| 4 | An experimental study of extracellular signal-regulated kinase and its interventional treatments in hepatic fibrosis显示文摘BACKGROUND: The pathogenesis of hepatic fibrosis and cirrhosis is still not fully understood. The extracellular signal-regulated kinase (ERK) pathway is involved in the regulation of cell proliferation and differentiation. The aim of this study was to investigate the effects of PD98059, a specific inhibitor of ERK, on the cell cycle, cell proliferation, secretion of type Ⅰ collagen and expression of cyclin D1 mRNA, CDK4 mRNA and transforming growth factor-β1 (TGF-β1) mRNA in rat hepatic stellate cells (HSCs) stimulated by acetaldehyde. METHODS: Rat HSCs stimulated by acetaldehyde were incubated with PD98059 at different concentrations. The cell cycle was analysed by flow cytometry. Cell proliferation was assessed by the methyl thiazolyl tetrazolium colorimetric assay. The mRNA expression of cyclin D1, CDK4 and TGF-β1 was examined using the reverse transcriptase-polymerase chain reaction. Type Ⅰ collagen in the culture medium was detected by enzyme- linked immunosorbent assay. RESULTS: 20, 50 and 100 μmol/L PD98059 significantly inhibited the proliferation and provoked a G0/G1- phase arrest of acetaldehyde-induced HSCs in a dose- dependent manner. | 6 | Giant submucosal lipoma located in the descending colon: A case report and review of the literature显示文摘Colonic lipoma is an uncommon tumor of the gastrointestinal tract.Most cases are asymptomatic,with a small tumor size,and do not need any special treatment.However,we encountered one patient with a giant submucosal lipoma,with a maximum diameter of 8.5 cm,which exhibited symptoms such as intermittent lower abdominal pain,changes in bowel habits with passage of fresh blood and mucus per rectum,abdominal distension,anorexia and weight loss.Unfortunately,the possibility of colonic malignancy could not be precluded and left hemicolectomy was planned.The exact diagnosis of this special case was accomplished by intraoperative pathology.In the end,local resection was performed instead of left hemicolectomy.To the best of our knowledge,colonic lipoma exceeding 8 cm in diameter has not been previously reported.We,therefore,present this case and discuss age and sex factors,clinical and histopathological findings,diagnostic methods and treatment by reviewing the available literature,to serve as a reminder that colonic lipoma can also exist in patients with significant symptoms.In addition,intraoperative pathology should be investigated in those doubtful cases,so as to guide the exact diagnosis and treatment plan. | Li Jiang Li-Sheng Jiang Fu-Yu Li Hui Ye Ning Li Nan-Sheng Cheng Yong Zhou | 2007 | World Journal of Gastroenterology2007,13,42: | 11 |
| 7 | Chinese Herbal Medicine Combined with Entecavir for HBeAg Positive Chronic Hepatitis B:Study Protocol for a Multi-Center,Double-Blind Randomized-Controlled Trial显示文摘Background: The domestic prevalence of chronic hepatitis B(CHB) in China is 7.18% in 2006, imposing great societal healthcare burdens. Nucleot(s)ide analogues(NUCs) anti-hepatitis B virus(HBV) therapies are widely applied despite the relatively low rate of seroconversion and high risk of drug-resistant mutation. More effective treatments for CHB deserve further explorations. Combined therapy of NUCs plus Chinese herbal medicine(CHM) is widely accepted in China, which is recognized as a prospective alternative approach. The study was primarily designed to confirm the hypothesis that Tiaogan-Yipi Granule(调肝益脾颗粒, TGYP) or Tiaogan-Jianpi-Jiedu Granule(调肝健脾解毒颗粒, TGJPJD) plus entecavir tablet(ETV) was superior over ETV monotherapy in enhancing HBeA g loss rate. Methods: The study was a nationwide, large-scale, multi-center, double-blind, randomized, placebo-control ed trial with a designed duration of 108 weeks. A total of 16 hospitals and 596 eligible Chinese HBeA g positive CHB patients were enrolled from November 2012 to September 2013 and randomly allocated into 2 groups in 1:1 ratio via central randomization system: experimental group(EG) and control group(CG). Subjects in EG received CM formulae(TGYP or TGJPJD, 50 g per dose, twice daily) plus ETV tablet(or ETV placebo) 0.5 mg per day in the first 24 weeks(stage 1), and CHM granule plus ETV tablet(0.5 mg per day) from week 25 to 108(stage 2). Subjects in CG received CHM Granule placebo plus ETV tablet(0.5 mg per day) for 108 weeks throughout the trial. The assessments of primary outcomes(HBV serum markers and HBV-DNA) were conducted by a third-party College of American Pathologists(CAP) qualified laboratory. Adverse effects were observed in the hospitals of recruitment. Discussion: The study was designed to compare the curative effect of CM plus ETV and ETV monotherapy in respect of HBe Ag loss, which is recognized by the European Association for the Study of the Liver as 'a valuable endpoint'. We believe this trial could provide a reliable status for patients' 'journey' towards durable responses after treatment discontinuation. | 8 | Correlation of Seven Biological Factors(Hsp90α,p53,MDM2,Bcl-2,Bax,Cytochrome C,and Cleaved caspase3)with Clinical Outcomes of ALK+ Anaplastic Large-cell Lymphoma显示文摘Objective To explore correlation of seven apoptosis‐related proteins (Hsp90α,p53,MDM2,Bcl‐2,Bax,Cytochrome C,and Cleaved caspase3) with clinical outcomes of ALK+ anaplastic large‐cell lymphoma (ALCL).Methods Using immunohistochemistry and immunofluorescence double staining methods,the expressions of these seven apoptosis‐associated proteins were studied to clarify their relationship with clinical outcomes of 36 ALK+ and 25 ALK‐ systemic ALCL patients enrolled between 1996 and 2006.The relationship of these apoptosis‐regulating proteins with NPM‐ALK status was also evaluated with the tyrosine inhibitor herbimycin A (HA) in vitro by immunocytochemistry,Western blotting and flow cytometric assays.Results The presence of Hsp90α‐,MDM2‐,Bax‐,Cytochrome C,and Cleaved caspase3‐positive tumor cells was found significantly different in ALK+ and ALK‐ ALCLs ,which was correlated with highly favorable clinical outcome.The Bcl‐2‐ and p53‐positive tumor cells were found in groups of patients with unfavorable prognosis.Inhibition of NPM‐ALK by HA could reactivate the p53 protein and subsequent apoptosis‐related proteins and therefore induced apoptosis in ALK+ ALCL cells.Conclusion Our results suggest that these seven proteins might be involved in apoptosis regulation and associated with clinical outcome of ALK+ systemic ALCLs.We also reveal a dynamic chain relation that NPM‐ALK regulates p53 expression and subsequent apoptosis cascade in ALK+ ALCLs. | LI Hui Ling HUANG Xue Ping ZHOU Xin Hua JI Tian Hai WU Zi Qing WANG Zhi Qiang JIANG Hui Yong LIU Fan Rong ZHAO Tong | 2011 | Biomedical and Environmental Sciences2011,24,6: | 9 |

| 10 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 13 | Autophagy enhanced by curcumin ameliorates inflammation in atherogenesis via the TFEB-P300-BRD4 axis显示文摘Disturbance of macrophage-associated lipid metabolism plays a key role in atherosclerosis.Crosstalk between autophagy deficiency and inflammation response in foam cells(FCs) through epigenetic regulation is still poorly understood. Here, we demonstrate that in macrophages, oxidized lowdensity lipoprotein(ox-LDL) leads to abnormal crosstalk between autophagy and inflammation,thereby causing aberrant lipid metabolism mediated through a dysfunctional transcription factor EB (TFEB)-P300-bromodomain-containing protein 4(BRD4) axis. ox-LDL led to macrophage autophagy deficiency along with TFEB cytoplasmic accumulation and increased reactive oxygen species generation. This activated P300 promoted BRD4 binding on the promoter regions of inflammatory genes, consequently contributing to inflammation with atherogenesis. Particularly, ox-LDL activated BRD4-dependent super-enhancer associated with liquid-liquid phase separation(LLPS) on the regulatory regions of inflammatory genes. Curcumin(Cur) prominently restored FCs autophagy by promoting TFEB nuclear translocation, optimizing lipid catabolism, and reducing inflammation. The consequences of P300 and BRD4 on super-enhancer formation and inflammatory response in FCs could be prevented by Cur. Furthermore, the anti-atherogenesis effect of Cur was inhibited by macrophage-specific Brd4 overexpression or Tfeb knock-out in Apoe knock-out mice via bone marrow transplantation. | 14 | Antibody response and therapy in COVID-19 patients: what can be learned for vaccine development?显示文摘The newly emerged severe acute respiratory syndrome coronavirus 2(SARS-CoV-2) has infected millions of people and caused tremendous morbidity and mortality worldwide. Effective treatment for coronavirus disease 2019(COVID-19) due to SARSCoV-2 infection is lacking, and different therapeutic strategies are under testing. Host humoral and cellular immunity to SARSCoV-2 infection is a critical determinant for patients’ outcomes. SARS-CoV-2 infection results in seroconversion and production of anti-SARS-CoV-2 antibodies. The antibodies may suppress viral replication through neutralization but might also participate in COVID-19 pathogenesis through a process termed antibody-dependent enhancement. Rapid progress has been made in the research of antibody response and therapy in COVID-19 patients, including characterization of the clinical features of antibody responses in different populations infected by SARS-CoV-2, treatment of COVID-19 patients with convalescent plasma and intravenous immunoglobin products, isolation and characterization of a large panel of monoclonal neutralizing antibodies and early clinical testing, as well as clinical results from several COVID-19 vaccine candidates. | 18 | Improving tuberculosis case detection in underdeveloped multi-ethnic regions with high disease burden:a case study of integrated control program in China显示文摘Background:In the underdeveloped multi-ethnic regions of China,high tuberculosis(TB)burden and regional inequity in access to healthcare service increase the challenge of achieving the End TB goals.Among all the provinces,the highest TB burden is reported in Xinjiang,where ethnic minorities and older people have suffered most.However,current case-finding strategy is inadequate given the complex social determinants and suboptimal case detection rates.Thus,we developed an integrated TB control program to improve case detection and conducted a pilot in Xinjiang from 2014 to 2015.In this case study,we summarized the activities and key findings.We also shared the experiences and challenges of implementing interventions and provided recommendations to inform the TB control program in the future.Case presentation:The pilot interventions were implemented in one selected town in Yining based on local TB control programs.By applying tailor-made educational materials,outreach TB educational activities were conducted in diverse ways.In 22 Masjids,the trained imams promoted TB education to the Muslims,covering 20,440 persontimes in 88 delivered preaching sessions.In seven schools,1944 students were educated by the teachers and contributed to educating 6929 family members.In the village communities,13,073 residents participated in household education and screening.Among them,12,292 people aged under 65 years were investigated for suspicious pulmonary TB symptoms,where six TB patients were diagnosed out of 89 TB suspects;781 older people were mobilized for screening directly by chest X-ray,where 10 patients were diagnosed out of 692 participants.Supportive healthcare system,multi-sectoral cooperation and multi-channel financing mechanism were the successful experiences of implementation.The interventions were proved to be more effective than the previous performance:the number of TB suspects consulting doctors and patients detected increased by 50%and 26%,respectively.The potential challenges,implications and recommendations should been taken into account for further program improvement.Conclusions:In underdeveloped multi-ethnic regions with high TB burden,improving case detection is necessary and the interventions can be feasible and effective within a supportive system.More intensive educational and training approaches,a high index of TB suspicion and prioritization of older people in screening are recommended.To sustain and scale up the program,the impacts,cost-effectiveness,feasibility and acceptability of interventions warrant further research and evaluation in each specific context. | Jun Li Xiao-Qiu Liu Shi-Wen Jiang Xue Li Fei Yu Yan Wang Yong Peng Xiao-Ming Gu Yan-Ni Sun Hui Zhang Li-Xia Wang | 2017 | Infectious Diseases of Poverty2017,6,1: | 3 |
| 19 | The first data release(DR1) of the LAMOST regular survey显示文摘The Large sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST) general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio(SNR) ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters: late A,FGK-type stars with high quality spectra(1 061 918 entries), A-type stars(100 073 entries), and M-type stars(121 522 entries). This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats.
