wu Zhemin Li Xian Li 2020Horticultural Plant Journal2020,6,4:18
5Beclin 1 cleavage by caspase-3 inactivates autophagy and promotes apoptosis显示文摘Autophagy and apoptosis are both highly regulated biological processes that play essential roles in tissue homeostasis,development and diseases.Autophagy is also described as a mechanism of death pathways,however,the precise mechanism of how autophagy links to cell death remains to be fully understood.Beclin 1 is a dual regulator for both autophagy and apoptosis.In this study we found that Beclin 1 was a substrate of caspase-3 with two cleavage sites at positions 124 and 149,respectively.Furthermore,the autophagosome formation occurred,followed by the appearance of morphological hallmarks of apoptosis after staurosporine treatment.The cleavage products of Beclin 1 reduced autophagy and promoted apoptosis in HeLa cells and the cells in which Beclin 1 was stably knocked down by specific shRNA.In addition,the cleavage of Beclin 1 resulted in abrogating the interaction between Bcl-2 with Beclin 1,which could be blocked by z-VAD-fmk.Thus,our results suggest that the cleavage of Beclin 1 by caspase-3 may contribute to inactivate autophagy leading towards augmented apoptosis.Yushan Zhu Lixia Zhao Lei Liu Ping Gao Weili Tian Xiaohui Wang Haijing Jin Haidong Xu Quan Chen 2010Protein & Cell2010,1,5:17
6Epigenetic regulation in B-cell maturation and its dysregulation in autoimmunity显示文摘B cells have a critical role in the initiation and acceleration of autoimmune diseases, especially those mediated byautoantibodies. In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. 9Gene admixture in ethnic populations in upper part of Silk Road revealed by mtDNA polymorphism显示文摘To evaluate the gene admixture on the current genetic landscape in Gansu Corridor (GC) in China, the upper part of the ancient Silk Road which connects the Eastern and Central Asia, we examined mitochondrial DNA (mtDNA) polymorphisms of five ethnic populations in this study. Using PCR-RFLP and sequencing, we analyzed mtDNA haplotypes in 242 unrelated samples in three ethnic populations from the GC region and two ethnic populations from the adjacent Xinjiang Uygur Autonomous Region of China. We analyzed the data in comparison with the previously reported data from Eastern, Central and Western Asia and Europe. We found that both European-specific haplogroups and Eastern Asian-specific haplogroups exist in the Gansu Corridor populations, while a modest matrilineal gene flow from Europeans to this region was revealed. The Gansu Corridor populations are genetically located between Eastern Asians and Central Asians, both of who contributed significantly to the maternal lineages of the GC populations. 13Rational design of novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructures for all-solid-state asymmetric supercapacitors显示文摘Constructing graphene-based heterostructures with large interfacial area is an efficient approach to enhance the electrochemical performance of supercapacitors but remains great challenges in their synthesis.Herein,a novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructure(a-Fe_(2)O_(3)NDs/RGO)aerogel was facilely synthesized via excessive metal-ion-induced self-assembly and subsequent calcination route using Prussian blue/graphene oxide(PB/GO)composite aerogel as precursors.The deliberately designed a-Fe_(2)O_(3)NDs/RGO heterostructure offers a highly interconnected porous conductive network,large heterostructure interfacial area,and plenty of accessible active sites,greatly facilitating the electron transfer,electrolyte diffusion,and pseudocapacitive reactions.The obtained a-Fe_(2)O_(3)NDs/RGO aerogel could be used as flexible free-standing electrodes after mechanical compression,which exhibited a significantly enhanced specific capacitance of 347.4 F·g^(-1)at 1 A·g^(-1),extraordinary rate capability of 184 F·g^(-1)at 10 A·g^(-1),and decent cycling stability.With the as-prepared a-Fe_(2)O_(3)NDs/RGO as negative electrodes and the Co_(3)O_(4)NDs/RGO as positive electrodes,an all-solid-state asymmetric supercapacitor(a-Fe_(2)O_(3)NDs/RGO//Co_(3)O_(4)NDs/RGO asymmetric supercapacitor(ASC))was assembled,which delivered a high specific capacitance of 69.1 F·g^(-1)at 1 A·g^(-1)and an impressive energy density of 21.6 W·h·k·g^(-1)at 750 W·k·g^(-1),as well as good cycling stability with a capacity retention of 94.3%after 5,000 cycles.This work provides a promising avenue to design high-performance graphene-based composite electrodes and profound inspiration for developing advanced flexible energy-storage devices.Chenxiao Wu Zhifang Zhang Zhonghui Chen Zuimin Jiang Huiyu Li Haijing Cao Yongsheng Liu Yanyan Zhu Zebo Fang Xiangrong Yu 2021Nano Research2021,14,4:6

16Denitrification Rate and Controlling Factors for Accumulated Nitrate in the Deep Subsoil of Intensive Farmlands: A Case Study in the North China Plain显示文摘Denitrification in subsoil(to a depth of 12 m) is an important mechanism to reduce nitrate(NO3^-) leaching into groundwater.However, regulating mechanisms of subsoil denitrification, especially those in the deep subsoil beneath the crop root zone, have not been well documented. In this study, soil columns of 0–12 m depth were collected from intensively farmed fields in the North China Plain. The fields had received long-term nitrogen(N) fertilizer inputs at 0(N0), 200(N200) and 600(N600) kg N ha^-1 year^-1. Main soil properties related to denitrification, i.e., soil water content, NO3^-, dissolved organic carbon(DOC), soil organic carbon(SOC),pH, denitrifying enzyme activity(DEA), and anaerobic denitrification rate(ADR), were determined. Statistical comparisons among the treatments were performed. The results showed that NO3^- was more heavily accumulated in the entire soil profile of the N600 treatment, compared to the N0 and N200 treatments. The SOC, DOC, and ADR decreased with increasing soil depth in all treatments,whereas considerable DEA was observed throughout the subsoil. The long-term fertilizer rates affected ADR only in the upper 4 m soil layers. The ADRs in the N200 and N600 treatments were significantly correlated with DOC. Multiple regression analysis indicated that DOC rather than DEA was the key factor regulating denitrification beneath the root zone. 17Variation in Plant Morphology and Sinigrin Content in Ethiopian Mustard(Brassica carinata L.)显示文摘Ethiopian mustard(Brassica carinata) is mainly grown as a leafy vegetable and oilseed crop. Sinigrin is predominant glucosinolate in the leaves of Ethiopian mustard. It is hydrolyzed by enzyme myrosinase to produce bioactive product specifically allyl-isothiocyanates. Due to sinigrin content, Ethiopian mustard has a mild flavor and it is a healthy addition to human diet. However, sinigrin concentration depends on genotype.Understanding variations of plant morphology and sinigrin content may be the first step for crop improvement. According to the analyses of various plant characteristics among ninety nine Ethiopian mustard accessions, we found significant differences in plant height, chlorophyll content, and petiole length. In addition, sinigrin content was determined using ultra-performance liquid chromatography tandem quadrupole mass spectrometers/tunable ultraviolet detector(UPLC-TQS/TUV). We found a significant difference in sinigrin content among leaves of 94 Ethiopian mustard accessions. Except for petiole length, the sinigrin content was negatively correlated with leaf area, leaf width, number of primary branches, and plant height. 20Woody species Rhus chinensis Mill. seedlings tolerance to Pb:Physiological and biochemical response显示文摘Screening potential plant species is a crucial consideration in phytoremediation technology.Our previous study demonstrated that Rhus chinensis Mill. seedlings had potentials for phytoremediation of Pb contaminated soil. However, its bioaccumulation and tolerance characteristics remain unclear. Seedling growth, LMWOAs secreted by roots, Pb subcellular distribution and chemical forms, and mineral elements in R. chinensis tissues were evaluated under different Pb concentrations(0, 25, 50, 100, 200 and 400 mg/L) in culture solution at14 days after planting. R. chinensis did not show visual symptoms of Pb toxicity under lower Pb treatments; however, Pb significantly declined the growth of seedlings under higher Pb treatments. Higher Pb stress also decreased the concentrations of nitrogen in leaves, but increased the concentrations of P and K in roots. Pb stress also decreased Mn concentrations in leaves. A great quantity of Pb was uptake and mostly retained in R. chinensis roots.Nonetheless, R. chinensis can still concentrate 459.3 and 1102.7 mg/kg Pb in leaves and stems,respectively. Most of Pb in R. chinensis tissues was stored in the cell wall with HAc-, HCl-, and NaC l-extractable form. LMWOAs secreted by R. chinensis roots showed a strong positive correlation with Pb concentrations in all plant tissues and with P in roots.
