2The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation.Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu 2021Horticultural Plant Journal2021,7,2:3
3Establishment of Self-incompatibility Gene cDNA Microarray to Identify S-genotypes of Pyrus pyrifolia显示文摘Based on the c DNA sequences from hyper variable(HV) regions of identified 52 S-alleles in Oriental pear cultivars, S-RNase c DNA probes were designed, and a c DNA microarray for S-RNase detections was established. Each microarray contained 240 sites from 55 c DNA probes, including all specific c DNA sequences from the HV regions of the S-alleles. Using the c DNA of pistils of tested pear cultivars as template and Cy3 fluorescently labeling primers by PCR amplification, microarray hybridization detected the S-genotype of each pear cultivar. The genotypes inferred from the c DNA microarray hybridization signals of pear cultivars such as ‘Lijiang Huangsuanli', ‘Xiuyu', ‘Midu Yuli', ‘Baimianli', and ‘Deshengxiang' were similar to the known genotypes of all tested cultivars. The S-RNase c DNA microarrays and the oligonucleotide gene chips were then used to conduct parallel testing of 24 P. pyrifolia cultivars with unknown S-genotypes. 4Breeding of a New Clubroot-resistant Chinese Cabbage Variety, CCR11242显示文摘In this study,using Japanese radish cytoplasmic sterile line ' Half Green Cabbage'( HGC) and clubroot-resistant South Korean cabbage variety ' CR Cabbage King'( CCK) as experimental materials,self-incompatible DH lines were obtained by microspore culture.Through five generations of backcross,CCR11239,a clubroot-resistant radish cytoplasmic sterile line,was obtained.Through six generations of self-crossing,CCR11240,the maintainer line of CCR11239,was bred.After the cross of CCK and Luchunbai 1( 83- 1) and five generations of self-crossing,self-compatible line CCR11241 was obtained.Through crossing CCR11240 with CCR11241,the new cabbage variety CCR11242 was obtained.After variety comparison test,provincial regional test and production demonstration test,the new variety was registered in 2012.The average yield of CCR11242 reached 60 975 kg / hm2,which was improved by 142% compared with Luchunbai 1( 83- 1).The disease index of CCR11242 was 5.63,which was 88.13 lower than the control,indicating high resistance( HR) to clubroot.Xuezhong XU Zhimei TIAN Hongli YANG Jingfeng HU Liqin ZHANG Mei LAN Hongju DUAN Jiangming HE 2016Agricultural Biotechnology2016,5,5:1
5Grain Boundaries Engineering of Hollow Copper Nanoparticles Enables Highly Efficient Ammonia Electrosynthesis from Nitrate显示文摘Electrochemical nitrate reduction reaction(NO_(3)−RR)is an ideal route to produce ammonia(NH_(3))under ambient conditions.Although a markedly improved NH3 production rate has been achieved on the NO_(3)−RR compared with the nitrogen reduction reaction(NRR),the NH_(3) production rate of NO_(3)−RR is still well below the industrial Haber-Bosch route due to the lack of robust electrocatalysts for yielding high current densitieswith concurrently good suppression of hydrogen evolution reaction(HER).Herein,we describe an in situ electrochemical strategy for the synthesis of hollow carbon-coated Cu nanoparticles(NPs)(HSCu@C)with abundant grain boundaries(HSCu-AGB@C)for highly efficient NO_(3)−RR in both alkaline and neutral media.Impressively,in alkaline media,the HSCu-AGB@C can achieve a maximum NH3 Faradaic efficiency of 94.2% with an ultrahigh NH_(3) rate of 487.8 mmol g^(−1) cat h^(−1) at−0.2 V versus a reversible hydrogen electrode,more than 2.4-fold of the rate obtained in the Haber-Bosch.Both theoretic computations and experimental results uncover that the grain boundaries play the key to improve the NO_(3)−RR performance.Herein,the industrial-scale NH_(3) production ratemay open exciting opportunities for the practical electrosynthesis NH_(3) under ambient conditions.Qi Hu Yongjie Qin Xiaodeng Wang Hongju Zheng Keru Gao Hengpan Yang Peixin Zhang Minhua Shao Chuanxin He 2022CCS Chemistry2022,4,6:1
6Effects of fresh-cut and storage on glucosinolates profile using broccoli as a case study显示文摘Glucosinolates(GLS) contribute to the unique flavour, nutrition, and plant defence of the Cruciferous vegetables. Understanding the GLS changes through postharvest processing is essential for defined preservation. In this study, four different fresh-cut types, whole flower(W),floret(F), quarterly cut floret(QF) and shredded floret(FS) of broccoli, were stored for 0, 1, 2 and 3 day(s) to explore GLS responses to postharvest treatments. As a result, seven GLS were identified, mainly including glucoraphanin(RAA), neoglucobrassicin(NEO), and glucobrassicin(GBC)and accounting for 52.69%, 20.12% and 14.99% of the total GLS(21.92 ± 0.48) μmol · g ^(-1 )DW, respectively. FS had the sharpest decrease in GLS after three days of storage(6.55 ± 0.37) μmol · g-1DW, while QF had the least(10.16 ± 0.33) μmol · g ^(-1 )DW. All GLS components decreased over storage, except for 4-methoxyglucobrassicin(4 ME) in FS and QF, suggesting its key role in serious wound defence. 9Chip-based visual detection of micro RNA using DNA-functionalized gold nanoparticles显示文摘In the present study, we developed a highly sensitive and convenient biosensor consisting of gold nanoparticle(Au NP) probes and a gene chip to detect micro RNAs(mi RNAs). Specific oligonucleotides were attached to the glass surface as capture probes for the target mi RNAs, which were then detected via hybridization to the Au NP probes. The signal was amplified via the reduction of HAu Cl4 by H2O2. The use of a single Au NP probe detected 10 pmol L?1 of target mi RNA. The recovery rate for mi R-126 from fetal bovine serum was 81.5%–109.1%. The biosensor detection of mi R-126 in total RNA extracted from lung cancer tissues was consistent with the quantitative PCR(q PCR) results. The use of two Au NP probes further improved the detection sensitivity such that even 1 fmol L?1 of target mi R-125a-5p was detectable. This assay takes less than 1 h to complete and the results can be observed by the naked eye. The platform simultaneously detected lung cancer related mi R-126 and mi R-125a-5p. 10Analysis on Composition and Content of Glucosinolates in Broccoli Flowers and Leaves显示文摘Glucosinolate composition and content were evaluated in flowers and leaves of 12 different broccoli varieties. The results indicated that there were 9 glucosinolates in broccoli,namely Glucoiberin(IBE); Progoitrin(PRO);Sinigrin(SIN);Glucoraphanin(RAA);Gluconapin(NAP);4-Hydroxyglucobrassicin(4OH);Glucobrassicin(GBC);4-Methoxyglucobrassicin(4ME);Neoglucobrassicin(NEO).Total glucosinolate content in flowers was 1-5 times higher than in leaves. 11Studies on the Simultaneous Determination of Cr(Ⅲ)and Cr(Ⅵ)by Ion Chromatography(IC)显示文摘The research aimed to develop and validate an ion chromatography method for the simultaneous analysis of Cr(Ⅲ) and Cr(Ⅵ)from pumpkin. A new analytical method based on ion chromatography techniques was developed by the Cr(Ⅲ) pre-column derivatization and Cr(Ⅵ) post-column derivatization. The ion chromatography condition was optimized and the detection sensitivity was improved. Cr(Ⅲ) and Cr(Ⅵ) were determined by 365 and 530 nm,respectively. The temperature of water bath,the heating time for pre-column derivatization,and the flow rate of post-column derivative liquid were screened on the basis of single factor experiment,the effects of various factors were determined by the method of L16( 43) orthogonal experiment design. Considering the results of orthogonal experiments and the variation tendency of peak area under different factors,the optimum derivatization conditions were chose as follows: the flow rate of post-column derivative liquid is0. 5 m L/min,the temperature of water bath for pre-column derivatization is 100 ℃ and the heating time is 5 min. The conditions were optimized by means of orthogonal experiments under the p H of leacheate ranged from 6. 5 to 6. 8 and the I-concentration of 5 mmol/L. Under optimized derivatization conditions,the detection limits for Cr(Ⅲ) and Cr(Ⅵ) were 0. 17 and 0. 019 mg,respectively. And the average recoveries of Cr(Ⅲ) in pumpkin were in the range of 82%-85%.
