1Phytophthora sojae Effector PsAvh240 Inhibits Host Aspartic Protease Secretion to Promote Infection显示文摘Plants secrete defense molecules into the extracellular space (the apoplast) to combat attacking microbes. However, the mechanisms by which successful pathogens subvert plant apoplastic immunity remain poorly understood. In this study, we show that PsAvh240, a membrane-localized effector of the soybean pathogen Phytophthora sojae, promotes P. sojae infection in soybean hairy roots. We found that PsAvh240 interacts with the soybean-resistant aspartic protease GmAP1 in planta and suppresses the secretion of GmAP1 into the apoplast. By solving its crystal structure we revealed that PsAvh240 contain six a helices and two WY motifs. The first two a helices of PsAvh240 are responsible for its plasma membrane-localization and are required for PsAvh240's interaction with GmAP1. The second WY motifs of two PsAvh240 molecules form a handshake arrangement resulting in a handshake-like dimer. This dimerization is required for the effector's repression of GmAP1 secretion. 2Monitoring the impact of climate change and human activities on grassland vegetation dynamics in the northeastern Qinghai-Tibet Plateau of China during 2000–2015显示文摘Climate change and human activities can influence vegetation net primary productivity(NPP), a key component of natural ecosystems. The Qinghai-Tibet Plateau of China, in spite of its significant natural and cultural values, is one of the most susceptible regions to climate change and human disturbances in the world. To assess the impact of climate change and human activities on vegetation dynamics in the grassland ecosystems of the northeastern Qinghai-Tibet Plateau, we applied a time-series trend analysis to normalized difference vegetation index(NDVI) datasets from 2000 to 2015 and compared these spatiotemporal variations with trends in climatic variables over the same time period. The constrained ordination approach(redundancy analysis) was used to determine which climatic variables or human-related factors mostly influenced the variation of NDVI. Furthermore, in order to determine whether current conservation measures and programs are effective in ecological protection and reconstruction, we divided the northeastern Qinghai-Tibet Plateau into two parts: the Three-River Headwater conservation area(TRH zone) in the south and the non-conservation area(NTRH zone) in the north. The results indicated an overall(73.32%) increasing trend of vegetation NPP in grasslands throughout the study area. During the period 2000–2015, NDVI in the TRH and NTRH zones increased at the rates of 0.0015/a and 0.0020/a, respectively. Specifically, precipitation accounted for 9.2% of the total variation in NDVI, while temperature accounted for 13.4%. In addition, variation in vegetation NPP of grasslands responded not only to long-and short-term changes in climate, as conceptualized in non-equilibrium theory, but also to the impact of human activities and their associated perturbations. The redundancy analysis successfully separated the relative contributions of climate change and human activities, of which village population and agricultural gross domestic product were the two most important contributors to the NDVI changes, explaining 17.8% and 17.1% of the total variation of NDVI(with the total contribution >30.0%), respectively. The total contribution percentages of climate change and human activities to the NDVI variation were 27.5% and 34.9%, respectively, in the northeastern Qinghai-Tibet Plateau. 3Combined effects of cadmium and fluoranthene on germination, growth and photosynthesis of soybean seedlings显示文摘The single and combinational effects of cadmium (Cd) and fluoranthene (FLT) on germination, growth and photosynthesis of soybean seedlings were investigated. Exposure to 5, 10, or 15 mg Cd/L or 1, 5, or 10 mg FLT/L individually or in combination significantly decreased germination vigor (3 days) and final germination rate of soybean seeds, except at 1 and 5 mg FLT/L. The results of two-way ANOVA analysis and the Bliss independence model showed that at lower concentrations of FLT (1 mg/L), the interaction between Cd and FLT on germination was antagonistic, whereas the interaction was synergistic when the concentration of FLT was 5 or 10 mg/L and the concentration of Cd was 15 mg/L. Growth, expressed as dry weight, length of shoot and root, leaf area, and photosynthesis, expressed as net photosynthetic rate, intercellular CO2 concentration, chlorophyll contents and fluorescence of soybean seedlings were also reduced by exposure to 5 or 10 mg Cd/L or 1 or 5 mg FLT/L, singly or jointly. 4Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8
5Evaluating Suitability of Multiple Precipitation Products for the Lancang River Basin显示文摘Global reanalysis precipitation products could provide valuable meteorological information for flow forecasting in poorly gauged areas, helping to overcome a long-standing challenge in the field. But not all data sources are suitable for all regions or perform the same way in hydrological modeling, so it is essential to test the suitability of precipitation products before applying them. In this study, five widely used global high-resolution precipitation products—Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation of Water Resources(APHRODITE), National Centers for Environmental Prediction Climate Forecast System Reanalysis(NCEP-CFSR), Climate Hazards Group InfraRed Precipitation with Station Data(CHIRPS), China Gauge-based Daily Precipitation Analysis developed by China Meteorological Administration(CMA) and Agricultural Model Intercomparison and Improvement Project based on the NASA Modern-Era Retrospective Analysis for Research and Applications(AgMERRA)—were evaluated using statistical methods and a hydrological approach for their suitability for the Lancang River Basin. The results indicated that APHRODITE, CMA, AgMERRA and CHIRPS were more accurate precipitation indicators than NCEP-CFSR in terms of the multiyear average and seasonal spatial distribution pattern, all of the CHIRPS, Ag MERRA and APHRODITE perform better than CMA and NCEP-CFSR at the small, medium and high precipitation intensities ranges in subbasin11 and sunbabsin46. All five products performed better in subbasin46(a low-altitude region) than in subbasin11(a high-altitude region) on the daily and monthly scales. In addition to NCEP-CFSR, the other four products all presented encouraging potential for streamflow simulation at daily(Yunjinghong) and monthly(Yunjinghong, Jiuzhou and Gajiu) scale. Hydrological simulations forced with APHRODITE were the best of the five for the Yunjinghong station in capturing daily and monthly measured streamflow. 6Characteristics of Hydrogen Storage Alloy Mg_2Ni Produced by Hydriding Combustion Synthesis显示文摘A high activity and large capacity of hydrogen storage alloy Mg2Ni by hydriding combustion synthesis was investigatedby means of pressure composition isotherms, X-ray diffraction and scanning electron microscopy. The results showedthat the maximum hydrogen absorption capacity of Mg2Ni is 3.25 mass fraction at 523 K, just after synthesis withoutany activation. The relationships between the equilibrium plateau pressure and the temperature for Mg2Ni were lgp(0.1 Mpa)=3026/T+5.814 (523 K≤ T ≤623 K) for hydriding and Igp (0.1 Mpa)=-3613/T+6.715 (523 K≤T ≤623 K) for dehydriding. 7One Step Ball-Milling Synthesis of LiFePO_ 4 Nanoparticles as the Cathode Material of Li-Ion Batteries显示文摘A one-step synthetic method was used to synthesize Olivline LiFePO_ 4 powders by direct ball milling the stoichiometric mixture of Fe, Li_ 3 PO_ 4 , and FePO_ 4 powders. XRD and TEM measurements revealed that the as-prepared LiFePO_ 4 powder have a homogeneous Olivine structure and a uniform size distribution of ca.50 nm. Based on this material, a LiFePO_ 4 /C composite was prepared and used for the cathode material of Li-ion batteries. The charge-discharge experiments demonstrated that the LiFePO_ 4 /C composite material has a high capacity of 132 mAh/g at 0.1 C and a quite high-rate capability of 95 mAh/g at 1 C. 8Effect of AAPH oxidation on digestion characteristics of seed watermelon(Citrullus lanatus var)kernels protein isolates显示文摘Seed watermelon kernel is a typical complex food with high fat and protein contents.During storage and processing,it is often affected by various factors to undergo interactions between components,which lead to its quality change.In this experiment,seed watermelon kernels were used as research objects,and the effects of 2-Azobis(2-amidinopropane)dihydrochloride(AAPH)on seed watermelon kernel protein isolates(WMP)were investigated.The structure and digestion characteristics of WMP after oxidation were studied.The results showed that with the increase of AAPH concentration(0.05−5 mol/L),WMP showed obvious aggregation,and its solubility decreased from 6.76 mg/mL to 9.59 mg/mL.The free sulfhydryl content of WMP was 18.24 mmol/g decreased to 11.25 mmol/g,α-helix decreased andβ-sheet decreased in secondary structure,and its disulfide bond increased by 43.06 mmol/g from 39.57 mmol/g,enthalpy(H)and denaturation temperature increased(Td)(P<0.05).By mass spectrometry results of simulated gastric digestion products,it was found that oxidation adversely affected the digestion characteristics of WMP.It can be seen that the lipid oxidation product APPH of seed watermelon kernel can significantly affect the structure and function of the protein extracted from the seed kernel.Shugang Li Zhihao Li Xiuting Li Ping Wang Xiongwei Yu Qinli Fu Sihai Gao 2020Food Science and Human Wellness2020,9,4:3
