1First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China.However,the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids.This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides,and elucidate the relationship between the resistant phenotype and kdr mutations.Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China.The larval susceptibility to deltamethrin,permethrin and beta-cypermethrin was tested by larval bioassays,and adult susceptibility to deltamethrin and DDT was determined by adult bioassays.The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult.The kdr alleles at codon 1534 of the VGSC gene were genotyped.The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test.Results:Out of five populations,assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids,and FM showed the highest degree of resistance,with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected,including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China.For the first time,the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus.Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma 2016Infectious Diseases of Poverty2016,5,1:19
2Direct generation of an ultrafast vortex beam in a CVD-graphene-based passively mode-locked Pr:LiYF4 visible laser显示文摘We report on the direct generation of passively mode-locked vortex lasers in the visible spectral region, for the first time to the best of our knowledge, using a Pr :Li YF4(Pr:YLF) crystal as the gain medium. A stable mode-locked TEM00 mode has been achieved with a maximum average output power of 75 m W using a graphene saturable absorber mirror. The mode-locked pulse width is measured to be as short as about 73.4 ps at a repetition rate of about 140 MHz, and the laser wavelength is at about 721 nm with spectral width of about 0.5 nm. By slightly misaligning the laser resonator, a first-order Laguerre-Gaussian mode(LG0,1) has also been obtained with output power reduced to about 22 m W. The achieved LG0,1 mode has been verified via a home made improved Fizeau interferometer. 4Cognitive improvement following transvenous adipose-derived mesenchymal stem cell transplantation in a rat model of traumatic brain injury显示文摘The effects of adipose-derived mesenchymal stem cell (ADMSC) transplantation for the repair of traumatic brain injury remain poorly understood. The present study observed neurological functional changes in a rat model of traumatic brain injury following ADMSC transplantation via the tail vein. Cell transplants were observed in injured cerebral cortex, and expression of brain-derived nerve growth factor was significantly increased in the injured hippocampus following transplantation. 6Effects of grassland management on the community structure, aboveground biomass and stability of a temperate steppe in Inner Mongolia, China显示文摘Plant community structure responds strongly to anthropogenic disturbances, which greatly influence community stability. The changes in community structure, aboveground biomass(AGB), biodiversity and community stability associated with different management practices were studied with a three-year field investigation in a temperate steppe of Inner Mongolia, China. The species richness, Shannon-Wiener index, evenness, plant functional type abundance, AGB, temporal community stability, summed covariance, scaling coefficient and dominant species stability were compared among areas subjected to long-term reservation(R), long-term grazing(G), mowing since enclosure in 2008(M) and grazing enclosure since 2008(E). Site R had higher perennial grass abundance and lower species richness than sites G, M and E, although the AGB was not significantly different among the four sites. The species structure varied from a single dominant species at site R to multiple dominant species at sites G, M and E. The long-term reservation grassland had lower biodiversity but higher stability, whereas the enclosed grassland with/without mowing had higher biodiversity but lower stability. Different stability mechanisms, such as the compensatory dynamics, mean-variance scaling and dominant species stability were examined. 7Carboniferous Arc Setting in Central Hainan: Geochronological and Geochemical Evidences on the Andesitic and Dacitic Rocks显示文摘Volcanic rocks in the Bangxi-Chenxing tectonic zone provide important carries for better understanding the Late Paleozoic tectonic evolution in Hainan and its temporal-spatial pattern of the eastern Paleotethyan evolution. This paper presents a set of new geochronological and geochemical data on the andesitic and dacitic rocks along the Bangxi-Chenxing tectonic zone in central Hainan. The representative andesitic and dacitic samples yield similar zircon U-Pb ages of 353±3 and 351±7 Ma, respectively, being of Early Carboniferous origin. These volcanic rocks are characterized by low TiO_2 and high Al_2O_3 contents and are enriched in LILEs and LREEs but depletion in HFSEs, along with negative ε_(Nd)(t) values of-1.4–-4.7 and high ^(87)Sr/^(86)Sr(i) ratios of 0.707 2–0.710 1. Geochemical signatures suggest that the andesitic and dacitic samples might originate from a metasomatized wedge modified by the slab-derived component in a continental arc setting. In combination with the available data, it is proposed for the development of a Carboniferous continental arc in response to the eastern Paleotethyan evolution. 8Engineering Escherichia coli to improve tryptophan production via genetic manipulation of precursor and cofactor pathways显示文摘Optimizing the supply of biosynthetic precursors and cofactors is usually an effective metabolic strategy to improve the production of target compounds.Here,the combination of optimizing precursor synthesis and balancing cofactor metabolism was adopted to improve tryptophan production in Escherichia coli.First,glutamine synthesis was improved by expressing heterologous glutamine synthetase from Bacillus subtilis and Bacillus megaterium in the engineered Escherichia coli strain KW001,resulting in the best candidate strain TS-1.Then icd and gdhA were overexpressed in TS-1,which led to the accumulation of 1.060 g/L tryptophan.Subsequently,one more copy of prs was introduced on the chromosome to increase the flux of 5-phospho-α-D-ribose 1-diphosphate followed by the expression of mutated serA and thrA to increase the precursor supply of serine,resulting in the accumulation of 1.380 g/L tryptophan.Finally,to maintain cofactor balance,sthA and pntAB,encoding transhydrogenase,were overexpressed.With sufficient amounts of precursors and balanced cofactors,the engineered strain could produce 1.710 g/L tryptophan after 48 h of shake-flask fermentation,which was 2.76-times higher than the titer of the parent strain.Taken together,our results demonstrate that the combination of optimizing precursor supply and regulating cofactor metabolism is an effective approach for high-level production of tryptophan.Similar strategies could be applied to the production of other amino acids or related derivatives.Zhu Li Dongqin Ding Huiying Wang Linxia Liu Huan Fang Tao Chen Dawei Zhang 2020Synthetic and Systems Biotechnology2020,5,3:5
9The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy.Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu 2021Cancer Communications2021,41,1:5
10Drug-nanoencapsulated PLGA microspheres prepared by emulsion electrospray with controlled release behavior显示文摘The development of modern therapeutics has raised the requirement for controlled drug delivery system which is able to efficiently encapsulate bioactive agents and achieve their release at a desired rate satisfying the need of the practical system.In this study,two kind of aqueous model drugs with different molecule weight,Congo red and albumin from bovine serum(BSA)were nanoencapsulated in poly(DL-lactic-co-glycolic acid)(PLGA)microspheres by emulsion electrospray.In the preparation process,the aqueous phase of drugs was added into the PLGA chloroform solution to form the emulsion solution.The emulsion was then electrosprayed to fabricate drugnanoencapsulated PLGA microspheres.The morphology of the PLGA microspheres was affected by the volume ratio of aqueous drug phase and organic PLGA phase(V_(w)/V_(o))and the molecule weight of model drugs.Confocal laser scanning microcopy showed the nanodroplets of drug phase were scattered in the PLGA microspheres homogenously with different distribution patterns related to V_(w)/V_(o).With the increase of the volume ratio of aqueous drug phase,the number of nanodroplets increased forming continuous phase gradually that could accelerate drug release rate.Moreover,BSA showed a slower release rate from PLGA microspheres comparing to Congo red,which indicated the drug release rate could be affected by not only V_(w)/V_(o)but also the molecule weight of model drug.In brief,the PLGA microspheres prepared using emulsion electrospray provided an efficient and simple systemto achieve controlled drug release at a desired rate satisfying the need of the practices.Shenglian Yao Huiying Liu Shukui Yu Yuanyuan Li Xiumei Wang Luning Wang 2016Regenerative Biomaterials2016,3,5:5
11Development of an on-line measurement system for water-soluble organic matter in PM_(2.5) and its application in China显示文摘Water-soluble organic matter(WSOM) represents a critical fraction of fine particles(PM_(2.5))in the air, but its changing behaviors and formation mechanisms are not well understood yet, partly due to the lack of fast techniques for the ambient measurements. In this study,a novel system for the on-line measurement of water-soluble components in PM_(2.5), the particle-into-liquid sampler(PILS)–Nebulizer–aerosol chemical speciation monitor(ACSM), was developed by combining a PILS, a nebulizer, and an ACSM. High time resolution concentrations of WSOM, sulfate, nitrate, ammonium, and chloride, as well as mass spectra, can be obtained with satisfied quality control results. The system was firstly applied in China for field measurement of WSOM. The mass spectrum of WSOM was found to resemble that of oxygenated organic aerosol, and WSOM agreed well with secondary inorganic ions. All evidence collected in the field campaign demonstrated that WSOM could be a good surrogate of secondary organic aerosol(SOA). 12Separation and Manipulation of Rare-earth Oxide Particles by Dielectrophoresis显示文摘A challenge in chemical engineering is the separation and purification of rare-earth elements and their compounds. We report the design and manufacture of a dielectrophoresis(DEP) microchip of microelectrode arrays. This microchip device is constructed in order to use DEP to capture micro-particles of rare-earth oxides in petro-leum. Dielectrophoretic behavior of micro-particles of rare-earth oxides in oil media is explored. The dielectropho-retic effects of particles under different conditions are investigated. It is showed that the prepared microchip is suit-able for use in the investigation of dielectrophoretic responses of the rare-earth oxides in oil media. The factors such as frequency,particle size and valence of rare-earth metal are discussed. When the frequency is fixed,the transla-tion voltage decreases as particle size increases. Lower frequencies are more effective for manipulation of inorganic particles in oil media. Particles of the same rare-earth oxide with different size,as well as particles of different rare-earth oxides,are captured in different regions of the field by regulating DEP conditions. 144-Formyl-2-(2H-benzotriazol-2-yl)-phenol: an ESIPT chromophore显示文摘In the present work, excited state intramolecular proton transfer(ESIPT) emission properties of a new benzotriazole derivative 4-formyl-2-(2H-benzotriazol-2-yl)-phenol(C1) were studied. 4-Formyl-2-(2H-benzotriazol-2-yl)-methoxy-benzene(C2), 4-formyl-2-(2H-benzotriazol-2-yl)-phenyl acetate(C3) and 4-methyl-2-(2H-benzotriazol-2-yl)-phenol(C4) were used as the reference molecules.1H NMR chemical shift of hydroxy group in C1 was located at more down field than that of C4 or p-hydroxy-benzaldehyde(C5), respectively. C1 showed two absorption bands in the range of 260–400 nm zones in various solvents, while C2 and C3 exhibited single absorption band. The equal molar mixtures of C2/C5 or C3/C5 showed single absorption band.C2 and C3 displayed single fluorescence emission band in various solvents, while C1 exhibited dual emission bands in some strong polar solvents. Furthermore, the second emission band in these strong polar solvents showed the large Stokes shift. The results show that the second emission band of C1 was produced by ESIPT. C2 and C3 could not undergo ESIPT due to no hydroxy group. 15Diode-pumped wavelength-switchable visible Pr^(3+):YLF laser and vortex laser around 670 nm显示文摘Here we developed a novel wavelength-switchable visible continuous-wave(CW)Pr^(3+):YLF laser around 670 nm.In single-wavelength laser operations,the maximum output powers of 2.60 W,1.26 W,and 0.21 W,the maximum slope efficiencies of 34.7%,27.3%,and 12.3%were achieved with good beam qualities(M^(2)<1.6)at 670.4 nm,674.2 nm,and 678.9 nm,respectively.Record-high output power(2.6 W)and record-high slope efficiency(34.7%)were achieved for the Pr^(3+):YLF laser operation at 670.4 nm.This is also the first demonstration of longer-wavelength peaks beyond 670 nm in the^(3)P_(1)→^(3)F_(3)transition of Pr^(3+):YLF.In multi-wavelength laser operations,the dual-wavelength lasings,including 670.1/674.8 nm,670.1/679.1 nm,and 675.0/679.4 nm,were obtained by fine adjustment of one/two etalons within the cavity.Furthermore,the triple-wavelength lasings,e.g.672.2/674.2/678.6 nm and 670.4/674.8/679.4 nm,were successfully demonstrated.Moreover,both the first-order vortex lasers(LG_(0)^(+1)and LG_(0)^(-1)modes)at 670.4 nm were obtained by off-axis pumping.Xiuji Lin Qichen Feng Yao Zhu Shuaihao Ji Bo Xiao Huiying Xu Wensong Li Zhiping Cai 2021Opto-Electronic Advances2021,4,4:4
16Intrinsic chemiluminescence production from the degradation of haloaromatic pollutants during environmentally-friendly advanced oxidation processes:Mechanism,structure-activity relationship and potential applications显示文摘The ubiquitous distribution of halogenated aromatic compounds(XAr) coupled with their carcinogenicity has raised public concerns on their potential risks to both human health and the ecosystem. Recently, advanced oxidation processes(AOPs) have been considered as an'environmentally-friendly' technology for the remediation and destruction of such recalcitrant and highly toxic XAr. During our study on the mechanism of metal-independent production of hydroxyl radicals(UOH) by halogenated quinones and H_2O_2, we found, unexpectedly, that an unprecedented UOH-dependent two-step intrinsic chemiluminescene(CL) can be produced by H_2O_2 and tetrachloro-p-benzoquinone, the major carcinogenic metabolite of the widely used wood preservative pentachlorophenol. Further investigations showed that, in all UOH-generating systems, CL can also be produced not only by pentachlorophenol and all other halogenated phenols, but also by all XAr tested. A systematic structure–activity relationship study for all 19 chlorophenolic congeners showed that the CL increased with an increasing number of Cl-substitution in general. More importantly, a relatively good correlation was observed between the formation of quinoid/semiquinone radical intermediates and CL generation. Based on these results, we propose that UOH-dependent formation of quinoid intermediates and electronically excited carbonyl species is responsible for this unusual CL production; and a rapid, sensitive,simple, and effective CL method was developed not only to detect and quantify trace amount of XAr, but also to provide useful information for predicting the toxicity or monitoring real-time degradation kinetics of XAr. 18Mid‐infrared all‐fiber gain‐switched pulsed laser at 3μm显示文摘Mid-infrared(MIR)fiber pulsed lasers are of tremendous application interest in eye-safe LIDAR,spectroscopy,chemical detection and medicine.So far,these MIR lasers largely required bulk optical elements,complex free-space light alignment and large footprint,precluding compact all-fiber structure.Here,we proposed and demonstrated an all-fiberized structured gain-switched Ho3+-doped ZBLAN fiber laser operating around 2.9μm.A home-made 1146 nm Raman fiber pulsed laser was utilized to pump highly concentrated single-cladding Ho3+-doped ZBLAN fiber with different lengths of 2 m or 0.25 m.A home-made MIR fiber mirror and a perpendicular-polished ZBLAN fiber end construct the all-fiberized MIR cavity.Stable gain-switched multiple states with a sub-pulse number tuned from 1 to 8 were observed.The effects of gain fiber length,pump power,pump repetition rate and output coupling ratio on performance of gain-switched pulses were further investigated in detail.The shortest pulse duration of 283 ns was attained with 10 kHz repetition rate.The pulsed laser,centered at 2.92μm,had a maximum average output power of 54.2 mW and a slope efficiency of 10.12%.It is,to the best of our knowledge,the first time to demonstrate a mid-infrared gain-switched Ho3+:ZBLAN fiber laser with compact all-fiber structure.Xiaojin Zhang Weiwei Li Jin Li Huiying Xu Zhiping Cai Zhengqian Luo 2020Opto-Electronic Advances2020,3,5:4
19Depression accelerates gastric cancer invasion and metastasis by inducing a neuroendocrine phenotype via the catecholamine/β_(2)-AR/MACC1 axis显示文摘Background:Depression is a common,easily ignored,accompanied disease of gastric cancer(GC)patients and is often observed with elevated plasma catecholamine levels.Depression frequently promotes GC progression and leads to poor clinical outcomes;however,the molecular mechanisms underlying depression-induced GC progression remain poorly understood.We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression-associated catecholamines on GC.Methods:Depression states of GC patients were graded using the Patient Health Questionnaire-9,and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry.Migrative and invasive GC cells were examined using transwell assays,and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress.Mouse depression-like behaviors were assessed through sucrose preference,forced swimming,and tail suspension tests.Characteristics of the neuroendocrine phenotype were observed via RT-PCR,Western blotting,flow cytometry,and transmission electron microscopy.Results:Fifty-one GC patients(age:53.61±1.79 years;cancer duration:3.71±0.33 months;depression duration:2.37±0.38 months;male-to-female ratio:1.55:1)were enrolled in the study.Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines(epinephrine:P=0.018;noradrenaline:P=0.009),higher oncogene metastasis-associated in colon cancer-1(MACC1)level(P=0.018),and metastasis(P<0.001).Further,depression-associated catecholamine specifically bound to the beta-2 adrenergic receptor(β_(2)-AR)and upregulated MACC1 expression,and thus promoting neuroendocrine phenotypic transformation through direct binding betweenMACC1 and synaptophysin.Eventually,the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo.However,β_(2)-AR antagonist ICI-118,551 or MACC1 silencing effectively blocked the catecholamineinduced neuroendocrine phenotypic transformation and eliminated depressionenhanced GC migration and invasion.Moreover,β_(2)-AR blocking or MACC1 silencing prevented GC metastasis attributed to a neuroendocrine phenotype in a depression mouse model.Conclusions:Catecholamine-induced neuroendocrine phenotypes of GC cells led to depression-accelerated GC invasion and metastasis via the β_(2)-AR/MACC1 axis,while β_(2)-AR antagonist or MACC1 silencing could reverse it,showing promising potential therapeutic strategies for improving the outcome of GC patients with comorbid depression.Changqie Pan Jianhua Wu Siting Zheng Huiying Sun Yisheng Fang Zhenhua Huang Min Shi Li Liang Jianping Bin Yulin Liao Jinzhang Chen Wangjun Liao 2021Cancer Communications2021,41,10:4
20Microstructure and properties of in situ synthesized TiB_2+WC reinforced composite coatings显示文摘Nickel-based composite coatings reinforced by in situ synthesized TiB2 and WC particles were deposited on stainless steel by laser cladding, and their microstructure and mechanical properties were investigated. The results show that the coatings are mainly composed of γ-Ni cellular dendrites and dispersed spherical/strip/network shaped TiB2 and equiaxial WC particles. The initial WC particles are dissolved to become fine and mostly dispersed within γ-Ni cellular dendrites. The coating prepared at a special laser energy of 0.225 kJ·mm-2 is uniform, continuous, and free of pores and cracks. With the decrease in special energy density, TiB2 phase changes from fine spherical particles which cluster together to strip shape with different morphologies and further crystallizes to form network structure, and the dispersion zone also gradually changes from intragranular to intergranular phase.
