| 1 | Calcium silicate accelerates cutaneous wound healing with enhanced re-epithelialization through EGF/EGFR/ERK-mediated promotion of epidermal stem cell functions显示文摘Background:Human epidermal stem cells(hESCs)play an important role in re-epithelialization and thereby in facilitating wound healing,while an effective way to activate hESCs remains to be explored.Calcium silicate(CS)is a form of bioceramic that can alter cell behavior and promote tissue regeneration.Here,we have observed the effect of CS on hESCs and investigated its possible mechanism.Methods:Using a mouse full-thickness skin excision model,we explored the therapeutic effect of CS on wound healing and re-epithelialization.In vitro,hESCs were cultured with diluted CS ion extracts(CSIEs),and the proliferation,migration ability and stemness of hESCs were evaluated.The effects of CS on the epidermal growth factor(EGF),epidermal growth factor receptor(EGFR)and extracellular signal-related kinase(ERK)signaling pathway were also explored.Results:In vivo,CS accelerated wound healing and re-epithelialization.Immunohistochemistry demonstrated that CS upregulated cytokeratin 19 and integrinβ1 expression,indicating that CS improved hESCs stemness.In vitro studies confirmed that CS improved the biological function of hESCs.And the possible mechanism could be due to the activation of the EGF/EGFR/ERK signaling pathway.Conclusion:CS can promote re-epithelialization and improve the biological functions of hESCs via activating the EGF/EGFR/ERK signaling pathway. | Bingmin Li Haowen Tang Xiaowei Bian Kui Ma Jiang Chang Xiaobing Fu Cuiping Zhang | 2021 | Burns & Trauma2021,9,1: | 3 |
| 2 | Floating Escherichia coli by Expressing Cyanobacterial Gas Vesicle Genes显示文摘Gas vesicles are hollow, air-filled polyprotein structures that provide the buoyancy to cells. They are found in a variety of prokaryotes. In this study, we isolated a partial gas vesicle protein gene cluster containing gvp A and gvp C20Ψ from Planktothrix rubescens, and inserted it into an expression vector and expressed it in E. coli. The gas vesicle was developed in bacterial cells, which made bacterial cells to float on medium surface. We also amplified gvp A and gvp C20Ψ separately and synthesized an artificial operon by fusing these two genes with the standardized gene expression controlling elements of E. coli. The artificial operon was expressed in E. coli, forming gas vesicles and floating bacteria cells. Our findings verified that the whole set of genes and the overall structure of gas vesicle gene cluster are not necessary for developing gas vesicles in bacteria cells. Two genes, gvp A and gvp C20Ψ, of the gas vesicle gene cluster are sufficient for synthesizing an artificial operon that can develop gas vesicles in bacteria cells. | 3 | Slope stability prediction based on a long short-term memory neural network:comparisons with convolutional neural networks,support vector machines and random forest models显示文摘The numerical simulation and slope stability prediction are the focus of slope disaster research.Recently,machine learning models are commonly used in the slope stability prediction.However,these machine learning models have some problems,such as poor nonlinear performance,local optimum and incomplete factors feature extraction.These issues can affect the accuracy of slope stability prediction.Therefore,a deep learning algorithm called Long short-term memory(LSTM)has been innovatively proposed to predict slope stability.Taking the Ganzhou City in China as the study area,the landslide inventory and their characteristics of geotechnical parameters,slope height and slope angle are analyzed.Based on these characteristics,typical soil slopes are constructed using the Geo-Studio software.Five control factors affecting slope stability,including slope height,slope angle,internal friction angle,cohesion and volumetric weight,are selected to form different slope and construct model input variables.Then,the limit equilibrium method is used to calculate the stability coefficients of these typical soil slopes under different control factors.Each slope stability coefficient and its corresponding control factors is a slope sample.As a result,a total of 2160 training samples and 450 testing samples are constructed.These sample sets are imported into LSTM for modelling and compared with the support vector machine(SVM),random forest(RF)and convo-lutional neural network(CNN).The results show that the LSTM overcomes the problem that the commonly used machine learning models have difficulty extracting global features.Furthermore,LSTM has a better prediction performance for slope stability compared to SVM,RF and CNN models. | Faming Huang Haowen Xiong Shixuan Chen Zhitao Lv Jinsong Huang Zhilu Chang Filippo Catani | 2023 | International Journal of Coal Science & Technology2023,10,2: | 1 |
| 4 | Genetic parameters estimation for growth traits in cultured tiger pufferfish(fugu),Takifugu rubripes显示文摘The aim of this study was to evaluate the genetic parameters of the growth performance of Takifugu rubripes.Heritabilities and genetic correlations were estimated for body weight(BW),body length(BL),total length(TL),chest measurement(CM)and trunk length(TKL)of T.rubripes from measurements of progeny at 6 months and 12 months.The results showed that the heritability was 0.37 for BW6,0.19 for BL6,0.35 for TL6,0.29 for CM6,0.26 for TKL6,0.36 for BW12,0.26 for BL12,0.25 for TL12,0.31 for CM12 and 0.15 for TKL12.The range of genetic correlations estimated at 6 months was 0.025–0.725 and−0.002–0.706 at 12 months.The results indicated that genetic improvement for faster growth rate or increased body weight in cultured T.rubripes was effective.Based on selection theory,the selection strategy for traits with medium heritability is flexible.Considering that these growth traits do not reach the high level of heritability,family selection should be expected.Given positive genetic correlations among BW,BL,TL,CM,and TKL at 6 months,the five traits could be improved simultaneously through selective breeding.As there was high genetic correlation only between BW12,BL12 and TL12,and negative correlations between TKL12 and BL12 as well as between CM12 and TL12,and only BW,BL and TL at 12 months could be improved simultaneously. | Xin’an Wang Aijun Ma Zhifeng Liu Zhibin Sun Liguang Zhu Haowen Chang | 2022 | Acta Oceanologica Sinica2022,41,12: | 0 |
| 5 | Submilliwatt, widely tunable coherent microcomb generation with feedback-free operation显示文摘Microcombs are revolutionizing optoelectronics by providing parallel, mutually coherent wavelengthchannels for time-frequency metrology and information processing. To implement this essential function inintegrated photonic systems, it is desirable to drive microcombs directly with an on-chip laser in a simpleand flexible way. However, two major difficulties have prevented this goal: (1) generating mode-lockedcomb states usually requires a significant amount of pump power and (2) the requirement to align laser andresonator frequency significantly complicates operation and limits the tunability of the comb lines. Here, weaddress these problems by using microresonators on an AlGaAs on-insulator platform to generate dark-pulsemicrocombs. This highly nonlinear platform dramatically relaxes fabrication requirements and leads to arecord-low pump power of <1 mW for coherent comb generation. Dark-pulse microcombs facilitated bythermally controlled avoided mode crossings are accessed by direct distributed feedback laser pumping.Without any feedback or control circuitries, the comb shows good coherence and stability. With around150 mW on-chip power, this approach also leads to an unprecedentedly wide tuning range of over one freespectral range (97.5 GHz).
