1Asian summer monsoon precipitation recorded by stalagmite oxygen isotopic composition in the western Loess Plateau during AD1875―2003 and its linkage with ocean-atmosphere system显示文摘Based on 5 high-precision 230Th dates and 103 stable oxygen isotope ratios (δ18O) obtained from the top 16 mm of a stalagmite collected from Wanxiang Cave,Wudu,Gansu,variation of monsoonal precipita-tion in the modern Asian Monsoon (AM) marginal zone over the past 100 years was reconstructed. Comparison of the speleothem δ18O record with instrumental precipitation data at Wudu in the past 50 years indicates a high parallelism between the two curves,suggesting that the speleothem δ18O is a good proxy for the AM strength and associated precipitation,controlled by 'amount effect' of the pre-cipitation. Variation of the monsoonal precipitation during the past 100 years can be divided into three stages,increasing from AD 1875 to 1900,then decreasing from AD 1901 to 1946,and increasing again thereafter. This variation is quite similar to that of the Drought/Flooding index archived from Chinese historical documents. This speleothem-derived AM record shows a close association with the Pacific Decadal Oscillation (PDO) between AD 1875 and 1977,with higher monsoonal precipitation corre-sponding to cold PDO phase and vice versa at decadal timescale. The monsoonal precipitation varia-tion is out of phase with the PDO after AD 1977,probably resulting from the decadal climate jump in the north Pacific occurring at around AD 1976/77. These results demonstrate a strong linkage between the AM and associated precipitation and the Pacific Ocean via ocean/atmosphere interaction. 2Association of three single nucleotide polymorphisms at the SIX1-SIX6 locus with primary open angle glaucoma in the Chinese population显示文摘This study investigated the genetic association of three single nucleotide polymorphisms(SNPs; rs10483727, rs33912345, and rs146737847) at the SIX1-SIX6 locus with primary open angle glaucoma(POAG) in the Chinese population. A total of 866 subjects with POAG(685 high-tension glaucoma(HTG) and 181 normal-tension glaucoma(NTG)) and 266 control individuals were included. Significant genetic association was identified for rs10483727 in HTG(P=0.02; odds ratio(OR)=1.31), NTG(P=7.41×10^(-6); OR=2.71), and POAG(i.e., HTG and NTG combined; P=0.001; OR=1.44). rs33912345 was also significantly associated with HTG(P=0.008; OR=1.36), NTG(P=2.72×10^(-6); OR=2.27), and POAG(P=3.84×10^(-4); OR=1.49). The rare SIX6 mutation, rs146737847, was not found in the subjects enrolled in this study. Stratification by patient age identified that both rs10483727 and rs33912345 were significantly associated with NTG in patients aged above 40 years(P=2.08×10^(-5); OR=2.28), whereas in patients aged between 20–40 years, rs33912345 was significantly associated with NTG(P=0.017; OR=2.06). In HTG, the genetic associations for both rs10483727 and rs33912345 were significant in patients aged between 20–40 years(P=0.006; OR=1.56) but not in those aged above 40 years(P=0.118, OR=1.21 and P=0.042, OR=1.29, respectively). 3Implications of stalagmite density for past climate change:An example from stalagmite growth during the last deglaciation from Wanxiang Cave,western Loess Plateau显示文摘The density of a stalagmite(WX42A)from Wanxiang Cave,Wudu County,Gansu Province,China,in the western margin of the Asian summer monsoon region,presents regular fluctuations in different deposition periods during the last deglaciation.Over long timescales,high-precision 230Th dating and high-resolution stalagmite density data indicate that the density-time series between 17644 a BP and 12758 a BP is quite similar to the stalagmite δ18O record which reflects Asian monsoon intensity.Strengthening/weakening(lighter/heavier stalagmite δ18O values)of the Asian monsoon is accompanied by increase/decrease in stalagmite density.Over short timescales,decrease in stalagmite density correlates to monsoon-retreat events such as the Inter-AllerФd Cold Period(IACP),Older Dryas(OD)and Inter-BФlling Cold Period(IBCP).Generally,this kind of decrease in stalagmite density reflects precipitation decrease with weakening of the Asian monsoon,which in turn slows cave dripwater rate and decreases crystal nuclei,leading to enlargement of calcite crystals,weakened biological activity and decreased soil pCO2 which increases the abundance of impure detrital materials in stalagmites.However,during the period of large amplitude reduction of precipitation and biological activity which resulted from extreme monsoon-retreat events,temperature variation would dominate fluctuation of stalagmite density.For example,stalagmite density increased suddenly when temperature dropped suddenly in the north Atlantic during Heinrich event 1(H-1).This may be caused by low seepage water temperature,more dissolved calcium carbonate,compact regular crystals forming under higher supersaturation,leading to the stalagmite increased density.Stalagmite density fluctuation sensitively recorded stalagmite growth history and environment,demonstrating that stalagmite density can be used as a proxy for paleoclimatic research.ZHANG DeZhong ZHANG PingZhong SANG WenCui CHENG Hai WU XiuPing YUAN Ye BAI YiJun WANG JiangLin JIA JiHong 2010Chinese Science Bulletin2010,55,34:7
4Mitochondrial localization of ORF346 causes pollen abortion in alloplasmic male sterility显示文摘The Nsa cytoplasmic male sterility(CMS) system confers stable male sterility and offers great potential for production of hybrid seeds in oilseed rape. However, genes responsible for male sterility in Nsa CMS have not been identified. By mitochondrial genome sequencing of Nsa CMS and its maintainer line,we identified in an Nsa CMS line several chimeric genes encoding hypothetical proteins harboring transmembrane domains. One novel chimeric gene orf346 showed high identity with cox1 at the 50 terminal region and was co-transcribed with nad3 and rps12 genes. Transgenic plants of orf346 fused with or without mitochondrial targeting peptide conferred complete male sterility in Arabidopsis. ORF346 was mitochondrion-localized. Expression of orf346 in Escherichia coli inhibited bacterial growth, with excessive accumulation of reactive oxygen species and decreased ATP content. These results reveal a link between the newly identified mitochondrial gene orf346 and the abortion of Nsa CMS. 5α-Glucosidase Inhibitory Activities of Lutein and Zeaxanthin Purified from Green Alga Chlorella ellipsoidea显示文摘α-Glucosidase inhibitors are used therapeutically to treat type-2 diabetes mellitus. Through a bioassay-guided fractionation technique, three carotenoids,(all-E)-lutein,(all-E)-zeaxanthin and(9-Z)-zeaxanthin, were purified from the green alga Chlorella ellipsoidea, in which(all-E)-lutein and(9-Z)-zeaxanthin had potent α-glucosidase inhibitory activity. IC_(50) values of(all-E)-lutein and(9-Z)-zeaxanthin were 70 and 53.5 μmol L^(-1) against Saccharomyces cerevisiae α-glucosidase, respectively, with non-competitive inhibition. In addition, IC_(50) values of(9-Z)-zeaxanthin against Bacillus stearothermophilus and rat-intestinal α-glucosidase were 805.1 and 671.2 μmol L^(-1), respectively. The K_i values of(all-E)-lutein and(9-Z)-zeaxanthin against S. cerevisiae α-glucosidase were 78.1 and 16.5 μmol L^(-1), respectively. 6A large-scale fatal outbreak of Pasteurella multocida among wild rodents in southwestern China显示文摘Background:Pasteurella multocida is an important and old zoonotic pathogen worldwide which has an impressive host spectrum including numerous domestic and wild animals as well as birds,causing specific diseases or outbreak with great economic impact.It has never been reported that P.multocida can cause an epidemic in wild rodents.In June 5–17,2016,more than 1000 rodent deaths of an unknown cause quickly spread in the PuEr City,Yunnan province,southwestern China.Methods:The rodents in affected areas and outside of the epidemic areas were collected and screened for possible known pathogens including Yersinia pestis,rabies virus and hantavirus as well as other bacteria.The possible bacterial pathogens were isolated both by culture medium and by mouse inoculation in parallel.The isolates were identified by the Vitek GNI card and PCR assays for 16S rRNA genes.The pathogen strains were selected for whole genome sequencing analysis.Results:A total of 123 rodents were collected from 25 sample sites at affected area,among of which,all 119 dead rodents were negative for the pathogen under consideration except P.multocida,and all four live rodents were negative for P.multocida.In addition,480 rodents collected from other 23 counties outside of the epidemic area in Yunnan were negative for with P.multocida.A total of 14 strains of P.multocida(six directly isolated from the field rodents and eight from the experimental mice that were injected with the organ substrates from the dead rodents)belonged to serogroup A and serogroup F represented by 9 N and 20 N were identified in these epidemic areas.Whole genome sequencing revealed that the serogroup F strain shared 99%similarity to P.multocida Pm70 from chicken,but contained a 50 k bp insertion sequence.The serogroup A strain shared 95%similarity to P.multocida FDAARGOS_385 from a human patient,but contained four large structural differences.Histological abnormalities were identified in the livers,lungs,hearts and brains of the inoculated mice.Conclusions:The simultaneous occurrence of both serotypes of P.multocida may have caused this sudden onset of mortality across the local rodent population in Yunnan Province,China.Further attention should be paid to this old bacterium in the world.Chun-Hong Du Yong Huang Jia-Fu Jiang Zi-Hou Gao Na Jia Shan Yang Chao-Nian Chen Shun-Gao Ma Yu-Ying Liang Bao-Gui Jiang Wei-Hong Sang Peng Wang Yun Liang Yun-Zhi Zhang Ji-Hua Zhou Shui-Ping Chen Frans Jongejan Zhi-Zhong Song Yi-Gang Tong Wu-Chun Cao 2020Journal of Biosafety and Biosecurity2020,2,2:3
7Catheter ablation for atrial fibrillation is associated with reduced risk of mortality in the elderly:a prospective cohort study and propensity score analysis显示文摘Background It is unclear whether catheter ablation(CA)for atrial fibrillation(AF)affects the long-term prognosis in the elderly.This study aims to evaluate the relationship between CA and long-term outcomes in elderly patients with AF.Methods Patients more than 75 years old with non-valvular AF were prospectively enrolled between August 2011 and December 2017 in the Chinese Atrial Fibrillation Registry Study.Participants who underwent CA at baseline were propensity score matched(1:1)with those who did not receive CA.The outcome events included all-cause mortality,cardiovascular mortality,stroke/transient ischemic attack(TIA),and cardiovascular hospitalization.Results Overall,this cohort included 571 ablated patients and 571 non-ablated patients with similar characteristics on 18 dimensions.During a mean follow-up of 39.75±19.98 months(minimum six months),24 patients died in the ablation group,compared with 60 deaths in the non-ablation group[hazard ratio(HR)=0.49,95%confidence interval(CI):0.30-0.79,P=0.0024].Besides,6 ablated and 29 non-ablated subjects died of cardiovascular disease(HR=0.25,95%CI:0.11-0.61,P=0.0022).A total of 27 ablated and 40 non-ablated patients suffered stroke/TIA(HR=0.79,95%CI:0.48-1.28,P=0.3431).In addition,140 ablated and 194 non-ablated participants suffered cardiovascular hospitalization(HR=0.84,95%CI:0.67-1.04,P=0.1084).Subgroup analyses according to gender,type of AF,time since onset of AF,and anticoagulants exposure in initiation did not show significant heterogeneity.Conclusions In elderly patients with AF,CA may be associated with a lower incidence of all-cause and cardiovascular mortality.Xin SU Xin DU Shang-Xin LU Chao JIANG Jing DU Shi-Jun XIA Zhao-Jie DONG Zhao-Xu JIA De-Yong LONG Cai-Hua SANG Ri-Bo TANG Nian LIU Song-Nan LI Rong BAI Jian-Zeng DONG Chang-Sheng MA 2020Journal of Geriatric Cardiology2020,17,12:2
8Psoriasis treatment using minimally manipulated umbilical cordderived mesenchymal stem cells:A case report显示文摘BACKGROUND Psoriasis is a chronic autoimmune disease that usually manifests as a red scaly epidermis,induration,and hyperproliferation of basal keratinocytes.About 2%of the world’s population suffers from psoriasis but there are no clear therapeutics yet.Recently,mesenchymal stem cells(MSCs)have been regarded as a therapeutic alternative for autoimmune diseases,as they possess immunosuppressive effects without risks.Human umbilical cord-derived MSCs effectively regulate immune cells and are characterized by low immunogenicity,which has many advantages in treating immune diseases.CASE SUMMARY The patient was a 47-year-old male,diagnosed with psoriasis in 1995.He had received various treatments for 25 years,but the psoriatic condition was not significantly improved.He was given three rounds of minimally manipulated umbilical cord-derived MSCs over 2 wk.The erythema gradually disappeared.Three months after the 1st round,all erythema completely disappeared,and the psoriasis did not recur.CONCLUSION Minimally manipulated umbilical cord-derived MSC transplantation can potentially treat patients who suffer from psoriasis.Hyunjun Ahn Sang Yeon Lee Won-Ju Jung Jia Pi Kye-Ho Lee 2021World Journal of Clinical Cases2021,9,23:2

10Highly coordinated Pd overlayers on nanoporous gold for efficient formic acid electro-oxidation显示文摘Design and fabrication of highly efficient and stable electrocatalysts remain key challenges in green energy technologies such as low-temperature direct liquid fuel cells.Based on in-depth theoretical calculations,here we demonstrate that surface Pd atoms with high coordination numbers(HCNs)can effectively modulate their adsorption energies for CO and OH,and thus achieve very high performance for formic acid electro-oxidation reaction(FAOR).Based on epitaxial coating Pd atomic layers onto nanoporous gold(NPG)thin membranes and a slight further decoration of Au clusters on top,the resulted core-shell structured NPG-Pd-Au electrocatalyst can demonstrate Pd intrinsic and mass activities of 8.62 mA·cm^(-2)and 27.25 A·mg^(-1)respectively at the peak potential around 0.33 V versus saturated calomel electrode toward FAOR,which are far better than those of commercial Pd/C catalysts(1.09 mA·cm^(-2)and 0.32 A·mg^(-1))tested under the same conditions.Moreover,the membrane electrode assemblies based on these low precious metal loading electrodes can achieve an anode Pd power efficiency over 10 W·mg^(-1)in a direct formic acid fuel cell,which is two orders of magnitude higher than that of the commercial Pd/C.These results provide new inspirations for the development of revolutionary electrodes for energy technologies in a rational manner.Qinqin Sang Shuai Yin Feng Liu Huiming Yin Jia He Yi Ding 2021Nano Research2021,14,10:2
11Desensitization of G-protein-coupled receptors induces vascular hypocontractility in response to norepinephrine in the mesenteric arteries of cirrhotic patients and rats显示文摘BACKGROUND: The increased β-arrestin-2 and its combination with G-protein-coupled receptors (GPCRs) lead to GPCRs desensitization. The latter may be responsible for decreased contractile reactivity in the mesenteric arteries of cirrhotic patients and rats. The present study is to investigate the machinery changes of α-adrenergic receptors and G proteins and their roles in the contractility of mesenteric arteries of cirrhotic patients and animal models. METHODS: Patients with cirrhosis due to hepatitis B and cirrhotic rats induced by CCl 4 were studied. Mesenteric artery contractility in response to norepinephrine was determined by a vessel perfusion system. The contractile effect of G protein-coupled receptor kinase-2 (GRK-2) inhibitor on the mesenteric artery was evaluated. The protein expression of the α 1 adrenergic receptor, G proteins, β-arrestin-2, GRK-2 as well as the activity of Rho associated coiled-coil forming protein kinase-1 (ROCK-1) were measured by Western blot. In addition, the interaction of α 1 adrenergic receptor with β-arrestin-2 was assessed by co-immunoprecipitation. RESULTS: The portal vein pressure of cirrhotic patients and rats was significantly higher than that of controls. The doseresponse curve to norepinephrine in mesenteric arteriole was shifted to the right, and EC 50 was significantly increased in cirrhotic patients and rats. There were no significant differences in the expressions of the α 1 adrenergic receptor and G proteins in the cirrhotic group compared with the controls. However, the protein expressions of GRK-2 and β-arrestin-2 were significantly elevated in cirrhotic patients and rats compared with those of the controls. The interaction of the α 1 adrenergic receptor and β-arrestin-2 was significantly aggravated. This interaction was significantly reversed by GRK-2 inhibitor. Both the protein expression and activity of ROCK-1 were significantly decreased in the mesenteric artery in patients with cirrhosis compared with those of the controls, and this phenomenon was not shown in the cirrhotic rats. Norepinephrine significantly increased the activity of ROCK-1 in normal rats but not in cirrhotic ones. Norepinephrine significantly increased ROCK-1 activity in cirrhotic rats when GRK-2 inhibitor was used. CONCLUSIONS: β-arrestin-2 expression and its interaction with GPCRs are significantly upregulated in the mesenteric arteries in patients and rats with cirrhosis. These upregulations result in GPCR desensitization, G-protein dysfunction and ROCK inhibition. 16Chiral mesoporous silica nano-screws as an efficient biomimetic oral drug delivery platform through multiple topological mechanisms显示文摘In the microscale, bacteria with helical body shapes have been reported to yield advantages in many bio-processes. In the human society, there are also wisdoms in knowing how to recognize and make use of helical shapes with multi-functionality. Herein, we designed atypical chiral mesoporous silica nano-screws(CMSWs) with ideal topological structures(e.g., small section area, relative rough surface,screw-like body with three-dimension chirality) and demonstrated that CMSWs displayed enhanced bioadhesion, mucus-penetration and cellular uptake(contributed by the macropinocytosis and caveolaemediated endocytosis pathways) abilities compared to the chiral mesoporous silica nanospheres(CMSSs)and chiral mesoporous silica nanorods(CMSRs), achieving extended retention duration in the gastrointestinal(GI) tract and superior adsorption in the blood circulation(up to 2.61-and 5.65-times in AUC).After doxorubicin(DOX) loading into CMSs, DOX@CMSWs exhibited controlled drug release manners with pH responsiveness in vitro. Orally administered DOX@CMSWs could efficiently overcome the intestinal epithelium barrier(IEB), and resulted in satisfactory oral bioavailability of DOX(up to 348%).CMSWs were also proved to exhibit good biocompatibility and unique biodegradability. 17The study on exploitation potential of original low-oil-saturation reservoirs显示文摘It is preliminary estimated that the proved geological reserves of original low-oil-saturation reservoirs(OLOSRs)of 54 blocks in China are more than 820 million tons.As same as the high water-cut stage of a reservoir after water flooding,the initial water cut of OLOSRS is higher than 80%.The industrial application of EOR technology could efficiently enhance oil recovery rate from 8.4% to 20.1% of reservoirs after water flooding.The suitable measurements of EOR to the OLOSRs probably have a profound impact on achieving higher oilfield reserves and production.In this paper,the main characteristics and reserves potential of OLOSRs have been studied by investigations and laboratory experiments.The relationship between initial oil saturation and displacement efficiency is also analyzed.The feasibility of EOR technologies for different OLOSRs is discussed.The results show that the OLOSRs could be divided into two categories(primary and secondary)by origin of low oil saturation.The two categories contain abundant reserves in China’s major oilfields,but their occurrence states of crude oil are quite different.If the average oil saturation of the OLOSR is defined at 40%,there are 13.1%-54.2%crude oil that could be recovered by using the existing EOR technology,especially in the primary OLOSRs with low permeability and the secondary OLOSRs with high permeability.Sun Yingying Zhang Shanyan Wei Xiaofang Sang Guoqiang Minghui Zhou Huang Jia Yang Jiru 2020Petroleum Research2020,5,1:1
18Complete organelle genomes of Sinapis arvensis and their evolutionary implications显示文摘Sinapis arvensis,belonging to the genus Sinapis of the family Brassicaceae,has good agronomic characters that make it a valuable genetic resource for crop improvement and is a cytoplasmic source of heterologous cytoplasmic male sterility(CMS).In addition,S.arvensis has played an important role in the evolution of the six major cultivated Brassica species involved in the triangle of U.Using next-generation sequencing,we assembled and revealed the gene composition of S.arvensis cytoplasmic genome.The chloroplast genome comprises 153,590 bp,with 112 individual genes,including 4 r RNA,29 t RNA,and 79 proteincoding genes.The mitochondrial genome comprises 240,024 bp,with 54 genes,including 18 t RNA,three r RNA and 33 protein-coding genes.Genome structure and evolutionary analysis indicated that the sequences of the S.arvensis organellar genomes were more similar to those of Brassica nigra and B.carinata than to those of other Brassicaceae species.Four mitochondrial open reading frames displaying chimeric structural features and encoding hypothetical proteins with transmembrane domains may account for the infertility of Nsa CMS previously derived from somatic cell hybridization between B.napus and S.arvensis.These results will not only contribute to utilize the germplasm resource of S.arvensis,and comprehend the evolution of organelle genomes within the Brassicaceae family,but also help to identify genes conditioning the alloplasmic male sterility of Nsa CMS in B.napus.Shifei Sang Hongtao Cheng Desheng Mei Li Fu Hui Wang Jia Liu Wenxiang Wang Qamar U.Zaman Kede Liu Qiong Hu 2020The Crop Journal2020,8,4:1
19Efficacy and safety of subsequent radiotherapy in patients with advanced-stage hepatocellular carcinoma treated with immune checkpoint inhibitors显示文摘Background:The development of immunotherapy resistance is associated with a poor prognosis in patients diagnosed with hepatocellular carcinoma(HCC)who are undergoing treatment with immune checkpoint inhibitors(ICI).This study aimed to evaluate the efficacy and safety of subsequent radiotherapy(RT)for patients with advanced-stage HCC who had lesion enlargement or new lesions(NLs)during ICI therapy.Methods:This retrospective observational study enrolled 36 patients with advanced-stage HCC who underwent subsequent RT for lesion enlargement or NLs during ICI therapy from two centers.The primary endpoints were progression-free survival(PFS)and overall survival(OS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),1-and 2-year local control(LC)rates,in-field PFS(IFPFS),out-field PFS(OFPFS),and safety.Results:The median follow-up time was 15.3 months.The median PFS was 7.4 months[95%confidence interval(CI):3.1-11.7 months],and the median OS was 18.8 months(95%CI:17.1-20.5 months).ORR and DCR were 38.9%and 72.2%,respectively.In addition,the median IFPFS was 17.8 months(95%CI:11.5-24.2 months),median OFPFS was 7.9 months(95%CI:3.4-12.5 months),and estimated 1-and 2-year LC rates were 67.1%and 31.9%,respectively.The most common treatment-related adverse events(all grades)were diarrhea(33.3%),rash(30.6%),and malaise(27.8%);a total of 14(38.9%)patients developed grade 3-4 AEs.Conclusions:Subsequent RT showed reliable antitumor effects and an acceptable safety profile in patients with advanced-stage HCC who had unsatisfactory response to ICI therapy;therefore,it could serve as an optional salvage strategy.Cong Ning Jun Jia Xinmu Zhang Jing Sun Yunchao Wang Jingnan Xue Longhao Zhang Xiaorong Hou Xiaobo Yang Xinting Sang Xuezhang Duan Haitao Zhao 2023Hepatobiliary Surgery and Nutrition2023,12,6:0
20Less hairy leaf 1,an RNaseH-like protein,regulates trichome formation in rice through auxin显示文摘The trichomes of rice leaves are formed by the differentiation and development of epidermal cells.Plant trichomes play an important role in stress resistance and protection against direct ultraviolet irradiation.However,the development of rice trichomes remains poorly understood.In this study,we conducted ethylmethane sulfonate(EMS)-mediated mutagenesis on the wild-type(WT)indica rice‘Xida 1B’.Phenotypic analysis led to the screening of a mutant that is defective in trichome development,designated lhl1(less hairy leaf 1).We performed map-based cloning and localized the mutated gene to the 70-kb interval between the molecular markers V-9 and V-10 on chromosome 2.The locus LOC_Os02g25230 was identified as the candidate gene by sequencing.We constructed RNA interference(LHL1-RNAi)and overexpression lines(LHL1-OE)to verity the candidate gene.The leaves of the LHL1-RNAi lines showed the same trichome developmental defects as the lhl1 mutant,whereas the trichome morphology on the leaf surface of the LHL1-OE lines was similar to that of the WT,although the number of trichomes was significantly higher.Quantitative real-time PCR(RT-qPCR)analysis revealed that the expression levels of auxin-related genes and positive regulators of trichome development in the lhl1 mutant were down-regulated compared with the WT.Hormone response analysis revealed that LHL1 expression was affected by auxin.The results indicate that the influence of LHL1 on trichome development in rice leaves may be associated with an auxin pathway.CHEN Hong-yan ZHU Zhu WANG Xiao-wen LI Yang-yang HU Dan-ling ZHANG Xue-fei JIA Lu-qi CUI Zhi-bo SANG Xian-chun 2023Journal of Integrative Agriculture2023,22,1:0
