1CRISPR-Cas13a mediated nanosystem for attomolar detection of canine parvovirus type 2显示文摘Canine parvovirus type 2(CPV-2) infection is the most lethal disease of dogs with higher mortality in puppies worldwide.In today’s world,dogs are an integral part of our communities as well as dogs breeding and rearing has become a lucrative business.Therefore,a fast,accurate,portable,and costeffective CPV-2 detection method with the ability for on-site detection is highly desired.In this study,we for the first time proposed a nanosystem for CPV-2 DNA detection with RNA-guided RNA endonuclease Cas13 a,which upon activation results in collateral RNA degradation.We expressed LwCasl3 a in prokaryotic expression system and purified it through nickel column.Activity of Cas13 a was verified by RNA-bound fluorescent group while using a quenched fluorescent probe as signals.Further Cas13 a was combined with Recombinase polymerase amplification(RPA) and T7 transcription to establish molecular detection system termed specific high-sensitivity enzymatic reporter un-locking(SHERLOCK) for sensitive detection of CPV-2 DNA.This nanosystem can detect 100 amol/L CPV-2 DNA within 30 min.The proposed nanosystem exhibited high specificity when tested for CPV-2 and other dog viruses.This CRISPR-Cas13 a mediated sensitive detection approach can be of formidable advantage during CPV-2 outbreaks because it is time-efficient,less laborious and does not involve the use of sophisticated instruments.Haroon Khan Adeel Khan Yufeng Liu Su Wang Sumaira Bibi Hongpan Xu Yuan Liu Samran Durrani Lian Jin Nongyue He Tao Xiong 2019Chinese Chemical Letters2019,30,12:8
2Antiglycation and antioxidation properties of juglans regia and calendula officinalis:possible role in reducing diabetic complications and slowing down ageing显示文摘OBJECTIVE:Accumulation of advanced glycation end products(AGEs) in the body due to the non-enzymatic glycation of proteins and oxidation is associated with aging and diabetes mellitus.In this study we wanted to investigate the antiglycation and antioxidation potential of two medicinal plants:Juglans regia and Calendula officinalis.METHODS:In-vitro investigation was carried out to discover the antiglycation and antioxidation potential of J.regia and C.officinalis.Using an Ultraviolet Double-beam Spectrophotometer,we evaluated the antiglycation property of the crude methanolic extracts of J.regia and C.officinalis by assessing their ability to inhibit the Maillard reaction.Employing the same instrument we also measured the antioxidation potential of these plant extracts using the nitric oxide(NO) free radical-scavenging assay.RESULTS:J.regia had greater antiglycation ability,with a minimum inhibitory concentration(MIC 50) of 28 μg/mL as compared with that of C.officinalis(270 μg/mL).C.officinalis had greater antioxidation potential(26.10,22.07 and 16.06% at 0.5 mg,0.25 mg and 0.125 mg,respectively,as compared with 18.15,16.50 and 16.06% of J.regia,respectively).CONCLUSION:J.regia and C.officinalis inhibited the Maillard reaction and prevented oxidation in-vitro.Hence,the extracts of these plants could have therapeutic uses in curbing chronic diabetic complications and slowing down aging.Haroon Ahmad Ibrar Khan Abdul Wahid 2012Journal of Traditional Chinese Medicine2012,32,3:5
3Why more research needs to be done on echinococcosis in Pakistan显示文摘Background:Echinococcosis has a worldwide geographical distribution with endemic foci on every inhabited continent.Due to the frequent outbreaks in different parts of Pakistan in the recent past,echinococcosis is being described as a neglected tropical disease and is considered one of the most neglected parasitic diseases in the country.In endemic regions,predominantly settings with limited resources,there are high numbers of echinococcosis patients,as these communities do not have access to appropriate treatment.In Pakistan,there are limited reports on echinococcosis.The disease is prevalent in human and livestock,but this has not been sufficiently explored yet.Pakistan is an agricultural country and due to the disease’s zoonotic mode of transmission,there is a dire need of future research on it.The present paper is an effort to highlight the importance of echinococcosis in Pakistan.Discussion:There is a dire need for future research on echinococcosis in Pakistan as very few investigations had been carried out on this topic thus far.The prevalence of the disease in neighbouring countries highlights that Pakistan might be at severe risk of this zoonotic infection and further supports the need for more research.In Pakistan,the majority of the population lives in rural areas with limited acess to proper hygienic/sanitary facilities.These conditions favour the outbreak of diseases such as echinococcosis.The limited available data could result in higher outbreaks in the future,and thus cause the already weak healthcare system to overburden.The country has a meagre annual budget for health,which is being spent on known infections such as polio,dengue fever and hepatic viral infections.A proper surveillance system for echinococcosis is required across the country as treatment is usually expensive,complicated and may require extensive surgery and/or prolonged drug therapy.Development of new/novel drugs and other treatment modalities receives very little,if any,attention.Prognostic awareness programmes against this infection involve deworming of the infected animals,improved food inspection and slaughterhouse hygiene,and public education campaigns.Conclusion:Future research on echinococcosis is anticipated to demonstrate whether the epidemiology,diagnosis and recombinant vaccines/antibodies relating to echinococcosis can meet the quality standards(purity,potency,safety and efficacy)defined by the World Health Organization.Research work should be carried out on the epidemiology and serodiagnosis of echinocossis in the different areas of Pakistan,which will be useful for the proper eradication of echinococcosis in this region.The health department should implement awareness-raising campaigns for the general public in order to reduce the burden of disease.Haroon Ahmed Shahzad Ali Muhammad Sohail Afzal Abid Ali Khan Hamid Raza Zaheer Hussain Shah Sami Simsek 2017Infectious Diseases of Poverty2017,6,1:2

5In vivo screening of essential oils of Skimmia laureola leaves for antinociceptive and antipyretic activity显示文摘Objective:To study the screening of essential oils of Skimmia laureola leaves(SLO)for acute toxicity,antinociceptive,antipyretic and anticonvulsant activities in various animal models.Methods:SLO were extracted using modified Clevenger type apparatus.Acute toxicity test was used in mice to observe its safety level.Antinociceptive activity of SLO was evaluated in acetic acid induced writhing and hot plate tests.Yeast induced hyperthermic mice and pentylenetetrazole induced convulsive mice were used for the assessment of its antipyretic and anticonvulsant profile respectively.Results:Substantial safety was observed for SLO in acute toxicity test.SLO showed a high significant activity in acetic acid induced writhing test in a dose dependent manner with maximum pain attenuation of 68.48%at 200 mg/kg i.p.However,it did not produce any relief in thermal induced pain at test doses.When challenged against pyrexia evoked by yeast,SLO manifested marked amelioration in hyperthermic mice,dose dependently.Maximum anti-hyperthermic activity(75%)was observed at 200 mg/kg i.p.after 4 h of drug administration.Nevertheless,SLO had no effect on seizures control and mortality caused by pentylenetetrazole.Conclusions:In vivo studies of SLO showed prominent antinociceptive and antipyretic activities with ample safety profile and thus provided pharmacological base for the traditional uses of the plant in various painful conditions and pyrexia.Additional detail studies are required to ascertain its clinical application.Naveed Muhammad Barkatullah Muhammad Ibrar Haroon Khan Muhammad Saeed Amir Zada Khan Waqar Ahmad Kaleem 2013Asian Pacific Journal of Tropical Biomedicine2013,3,3:2
6Sedative and antinociceptive activities of two new sesquiterpenes isolated from Ricinus communis显示文摘Two new sesquiterpenes, trivially named ricinusoids A(1) and ricinusoids B(2), were isolated from ethyl acetate fraction of Ricinus communis. The structures of new compounds were elucidated by detailed spectroscopic techniques, including 1 D-and 2 D-NMR, UV, IR spectroscopy, and mass spectrometry. The compounds(1-2) were also assessed for in-vivo sedative and analgesic like effects in open field and acetic acid induced writhing tests respectively at 5, 10, and 20 mg·kg^(–1) i.p. Pretreatment of both test compounds caused significant(P ≤ 0.05) reduction in locomotive activity like sedative agents and abdominal constrictions like analgesics. 10Knowledge,attitudes and practices related to cystic echinococcosis endemicity in Pakistan显示文摘Background:Cystic echinococcosis(CE)is a human and animal health problem in many endemic areas worldwide.It is considered a neglected zoonotic disease caused by the larval form(hydatid cyst)of Echinococcus spp.tapeworm.There are limited studies on echinococcosis in Pakistan.Methods:A cross-sectional survey was conducted to find out recent knowledge,attitudes and practices on the occurrence of cystic echinococcosis in butchers and dog owners in both urban and rural areas of Rawalpindi/Islamabad regions,Pakistan.The quantitative data was collected in the form of questionnaires to investigate the knowledge and awareness of CE among community members and their routine practices that were behind the factors involved in hydatid cyst infection.The practices and infrastructure of abattoirs/butcher shops and their role in transmission of cystic echinococcosis were also evaluated in the present study.Results:The participants involved in the study were dog owners and people who kept animals.A total of 400 people were interviewed and 289 questionnaires were received.The results showed that only 4.1%of people have heard about the disease,and 58.1%were closely associated with dogs.Sixty-three percent of dogs in study area were consuming uncooked organs(e.g.liver,lung,etc.)of slaughtered animals,while 100%of dogs at butcher shops were consuming uncooked organs.Home slaughtering was common in 20.06%.Among butchers,32.3%had heard about zoonoses and 7.61%knew about CE.The statistical analysis showed that there was highly significant difference(P<0.05)among most of the practices that were associated with the prevalence of CE.Conclusions:It was concluded from the present study that,the knowledge and awareness of CE among people of Rawalpindi/Islamabad were low.Because of dogs and poor knowledge of CE among community members and butchers,the transmission of echinococcosis is facilitated.Therefore,there is urgent need to strengthen awareness and health education among people,as well as proper practices related to the CE not only in the study area,but also in other areas of Pakistan.Aisha Khan Kashf Naz Haroon Ahmed Sami Simsek Muhammad Sohail Afzal Waseem Haider Sheikh Saeed Ahmad Sumaira Farrakh Wu Weiping Guan Yayi 2018Infectious Diseases of Poverty2018,7,1:1

14Screening of solanum surrattense for antibacterial,antifungal,phytotoxic and haemagglutination显示文摘OBJECTIVE:Plants produce a variety of useful bioactive materials that have been used to cure different ailments of human beings.With the same idea in mind,the crude methanolic extract and various fractions of Solanum surrattense were screened for antibacterial,antifungal,phytotoxic and heamagglutination activities.METHODS:Standardized assays were followed for the determination of antibacterial,antifungal phytotoxic and heamagglutination activity.RESULTS:The results of the antibacterial showed that crude methanolic extract was significantly active against Staphylococcus aureus(86%) The nhexane fraction showed good activity against Pseudomonas aerogenosa(66.6%) and Bacillus subtilus(66.6%).The crude methanolic extract and various fractions were inactive against all test fungi.The crude methanolic extract,n-hexane,CHCl 3 and aqueous fractions showed moderate phytotoxic activities of 46.67%,40.00%,33.34% and 33.34% respectively at 1000 μg/mL.The crude methanolic extract and various fractions S.surrattense were unable to agglutinate RBCs of the human blood indicating that this species lack phytolectins.CONCLUSION:The test sample showed significant antibacterial activity,no antifungal and heamagglutination activity while moderate phytotoxic was observed against Lemna minor L.Haroon Ahmad Nasar Ali Bashir Ahmad Ibrar Khan 2012Journal of Traditional Chinese Medicine2012,32,4:1
15In vivo sedative and muscle relaxants activity of Diospyros lotus L显示文摘Objective: To evaluate the sedative effect of Diospyros lotus L(D. lotus) extract in mice using the open field and Rota rod tests.Methods: For the sedative and muscle relaxants activities of extract/fractions of the plant, invivo open field and phenobarbitone-induced sleeping time were used, while the Roda rod test was employed in animals for the assessment of muscle relaxant activity.Results: Results from this investigation revealed that the extracts of D. lotus have exhibited significant sedative effect in mice(45.98%) at 100 mg/kg i.p. When the extract was partitioned with different solvents, the n-hexane fraction was inactive whereas the chloroform fraction was the most active with 82.67% sedative effect at 50 and 100 mg/kg i.p. On the other hand,the ethyl acetate and n-butanol fractions displayed significant sedative effects(55.65% and40.87%, respectively) at 100 mg/kg i.p. 16Estimation of Genetic Divergence and Character Association Studies in Local and Exotic Diversity Panels of Soybean(Glycine max L.)Genotypes显示文摘The availability of favorable genetic diversity is a thriving vitality for the success of a breeding program.It provides a firm basis of selecting superior breeding lines for the development of high yielding crop genotypes.In this context,present investigation aimed to generate information on genetic divergence and character association in a diversity panel of 123 local and exotic soybean genotypes.Analysis of variance revealed significant response of the evaluated genotypes based on studied attributes.It depicted the probability of selecting desirable soybean genotypes by focusing on character association studies and genetic diversity analysis.Correlation analysis revealed that seed yield per plant showed high positive correlation with 100-seed weight followed by pods per plant and plant height.Furthermore,path coefficient analysis exposed that pods per plant had maximum direct contribution in seed yield per plant followed by 100-seed weight,days to flowering and SPAD measurement.Genotype named“G-10”showed maximum yield per plant followed by 24607,G-52,24593,Arisoy,24566,17426,A-3127,24570 and 24567.Genetic diversity analysis grouped the evaluated germplasm into 17 clusters.All clusters showed zero intra-cluster variability;while inter-cluster divergence ranged from 9.00 to 91.11.Cluster V showed maximum inter-cluster distance with cluster XII followed by that of between V and VIII.Moreover,cluster IV with superior genotypes(G-10,24607,24593 and 24566),VI(17426 and 24567),XIII(24570)and X(Arisoy and G-52)showed above mean values for most of the studied characters.Overall,the results of hybridization between the superior genotypes of these cluster pairs might be useful for soybean breeding with improving agronomic traits and adaptability.Syed Ali Zafar Muhammad Aslam Haroon Zaman Khan Sehrish Sarwar Rao Saad Rehman Mariam Hassan Ramala Masood Ahmad Rafaqat AGill Basharat Ali Ibrahim Al-Ashkar Abdullah Ibrahim Md Atikur Rahman Ayman El Sabagh 2023Phyton-International Journal of Experimental Botany2023,92,6:0
17Antipyretic and anticonvulsant activity of n-hexane fraction of viola betonicifolia显示文摘Objective:To investigate the antipyretic and anticonvulsant activities of n-hexane fraction of Viola betonicifolia(V.betonicifolia).Methods:The antipyretic effect was scrutinized using brewer's yeast induced pyrexia and anticonvlsion effect was tested using pentylenetetrazol and strychnine induced convulsion in mice.Results:N-hexane fraction of V.betonicifolia demonstrated highly significant antipyretic activity during various assessment times(1-5 h)when challenged in yeast induced pyrexia test.The effect was in a dose dependent manner with maximum attenuation(82.50%)observed at 300 mg/kg i.p.When tested in pentylenetetrazol induced convulsion test,the 1st stage(Ear and facial twitching)and 2nd stage(Convulsive wave through the body)was 100%protected during 24 h at all the test doses(300,400 and 500 mg/kg i.p.),while the latency time of remaining stages was significantly increased.The maximum effect was observed by n-hexane fraction of V.betonicifolia at 400 and 500 mg/kg i.p.,as the latency time for generalized clonic-tonic seizure(5th stage)was increased up to 25.34 min.However,n-hexane fraction of V.betonicifolia had no protection in strychnine induced convulsion test.Conclusions:In conclusion,phytopharmacological studies provide scientific foundation to the folk uses of the plant in the treatment of pyrexia and neurological disorders.Naveed Muhammad Muhammad Saeed Haroon Khan Naila Raziq Syed Muhammad Ashhad Halimi 2013Asian Pacific Journal of Tropical Biomedicine2013,3,4:0
18Reversal of multidrug resistance and antitumor promoting activity of 3-oxo-6β-hydroxy-β-amyrin isolated from Pistacia integerrima显示文摘The bioactive triterpenoid 3-oxo-6-β-hydroxy-β-amyrin(1)has been isolated from multiple plant sources.In this study,chloroform fraction of Pistacia integerrima extract was processed for the isolation of the compound.The compound identity was confirmed by advanced spectroscopy technique.X-ray crystallography was applied for molecular structure confirmation.In addition,compound 1 was screen for its activity on reversal of MDR(multidrug resistance)mediated by P-gp(P-glycoprotein).This was accomplished by using rhodamine123 exclusion on multidrug-resistant human ABCB1 gene transfected mouse T-lymphoma cell line.Outcomes revealed that MDR reversing effect was comparable to verapamil as positive control in vitro.Treatment of TPA-induced tumor promotion with 3-oxo-6β-hydroxy-β-amyrin led to reduction in the applied anti-tumor promotion experiment.The chemo-preventive effect of 3-oxo-6β-hydroxy-β-amyrin was comparable to curcumin as positive control based on the reduction of immediate early tumor antigen expression.Molecular docking by applying Autodock Vina 1 and i-GEMDOCK v 2.1 tools indicated that compound 1 gives good docking results,as determined by their fitness score and specificity.Moreover,results showed that compound 1 isolated from Pistacia integerrima precisely attached to a region where co-crystallized ligand for receptor previously existed.Our findings may explain the use of Pistacia integerrima plant extracts as an anticancer agent in folk medicine.ABDUR RAUF SAUD BAWAZEER MUSLIM RAZA EMAN EL-SHARKAWY MD.HABIBUR RAHMAN MOHAMED A.EL-ESAWI GHIAS UDDIN BINA SSIDDIQUI ANEES AHMED KHALIL JOSEPH MOLNAR AKOS CSONKA DIÁNA SZABÓ HAROON KHAN MOHAMMAD SMUBARAK TAIBI BEN HADDA MUDYAWATI KAMARUDDIN SEEMA PATEL 2021BIOCELL2021,45,1:0
19Toxic and Antifeedant Effects of Different Pesticidal Plant Extracts against Beet Armyworm (Spodoptera exigua)显示文摘The beet armyworm(BAW),Spodoptera exigua(Lepidoptera:Noctuidae)is a highly destructive pest of vegetables and field crops.Management of beet armyworm primarily relies on synthetic pesticides,which is threatening the beneficial community and environment.Most importantly,the BAW developed resistance to synthetic pesticides with making it difficult to manage.Therefore,alternative and environment-friendly pest management tactics are urgently required.The use of pesticidal plant extracts provides an effective way for a sustainable pest management program.To evaluate the use of pesticidal plant extracts against BAW,we selected six plant species(Lantana camara,Aloe vera,Azadirachta indica,Cymbopogon citratus,Nicotiana tabacum,and Ocimum basilicum)for initial screening experiment.Four out of six plant species such as A.indica,N.tabacum,C.citratus and O.basilicum showed promising mortality of more than 50%.Therefore,we selected these four plant extracts for the subsequent experiments.Through contact bioassay,A.indica showed high mortality 66.63%,followed by the N.tabacum 53.33%,at 10%w/v concentration.Similarly,N.tabacum showed the highest mortality rate,66%at 10%w/v concentration,followed by the A.indica 46%through feeding bioassay.Furthermore,the feeding deterrence assay showed that C.citratus had a high antifeedant index(−50)followed by A.indica(−39),and N.tabacum(−28).In living plant assay,the N.tabacum extract showed a low mean damage score 3.6 on living cotton plant followed by C.citratus 4.5 and A.indica 5.5.Hence,extracts of three plant species provided promising results against the BAW,which can minimize the use of synthetic chemicals,particularly for small landholding farmers.Further studies are also required to evaluate the effects of these plant extract against BAW on cotton plants under field conditions to optimize the further use.Muhammad Asad Rashad Rasool Khan Ahmed B.Aljuboory Muhammad Haroon URashid Uttam Kumar Inzamam Ul Haq Aqsa Hafeez Ahmed Noureldeen Khadiga Alharbi 2023Phyton-International Journal of Experimental Botany2023,92,4:0
20The role of green industrial transformation in mitigating carbon emissions:Exploring the channels of technological innovation and environmental regulation显示文摘The industrial sector is vital to economic progress,yet industrial pollution poses environmental and economic concerns.The purpose of the study was to investigate the influence of green industrial transformation in re-ducing Pakistan’s carbon intensity between 1975 and 2020.Carbon emissions are considered an endogenous construct,while foreign direct investment(FDI)inflows,technological innovation,green industrial transforma-tion,environmental legislation,and research and development(R&D)investment are possible mediators.The association between variables is assessed using the robust least-squares approach.Green industrial transforma-tion is connected with lower carbon emissions,yet technical innovation,R&D investment,and inbound FDI raise a country’s carbon emissions.The findings support the pollution haven hypothesis in a country.The causality esti-mates indicate that inward FDI contributes to environmental regulations;green industrial transformation directly relates to inbound FDI and R&D expenditures;and technological innovations correspond to inbound FDI,R&D ex-penditures,industrial ecofriendly progression,and environmental standards.According to the impulse response function,environmental policies are anticipated to have a differential effect on carbon emissions in 2023,2024,2028-2030,while they are likely to decrease in the years 2025-2027 and 2031 forward.Additionally,inward FDI and technology advancements would almost certainly result in a rise in carbon emissions over time.Green industrial transitions are projected to result in a ten-year reduction in carbon emissions.The variance decomposi-tion analysis indicates that eco-friendly industrial adaptations would likely have the largest variance error shock on carbon emissions(11.747%),followed by inbound FDI,technological advancements,and regulatory changes,with R&D spending having a minimal impact over time.Pakistan’s economy should foster a green industrial revolution to avoid pollution and increase environmental sustainability to meet its environmental goals.Saima Mehmood Khalid Zaman Shiraz Khan Zohaib Ali Haroon ur Rashid Khan 2024Energy and Built Environment2024,5,3:0
