| 1 | Recombinant Newcastle disease virus expressing African swine fever virus protein 72 is safe and immunogenic in mice显示文摘African swine fever(ASF) is a lethal hemorrhagic disease that affects wild and domestic swine. The etiological agent of ASF is African swine fever virus(ASFV). Since the first case was described in Kenya in 1921, the disease has spread to many other countries. No commercial vaccines are available to prevent ASF. In this study, we generated a recombinant Newcastle disease virus(r NDV) expressing ASFV protein 72(p72) by reverse genetics and evaluated its humoral and cellular immunogenicity in a mouse model. The recombinant virus, r NDV/p72, replicated well in embryonated chicken eggs and was safe to use in chicks and mice. The p72 gene in r NDV/p72 was stably maintained through ten passages. Mice immunized with r NDV/p72 developed high titers of ASFV p72 specific Ig G antibody, and had higher levels of Ig G1 than IgG2 a. Immunization also elicited T-cell proliferation and secretion of IFN-γ and IL-4. | 3 | High-performance Zn battery with transition metal ions co-regulated electrolytic MnO_(2)显示文摘Electrolytic MnO_(2)/Zn batteries have attracted extensive attention for use in large-scale energy storage applications due to their low cost,high output voltage,safety,and environmental friendliness.However,the poor electrical conductivity of MnO_(2)limits its deposition and dissolution at large capacities,which leads to sluggish reaction kinetics and drastic capacity decay.Here,we report a theory-guided design principle for an electrolytic MnO_(2)/Zn battery co-regulated with transition metal ions that has improved electrochemical performance in terms of deposition and stripping chemistries.We start with first-principles calculations to predict the electrolytic effects of regulating transition metal ions in the deposition/stripping chemistry of the MnO_(2)cathode.The results indicate that with the simultaneous incorporation of strongly electronegative Co and Ni,the MnO_(2)cathode tends to possess more active electron states,faster charge-transfer kinetics,and better electrical conductivity than either MnO_(2)regulated with Co or Ni on their own,or pristine MnO_(2);hence,this co-regulation is beneficial for the cathode solid/liquid MnO_(2)/Mn2t reactions.We then fabricate and demonstrate a novel Co2t and Ni2t coregulated MnO_(2)/Zn(Co-Ni-MnO_(2)/Zn)battery that yields significantly better electrochemical performance,finding that the synergistic regulation of Co and Ni on MnO_(2)can significantly increase its intrinsic conductivity and achieve high rates and Coulombic efficiencies at large capacities.The aqueous Co-Ni-MnO_(2)/Zn battery exhibits a high rate(10C,100 mA cm^(-2)),high Coulombic efficiency(91.89%),and excellent cycling stability(600 cycles without decay)at a large areal capacity of 10 mAh cm^(-2).Our proposed strategy of co-regulation with transition metal ions offers a versatile approach for improving the electrochemical performance of aqueous electrolytic MnO_(2)/Zn batteries in large-scale energy storage applications. | Mingyan Chuai Jinlong Yang Mingming Wang Yuan Yuan Zaichun Liu Yan Xu Yichen Yin Jifei Sun Xinhua Zheng Na Chen Wei Chen | 2021 | eScience2021,1,2: | 8 |

| 5 | Aerodynamic configuration integration design of hypersonic cruise aircraft with inward-turning inlets显示文摘In this work, a novel airframe/propulsion integration design method of the wing-body configuration for hypersonic cruise aircraft is proposed, where the configuration is integrated with inward-turning inlets. With the help of this method, the major design concern of balancing the aerodynamic performance against the requirements for efficient propulsion can be well addressed. A novel geometric parametrically modelling method based on a combination of patched class and shape transition(CST) and COONs surface is proposed to represent the configuration, especially a complex configuration with an irregular inlet lip shape. The modelling method enlarges the design space of components on the premise of guaranteeing the configuration integrity via special constraints imposed on the interface across adjacent surfaces. A basic flow inside a cone shaped by a dual-inflection-point generatrix is optimized to generate the inward-turning inlet with improvements of both compression efficiency and flow uniformity. The performance improvement mechanism of this basic flow is the compression velocity variation induced by the variation of the generatrix slope along the flow path. At the design point, numerical simulation results show that the lift-to-drag ratio of the configuration is as high as 5.2 and the inlet works well with a high level of compression efficiency and flow uniformity. The design result also has a good performance on off-design conditions. | 7 | Toward dendrite-free and anti-corrosion Zn anodes by regulating a bismuth-based energizer显示文摘Aqueous rechargeable zinc metal batteries display high theoretical capacity along with economical effectiveness,environmental benignity and high safety.However,dendritic growth and chemical corrosion at the Zn anodes limit their widespread applications.Here,we construct a Zn/Bi electrode via in-situ growth of a Bi-based energizer upon Zn metal surface using a replacement reaction.Experimental and theoretical calculations reveal that the Bi-based energizer composed of metallic Bi and ZnBi alloy contributes to Zn plating/stripping due to strong adsorption energy and fast ion transport rates.The resultant Zn/Bi electrode not only circumvents Zn dendrite growth but also improves Zn anode anti-corrosion performance.Specifically,the corrosion current of the Zn/Bi electrode is reduced by 90%compared to bare Zn.Impressively,an ultra-low overpotential of 12mV and stable cycling for 4000h are achieved in a Zn/Bi symmetric cell.A Zn–Cu/Bi asymmetric cell displays a cycle life of 1000 cycles,with an average Coulombic efficiency as high as 99.6%.In addition,an assembled Zn/Bi-activated carbon hybrid capacitor exhibits a stable life of more than 50,000 cycles,an energy density of 64Wh kg−1,and a power density of 7kWkg−1.The capacity retention rate of a Zn/Bi–MnO_(2)full cell is improved by over 150%compared to a Zn–MnO_(2)cell without the Bi-based energizer.Our findings open a new arena for the industrialization of Zn metal batteries for large-scale energy storage applications. | Mingming Wang Yahan Meng Ke Li Touqeer Ahmad Na Chen Yan Xu Jifei Sun Mingyan Chuai Xinhua Zheng Yuan Yuan Chunyue Shen Ziqi Zhang Wei Chen | 2022 | eScience2022,2,5: | 4 |
| 8 | Interaction of single-pulse laser energy with bow shock in hypersonic flow显示文摘Pressure sensing and schlieren imaging with high resolution and sensitivity are applied to the study of the interaction of single-pulse laser energy with bow shock at Mach 5. An Nd:YAG laser operated at 1.06 lm, 100 mJ pulse energy is used to break down the hypersonic flow in a shock tunnel. Three-dimensional Navier–Stokes equations are solved with an upwind scheme to simulate the interaction. The pressure at the stagnation point on the blunt body is measured and calculated to examine the pressure variation during the interaction. Schlieren imaging is used in conjunction with the calculated density gradients to examine the process of the interaction. The results show that the experimental pressure at the stagnation point on the blunt body and schlieren imaging fit well with the simulation. The pressure at the stagnation point on the blunt body will increase when the transmission shock approaches the blunt body and decrease with the formation of the rarefied wave. Bow shock is deformed during the interaction. Quasi-stationary waves are formed by high rate laser energy deposition to control the bow shock. The pressure and temperature at the stagnation point on the blunt body and the wave drag are reduced to 50%, 75% and 81% respectively according to the simulation. | 11 | Co-transplantation of Schwann cells and bone marrow stromal cells versus single cell transplantation on repairing hemisected spinal cord injury of rats显示文摘BACKGROUND: Bone marrow stromal cells (BMSCs) or Schwann cells (SCs) transplantation alone can treat spinal cord injury. However, the transplantation either cell-type alone has disadvantages. The co-transplantation of both cells may benefit structural reconstruction and functional recovery of spinal nerves. OBJECTIVE: To verify spinal cord repair and related mechanisms after co-transplantation of BMSCs and SCs in a rat model of hemisected spinal cord injury. DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at the Department of Histology and Embryology, Mudanjiang Medical College from January 2008 to May 2009. MATERIALS: Rabbit anti-S-100, glial fibrillary acidic protein, neuron specific enolase and neurofilament-200 monoclonal antibodies were purchased from Sigma, USA. METHODS: A total of 100 Wistar rats were used in a model of hemisected spinal cord injury. The rats were randomly assigned to vehicle control, SCs transplantation, BMSCs transplantation, and co-transplantation groups; 25 rats per group. At 1 week after modeling, SCs or BMSCs cultured in vitro were labeled and injected separately into the hemisected spinal segment of SCs and BMSCs transplantation groups through three injection points [5 μL (1 × 107 cells/mL)] cell suspension in each point). In addition, a 15 μL 1 × 107 cells/mL SCs suspension and a 15 μL 1 × 107 cells/mL BMSC suspension were injected into co-transplantation group by the above method. MAIN OUTCOME MEASURES: The Basso-Beattie-Bresnahan (BBB) locomotor rating scale and somatosensory evoked potential (SEP) tests were used to assess the functional recovery of rat hind limbs following operation. Structural repair of injured nerve tissue was observed by light microscopy, electron microscopy, immunohistochemistry, and magnetic resonance imaging (MRI). In vivo differentiation, survival and migration of BMSCs were evaluated by immunofluorescence. RESULTS: BBB scores were significantly greater in all three transplantation groups compared with vehicle control group 8 weeks after transplantation. In particular, the co-transplantation group displayed the highest scores among the groups (P < 0.05). Moreover, recovery of SEP latency and amplitude was observed in all the transplantation groups, particularly after 8 weeks. Again, the co-transplantation group exhibited the greatest improvement (P < 0.05). In the co-transplantation group, imaging showed a smooth surface and intact inner structure at the injury site, with no scar formation, and a large number of orderly cells at the injured site. Axonal regeneration, new myelination, and a large amount of cell division were detected in the co-transplantation group by electron microscopy. Neuron specific enolase (NSE)- and glial fibrillary acidic protein (GFAP)-positive cells were observed in the spinal cord sections 1 week following co-transplantation by immunofluorescence staining. CONCLUSION: Co-transplantation of SCs and BMSCs effectively promoted functional recovery of injured spinal cord in rats compared with SCs or BMSCs transplantation alone. | 14 | OGG1 inhibition suppresses African swine fever virus replication显示文摘African swine fever virus(ASFV)is an important pathogen that causes a highly contagious and lethal disease in swine,for which neither a vaccine nor treatment is available.The DNA repair enzyme 8-oxoguanine DNA glycosylase 1(OGG1),which excises the oxidative base lesion 8-oxo-7,8-dihydroguanine(8-oxoG),has been linked to the pathogenesis of different diseases associated with viral infections.However,the role of OGG1-base excision repair(BER)in ASFV infection has been poorly investigated.Our study aimed to characterize the alteration of host reactive oxygen species(ROS)and OGG1 and to analyse the role of OGG1 in ASFV infection.We found that ASFV infection induced high levels and dynamic changes in ROS and 8-oxoG and consistently increased the expression of OGG1.Viral yield,transcription level,and protein synthesis were reduced in ASFV-infected primary alveolar macrophages(PAMs)treated by TH5487 or SU0268 inhibiting OGG1.The expression of BER pathway associated proteins of ASFV was also suppressed in OGG1-inhibited PAMs.Furthermore,OGG1 was found to negatively regulate interferonβ(IFN-β)production during ASFV infection and IFN-βcould be activated by OGG1 inhibition with TH5487 and SU0268,which blocked OGG1 binding to 8-oxoG.Additionally,the interaction of OGG1 with viral MGF360-14-L protein could disturb IFN-βproduction to further affect ASFV replication.These results suggest that OGG1 plays the crucial role in successful viral infection and OGG1 inhibitors SU0268 or TH5487 could be used as antiviral agents for ASFV infection. | Jie Fan Xinqian Lv Saixia Yang Shuxian Geng Jifei Yang Yaru Zhao Zhonghui Zhang Zhijie Liu Guiquan Guan Jianxun Luo Qiaoying Zeng Hong Yin Qingli Niu | 2023 | Virologica Sinica2023,38,1: | 1 |

| 20 | Improving the accuracy of genomic prediction for meat quality traits using whole genome sequence data in pigs显示文摘Background Pork quality can directly affect customer purchase tendency and meat quality traits have become valu-able in modern pork production.However,genetic improvement has been slow due to high phenotyping costs.In this study,whole genome sequence(WGS)data was used to evaluate the prediction accuracy of genomic best linear unbiased prediction(GBLUP)for meat quality in large-scale crossbred commercial pigs.Results We produced WGS data(18,695,907 SNPs and 2,106,902 INDELs exceed quality control)from 1,469 sequenced Duroc×(Landrace×Yorkshire)pigs and developed a reference panel for meat quality including meat color score,marbling score,L*(lightness),a*(redness),and b*(yellowness)of genomic prediction.The prediction accuracy was defined as the Pearson correlation coefficient between adjusted phenotypes and genomic estimated breeding values in the validation population.Using different marker density panels derived from WGS data,accuracy differed substantially among meat quality traits,varied from 0.08 to 0.47.Results showed that MultiBLUP outperform GBLUP and yielded accuracy increases ranging from 17.39%to 75%.We optimized the marker density and found medium-and high-density marker panels are beneficial for the estimation of heritability for meat quality.Moreover,we conducted genotype imputation from 50K chip to WGS level in the same population and found average concord-ance rate to exceed 95%and r^(2)=0.81.Conclusions Overall,estimation of heritability for meat quality traits can benefit from the use of WGS data.This study showed the superiority of using WGS data to genetically improve pork quality in genomic prediction. | Zhanwei Zhuang Jie Wu Yibin Qiu Donglin Ruan Rongrong Ding Cineng Xu Shenping Zhou Yuling Zhang Yiyi Liu Fucai Ma Jifei Yang Ying Sun Enqin Zheng Ming Yang Gengyuan Cai Jie Yang Zhenfang Wu | 2023 | Journal of Animal Science and Biotechnology2023,14,5: | 1 |
