1A more consistent intraluminal rhesus monkey model of ischemic stroke显示文摘Endovascular surgery is advantageous in experimentally induced ischemic stroke because it causes fewer cranial traumatic lesions than invasive surgery and can closely mimic the pathophysiology in stroke patients. However, the outcomes are highly variable, which limits the accuracy of evaluations of ischemic stroke studies. In this study, eight healthy adult rhesus monkeys were randomized into two groups with four monkeys in each group: middle cerebral artery occlusion at origin segment(M1) and middle cerebral artery occlusion at M2 segment. The blood flow in the middle cerebral artery was blocked completely for 2 hours using the endovascular microcoil placement technique(1 mm × 10 cm)(undetachable), to establish a model of cerebral ischemia. The microcoil was withdrawn and the middle cerebral artery blood flow was restored. A reversible middle cerebral artery occlusion model was identified by hematoxylin-eosin staining, digital subtraction angiography, magnetic resonance angiography, magnetic resonance imaging, and neurological evaluation. The results showed that the middle cerebral artery occlusion model was successfully established in eight adult healthy rhesus monkeys, and ischemic lesions were apparent in the brain tissue of rhesus monkeys at 24 hours after occlusion. The rhesus monkeys had symptoms of neurological deficits. Compared with the M1 occlusion group, the M2 occlusion group had lower infarction volume and higher neurological scores. These experimental findings indicate that reversible middle cerebral artery occlusion can be produced with the endovascular microcoil technique in rhesus monkeys. 2Longitudinal virological changes and underlying pathogenesis in hospitalized COVID-19 patients in Guangzhou,China显示文摘Prolonged viral RNA shedding and recurrence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in coronavirus disease 2019(COVID-19)patients have been reported.However,the clinical outcome and pathogenesis remain unclear.In this study,we recruited 43 laboratory-confirmed COVID-19 patients.We found that prolonged viral RNA shedding or recurrence mainly occurred in severe/critical patients(P<0.05).The average viral shedding time in severe/critical patients was more than 50 days,and up to 100 days in some patients,after symptom onset.However,chest computed tomography gradually improved and complete absorption occurred when SARS-CoV-2 RT-PCR was still positive,but specific antibodies appeared.Furthermore,the viral shedding time significantly decreased when the A1,430G or C12,473T mutation occurred(P<0.01 and FDR<0.01)and increased when G227A occurred(P<0.05 and FDR<0.05).High IL1R1,IL1R2,and TNFRSF21 expression in the host positively correlated with viral shedding time(P<0.05 and false discovery rate<0.05).Prolonged viral RNA shedding often occurs but may not increase disease damage.Prolonged viral RNA shedding is associated with viral mutations and host factors.Zhengtu Li Yinhu Li Ruilin Sun Shaoqiang Li Lingdan Chen Yangqing Zhan Mingzhou Xie Jiasheng Yang Yanqun Wang Airu Zhu Guoping Gu Le Yu Shuaicheng Li Tingting Liu Zhaoming Chen Wenhua Jian Qian Jiang Xiaofen Su Weili Gu Liyan Chen Jing Cheng Jincun Zhao Wenju Lu Jinping Zheng Shiyue Li Nanshan Zhong Feng Ye 2021Science China(Life Sciences)2021,64,12:1

6Inkjet printing-based fabrication of microscale 3D ice structures显示文摘This study proposed a method for fabricating 3D microstructures of ice without a supporting material.The inkjet printing process was performed in a low humidity environment to precisely control the growth direction of the ice crystals.In the printing process,water droplets(volume=hundreds of picoliters)were deposited onto the previously formed ice structure,after which they immediately froze.Different 3D structures(maximum height=2000µm)could be formed by controlling the substrate temperature,ejection frequency and droplet size.The growth direction was dependent on the landing point of the droplet on the previously formed ice structure;thus,3D structures could be created with high degrees of freedom.Fengyi Zheng Zhongyan Wang Jiasheng Huang Zhihong Li 2020Microsystems & Nanoengineering2020,6,1:1

8New perspectives on the genetic structure of dotted gizzard shad(Konosirus punctatus)based on RAD-seq显示文摘To maintain,develop and rationally utilize marine organisms,understanding their genetic structure and habitat adaptation pattern is necessary.Konosirus punctatus,which is a commercial fish species inhabiting the Indo-west Pacific Ocean,has shown an obvious annual global capture and aquaculture production decline due to climate changes and human activities.In the present study,restriction-site associated DNA sequencing(RAD-seq)was used to describe its genome-wide single nucleotide polymorphisms panel(SNPs).Among 146 individuals collected at nine locations scattered in China,Korea and Japan,a set of 632,090 SNPs were identified.Population genetic analysis showed that K.punctatus individuals were divided into two significant genetic clusters.Meanwhile,potential genetic differentiation between northern and southern population of K.punctatus was found.Treemix results indicated that gene flow existed among sampling locations of K.punctatus,especially from southern Japan to others.Moreover,candidate genes associated with habitat adaptations of K.punctatus were identified,which are involved in diverse physiological processes of K.punctatus including growth and development(e.g.,KIDINS220,PAN3),substance metabolism(e.g.,PGM5)and immune response(e.g.,VAV3,CCT7,HSPA12B).Our findings may aid in understanding the possible mechanisms for the population genetic structure and local adaptation of K.punctatus,which is beneficial to establish the management and conservation units of K.punctatus,guiding the rational use of resources,with reference significance for a profound understanding of the adaptative mechanisms of other marine organisms to the environment.Ying Peng Yifan Liu Jiasheng Li Kun Zhang Xun Jin Sixu Zheng Yunpeng Wang Zhenming Lu Liqin Liu Li Gong Bingjian Liu 2024Marine Life Science & Technology2024,6,1:0
9The complete mitochondrial genome of Parnassius actius(Lepidoptera: Papilionidae: Parnassinae) with the related phylogenetic analysis显示文摘In this study, the complete mitochondrial genome of Parnassius actius(Lepidoptera: Papilionidae: Parnassinae) was reported. Meanwhile, the phylogenetic position of this Apollo species was inferred from this sequence combined with other available related sequence data. The results showed that the whole mitogenome is 15,386 bp in length, containing 13 typical protein-coding genes(PCGs), 22 transfer RNA genes(t RNAs), two ribosomal RNA genes(r RNAs) and a noncoding control region(A+T-rich region). The mitogenomic gene orientation and arrangement are the same with other sequenced Parnassius species. Thirteen protein coding genes is 11,195 bp in size, encoding 3,720 amino acids totally; the lr RNA and sr RNA genes are 1,351 bp and 779 bp, respectively. Eleven intergenic spacers ranging from 1 to 40 bp(129 bp in total), 10 overlapping regions from 1 to 8 bp(26 bp in total), and a 504 bp A+T-rich region harboring some Parnassius-specific sequences are scattered throughout the whole mitogenome. 11Metabolite profiling and transcriptome analyses reveal novel regulatory mechanisms of melatonin biosynthesis in hickory显示文摘Studies have shown that melatonin regulates the expression of various elements in the biosynthesis and catabolism of plant hormones.In contrast,the effects of these different plant hormones on the biosynthesis and metabolism of melatonin and their underlying molecular mechanisms are still unclear.In this study,the melatonin biosynthesis pathway was proposed from constructed metabolomic and transcriptomic libraries from hickory(Carya cathayensis Sarg.)nuts.The candidate pathway genes were further identified by phylogenetic analysis,amino-acid sequence alignment,and subcellular localization.Notably,most of the transcription factor-related genes coexpressed with melatonin pathway genes were hormone-responsive genes.Furthermore,dual-luciferase and yeast one‐hybrid assays revealed that CcEIN3(response to ethylene)and CcAZF2(response to abscisic acid)could activate melatonin biosynthesis pathway genes,a tryptophan decarboxylase coding gene(CcTDC1)and an N-acetylserotonin methyltransferase coding gene(CcASMT1),by directly binding to their promoters,respectively.Our results provide a molecular basis for the characterization of novel melatonin biosynthesis regulatory mechanisms and demonstrate for the first time that abscisic acid and ethylene can regulate melatonin biosynthesis.Wenchao Chen Jiaqi Zhang Shan Zheng Zhanqi Wang Chuanmei Xu Qixiang Zhang Jiasheng Wu Heqiang Lou 2021Horticulture Research2021,8,1:0
12Preparation and Mechanical and Thermal Properties of Chlorinated Polyethylene Reinforced Polypropylene Foam显示文摘The foamed polypropylene(PP) with excellent mechanical and thermal insulating properties was reinforced by blending chlorinated polyethylene(CPE), followed by compression molding foaming in cross-link. The effects of CPE on the foaming behavior, thermal and mechanical properties of the foamed PP were studied by the measurements of density, thermal conductivity, Vicat softening temperature, tensile strength, impact strength and SEM. The results showed that the foamed PP got the best properties when the weight ratio of CPE/PP was 20 wt%.
