| 1 | Fault diagnosis of wind turbine bearing based on stochastic subspace identification and multi-kernel support vector machine显示文摘In order to accurately identify a bearing fault on a wind turbine, a novel fault diagnosis method based on stochastic subspace identification(SSI) and multi-kernel support vector machine(MSVM) is proposed. Firstly, the collected vibration signal of the wind turbine bearing is processed by the SSI method to extract fault feature vectors. Then, the MSVM is constructed based on Gauss kernel support vector machine(SVM) and polynomial kernel SVM. Finally, fault feature vectors which indicate the condition of the wind turbine bearing are inputted to the MSVM for fault pattern recognition. | 2 | TRAF3IP3 at the trans-Golgi network regulates NKT2 maturation via the MEK/ERK signaling pathway显示文摘Thymic natural killer T(NKT)2 cells are a subset of invariant NKT cells with PLZF^(hi)GATA3^(hi)IL-4^(+).The differentiation of NKT2 cells is not fully understood.In the present study,we report an important role of TRAF3-interacting protein 3(TRAF3IP3)in the functional maturation and expansion of committed NKT2s in thymic medulla.Mice with T-cell-specific deletion of TRAF3IP3 had decreased thymic NKT2 cells,decreased IL-4-producing peripheral iNKTs,and defects in response toα-galactosylceramide.Positive selection and high PLZF expression in CD24^(+)CD44^(−) and CCR7^(+)CD44^(−) immature iNKTs were not affected.Only CD44^(hi)NK1.1^(−) iNKTs in Traf3ip3^(−/−) mice showed reduced expression of Egr2,PLZF,and IL-17RB,decreased proliferation,and reduced IL-4 production upon stimulation.This Egr2 and IL-4 expression was augmented by MEK1/ERK activation in iNKTs,and TRAF3IP3 at the trans-Golgi network recruited MEK1 and facilitated ERK phosphorylation and nuclear translocation.LT βR-regulated bone marrow-derived nonlymphoid cells in the medullary thymic microenvironment were required for MEK/ERK activation and NKT2 maturation.These data demonstrate an important functional maturation process in NKT2 differentiation that is regulated by MEK/ERK signaling at the trans-Golgi network. | Xinwei Zhang Ke Wang Weijia Zhao Li Cao Shusong Zhang Rong Jin Xiuyuan Sun Jie Hao Xiaojun Huang Mingzhao Zhu Hounan Wu Hongshan Zhao Qing Ge | 2020 | Cellular & Molecular Immunology2020,17,4: | 2 |
| 3 | Homeobox B8 Targets Sterile Alpha Motif Domain-Containing Protein 9 and Drives Glioma Progression显示文摘Gliomas are the most commonly occurring tumors of the central nervous system. Glioblastoma multiforme (GBM) is the most malignant and aggressive brain cancer in adults. Further understanding of the mechanisms underlying the aggressive nature of GBM is urgently needed. Here we identified homeobox B8(HOXB8), a member of the homeobox family, as a crucial contributor to the aggressiveness of GBM. Data mining of publicly accessible RNA sequence datasets and our patient cohorts confirmed a higher expression of HOXB8 in the tumor tissue of GBM patients, and a strong positive correlation between the expression level and pathological grading of tumors and a negative correlation between the expression level and the overall survival rate. We next showed that HOXB8 promotes the proliferation and migration of glioblastoma cells and is crucial for the activation of the PI3K/AKT pathway and expression of epithelial–mesenchymal transition-related genes, possibly through direct binding to the promoter of SAMD9 (Sterile Alpha Motif Domain-Containing Protein 9) and activating its transcription. | 6 | High-yield and low-cost separation of high-purity semiconducting single-walled carbon nanotubes with closed-loop recycling of raw materials and solvents显示文摘Semiconducting single-walled carbon nanotubes (s-SWCNTs) are the foundation of CNT-based electronics and optoelectronics. For practical applications, s-SWCNTs should be produced with high purity, high structural quality, low cost, and high yield. Currently conjugated polymer wrapping method shows great potential to fulfill these requirements due to its advantages of simple operation process, high purity separation, and easy scaling-up. However, only a small portion of both CNTs and polymers go into the final solution, and most of them are discarded after a single use, resulting in high cost and low yield. In this paper, we introduce a closed-loop recycling strategy, in which raw materials (CNTs and polymers) and solvents were all recycled and reused for multiple separation cycles. In each cycle, high-purity (> 99.9%) s-SWCNTs were obtained with no significant change of structural quality. After 7 times of recycling and separation, the material cost was reduced to ∼ 1% in comparison with commercially available products, and total yield was increased to 36% in comparison with 2%–5% for single cycle separation. | 15 | Improving cis-regulatory elements modeling by consensus scaffolded mixture models显示文摘A position weight matrix(PWM) is widely accepted as a probabilistic representation for modeling protein-DNA binding specificity.Previous studies showed that for factors which bind to divergent binding sites,mixtures of multiple PWMs improve performance.We propose a consensus scaffolded mixutre PWM(CSM) model to improve cis-regulatory elements modeling by allowing overlapping components represented by a set of PWMs,each of which corresponds to a binding pattern and is scaffolded by a degenerate consensus.In addition,we propose a learning algorithm that involves an initial structure learning stage based on the frequent pattern mining and a refining stage based on the expectation maximization(EM) algorithm.We assess the merits of CSM using three independent criteria.In a case-study of transcription factor Leu3,the derived CSM models agree with conventional mixtures but show better fitness according to Fermi-Dirac distribution.Analysis of the human-mouse conservation of predicted binding sites of 83 JASPAR transcription factors(TFs) shows that the CSM is as good as or better than the simple mixture,the context-specific independent(CSI) mixture,and the single PWM model,for 83%,84%,and 75% of the cases,respectively.Five-fold cross validation on 46 TRANSFAC datasets shows that CSM model has better generality than other mixture models. | JIANG HongShan ZHAO Ying CHEN WenGuang ZHANG XueGong ZHENG WeiMin | 2013 | Science China(Information Sciences)2013,56,1: | 0 |
