| 1 | A Kelch Motif-Containing Serine/Threonine Protein Phosphatase Determines the Large Grain QTL Trait in Rice显示文摘A thorough understanding of the genetic basis of rice grain traits is critical for the improvement of rice (Oryza sativa L.) varieties.In this study,we generated an F 2 population by crossing the large-grain japonica cultivar CW23 with Peiai 64 (PA64),an elite indica small-grain cultivar.Using QTL analysis,17 QTLs for five grain traits were detected on four different chromosomes.Eight of the QTLs were newly-identified in this study.In particular,qGL3-1,a newly-identified grain length QTL with the highest LOD value and largest phenotypic variation,was fine-mapped to the 17 kb region of chromosome 3.A serine/threonine protein phosphatase gene encoding a repeat domain containing two Kelch motifs was identified as the unique candidate gene corresponding to this QTL.A comparison of PA64 and CW23 sequences revealed a single nucleotide substitution (C→A) at position 1092 in exon 10,resulting in replacement of Asp (D) in PA64 with Glu (E) in CW23 for the 364th amino acid.This variation is located at the D position of the conserved sequence motif AVLDT of the Kelch repeat.Genetic analysis of a near-isogenic line (NIL) for qGL3-1 revealed that the allele qGL3-1 from CW23 has an additive or partly dominant effect,and is suitable for use in molecular marker-assisted selection. | Zejun Hu Haohua He Shiyong Zhang Fan Sun Xiaoyun Xin Wenxiang Wang Xi Qian Jingshui Yang Xiaojin Luo | 2012 | Journal of Integrative Plant Biology2012,54,12: | 41 |
| 2 | Additive and Over-dominant Effects Resulting from Epistatic Loci Are the Primary Genetic Basis of Heterosis in Rice显示文摘A set of 148 F9 recombinant inbred lines(RILs) was developed from the cross of an indica cultivar 93-11 and japonica cultivar DT713,showing strong F1 heterosis.Subsequently,two backcross F1(BCF1) populations were constructed by backcrossing these 148 RILs to two parents,93-11 and DT713.These three related populations(281BCF1 lines,148 RILs) were phenotyped for six yield-related traits in two locations.Significant inbreeding depression was detected in the population of RILS and a high level of heterosis was observed in the two BCF1 populations.A total of 42 main-effect quantitative trait loci(M-QTLs) and 109 epistatic effect QTL pairs(E-QTLs) were detected in the three related populations using the mixed model approach.By comparing the genetic effects of these QTLs detected in the RILs,BCF1 performance and mid-parental heterosis(HMP),we found that,in both BCF1 populations,the QTLs detected could be classified into two predominant types:additive and over-dominant loci,which indicated that the additive and over-dominant effect were more important than complete or partially dominance for M-QTLs and E-QTLs.Further,we found that the E-QTLs detected collectively explained a larger portion of the total phenotypic variation than the M-QTLs in both RILs and BCF1 populations.All of these results suggest that additive and over-dominance resulting from epistatic loci might be the primary genetic basis of heterosis in rice. | Xiaojin Luo Yongcai Fu Peijiang Zhang Shuang Wu Feng Tian Jiayong Liu Zuofeng Zhu Jinshui Yang Chuanqing Sun | 2009 | Journal of Integrative Plant Biology2009,51,4: | 20 |
| 3 | Genomic Survey and Expression Profiling of the MYB Gene Family in Watermelon显示文摘Myeloblastosis(MYB)proteins constitute one of the largest transcription factor(TF)families in plants.They are functionally diverse in regulating plant development,metabolism,and multiple stress responses.However,the function of watermelon MYB proteins remains elusive to date.Here,a genome-wide identification of watermelon MYB TFs was performed by bioinformatics analysis.A total of 162 MYB genes were identified from watermelon(Cla MYB).A comprehensive overview of the Cla MYB genes was undertaken,including the gene structures,chromosomal distribution,gene duplication,conserved protein motif,and phylogenetic relationship.According to the analyses,the watermelon MYB genes were categorized into three groups(R1R2R3-MYB,R2R3-MYB,and MYB-related).Amino acid alignments for all MYB motifs of Cla MYBs demonstrated high conservation.Investigation of their chromosomal localization revealed that these Cla MYB genes distributed across the 11watermelon chromosomes.Gene duplication analyses showed that tandem duplication events contributed predominantly to the expansion of the MYB gene family in the watermelon genome.Phylogenetic comparison of the Cla MYB proteins with Arabidopsis MYB proteins revealed that watermelon MYB proteins underwent a more diverse evolution after divergence from Arabidopsis.Some watermelon MYBs were found to cluster into the functional clades of Arabidopsis MYB proteins.Expression analysis under different stress conditions identified a group of watermelon MYB proteins implicated in the plant stress responses.The comprehensive investigation of watermelon MYB genes in this study provides a useful reference for future cloning and functional analysis of watermelon MYB proteins. | XU Qing HE Jie DONG Jianhui HOU Xiaojin ZHANG Xian | 2018 | Horticultural Plant Journal2018,4,1: | 14 |

| 9 | An AIEgens and exonuclease Ⅲ aided quadratic amplification assay for detecting and cellular imaging of telomerase activity显示文摘Monitoring telomerase activity with high sensitive and reliable is of great importance to cancer analysis.In this paper, we report a sensitive and facile method to detect telomerase activity using AIEgens modified probe(TPE-Py-DNA) as a fluorescence reporter and exonuclease Ⅲ(Exo Ⅲ) as a signal amplifier.With the aid of telomerase, repeat units(TTAGGG)n are extended from the end of template substrate oligonucleotides(TS primer) that form duplex DNAs with TPE-Py-DNA. Then, Exo Ⅲ catalyzes the digestion of duplex DNAs, liberating elongation product and releasing hydrophobic TPE-Py. The released hydrophobic TPE-Py aggregate together and produce a telomerase-activity-related fluorescence signal.The liberated product hybridizes with another TPE-Py-DNA probe, starting the second cycle. Finally,we obtain the target-to-signal amplification ratio of 1:N^2. | 10 | Tribological Properties and Failure Analysis of PTFE Composites used for Seals in the Transmission Unit显示文摘The sealing rings are one of the most important components as the sealing devices in the wet clutch unit of a heavy vehicle. The sealing ring, made from PTFE composites, was subjected to serious wear on the sealing surface, but the mating metal surface only had slight abrasion. A specialized test rig was designed for wear research and failure analysis of the sealing ring. The composition analyses of the ring material, working conditions and wear surface characteristics by visual inspection and tribological properties as well as microscopic analysis with scanning electron microscope was performed to determine the wear mechanism and failure causes. Results revealed that the wear of PTFE composites was characterized by abrasion and adhesion after a certain duration testing, and the wear mechanism changed to thermal fatigue and abrasive wear in the stage of intense wear. | 11 | Mid‐infrared all‐fiber gain‐switched pulsed laser at 3μm显示文摘Mid-infrared(MIR)fiber pulsed lasers are of tremendous application interest in eye-safe LIDAR,spectroscopy,chemical detection and medicine.So far,these MIR lasers largely required bulk optical elements,complex free-space light alignment and large footprint,precluding compact all-fiber structure.Here,we proposed and demonstrated an all-fiberized structured gain-switched Ho3+-doped ZBLAN fiber laser operating around 2.9μm.A home-made 1146 nm Raman fiber pulsed laser was utilized to pump highly concentrated single-cladding Ho3+-doped ZBLAN fiber with different lengths of 2 m or 0.25 m.A home-made MIR fiber mirror and a perpendicular-polished ZBLAN fiber end construct the all-fiberized MIR cavity.Stable gain-switched multiple states with a sub-pulse number tuned from 1 to 8 were observed.The effects of gain fiber length,pump power,pump repetition rate and output coupling ratio on performance of gain-switched pulses were further investigated in detail.The shortest pulse duration of 283 ns was attained with 10 kHz repetition rate.The pulsed laser,centered at 2.92μm,had a maximum average output power of 54.2 mW and a slope efficiency of 10.12%.It is,to the best of our knowledge,the first time to demonstrate a mid-infrared gain-switched Ho3+:ZBLAN fiber laser with compact all-fiber structure. | Xiaojin Zhang Weiwei Li Jin Li Huiying Xu Zhiping Cai Zhengqian Luo | 2020 | Opto-Electronic Advances2020,3,5: | 4 |

Methods: Full length of HSP27 cDNA which got by RT-PCR was constructed into pCDNA3.1+. The recombinant was transfected into rat cardiomyocyte cell line H9c2 and the stable transfection cell line was selected by G418. Then we observe the effects of HSP27 over-expression on LDH release and apoptosis induced H2O2 in H9c2. Results:①pCDNA3.1+/HSP27 provided a sound expression of HSP27 in both 293T and H9c2. ②LDH releasing induced by 0, 100,250,500,1000 μmol/L H2O2 in HSP27 over-expression group and wild type group were 0.396±0.017 vs. 0.390±0.009 (p>0.05), 0.437±0.014 vs. 0.416±0.015 (P<0.05), 0.471±0.018 vs. 0.417±0.009 (P<0.001), 0.505±0.030 vs. 0.657±0.022(P<0.001),0.547±0.027 and 0.661±0.011(P<0.001),respectively. ③Apoptosis induced by 150 μmol/L H2O2 in HSP27 over-expression group and wild type group were (10.693±1.122)% vs. (4.027±1.628)%(P<0.01). | 13 | Ginsenoside Rb1 relieves glucose fluctuationinduced oxidative stress and apoptosis in Schwann cells显示文摘Cultured Schwann cells were treated with 5.6 mM and 50 mM glucose alternating every 8 hours to simulate intermittent high glucose. The present study analyzed the neuroprotective effects of 1, 10 and 100 μM ginsenoside Rb1 on oxidative damage and apoptosis in Schwann cells induced by intermittent high glucose. Flow cytometry demonstrated that ginsenoside Rb1 reduced intermittent high glucose-mediated reactive oxygen species production. Enzyme linked immunosorbent assay showed that 8-hydroxy-2-deoxy guanosine levels in Schwann cells decreased following ginsenoside Rb1 treatment. Quantitative real-time reverse transcription-PCR and western blot assay results revealed that ginsenoside Rb1 inhibited intermittent high glucose-upregulated Bax expression, but antagonized intermittent high glucose-downregulated Bcl-2 expression in Schwann cells. These effects were most pronounced with 100 μM ginsenoside Rb1. | 18 | Spleen-kidney supplementing formula alleviates insulin resistance via regulating AKT/glycogen synthase kinase 3β pathway in rats with type 2 diabetic induced by high-fat diet显示文摘OBJECTIVE: To explore the molecular mechanism underpinning the action by investigating its effect on glycogen content and AKT(also known as protein kinase B)/glycogen synthase kinase 3β(GSK-3β) pathway in the liver of rats with type 2 diabetic induced by high-fat diet.METHODS: The rat model of type 2 diabetes was induced by high-fat diet and multiple low-dose streptozotocin injection. Diabetic rats were divided into five groups: the model control group, the Metformin group, spleen-kidney supplementing formula groups of low, medium and high doses. Fasting blood glucose(FBG) levels were measured before treatment and every two weeks during treatment.After the treatment, oral glucose tolerance test was performed, and hemoglobin A1c (HbA1c) and C-peptide were measured to assess the formula's effect on glucose metabolism and insulin resistance. The protein expression levels of AKT, GSK-3βand their phosphorylated forms in the liver were also measured to study the formula's role in insulin signaling pathway.RESULTS: Spleen-kidney supplementing formula significantly relieved the symptoms of polydipsia,polyuria and weight loss in type 2 diabetic rats, reduced FBG and HbA1c levels, increased glycogen content, and improved insulin sensitivity. The anti-diabetic effects of spleen-kidney supplementing formula are dose dependent.
